Problem definition: We examine the impact of logistics performance metrics such as delivery time and customer’s requested delivery speed on logistics service ratings and third-party sellers’ sales on an e-commerce platform. Academic/practical relevance: Although e-commerce retailers like Amazon have recently invested heavily in their logistics networks to provide faster delivery to customers, there is scant academic literature that tests and quantifies the premise that convenient and fast delivery will drive sales. In this paper, we provide empirical evidence on whether this relationship holds in practice by analyzing a mechanism that connects delivery performance to sales through logistics ratings. Prior academic work on online ratings in e-commerce platforms has mostly analyzed customers’ response to product functional performance and biases that exist within. Our study contributes to this stream of literature by examining customer experience from a service quality perspective by analyzing logistics service performance, logistics ratings, and its impact on customer purchase probability and sales. Methodology: Using an extensive data set of more than 15 million customer orders on the Tmall platform and Cainiao network (logistics arm of Alibaba), we use the Heckman ordered regression model to explain the variation in customers’ rating of logistics performance and the likelihood of customers posting a logistics rating. Next, we develop a generic customer choice model that links the customer’s likelihood of making a purchase to the logistics ratings provided by prior customers. We implement a two-step estimation of the choice model to quantify the impact of logistics ratings on customer purchase probability and third-party seller sales. Results: We surprisingly find that even customers with no promise on delivery speed are likely to post lower logistics ratings for delivery times longer than two days. Although these customers are not promised an explicit delivery deadline, they seem to have a mental threshold of two days and expect deliveries to be made within that time. Similarly, we find that priority customers (those with two-day and one-day promise speed) provide lower logistics ratings for delivery times longer than their anticipated delivery date. We estimate that reducing the delivery time of all three-day delivered orders on this platform (which makeup [Formula: see text] 35% of the total orders) to two days would improve the average daily third-party seller sales by 13.3% on this platform. The impact of delivery time performance on sales is more significant for sellers with a higher percentage of three-day delivered orders and a higher spend per order. Managerial implications: Our study emphasizes that delivery performance and logistics ratings, which measure service quality, are essential drivers of the customer purchase decision on e-commerce platforms. Furthermore, by quantifying the impact of delivery time performance on sales, our study also provides a framework for online retailers to assess if the increase in sales because of improved logistics performance can offset the increase in additional infrastructure costs required for faster deliveries. Our study’s insights are relevant to third-party sellers and e-commerce platform managers who aim to improve long-term online customer traffic and sales.

The thesis proposes a method for introducing lean manufacturing using string diagram in an operating CNG high pressure storage tank manufacturing job shop at Jayfe Cylinder Ltd. Haryana. By applying lean manufacturing using process layout diagram to produce part families with similar manufacturing processes and stable demand, plants expect to reduce costs and lead-times and improve quality and delivery performance. The thesis outlines a method for assessing, designing, and implementing lean manufacturing using process layout diagram, and illustrates this process with an example. A manufacturing cell that produces high pressure steel tank container for commercial & automobile customers is implemented at cylinder tank Machining Centers. The conclusion of the thesis highlights the key lessons learned from this process.

AbstractMost Workload Control literature assumes that delivery performance is determined by tardiness related performance measures only. While this may be true for companies that directly deliver to end-customers, for make-to-stock companies or firms that are part of supply chains, producing early often means large inventories in the finished goods warehouse or penalties incurred by companies downstream in the supply chain. Some earlier Workload Control studies used a so-called time limit, which constrains the set of jobs that can be considered for order release, to reduce earliness. However, recent literature largely abandoned the time limit since it negatively impacts tardiness performance. This study revisits the time limit, assessing the use of different adaptive policies that restrict its use to periods of either low or high load. By using a simulation model of a pure job shop, the study shows that an adaptive policy allows to balance the contradictory objectives of delaying the release of orders to reduce earliness and to release orders early to respond to periods of high load as quick as possible. Meanwhile, only using a time limit in periods of high load was found to be the best policy.

The discussion paper analyzes international experience in megaproject management, identifying problems and delivery performance. The most important distortions are optimism bias and strategic misrepresentation. it explores the approaches of Norway and the United Kingdom. Both countries have strengthened the robustness of planning and appraisal for megaprojects and have established specific methods and institutional arrangements for these projects to strengthen governance and transparency. Finally, some lessons are put forward that could help other countries, especially in Latin America, when embarking on megaprojects.

The development of polymers that can replace engineered viral vectors in clinical gene therapy has proven elusive despite the vast portfolios of multifunctional polymers generated by advances in polymer synthesis. Functional delivery of payloads such as plasmids (pDNA) and ribonucleoproteins (RNP) to various cellular populations and tissue types requires design precision. Here, we systematically screen a combinatorially designed library of 43 well-defined polymers, ultimately identifying a lead polycationic vehicle (P38) for efficient pDNA delivery. Further, we demonstrate the versatility of P38 in co-delivering spCas9 RNP and pDNA payloads to mediate homology directed repair as well as in facilitating efficient pDNA delivery in ARPE-19 cells. P38 achieves nuclear import of pDNA and eludes lysosomal processing far more effectively than a structural analog that does not deliver pDNA as efficiently. To reveal the physicochemical drivers of P38's gene delivery performance, SHapley Additive exPlanations (SHAP) are computed for nine polyplex features, and a causal model is applied to evaluate the average treatment effect of the most important features selected by SHAP. Our machine learning interpretability and causal inference approach derives structure-function relationships underlying delivery efficiency, polyplex uptake, and cellular viability, and probes the overlap in polymer design criteria between RNP and pDNA payloads. Together, combinatorial polymer synthesis, parallelized biological screening, and machine learning establish that pDNA delivery demands careful tuning of polycation protonation equilibria while RNP payloads are delivered most efficaciously by polymers that deprotonate cooperatively via hydrophobic interactions. These payload-specific design guidelines will inform further design of bespoke polymers for specific therapeutic contexts.

Abstract Background Improving anti-cancer drug delivery performance can be achieved through designing smart and targeted drug delivery systems (DDSs). For this aim, it is important to evaluate overexpressed biomarkers in the tumor microenvironment (TME) for optimizing DDSs. Materials and methods Herein, we designed a novel DDS based on magnetic mesoporous silica core–shell nanoparticles (SPION@MSNs) in which release of doxorubicin (DOX) at the physiologic pH was blocked with gold gatekeepers. In this platform, we conjugated heterofunctional polyethylene glycol (PEG) onto the outer surface of nanocarriers to increase their biocompatibility. At the final stage, an epithelial cell adhesion molecule (EpCAM) aptamer as an active targeting moiety was covalently attached (Apt-PEG-Au@NPs-DOX) for selective drug delivery to colorectal cancer (CRC) cells. The physicochemical properties of non-targeted and targeted nanocarriers were fully characterized. The anti-cancer activity, cellular internalization, and then the cell death mechanism of prepared nanocarriers were determined and compared in vitro. Finally, tumor inhibitory effects, biodistribution and possible side effects of the nanocarriers were evaluated in immunocompromised C57BL/6 mice bearing human HT-29 tumors. Results Nanocarriers were successfully synthesized with a mean final size diameter of 58.22 ± 8.54 nm. Higher cytotoxicity and cellular uptake of targeted nanocarriers were shown in the EpCAM-positive HT-29 cells as compared to the EpCAM-negative CHO cells, indicating the efficacy of aptamer as a targeting agent. In vivo results in a humanized mouse model showed that targeted nanocarriers could effectively increase DOX accumulation in the tumor site, inhibit tumor growth, and reduce the adverse side effects. Conclusion These results suggest that corporation of a magnetic core, gold gatekeeper, PEG and aptamer can strongly improve drug delivery performance and provide a theranostic DDS for efficient CRC therapy. AbstractIn the North Nile Delta of Egypt, the impacts of overplanting paddy rice on water delivery performance have not been discussed quantitatively. Further, the amount of water that could have been saved if farmers would follow the planned area is unknown. In this study, water delivery performance was assessed by comparison of actual paddy rice planting and the government’s planned conditions. For both conditions, performance indicators relating to adequacy, equity, and dependability were analyzed across six locations in conjunction with the branch canal water level in 2013 and 2014. Based on the difference between the actual water supply and planned water demand, the amount of water that could have been saved for downstream uses was calculated. The average adequacy for the investigation period was good at one location, fair at 2 locations, and poor at 3 locations in both years. Further, adequacy under both actual and planned conditions was poor in late July at all locations. The planned adequacy and dependability downstream and equity among locations improved compared to the actual condition in both years. Under the condition that paddy rice area is the upper limit planned by the government, about 12.3% and 9.6% of water could be potentially saved in each year. The difference between actual and planned water delivery performance is caused by the branch canal’s low water level. Control of overplanting paddy rice and coordination of water distribution among water user associations would improve stable water level in the canal and, eventually, water delivery performance.
