Foregut Issues After Bariatric Surgery

Weight loss surgery has overall been shown to be very safe and effective. However, long-term outcomes data has allowed codification of post-operative complications specific to the type of weight loss surgery performed. This review focuses specifically on foregut-related postoperative complications after weight loss surgery, most of which are not discussed on a broad scale in the literature yet whose prevalence continues to rise. Clinicians should maintain a broad differential when treating patients with complications after bariatric surgery in order to perform a thorough and precise workup to identify the diagnosis and guide management.

Barking at the Wrong Tree: Articles 31-31bis TRIPs Revisited in Light of COVID-19 Lessons

The COVID-19 reality challenged the assumption underlying Articles31-31bisof the TRIPS agreement. It illustrated that the major obstacle to access of developing countries and LDCs to medicines and/or vaccines in cases of broad-scale, global pandemics is global production capacity and distribution priorities, namely: availability, rather than price. This article examines the future implications of this understanding.

What Would You Do? Types of Ethical Challenging Situations Depicted in Vignettes Published in the Veterinary Literature from 1990 to 2020

Veterinary team members encounter a wide range of ethically challenging situations (ECS) in their work. Inability to resolve ECS in accordance with their values may negatively impact the wellbeing of veterinary team members. We sought to determine the types of ECS described in published ethical vignettes in the veterinary literature. We performed a strategic literature search, followed by a thematic analysis of vignettes published in the veterinary literature from 1990–2020. We identified 567 published vignettes in 544 publications. In the majority of vignettes, the protagonist was a veterinarian (61.6%) and the most common categories of animal involved were dogs (28.0%), livestock in general (10.8%), and cattle (10.6%). The primary type of ECS was coded for each scenario, generating 29 themes. These findings extend knowledge about types of ECS that may be encountered by veterinary team members. These themes can help to inform curricula and better prepare veterinary team members to navigate ECS. They may also highlight factors that contribute to ECS that can be addressed on a broad scale, such as through regulation, continuing professional development, or stakeholder education. Knowing that others may experience similar ECS may help veterinary team members feel part of a moral community.

Forest Fires Reduce Snow-Water Storage and Advance the Timing of Snowmelt across the Western U.S.

As climate warms, snow-water storage is decreasing while forest fires are increasing in extent, frequency, and duration. The majority of forest fires occur in the seasonal snow zone across the western US. Yet, we do not understand the broad-scale variability of forest fire effects on snow-water storage and water resource availability. Using pre- and post-fire data from 78 burned SNOTEL stations, we evaluated post-fire shifts in snow accumulation (snow-water storage) and snowmelt across the West and Alaska. For a decade following fire, maximum snow-water storage decreased by over 30 mm, and the snow disappearance date advanced by 9 days, and in high severity burned forests snowmelt rate increased by 3 mm/day. Regionally, forest fires reduced snow-water storage in Alaska, Arizona, and the Pacific Northwest and advanced the snow disappearance date across the Rockies, Western Interior, Wasatch, and Uinta mountains. Broad-scale empirical results of forest fire effects on snow-water storage and snowmelt inform natural resource management and modeling of future snow-water resource availability in burned watersheds.

Understanding the distribution of introduced mammalian predators in an urban environment using monitoring tools and community trapping

<p>Introduced mammalian predators are one of the largest conservation threats to New Zealand native flora and fauna, and there is an increasing concern about their presence in urban environments, coupled with a recognition that cities present a unique opportunity for ecological restoration, due to the availability of a large number of volunteers and options for intensive management of green spaces and gardens. Predator control is an essential step towards the ecological restoration of urban environments, however, it requires an understanding of the factors influencing the distribution of these mammalian predators before successful control operations can be implemented. Few studies have investigated mammalian predators in urban environments, and there is little certainty about what drives their distribution in these environments. This thesis used simple mammal monitoring techniques and trapping data to investigate the distribution of mammalian predators within broad scale urban environments, with the aim of identifying drivers of their distribution. Chew cards and tracking tunnels collected across three New Zealand cities were assessed for their efficacy as accurate monitoring devices in urban environments. In Chapter 2, monitoring devices were cross-checked between observers to assess the level of consistency in interpretation of chew and tracking marks. The consistency of chew card and tracking tunnel identifications was relatively high overall and were not substantially influenced by the city of identification, or the duration of card exposures. Monitoring devices were also assessed for their change in sensitivity between one and six-night exposures. Both devices were effective at detecting rats, however, tracking tunnels showed greater sensitivity and consistency in detecting mice and hedgehogs, whereas chew cards were better suited to the monitoring of possums. Neither device was particularly effective at detecting mustelids or cats. In Chapter 3, mammalian predators were monitored across 24 monitoring lines in autumn, 2018, and results were compiled with spring 2017 and autumn 2018 data, pre-collected in two other cities, following the same procedures. There were distinct differences in the broad-scale habitat utilisation of rats, mice, hedgehogs, with possums being the only species to show a strong preference for urban forests. Only two of the tested microhabitat variables had an influence on species distributions. Detection of rats declined with increasing distance to the coast, and the increase in human population size was related to a significant increase in hedgehogs. There was a strong seasonal difference on the influence of local trap density and the detection of mammals. The increase in trap density within 25-50m radii was significantly related to a decrease in rat and hedgehog detections. Overall, there are substantial differences between the distributions of species in an urban environment. Trapping is one of the main methods of predator control in New Zealand, and is already widespread within urban and suburban Wellington. In Chapter 4, I compiled trap data from 22 community trapping groups operating in residential and reserve areas in Wellington City. Residential groups (“backyard trappers”) used a high proportion of Victor and various rat and mouse traps, which was strongly linked to their high number of rat and mouse catches. Groups trapping in reserves used a high proportion of DOC 200, Victor and A24 traps, however, fewer hedgehogs were caught compared to residential areas. Catches were significantly influenced by various landscape variables. An increased distance of traps to streams led to significantly higher catches of rats, conversely, proximity to streams resulted in significantly higher catches of mice and hedgehogs. Although few catches of weasels were reported, traps closer to the coast and to forest fragments caught significantly more individuals. The research in this thesis contributes to the small body of research conducted on mammalian predators within urban environments. The findings in this thesis can assist with the current and future predator management programmes, by highlighting areas of potential significance, particularly in Wellington.</p>

NamZ1 and NamZ2 from the oral pathogen Tannerella forsythia are peptidoglycan processing exo-β-N-acetylmuramidases with distinct substrate specificity

ABSTRACTThe Gram-negative periodontal pathogen Tannerella forsythia is inherently auxotrophic for N-acetylmuramic acid (MurNAc), which is an essential carbohydrate constituent of the peptidoglycan (PGN) of the bacterial cell wall. Thus, to build up its cell wall, T. forsythia strictly depends on the salvage of exogenous MurNAc or sources of MurNAc, such as polymeric or fragmentary PGN, derived from cohabiting bacteria within the oral microbiome. In our effort to elucidate how T. forsythia satisfies its demand for MurNAc, we recognized that the organism possesses three putative orthologs of the exo-β-N-acetylmuramidase BsNamZ from Bacillus subtilis, which cleaves non-reducing end, terminal MurNAc entities from the artificial substrate pNP-MurNAc and the naturally-occurring disaccharide substrate MurNAc-β-1,4-N-acetylglucosamine (GlcNAc). TfNamZ1 and TfNamZ2 were successfully purified as soluble, pure recombinant His6-fusions and characterized as exo-lytic β-N-acetylmuramidases with distinct substrate specificities. The activity of TfNamZ1 was considerably lower compared to TfNamZ2 and BsNamZ, in the cleavage of pNP-MurNAc and MurNAc-GlcNAc. When peptide-free PGN glycans were used as substrates, we revealed striking differences in the specificity and mode of action of these enzymes, as analyzed by mass spectrometry. TfNamZ1, but not TfNamZ2 or BsNamZ, released GlcNAc-MurNAc disaccharides from these glycans. In addition, glucosamine (GlcN)-MurNAc disaccharides were generated when partially N-deacetylated PGN glycans from B. subtilis 168 were applied. This characterizes TfNamZ1 as a unique disaccharide-forming exo-lytic β-N-acetylmuramidase (exo-disaccharidase), and, TfNamZ2 and BsNamZ as sole MurNAc monosaccharide-lytic exo-β-N-acetylmuramidases.IMPORTANCETwo exo-β-N-acetylmuramidases from T. forsythia belonging to glycosidase family GH171 (www.cazy.org) were shown to differ in their activities, thus revealing a functional diversity within this family: NamZ1 releases disaccharides (GlcNAc-MurNAc/GlcN-MurNAc) from the non-reducing ends of PGN glycans, whereas NamZ2 releases terminal MurNAc monosaccharides. This work provides a better understanding of how T. forsythia may acquire the essential growth factor MurNAc by the salvage of PGN from cohabiting bacteria in the oral microbiome, which may pave avenues for the development of anti-periodontal drugs. On a broad scale, our study indicates that the utilization of PGN as a nutrient source, involving exo-lytic N-acetylmuramidases with different modes of action, appears to be a general feature of bacteria, particularly among the phylum Bacteroidetes.

Estimating the movements of terrestrial animal populations using broad-scale occurrence data

As human and automated sensor networks collect increasingly massive volumes of animal observations, new opportunities have arisen to use these data to infer or track species movements. Sources of broad scale occurrence datasets include crowdsourced databases, such as eBird and iNaturalist, weather surveillance radars, and passive automated sensors including acoustic monitoring units and camera trap networks. Such data resources represent static observations, typically at the species level, at a given location. Nonetheless, by combining multiple observations across many locations and times it is possible to infer spatially continuous population-level movements. Population-level movement characterizes the aggregated movement of individuals comprising a population, such as range contractions, expansions, climate tracking, or migration, that can result from physical, behavioral, or demographic processes. A desire to model population movements from such forms of occurrence data has led to an evolving field that has created new analytical and statistical approaches that can account for spatial and temporal sampling bias in the observations. The insights generated from the growth of population-level movement research can complement the insights from focal tracking studies, and elucidate mechanisms driving changes in population distributions at potentially larger spatial and temporal scales. This review will summarize current broad-scale occurrence datasets, discuss the latest approaches for utilizing them in population-level movement analyses, and highlight studies where such analyses have provided ecological insights. We outline the conceptual approaches and common methodological steps to infer movements from spatially distributed occurrence data that currently exist for terrestrial animals, though similar approaches may be applicable to plants, freshwater, or marine organisms.
