Brassica oleracea var. capitata L. (white cabbage) is a valuable vegetable with diverse nutraceutical benefit. Present study aimed to investigate the preventive effects of B. oleracea extract (BOE) standardized by vitamin U on indomethacin (IND)-induced acute gastric injury in Sprague-Dawley rats. Pre-administration of three different doses of BOE (12.5–50 mg/kg) for 14 days significantly decreased visible ulcerative lesions in the gastric tissue. In addition, BOE alleviated IND-mediated increase in histological score with inhibiting invaded percentage of lesion and restoring mucosa thickness in peri-ulcerative region. BOE increased the gastric tissue bound to Alcian blue and inhibited the decrease in hexose, sialic acid, and collagen levels by IND, suggesting that BOE protects the gastric tissue through preserving mucus and mucosal integrity. Moreover, BOE pre-administration blocked the reduction of prostaglandin E2 and down-regulated histamine and mRNA expression related to secret gastric acid. Furthermore, BOE mitigated inflammatory responses in the gastric tissue by decreasing activity of myeloperoxidase and expression of nuclear factor-κB-dependent inflammatory genes. BOE also suppressed malondialdehyde with preventing the reduction of glutathione, superoxide dismutase, and catalase in the gastric tissue. Therefore, results from present study suggest that BOE will have a potential for preventing gastric injury.

The aim: To investigate the efficacy of Doctovit, a combination of dexpanthenol (provitamin B5) and methylmethionine (vitamin U), in the treatment of patients with chronić pancreatitis in combination with chronic erosive gastritis associated with H. pylori by studying the dynamics of stomach lining morphological changes. Materials and methods: Forty-five outpatients with CP and H.pylori CG were examined. The degree of excretory insufficiency of the pancreas was determined by the level of fecal α-elastase-1. At the beginning of the study and two months after the treatment has started, esophagogastroduodenoscopy + urease test for H. pylori + biopsy from 5 sites with histological examination has been performed. Results: It was found that a significant decrease in lymphohistiocytic infiltration of stomach lining, restoration of the structure of glands which have not undergone atrophy, increased focal hyperplasia (proliferation) of the glandular epithelium as signs of morphological restoration of the epithelium, reduction of epithelial dysplasia signs against the complete absence of positive dynamics of epithelial dysplasia in the group of patients receiving standard treatment of CP and CG, are clear and reliable signs of the effectiveness of vitamin Doctovit in complex therapy of CG associated with H.pylori, which indicate the feasibility of using the medicine to restore SL, which is the basis for effective carcinoprevention. Conclusions: The effectiveness of both treatment complexes in the correction of exocrine insufficiency of the pancreas by the dynamics of fecal α-elastase-1 was proved and which was statistically significantly higher when using the program with the inclusion of Doctovit: respectively 28.12% vs. 20.74% (p< 0.05). The total dynamics of morphological state improvement of stomach lining in the 1st group of patients was 0.9 points against 1.6 points in the 2nd group of patients, which was 17% and 32%, respectively (p <0.05), which activates clinical data on the effectiveness and feasibility of using a combination of dexpanthenol and methylmethionine according to the suggested scheme in the treatment of patients with chronic pancreatitis in comorbidity with H. pylori erosive CG.

Aim. Assessment of efficacy and the mechanism of action of gastrointestinal mucosa (GM) protection in current treatment settings with methylmethionine-sulfonium chloride (vitamin U) to illustrate its applicability in erosive ulcerative lesions of various aetiologies.Key points. Aside to damage prevention in exposure to aggressive agents, gastroprotection implies healing promotion under the preserved level of hydrochloric acid secretion. Prostaglandins (PG) and SH-antioxidants are key mediators of gastroprotection in acute and chronic damage. SH-containing endogenous substances (L-cysteine, D,L-methionine, GSH) and exogenous molecules (methylmethionine-sulfonium chloride (MMSC), N-acetylcysteine) prevent damage due to the ability to absorb/neutralise free radicals released in xenobiotic-triggered cell damage, inhibit TNF-α expression, reduce the aspirin-induced leukocyte-endothelium adhesion and stimulate mucin release. In experiment, MMSC prevented the ethanol-induced GM damage, stimulated mucin release and its redistribution on the GM surface; in clinical trials, MMSC effectively facilitated remission in duodenal ulcer.Conclusion. Preparations exerting a protective effect on gastroduodenal mucosa, such as methylmethionine-sulfonium chloride (vitamin U), may improve basic treatment settings and facilitate remission in erosive ulcerative lesions of upper gastrointestinal tract.

Graviola leaves contain much vitamin U (vit U), but their sensory quality is not good enough for them to be developed as food ingredients. Addition of excipient natural ingredients formulated alongside vit U as active ingredients could enhance not only its sensory quality but also its bioavailability. The objectives of this study were to measure the bioaccessibility and intestinal cellular uptake of bioactive components, including rutin, kaempferol-rutinoside, and vit U, from steamed extract of graviola leaves (SGV) and SGV enriched with kale extract (SGK), and to examine how much they can detoxify nicotine in HepG2 cells. The bioaccessibility of vit U from SGV and SGK was 82.40% and 68.03%, respectively. The cellular uptake of vit U in SGK by Caco-2 cells was higher than that in SGV. Cotinine content converted from nicotine in HepG2 cells for 120 min was 0.22 and 0.25 μg/mg protein in 50 μg/mL of SGV and SGK, respectively, which were 2.86 and 3.57 times higher than the no-treatment control. SGK treatment of HepG2 cells upregulated CYP2A6 three times as much as did that of SGV. Our results suggest that graviola leaf extract enriched with excipient ingredients such as kale could improve vit U absorption and provide a natural therapy for detoxifying nicotine.

The management of patients in remission, rehabilitation after treatment of inflammatory and erosive-ulcerative lesions of gastroduodenal zone in spite of actively used schemes antihelikobakterna therapy require additional discussions regarding the issue of cytoprotective. The article substantiates the importance of the inclusion in the complex therapy of these diseases S-methylmethanesulfonamide ( vitamin U). The data of experimental and clinical studies, indicating the protective properties and clinical efficacy of this drug. The possibility of drug astrorex containing the active substance S-methylmethane to improve the course of disease, faster relief of symptoms, long disease-free. Pharmacological preparations containing vitamin U in their composition can be used in a complex of medical, preventive and rehabilitation programs with active participation and sufficient compliance in the implementation of these programs by the patient.
