Fear regulation changes as a function of age and adolescence is a key developmental period for the continued maturation of fear neural circuitry. The involvement of prelimbic proBDNF in fear memory extinction and its mediated signaling were reported previously. Given the inherent high level of proBDNF during juvenile period, we tested whether prelimbic proBDNF regulated synaptic and neuronal functions allowing to influencing retrieval-dependent memory processing. By examining freezing behavior of auditory fear conditioned rats, we found high levels of prelimbic proBDNF in juvenile rats enhanced destabilization of the retrieval-dependent weak but not strong fear memory through activating p75NTR-GluN2B signaling. This modification was attributed to the increment in proportion of thin type spine and promotion in synaptic function, as evidence by facilitation of NMDA-mediated EPSCs and GluN2B-dependent synaptic depression. The strong prelimbic theta- and gamma-oscillation coupling predicted the suppressive effect of proBDNF on the recall of post-retrieval memory. Our results critically emphasize the importance of developmental proBDNF for modification of retrieval-dependent memory and provide a potential critical targeting to inhibit threaten memories associated with neurodevelopment disorders.

The transposon theory of aging hypothesizes the activation of transposable elements (TEs) in somatic tissues with age, leading to a shortening of the lifespan. It is thought that TE activation in aging produces an increase in DNA double-strand breaks, contributing to genome instability and promoting the activation of inflammatory responses. To investigate how TE regulation changes in somatic tissues during aging, we analyzed the expression of some TEs, as well as a source of small RNAs that specifically silence the analyzed TEs; the Drosophila cluster named flamenco. We found significant variations in the expression levels of all the analyzed TEs during aging, with a trend toward reduction in middle-aged adults and reactivation in older individuals that suggests dynamic regulation during the lifespan.

Introduction: The Intensive Care Unit (ICU) is a treatment that can cause feelings of stress, anxiety, fear not only to the patient but also to the patient's family. The unfamiliar environment, intensive space regulation, changes in emotional status, and changes in daily activities are some of the factors that cause stress in the ICU patient's family. The purpose of this study is to reveal the experiences of families in facing hospitalization in intensive care units based on empirical studies in the last five years.Methods:Journal searches use indexed databases Scopus, Proquest, ScienceDirect and PubMed using keywords: hospitalitation, family, critical patient, icu. The Center for Review and Dissemination and The Joanna Briggs Institute was used to assess the quality of the study. The framework used is PICOS with inclusion criteria, namely journals in English and Indonesian, published years 2015 to 2020. Analyzes and tabulation of data on articles or journals, titles, abstracts, full text and methodology are assessed to determine the eligibility of articles or journals.Result:The family's experience in dealing with critical patient hospitalization in the ICU care room has an impact on the family who treats these patients both physically and psychologically. During patient hospitalization, the family plays a role in providing care, compassion, creating security and privacy, advocating for and ensuring that patients receive good care.Conclusion: The experience of hospitalization can disrupt the client's psychology and psychosocial condition, especially if the client is unable to adapt to his new environment at the hospital. published years 2015 to 2020. Connections to world markets facilitate local markets developments to support more efficient capital allocation and greater investment and growth opportunities. Under the framework of cross-market rebalancing theory, in this study, we aim to systematically examine the market connections among world financial, energy, renewable energy and European carbon markets by measuring the return spillovers from 2008 to 2021. We find that the renewable energy market is more closely connected to the world financial and energy markets in the sense of the return transmission, while the carbon market is less connected to them. However, due to improved market regulations and determinations related to fighting climate change, the connections between the carbon market and other markets have gradually intensified. Plotting the return spillover indexes, we observe that strong return spillovers from the renewable energy market to other markets occurred when large investment plans were announced. Regarding the carbon market, regulation changes introduced by the EU Commission to improve and stabilize market environment induced intensified return transmission from carbon market to other markets. Another interesting finding is that the highly intensified return transmission among markets due to the COVID-19 crisis started to loosen when COVAX published the first interim distribution forecast on 3 February 2021.

Previous research has shown that delay is a common problem in construction industry.  Weather condition and regulation changes, though being minor factors, are also known as external factors contributing to the delay of project construction. This study involved 118 respondents from the construction industry in Kuantan, Pahang. In order to obtain the data, copies of questionnaire were distributed widely among people in Pahang construction industry. The data were analysed by using Partial Least Squares Structural Equation Modelling version 3. A significant positive relationship between weather conditions and building projects in Pahang construction industry was identified. This study also found a negative relationship between regulation changes and building projects in Pahang construction industry. As expected, weather conditions were found to be one of the factors contributing to delay in the process of projects’ construction in Malaysia. However, weather conditions and the regulation changes did not significantly affect the construction projects in Malaysia. Out of the factors which contributed to delay in building projects, weather conditions, though a minor factor,    played to an extent a part in contributing to extension of the time of the projects in Malaysia. Nevertheless, regulation changes was not considered a vital factor leading to delay in building projects in Malaysia.

Purpose The purpose of this paper was to examine whether or not the sponsor lock-up ratio, lock-up period, regulation changes and interaction variable (oversubscription [OSR]) affected initial public offering (IPO) initial return. Design/methodology/approach A complete sample of 111 listed IPOs in Pakistan stock exchange from 1996 to 2018 was incorporated. Based on the cross-section data, this paper estimated using ordinary least square and quantile least square for robustness. In addition to that, this paper estimated the data using stepwise least square to inspect the signalling aspect of the lock-up ratio, lock-up period and regulation changes on IPO initial return. Findings This study showed that the lock-up ratio, lock-up period and regulatory changes had a positive impact on the IPO’s initial return. Furthermore, the assertion of interaction variable (regulation changes × OSR) and (lock-up period × OSR) was a negatively significant factor in influencing the IPO’s initial return. The results of this paper were robust to endogeneity bias. Practical implications The finding of this study proposed that sponsors of IPOs can be a strong signal of risk or quality, which was consistent with the signalling theory prediction. Concurrently, investors must be aware of the total proportions of lock-up ratio so that they can estimate the chances of getting the highest initial return on IPOs. From the regulators’ point of view, it is suggested that the lock-up ratio and the lock-up period should be determined with a deeper understanding and incorporated into the equity guidelines as it is evident that these factors are priced by the market. Originality/value Studies on the effect of sponsors have always been centred on well-recognized firms. Therefore, using the IPO samples listed in Pakistan, this paper contributes to the IPO literature by investigating the lock-up ratio of the sponsor, the lock-up period and the regulatory changes to the initial IPO return. Additionally, OSR has been introduced as an interaction variable among the sponsors’ lock-up period and regulations changes to explain the ongoing IPO initial return phenomenon.

Truck size and weight regulations have been a key instrument used to improve trucking productivity, safety, and operational performance in Canada. In response to these changes, bridge design codes undergo modifications to envelop the potential range of trucks in operation. A five-decade timeline is presented: (1) to document how bridge codes and their live load models have evolved, with a focus on the Manitoba-specific HSS-25 truck, and (2) to discuss how responsive bridge design codes have historically been to changes in truck size and weight regulations. While at times bridge codes are released in conjunction with expected regulation changes, there is often delay in the issuance of revised bridge design and evaluation codes. Assessments of the current truck fleet, which now includes long combination vehicles (LCVs), may be a consideration for future bridge design live load models.

AbstractAlternative splicing relies on the combinatorial recruitment of splicing regulators to specific RNA binding sites. Chromatin has been shown to impact this recruitment. However, a limited number of histone marks have been studied at a global level. In this work, a machine learning approach, applied to extensive epigenomics datasets in human H1 embryonic stem cells and IMR90 foetal fibroblasts, has identified eleven chromatin modifications that differentially mark alternatively spliced exons depending on the level of exon inclusion. These marks act in a combinatorial and position-dependent way, creating characteristic splicing-associated chromatin signatures (SACS). In support of a functional role for SACS in coordinating splicing regulation, changes in the alternative splicing of SACS-marked exons between ten different cell lines correlate with changes in SACS enrichment levels and recruitment of the splicing regulators predicted by RNA motif search analysis. We propose the dynamic nature of chromatin modifications as a mechanism to rapidly fine-tune alternative splicing when necessary.
