SUMMARY A total of twenty-three human pre-viable foetuses (7–400 g.) were removed by abdominal hysterotomy (legal abortions) and studied during survival in an anoxic state at different temperatures. The duration of survival, as judged by ECG waves, at 37° c was about 3 hr.; it was longer for female foetuses than for males, and longer for large foetuses than for small ones. General reduction of the body-temperature to 4° c during periods varying between 30 min. and 6 hr. with subsequent rewarming to and maintenance at 37° c increased the survival time by a further 1–2 hr. The blood sugar did not decrease either during the period of survival or during hypothermia, but the non-protein nitrogen increased, while acidosis and hyperkalaemia developed. The glycogen content of the liver decreased during the anoxic period of survival and diminished further during hypothermia. The adrenal glands produced adrenaline in addition to noradrenaline during the anoxic phase, and this may occur in a very early stage of intrauterine life.

X-ray Photoelectron Spectroscopy (XPS) has been used to investigate the oxidation of (011) Ge substrates. The sample surfaces were CP4-etched, then annealed in situ, at different temperatures, for various durations. Dry and wet atmospheres were used. The oxidation rate during the early stage was increased by the presence of moisture in the atmosphere. A simple model was used to define and determine an apparent thickness of the oxide film from XPS measurements. The time dependence of the apparent thickness is consistent with a partial coverage of the surface by oxide islands. The growth kinetics of the oxide islands obeys a nearly cubic law.

2- to 4-cell and morula- to blastocyst-stage mouse embryos were cultured for 1 h in tritiated leucine at two specific activities and their subsequent development followed in vitro and in vivo (after transfer to recipients), respectively. 2- to 4-cell embryos that incorporated an average of 42 d.p.m. per embryo were impaired in their ability to develop to the morula and blastocyst stage. Recipients receiving morulae and blastocysts that had incorporated an average of 384 d.p.m. per embryo failed to produce young. Reduction of the specific activity improved the viability of embryos both in vitro and in vivo but development was still less than that of unlabelled embryos. Protein degradation curves were different for both 2- to 4-cell and morulato blastocyst-stage embryos labelled at the two different specific activities. Most studies using tritiated amino acids have employed higher specific activities than those used here and they may have to be reevaluated due to the possibility of radiation-induced artifacts.

Gaucher disease (GD) is a lysosomal storage disorder that responds well to enzyme replacement therapy (ERT). Certain laboratory parameters, including blood concentration of glucosylsphingosine (Lyso-Gb1), the lyso-derivate of the common glycolipid glucocerebroside, correlate with clinical improvement and are therefore considered candidate-monitoring biomarkers. Whether they can indicate a reduction or loss of treatment efficiency, however, has not been systematically addressed for obvious reasons. We established and validated measurement of Lyso-Gb1 from dried blood spots (DBSs) by mass spectrometry. We then characterized the assay’s longitudinal performance in 19 stably ERT-treated GD patients by dense monitoring over a 3-year period. The observed level of fluctuation was accounted for in the subsequent development of a unifying data normalization concept. The resulting approach was eventually applied to data from Lyso-Gb1 measurements after an involuntary treatment break for all 19 patients. It enabled separation of the “under treatment” versus “not under treatment” conditions with high sensitivity and specificity. We conclude that Lyso-Gb1 determination from DBSs indicates treatment issues already at an early stage before clinical consequences arise. In addition to its previously shown diagnostic utility, Lyso-Gb1 thereby qualifies as a monitoring biomarker in GD patients.

The influence of temperature on atomic site occupation probability (SOP) in L12--Ni3(Al1-xFex) has been studied using microscopic phase-field simulation for Ni3Al-Fe alloy with the lower Fe content. Microscopic phase-field kinetics model was further extended to study the temporal evolution of atomic site occupation and atomic anti-sites behavior, which was revealed in detail at the different temperatures. The results show that the size, number and the precipitation mechanism of L12phases also present obvious changes with the increase of temperature. The SOP of each atom on its right-sites decreases slowly; while the SOP of Fe atoms are far lower than those of Al atoms. On the other hand, the SOP of each atom on its anti-sites increases gradually. The formation of NiAl(Ni atoms form anti-sites on Al sites) is more easily and the formation of NiAlis more sensitive to temperature than that of FeNi(Fe atoms form anti-sites on Ni sites). Atomic site occupation will not evolve during the early stage of precipitation, which becomes more and more obvious.

We have used microinjection of antisense oligonucleotides, monoclonal antibody, and the dominant negative Ras N-17 mutant to interfere with Ras expression and function in mouse oocytes and early embryos. Microinjection of either ras antisense oligonucleotides or anti-Ras monoclonal antibody Y13-259 did not affect normal progression of oocytes through meiosis and arrest at metaphase II. However, microinjection of fertilized eggs with constructs expressing Ras N-17 inhibited subsequent development through the two-cell stage. The inhibitory effect of Ras N-17 was overcome by simultaneous injection of a plasmid expressing an active raf oncogene, indicating that it resulted from interference with the Ras/Raf signaling pathway. In contrast to the inhibition of two-cell embryo development resulting from microinjection of pronuclear stage eggs, microinjection of late two-cell embryos with Ras N-17 expression constructs did not affect subsequent cleavages and development to morulae and blastocysts. It thus appears that the Ras/Raf signaling pathway, presumably activated by autocrine growth factor stimulation, is specifically required at the two-cell stage, which is the time of transition between maternal and embryonic gene expression in mouse embryos.

The direct nitridation process of silicon added with amorphous silicon nitride powder at atmospheric pressure was investigated and the product was analyzed by XRD and SEM. Based on the relationship between the conversion ratio of silicon and the reaction time at different temperatures, a physical and mathematical model was derived to describe the nitridation process of silicon particles. The results showed that the conversion ratio of silicon increased rapidly at the early stage of reaction. And the reaction would be accelerated by reducing the size of silicon particle and increasing the pressure of N2. At the range of experimental temperature, the conversion ratio of silicon increases with improving temperature.

Chronic inflammation and long-standing oxidative stress are potential predisposing factors for developing malignancies, including ovarian cancer (OVCA). Information on the association of ovarian chronic abnormal conditions, including polycystic ovarian syndrome (PCOS), with the development of OVCA is unknown. The goal of this study was to examine if polycystic ovarian conditions are associated with OVCA development. In the exploratory study, 3–4-year-old laying hens were randomly selected and examined for the presence of polycystic ovaries with cancer (PCOC). In the prospective study, hens were monitored by ultrasound scanning to detect the incidence of a polycystic ovaries and subsequent development of OVCA. Tissues from normal ovaries and PCOC were examined for macrophage infiltration, expression of interleukin-16, and superoxide dismutase 2. The exploratory study detected spontaneous PCOC at early and late stages in hens. PCOC in hens were accompanied with influx of macrophages (17.33 ± 2.26 in PCOC at the early stage and 24.24 ± 2.5 in PCOC at the late stage in 20 mm2 areas of tissue as compared with 6.77 ± 1.58 in normal hens). Expression of interleukin-16 was more than 2.5-fold higher and superoxide dismutase 2 was approximately 3-fold higher in PCOC hens than normal hens. The prospective study showed the development of OVCA in some hens with polycystic ovarian condition (PCO). PCOC development in hens was associated with chronic inflammation in the ovary. Laying hens may represent a potential model for the study of spontaneous PCOS and its long-term risk of PCOC development.

A nanostructured surface layer on a pure iron sample was prepared by surface mechanical attrition treatment (SMAT). The thermal stability of SMAT sample was investigated at different temperatures with or without a high magnetic field (H =12T). It was found that a high magnetically annealing enhanced grain growth at the early stage of annealing, and produced a uniform nanocrystalline grain structure. After homogeneous grains developed, further grain growth became restrained.
