In this research work, the antioxidant and metabolomic profiling of seven selected medicinally important herbs including Rauvolfia serpentina, Terminalia arjuna, Coriandrum sativum, Elettaria cardamom, Piper nigrum, Allium sativum, and Crataegus oxyacantha was performed. The in vivo cardioprotective potential of these medicinal plants was evaluated against surgically induced oxidative stress through left anterior descending coronary artery ligation (LADCA) in dogs. The antioxidant profiling of these plants was done through DPPH and DNA protection assay. The C. oxyacantha and T. arjuna showed maximum antioxidant potential, while the E. cardamom showed poor antioxidative strength even at its high concentration. Different concentrations of extracts of the said plants exhibited the protection of plasmid DNA against H2O2 damage as compared to the plasmid DNA merely treated with H2O2. The metabolomic profiling through LC-MS analysis of these antioxidants revealed the presence of active secondary metabolites responsible for their antioxidant potential. During in vivo analysis, blood samples of all treatment groups were drawn at different time intervals to analyze the cardiac and hemodynamic parameters. The results depicted that the group pretreated with HC4 significantly sustained the level of CK-MB, SGOT, and LDH as well as hemodynamic parameters near to normal. The histopathological examination also confirmed the cardioprotective potential of HC4. Thus, the HC4 being safe and inexpensive cardioprotective herbal combination could be considered as an alternate of synthetic drugs.

Medicinal plants have bioactive compounds which are used for curing of various human diseases and also play an important role in healing. Secondary constituents contain alkaloids, flavonoids, phenol, sapon in, steroids and tannins. Medicinal plants have anticancer, antimicrobial, antidiabetic, antidiuretic and anti-inflammation activities. The increasing interest in powerful biological activity of secondary metabolites outlined the necessity of determining their contents in medicinal plants. In Indian Ayurvedic system, Lagerstroemia Indica (Lythraceae) are well-known plants used for major and minor ailments. The aim of the present study is to examine leaf of Lagerstroemia Indica for phytochemical profile, antioxidant potential, antimicrobial including anti acne activity. Qualitative analysis of various phytochemical constituents and quantitative analysis of total phenolics and flavonoids were determined by the well-known test protocol available in the literature. Quantitative analysis of phenolic and flavonoids was carried out by FolinsCiocalteau reagent method and aluminium chloride method respectively. Phytochemical analysis revealed the presence of phenols, flavonoids, tannins, saponins, alkaloids, fixed oil and fats. The present study concluded that the crude extract of Lagerstroemia Indica is a rich source of secondary phytoconstituents which impart significant antioxidant potential.  The extract of Lagerstroemia Indica possess antimicrobial effectiveness againt P. acne and other microbes, In Vitro anti acne activity. Objective: The aim of this study is to evaluate the antioxidant potential, determination of total phenolic and flavonoid content in nine selected medicinal plants Spondias pinnata, Melia azedarach, Ageratina adenophora, Urtica dioica, Curcuma longa, Bauhinia variegata, Elaeocarpus angustifolius Blume, Achyranthes aspera, and Psidium guajava from Kavre district of Nepal using in vitro studies. Methods: Methanolic plant extracts were prepared by cold percolation method. The methanol extract of nine medicinal plants collected from Kavre district of Nepal, was screened for assessing bioactive phytoconstituents followed by antioxidant property, total phenolic, and flavonoid content. Different plants collected were powdered and extracted with methanol, concentrated by a rotatory evaporator and analyzed for the presence of phytochemicals. The antioxidant potential of the plant extracts was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Results: The phytochemical analysis of methanolic extracts of all nine medicinal plants displayed the presence of various secondary metabolites such as alkaloids, flavonoids, polyphenols, saponins, and quinones. The extract of S. pinnata showed the highest percentage of radical scavenging activity up to 87.94±1.88 with 50% inhibitory concentration (IC50) 17.51±1.27 μg/mL, followed by B. variegata, 80.63±1.06 with IC50 value 26.55±2.61 μg/mL. The standard, ascorbic acid has IC50 value of 20.13±1.17 μg/mL. Further, the ethyl acetate fraction of S. pinnata showed the maximum percentage of radical scavenging (85.92±1.37) with IC50 value of 46.95±1.17 μg/mL. Moreover, S. pinnata displayed the highest total phenolic content (TPC) 48.26±1.23 mg GAE/g (milligram gallic acid equivalent per gram) extract while the highest flavonoid content was displayed by Melia azedarach 41.07±1.53 mg QE/g (milligram quercetin equivalent per gram) extract measured by the Folin–Ciocalteu reagent method and aluminum chloride colorimetric method. Conclusions: The preliminary results of this study have put forward the extract of S. pinnata showed the highest percentage of radical scavenging activity and S. pinnata displayed the highest TPC while the highest flavonoid content was displayed by Melia azedarach methanolic extracts although the further studies are needed to assess its mechanism of action.

Free radical-mediated oxidation is often linked to various degenerative diseases. Biological substrates with lipids as major components are susceptible to oxygen-derived lipid peroxidation due to their composition. Lipid peroxide products act as biomarkers in evaluating the antioxidant potential of various plants and functional foods. The study focused on evaluation of the antioxidant potential of two extracts (methanol and 80% methanol) of four medicinal plants,Andrographis paniculata,Costus speciosus, Canthium parviflorum, andAbrus precatorius, against Fenton reaction-mediated oxidation of three biological lipid substrates; cholesterol, low-density lipoprotein, and brain homogenate. The antioxidant activity of the extracts was measured by thiobarbituric acid reactive substances method. Also, the correlation between the polyphenol, flavonoid content, and the antioxidant activity in biological substrates was analyzed. Results indicated highest antioxidant potential by 80% methanol extract ofCanthium parviflorum(97.55%), methanol extract ofAndrographis paniculata(72.15%), and methanol extract ofCanthium parviflorum(49.55%) in cholesterol, low-density lipoprotein, and brain, respectively. The polyphenol and flavonoid contents of methanol extract ofAndrographis paniculatain cholesterol (r=0.816) and low-density lipoprotein (r=0.948) andCostus speciosusin brain (r=0.977, polyphenols, andr=0.949, flavonoids) correlated well with the antioxidant activity. The findings prove the antioxidant potential of the selected medicinal plants against Fenton reaction in biological lipid substrates.

Evaluation of flavonoid contents from three selected medicinal plant species of capparidaceae family growing in North–Western Rajasthan was carried out. The leaves of Capparis decidua, Cleome gynandra and Cleome viscosa were analysed for flavonoid contents i.e. Quercetin and Kaempferol. Flavonoid contents like Quercetin and Kaempferol were isolated and identified. The maximum total flavonoid contents (1.16mg./gdw) was found in leaves of Capparis decidua while minimum (0.71mg./gdw) in leaves of Cleome viscosa.

Context: The medicinal plants have enormous pharmacological properties and having fewer side effects. Today there is increasing demand of medicinal plants as an anti-aging and anti-wrinkle agent. Objective: The aim of this study is to evaluate antioxidant, anti-aging and anti-wrinkle potential of Salvia officinalis. Materials and Methods: Salvia officinalis (Lamiaceae) is folk medicine of Asia and Latin America. Powdered crude drug 100 g were successively extracted in a soxhlet apparatus with petroleum ether (60-80ºC), chloroform and methanol. After successive solvents extraction methanolic extract was used for testing of antioxidant potential using DPPH assay. Further, antiaging potential of extract was investigated by inhibitory effect of various enzymatic estimations i.e. Col-I, Ela-I and Hya-I inhibitory assays on early aging human skin fibroblasts. Antiwrinkle potential of plant Salvia officinalis was done by using UV light induced photo aging model. Results: Phytochemical analysis showed the presence of glycosides, alkaloids flavonoids, and triterpenoids, saponins and Phenolic compounds in high level. Extract showed inhibitory concentration (IC50:24.65) and ascorbic acid the standard antioxidant showed inhibitory concentration (IC50:20.10). In enzymatic estimations assay, the Col-I, Ela-I and Hya-I of extract were assessed showing inhibitory concentration as Col-I (IC50:21.36), Ela-I (IC50:35.05) and Hya-I (IC50:23.44) respectively. Thus, MeOH extract of Salvia officinalis able to inhibit 50% of the activity of aging related enzymes Col-I, Ela-I and Hya-I. The wrinkle score of negative control i.e. UV treated group was 2.83±0.408 and MeOH extract of Salvia officinalis treated group is 1.83 ±0.753. Conclusion: This study concluded that MeOH extract of Salvia officinalis has confirmed the high antioxidant potential and In vitro and In vivo inhibitory potential of antiaging enzymes assessed, thus they could be used for further development of cosmetic products and nutraceuticals.
