Ralstonia Solanacearum is one of the most infectious soil-borne bacterial plant pathogens, causing tomato bacterial wilt (TBW). Nanotechnology is an emerging area of research, particularly the application of nanoparticles (NPs) as nanopesticides to manage plant disease is gaining attention nowadays. However, the interaction between NPs and rhizosphere bacterial communities remains largely elusive. This study indicated that metal NPs (CuO, ZnO, and FeO) reduced the incidence of bacterial wilt to varying degrees and affected the composition and structure of the rhizosphere bacterial community. The results revealed that the application of metal oxide NPs can improve the morphological and physiological parameters of TBW infected tomato plants. Among all, CuONPs amendments significantly increase the Chao1 and Shannon index. In the early stage (the second week), it significantly reduces the relative abundance of pathogens. However, the relative abundance of beneficial Streptomyces bacteria increased significantly, negatively correlated with the relative abundance of pathogenic bacteria. In addition, the nano-treatment group will enrich some potential beneficial bacteria such as species from Sphingomonadaceae, Rhizobiaceae, etc. In general, our research provides evidence and strategies for preventing and controlling soil-borne disease tomato bacterial wilt with metal oxide NPs.

Tomato (Solanum lycopersicum L.) is one of the most important vegetables in Togo. Unfortunately, tomatoes are susceptible to many diseases, among which bacterial wilt caused by Ralstonia solanacearum causes major yield losses. In this study, incidence of bacterial wilt and its distribution was evaluated in the central region of Togo, the major tomato producing area in the country. Overall, 16 localities were surveyed in four prefectures. In each locality, three fields were visited, and the incidence of the disease was recorded, and diseased samples were collected for laboratory investigation. The results showed that bacterial wilt occurred in all the fields visited, indicating a field incidence of 100%, whereas the plant incidence ranged from 10.00±00% to 43.33±3.33%, with an average of 20.94±1.77%. The antibody based Immunostrip test was positive for R. solanacearum in 100% of the visited fields. From 144 samples collected from fields, 45 R. solanacearum isolates were isolated on Modified SMSA media. This survey results show that tomato bacterial wilt is a real threat to tomato production in the central region of Togo.

(1) Background: Ralstonia solanacearum causes tomato bacterial wilt disease, one of the most serious tomato diseases. As the combination of Trichoderma virens (Tvien6) and Bacillus velezensis (X5) was more effective at controlling tomato bacterial wilt disease than a single agent, we investigated the synergistic effect of Tvien6 and X5 in controlling this disease; (2) Methods: The disease incidence, plant heights and weights, relative chlorophyll content (SPAD values), defensive enzymes (PPO, POD, and SOD) activities, and metabolome were estimated among four treatment groups (BR treatment, X5 + R. solanacearum (RS-15); TR treatment, Tvien6+ RS-15; TBR treatment, Tvien6 + X5 + RS-15; and R treatment, RS-15); (3) Results: The R treatment group had the highest disease incidence and lowest plant heights, plant weights, SPAD values, defensive enzyme activities, and D-fructose and D-glucose contents; the TBR treatment group had the lowest disease incidence and highest plant heights, plant weights, SPAD values, defensive enzyme activities, and D-fructose and D-glucose contents; (4) Conclusions: The results revealed that Tvien6 and X5 can both individually promote tomato plant growth, increase leaf chlorophyll content, enhance defensive enzyme activities, and induce the accumulation of D-fructose and D-glucose; however, they were more effective when combined.

Plant bacterial and fungal diseases cause significant agricultural losses and need to be controlled. Beneficial bacteria are promising candidates for controlling these diseases. In this study, Streptomyces sp. JCK-6131 exhibited broad-spectrum antagonistic activity against various phytopathogenic bacteria and fungi. In vitro assays showed that the fermentation filtrate of JCK-6131 inhibited the growth of bacteria and fungi with minimum concentration inhibitory (MIC) values of 0.31–10% and 0.31–1.25%, respectively. In the in vivo experiments, treatment with JCK-6131 effectively suppressed the development of apple fire blight, tomato bacterial wilt, and cucumber Fusarium wilt in a dose-dependent manner. RP-HPLC and ESI-MS/MS analyses indicated that JCK-6131 can produce several antimicrobial compounds, three of which were identified as streptothricin E acid, streptothricin D, and 12-carbamoyl streptothricin D. In addition, the disease control efficacy of the foliar application of JCK-6131 against tomato bacterial wilt was similar to that of the soil drench application, indicating that JCK-6131 could enhance defense resistance in plants. Molecular studies on tomato plants showed that JCK-6131 treatment induced the expression of the pathogenesis-related (PR) genes PR1, PR3, PR5, and PR12, suggesting the simultaneous activation of the salicylate (SA) and jasmonate (JA) signaling pathways. The transcription levels of PR genes increased earlier and were higher in treated plants than in untreated plants following Ralstonia solanacearum infection. These results indicate that Streptomyces sp. JCK-6131 can effectively control various plant bacterial and fungal diseases via two distinct mechanisms of antibiosis and induced resistance.

Bacterial wilt caused by Ralstonia solanacearum is a distributed and worldwide soil-borne disease. The application of biocontrol microbes or agricultural chemicals has been widely used to manage tomato bacterial wilt. However, whether and how agricultural chemicals affect the antagonistic ability of biocontrol microbes is still unknown. Here, we combined potassium phosphite (K-Phite), an environmentally friendly agricultural chemical, and the biocontrol agent Bacillus amyloliquefaciens QPF8 (strain F8) to manage tomato bacterial wilt disease. First, K-Phite at a concentration of 0.05% (w/v) could significantly inhibit the growth of Ralstonia solanacearum. Second, 0.05% K-Phite enhanced the antagonistic capability of B. amyloliquefaciens F8. Third, the greenhouse soil experiments showed that the control efficiency for tomato bacterial wilt in the combined treatment was significantly higher than that of the application of B. amyloliquefaciens F8 or K-Phite alone. Overall, our results highlighted a novel strategy for the control of tomato bacterial wilt disease via application and revealed a new integrated pattern depending on the enhancement of the antagonistic capability of biocontrol microbes by K-Phite.
