Mucosal-Associated Invariant T (MAIT) cells have been shown to play protective roles during infection with diverse pathogens through their propensity for rapid innate-like cytokine production and cytotoxicity. Among the potential applications for MAIT cells is to defend against Staphylococcus aureus, a pathogen of serious clinical significance. However, it is unknown how MAIT cell responses to S. aureus are elicited, nor has it been investigated whether MAIT cell cytotoxicity is mobilized against intracellular S. aureus. In this study, we investigate the capacity of human MAIT cells to respond directly to S. aureus. MAIT cells co-cultured with dendritic cells (DCs) infected with S. aureus rapidly upregulate CD69, express IFNγ and Granzyme B and degranulate. DC secretion of IL-12, but not IL-18, was implicated in this immune response, while TCR binding of MR1 is required to commence cytokine production. MAIT cell cytotoxicity resulted in apoptosis of S. aureus-infected cells, and reduced intracellular persistence of S. aureus. These findings implicate these unconventional T cells in important, rapid anti-S. aureus responses that may be of great relevance to the ongoing development of novel anti-S. aureus treatments.

IT is being increasingly used in most areas of life. With the IoT, this technology is set to be in a state of continuous evolution in urban and regional settings. The ongoing development of digitalization processes also increases the possibilities of abuse—both at the technical and interpersonal level. Better information security (IS) awareness (ISA) and knowledge about the dangers that accompany digitalization and the corresponding protective measures are important in private and work life. However, ISA is often overlooked. Training the relevant awareness and skills should also be included in urban and regional planning for citizens. This article thus provides a review of the scientific literature of leading academic journals in the area of IS and the transfer of scientific knowledge for practical purposes. The article presents Serious Games as a way to achieve a deeper understanding of how to promote sustainable ISA using creative methods. Furthermore, ideas of how to apply the Fun Theory and its practice to integrate awareness into modern urban and regional planning will be discussed.

The reviewed publication, published in 2019 by the British publishing house Archaeopress, is a collection of texts based on papers and posters presented at the international conference under the same title held in Prague in April 2018. This meeting brought together archaeologists from many European countries – including Croatia, Czechia, Germany, Norway, Poland, Portugal, Russia, Slovakia, Switzerland, Italy, and Hungary – who conduct research on the early-modern period and the pertaining material culture. Both this event and the book in question were a response to the needs of the academic community, due to the ongoing development of historical archaeology in Europe and an increase in research on artefacts and other evidence recovered during fieldwork related to this. The time-frame of the post-medieval period differs slightly across particular European countries, encompassing artefacts from between the 15th and 18th centuries.

AbstractThis paper critically discusses blind spots in AI ethics. AI ethics discourses typically stick to a certain set of topics concerning principles evolving mainly around explainability, fairness, and privacy. All these principles can be framed in a way that enables their operationalization by technical means. However, this requires stripping down the multidimensionality of very complex social constructs to something that is idealized, measurable, and calculable. Consequently, rather conservative, mainstream notions of the mentioned principles are conveyed, whereas critical research, alternative perspectives, and non-ideal approaches are largely neglected. Hence, one part of the paper considers specific blind spots regarding the very topics AI ethics focusses on. The other part, then, critically discusses blind spots regarding to topics that hold significant ethical importance but are hardly or not discussed at all in AI ethics. Here, the paper focuses on negative externalities of AI systems, exemplarily discussing the casualization of clickwork, AI ethics’ strict anthropocentrism, and AI’s environmental impact. Ultimately, the paper is intended to be a critical commentary on the ongoing development of the field of AI ethics. It makes the case for a rediscovery of the strength of ethics in the AI field, namely its sensitivity to suffering and harms that are caused by and connected to AI technologies.

AbstractGas therapy (GT) has attracted increasing attention in recent years as a new cancer treatment method with favorable therapeutic efficacy and reduced side effects. Several gas molecules, such as nitric oxide (NO), carbon monoxide (CO), hydrogen (H2), hydrogen sulfide (H2S) and sulfur dioxide (SO2), have been employed to treat cancers by directly killing tumor cells, enhancing drug accumulation in tumors or sensitizing tumor cells to chemotherapy, photodynamic therapy or radiotherapy. Despite the great progress of gas therapy, most gas molecules are prone to nonspecific distribution when administered systemically, resulting in strong toxicity to normal tissues. Therefore, how to deliver and release gas molecules to targeted tissues on demand is the main issue to be considered before clinical applications of gas therapy. As a specific and noninvasive stimulus with deep penetration, near-infrared (NIR) light has been widely used to trigger the cleavage and release of gas from nano-prodrugs via photothermal or photodynamic effects, achieving the on-demand release of gas molecules with high controllability. In this review, we will summarize the recent progress in cancer gas therapy triggered by NIR light. Furthermore, the prospects and challenges in this field are presented, with the hope for ongoing development.

AbstractOver the last decade, several publications have reported the safety and efficacy of liquid embolics in a wide range of clinical indications. The expansion of clinical scenarios in which liquid embolics are utilized has been in part driven by some of the advantages these materials hold over other embolic agents. For instance, liquid embolics do not rely on the patient's ability to form clot and can be delivered more distally then coils at times. While several liquid embolics are commercially available, and in some cases have been for decades, there are limitations to these materials as well. The potential benefits of liquid embolics combined with the limitations of available materials have led to the ongoing development of several new agents. This review aims to familiarize the reader with both available liquid embolics and those that are relatively new or in development.

As a result of the steadily ongoing development of microfluidic cultivation (MC) devices, a plethora of setups is used in biological laboratories for the cultivation and analysis of different organisms. Because of their biocompatibility and ease of fabrication, polydimethylsiloxane (PDMS)-glass-based devices are most prominent. Especially the successful and reproducible cultivation of cells in microfluidic systems, ranging from bacteria over algae and fungi to mammalians, is a fundamental step for further quantitative biological analysis. In combination with live-cell imaging, MC devices allow the cultivation of small cell clusters (or even single cells) under defined environmental conditions and with high spatio-temporal resolution. Yet, most setups in use are custom made and only few standardised setups are available, making trouble-free application and inter-laboratory transfer tricky. Therefore, we provide a guideline to overcome the most frequently occurring challenges during a MC experiment to allow untrained users to learn the application of continuous-flow-based MC devices. By giving a concise overview of the respective workflow, we give the reader a general understanding of the whole procedure and its most common pitfalls. Additionally, we complement the listing of challenges with solutions to overcome these hurdles. On selected case studies, covering successful and reproducible growth of cells in MC devices, we demonstrate detailed solutions to solve occurring challenges as a blueprint for further troubleshooting. Since developer and end-user of MC devices are often different persons, we believe that our guideline will help to enhance a broader applicability of MC in the field of life science and eventually promote the ongoing advancement of MC.

Adopting a creative yet novel autoethnographic approach, this study explores the experiences of the first author, a newly qualified footballing official. In doing so, the study provides a first person account to showcase the realities of refereeing whereby adding to a small pool of refereeing literature in the process. In providing an evocative account with a theoretical analysis, the research aims to both showcase and explain the demands associated with the position. Therefore, by constructing the said narratives in an easy to understand manner, the study looks to showcases the challenges associated with officiating to a broad audience. While, the study gives a viable explanation to why many newly qualified referees drop out from the role, the study hopes to inform and subsequently aid aspiring officials in their ongoing development.

AbstractThe ongoing development of eXtended Reality (XR) technologies is supporting a rapid increase of their performances along with a progressive decrease of their costs, making them more and more attractive for a large class of consumers. As a result, their widespread use is expected within the next few years. This may foster new opportunities for e-commerce strategies, giving birth to an XR-based commerce (x-commerce) ecosystem. With respect to web and mobile-based shopping experiences, x-commerce could more easily support brick-and-mortar store-like experiences. One interesting and consolidated one amounts to the interactions among customers and shop assistants inside fashion stores. In this work, we concentrate on such aspects with the design and implementation of an XR-based shopping experience, where vocal dialogues with an Amazon Alexa virtual assistant are supported, to experiment with a more natural and familiar contact with the store environment. To verify the validity of such an approach, we asked a group of fashion experts to try two different XR store experiences: with and without the voice assistant integration. The users are then asked to answer a questionnaire to rate their experiences. The results support the hypothesis that vocal interactions may contribute to increasing the acceptance and comfortable perception of XR-based fashion shopping.

Fibroblast growth factor 19 (FGF19) is a hormone with pleiotropic metabolic functions, leading to ongoing development of analogues for the treatment of metabolic disorders. On the other hand, FGF19 is overexpressed in a sub-group of hepatocellular carcinoma (HCC) patients and has oncogenic properties. It is therefore crucial to precisely define FGF19 effects, notably chronic exposure to elevated concentrations of the hormone. We used hydrodynamic gene transfer approach to generate a transgenic mouse model with long-term FGF19 hepatic overexpression. Here we describe a novel effect of FGF19, namely stimulation of water intake. This phenotype, lasting at least over a 6-month period, depends on signaling in the central nervous system and is independent of FGF21, although it mimics some of its features. We further show that HCC patients with high levels of circulating FGF19 have a reduced natremia, indicating dispogenic features. The present study provides evidence of a new activity for FGF19, which could be clinically relevant in the context of FGF19 overexpressing cancers and treatment of metabolic disorders by FGF19 analogues.
