AbstractSynthetic biologists use and combine diverse biological parts to build systems such as genetic circuits that perform desirable functions in, for example, biomedical or industrial applications. Computer-aided design methods have been developed to help choose appropriate network structures and biological parts for a given design objective. However, they almost always model the behavior of the network in an average cell, despite pervasive cell-to-cell variability. Here, we present a computational framework to guide the design of synthetic biological circuits while accounting for cell-to-cell variability explicitly. Our design method integrates a NonLinear Mixed-Effect (NLME) framework into an existing algorithm for design based on ordinary differential equation (ODE) models. The analysis of a recently developed transcriptional controller demonstrates first insights into design guidelines when trying to achieve reliable performance under cell-to-cell variability. We anticipate that our method not only facilitates the rational design of synthetic networks under cell-to-cell variability, but also enables novel applications by supporting design objectives that specify the desired behavior of cell populations.

AbstractThe use of orthogonal ribosomes in combination with dynamic resource allocation controllers is a promising approach for relieving the negative effects of cellular resource limitations on the modularity of synthetic gene circuits. Here, we develop a detailed mechanistic model of gene expression and resource allocation, which when simplified to a tractable level of complexity, allows the rational design of translational resource allocation controllers. Analysis of this model reveals a fundamental design trade-off; that reducing coupling acts to decrease gene expression. Through a sensitivity analysis of the experimentally tuneable controller parameters, we identify how each controller design parameter affects the overall closed-loop behaviour of the system, leading to a detailed set of design guidelines for optimally managing this trade-off. Based on our designs, we evaluated a number of alternative potential experimental implementations of the proposed system using commonly available biological components. Finally, we show that the controller is capable of dynamically allocating ribosomes as needed to restore modularity in a number of more complex synthetic circuits, such as the repressilator, and activation cascades composed of multiple interacting modules.

For several decades, disturbance observers (DOs) have been widely utilized to enhance tracking performance by reducing external disturbances in different industrial applications. However, although a DO is a verified control structure, a conventional DO does not guarantee stability. This paper proposes a stability-guaranteed design method, while maintaining the DO structure. The proposed design method uses a linear matrix inequality (LMI)-based H∞ control because the LMI-based control guarantees the stability of closed loop systems. However, applying the DO design to the LMI framework is not trivial because there are two control targets, whereas the standard LMI stabilizes a single control target. In this study, the problem is first resolved by building a single fictitious model because the two models are serial and can be considered as a single model from the Q-filter point of view. Using the proposed design framework, all-stabilizing Q filters are calculated. In addition, for the stability and robustness of the DO, two metrics are proposed to quantify the stability and robustness and combined into a single unified index to satisfy both metrics. Based on an application example, it is verified that the proposed method is effective, with a performance improvement of 10.8%.

Purpose Additive manufacturing (AM) is a rapidly evolving manufacturing process, which refers to a set of technologies that add materials layer-by-layer to create functional components. AM technologies have received an enormous attention from both academia and industry, and they are being successfully used in various applications, such as rapid prototyping, tooling, direct manufacturing and repair, among others. AM does not necessarily imply building parts, as it also refers to innovation in materials, system and part designs, novel combination of properties and interplay between systems and materials. The most exciting features of AM are related to the development of radically new systems and materials that can be used in advanced products with the aim of reducing costs, manufacturing difficulties, weight, waste and energy consumption. It is essential to develop an advanced production system that assists the user through the process, from the computer-aided design model to functional components. The challenges faced in the research and development and operational phase of producing those parts include requiring the capacity to simulate and observe the building process and, more importantly, being able to introduce the production changes in a real-time fashion. This paper aims to review the role of robotics in various AM technologies to underline its importance, followed by an introduction of a novel and intelligent system for directed energy deposition (DED) technology. Design/methodology/approach AM presents intrinsic advantages when compared to the conventional processes. Nevertheless, its industrial integration remains as a challenge due to equipment and process complexities. DED technologies are among the most sophisticated concepts that have the potential of transforming the current material processing practices. Findings The objective of this paper is identifying the fundamental features of an intelligent DED platform, capable of handling the science and operational aspects of the advanced AM applications. Consequently, we introduce and discuss a novel robotic AM system, designed for processing metals and alloys such as aluminium alloys, high-strength steels, stainless steels, titanium alloys, magnesium alloys, nickel-based superalloys and other metallic alloys for various applications. A few demonstrators are presented and briefly discussed, to present the usefulness of the introduced system and underlying concept. The main design objective of the presented intelligent robotic AM system is to implement a design-and-produce strategy. This means that the system should allow the user to focus on the knowledge-based tasks, e.g. the tasks of designing the part, material selection, simulating the deposition process and anticipating the metallurgical properties of the final part, as the rest would be handled automatically. Research limitations/implications This paper reviews a few AM technologies, where robotics is a central part of the process, such as vat photopolymerization, material jetting, binder jetting, material extrusion, powder bed fusion, DED and sheet lamination. This paper aims to influence the development of robot-based AM systems for industrial applications such as part production, automotive, medical, aerospace and defence sectors. Originality/value The presented intelligent system is an original development that is designed and built by the co-authors J. Norberto Pires, Amin S. Azar and Trayana Tankova.

In der Stückgutindustrie lassen sich die acht Gestaltungsrichtlinien der Wertstrommethode nach Erlach hervorragend anwenden. In der Prozessindustrie weist die Produktion jedoch häufig andere Merkmale (beispielsweise eine Kuppelproduktion) auf, die neue Herausforderungen an die Wertstrommethode stellen. Aufbauend auf den acht Gestaltungsrichtlinien des Wertstromdesigns werden in diesem Fachbeitrag Handlungsempfehlungen für die Anwendung der Wertstrommethode in der Prozessindustrie diskutiert.   The eight design guidelines of Erlach‘s Value Stream Design Method work well in streamlining operations in the piece-good industry. In the process and chemical industries, however, production systems exhibit different characteristics, like joint production, that present challenges for the eight step value stream design method. Building on the eight design guidelines, this article discusses the deficits of this method in the process industries and gives recommendations for its adaptatio.

Expert systems, or knowledge based systems, are programs in which the answer to a user-posed question is reached by logical or plausible inference rather than strictly by calculation, although calculation routines can form a major part of an expert system. Based on the integration of expert system technology and optimization technology, an intelligent computer aided design method for mine ventilation systems is proposed in this paper. Firstly, the structure and control algorithm of the intelligent design system are explored. Secondly, the knowledge types required for the mine ventilation expert system and the acquiring method of knowledge are discussed. Finally, the inference method of this expert system is put forward.

A computer-aided design system has been developed for hydraulic parts of pumps including impellers, bowl diffusers, volutes, and vaned return channels. The key technologies include three-dimensional (3-D) CAD modeling, automatic grid generation, CFD analysis, and a 3-D inverse design method. The design system is directly connected to a rapid prototyping production system and a flexible manufacturing system composed of a group of DNC machines. The use of this novel design system leads to a drastic reduction of the development time of pumps having high performance, high reliability, and innovative design concepts. The system structure and the design process of “Blade Design System” and “Channel Design System” are presented. Then the design examples are presented briefly based on the previous publications, which included a centrifugal impeller with suppressed secondary flows, a bowl diffuser with suppressed corner separation, a vaned return channel of a multistage pump, and a volute casing. The results of experimental validation, including flow fields measurements, were also presented and discussed briefly.

Abstract Additive Manufacturing processes are considered advanced manufacturing methods. It would be possible to produce complex shape components from a Computer-Aided Design model in a layer-by-layer manner. Lattice structures as one of the complex geometries could attract lots of attention for both medical and industrial applications. In these structures, besides cell size and cell type, the microstructure of lattice structures can play a key role in these structures' mechanical performance. On the other hand, heat treatment has a significant influence on the mechanical properties of the material. Therefore, in this work, the effect of the heat treatments on the microstructure and mechanical behaviour of Ti-6Al-4V lattice structures manufactured by EBM was analyzed. The main mechanical properties were compared with the Ashby and Gibson model. It is very interesting to notice that a more homogeneous failure mode was found for the heat-treated samples. The structures' relative density was the main factor influencing their mechanical performance of the heat-treated samples. It is also found that the heat treatments were able to preserve the stiffness and the compressive strength of the lattice structures. Besides, an increment of both the elongation at failure and the absorbed energy was obtained after the heat treatments. Microstructure analysis of the heat-treated samples confirms the increment of ductility of the heat-treated samples with respect to the as-built one.

Abstract Recently, there is an increasing interest in design of circular weft jacquard because of the pursuit of fashion and comfort. Aiming at the complexity of the computer-aided design method of the existing circular weft-knitted jacquard fabrics, which is not conducive to the rapid design and intelligible for designers, a design method was proposed to transform pattern notation into knitting diagram efficiently, which was based on knitting rules and its creation as a set of jacquard modules. Knitting characteristics of jacquard fabrics were studied as a precondition. On this basis, the design procedures of jacquard modules were analyzed and illustrated by taking tricolor bird's eye backing jacquard as an example. Jacquard modules with various jacquard effects were designed and stored in a jacquard module database. To mathematically describe pattern notation, knitting diagram, and jacquard module, two-dimensional matrixes were established by the method of mathematical modeling, and a corresponding algorithm for the transformation of the pattern to knitting information according to the knitting rules of jacquard modules, which can be applied to ordinary jacquard fabrics was summarized. The project of tricolor circular weft-knitted jacquard with bird's eye in the reverse and four-color air-layer jacquard were taken for instance to verify the models and algorithm. The results obtained show that the approach can efficiently and conveniently realize the designation and machine-knitting of weft-knitted jacquard fabric, which provide a theoretical basis and notation of modeling for the computer-aided design of circular weft-knitted jacquard fabrics.

With the objective of developing a computer-aided design method to seek the optimal semisubmersible’s form, hierarchical relationships among many design objectives and conditions are investigated first based on the interpretive structural modeling method. Then, an optimal design method is formulated as a nonlinear multiobjective optimization problem by adopting three mutually conflicting design objectives. A set of Pareto optimal solutions is derived numerically by adopting the generalized reduced gradient algorithm, and it is ascertained that the designer can determine the optimal form more rationally by investigating the trade-off relationships among design objectives.
