SummaryThe injection of PAP (6 μg/kg, i. v.) induced, in rats, haemoconcentration accompanied by an increase in the platelet number, as attested by the counts of platelets in blood samples diluted in formalin-free EDTA solution. This increase was significant at 15 min, peaked from 1 to 4 h and returned to basal levels 24 h after the lipid administration. The release of platelets induced by PAP was inhibited dose-dependently by specific PAP receptor antagonists such as WEB 2086 (0.5-2 mg/kg), BN 52021 and 48740 RP (5-25 mg/kg). Furthermore, platelet mobilization was clearly impaired in splenectomized animals stimulated by PAP, whereas thrombocytopenia and haemoconcentration by the same stimulus were intact. It was also noted that a second injection of PAP, 24 h after the initial stimulation with the lipid, failed to induce an increase in platelet counts, indicating autodesensitization. Desensitization to PAP or pretreatment with PAP antagonists clearly prevented the increase in the platelet counts after stimulation by adrenaline (15 μg/kg). These findings suggest that, in rats, PAP can induce release of platelets by a spleen-dependent mechanism and that this lipid may be relevant to the thrombocytosis triggered by adrenaline.

AbstractThe electrochemical behavior of gold dissolution in the Cu2+–NH3–S2O32−–EDTA solution has been investigated in detail by deriving and analyzing the Tafel polarization curve, as this method is currently widely implemented for the electrode corrosion analysis. The dissolution rate of gold in Cu2+–NH3–S2O32−–EDTA solution was determined based on the Tafel polarization curves, and the effects of various compound compositions in a Cu2+–NH3–S2O32−–EDTA mixture on the corrosion potential and corrosion current density were analyzed. The results showed that the corrosion potential and polarization resistance decreased, whereas the corrosion current density increased for certain concentrations of S2O32−–NH3–Cu2+ and EDTA, indicating that the dissolution rate of gold had changed. The reason for promoting the dissolution of gold is also discussed.

The changes in the membrane permeability to sodium, potassium, and chloride ions as well as the changes in the intracellular concentration of these ions were studied on frog sartorius muscles in Ca-free EDTA solution. It was found that the rate constants for potassium and chloride efflux became almost constant within 10 minutes in the absence of external calcium ions, that for potassium increasing to 1.5 to 2 times normal and that for chloride decreasing about one-half. The sodium influx in Ca-free EDTA solution, between 30 and 40 minutes, was about 4 times that in Ringer's solution. The intracellular sodium and potassium contents did not change appreciably but the intracellular chloride content had increased to about 4 times normal after 40 minutes. By applying the constant field theory to these results, it was concluded that (a) PCl did not change appreciably whereas PK decreased to a level that, in the interval between 10 and 40 minutes, was about one-half normal, (b) PNa increased until between 30 and 40 minutes it was about 8 times normal. The low value of the membrane potential between 30 and 40 minutes was explained in terms of the changes in the membrane permeability and the intracellular ion concentrations. The mechanism for membrane depolarization in this solution was briefly discussed.

Abstract Context.—In immunohistochemistry, nonstandardized antigen retrieval protocols and fluids, poor-quality antibodies, and the presence of endogenous biotin frequently lead to incorrect results. Recently, advanced reagents including bifunctional SkipDewax pretreatment solution (BSPS), rabbit monoclonal (RM) antibodies, and biotin-free polymer detection systems (PDSs) have been developed, which, it is claimed, resolve these problems. Objectives.—To determine whether BSPS, RM antibodies, and biotin-free PDSs improve the accuracy of immunohistochemistry; to optimize a new protocol consisting of a combination of BSPS, RM antibodies, and PDSs; and to compare it with a conventional protocol. Design.—The efficacies of BSPS, RM antibodies, and PDSs were compared with those of their respective conventional reagents using multitissue spring-roll sections. The new protocol was compared with a conventional protocol using Ki-67 immunostaining of 49 colorectal carcinoma specimens. Results.—For antigen retrieval, BSPS resulted in similar or better tissue staining than an EDTA solution, but the efficacy of BSPS decreased when it was reused. Most RM antibodies resulted in a greater proportion of positive cells than the corresponding non-RM antibodies, which did not produce satisfactory results in the absence of antigen retrieval. The PDSs Bond, ChemMate, and SuperPicture resulted in a high percentage of positive cells, good staining intensities, and low backgrounds. Other PDSs, except that from Ventana, resulted in high backgrounds and false positivity. The new combined protocol resulted in better Ki-67 staining than the conventional assay. Conclusions.—Bifunctional SkipDewax pretreatment solution, RM antibodies, and PDSs improve staining quality and diagnostic accuracy of immunohistochemistry assays and provide a foundation for standardization.

Chemically modified mesoporous silica material (SBA-15) was used for the construction of Tl(I) selective carbon paste electrode. The best response was found with the electrode containing 10% modifier as electrode material. The electrode has a lower detection limit of 6.0 × 10−9 M in a working concentration range of 1.0 × 10−8–1.0 × 10−1 M. The selectivity coefficient calculated by match potential method (MPM) shows the high selectivity of electrode towards Tl(I) over other tested ions. The electrode was successfully applied as an indicator electrode for the titration of 0.01 M TlNO3solution with standards EDTA solution and for sequential titration of mixture of different anions.

Objective: To evaluate the fracture resistance (RF) of Class II Glass-ionomer Cement (GIC) ART restorations with and without proximal retentions. Material and Methods: 20 freshly extracted human molars were used. Forty (40) standard Mesial-Occlusal (MO) and Distal-Occlusal (DO) preparations (20 for each material) were performed with a 245 bur. The unprepared surfaces of the teeth were protected with nail polish and the specimens submerged in 0.5Mol EDTA solution, pH 7.4 for 8h under stirring. The preparations were finished with dentine spoons and 50% received proximal retention with # 3 excavators. 20 cavities were restored with Chemfil Rock (10 with retention and 10 without retention) and 20 cavities were restored with Equia Fil (10 with retention and 10 with no retention) and were stored in an oven at 37ºC and 100% relative humidity for 24h and submitted to axial compression loading in Test Machine - EMIC at a rate of 0.5 mm / minute, until restoration fracture occurred. The values were analyzed by two-way ANOVA (p<0.05). Results: ChemFil Rock presented 300.84 (69.20) (without retention) and 361.70 (81.08) (with retention) and Equia Fil showed 314.60 (69.97) (without retention) and 366.67 (103.38) (with retention). Data obtained with retention were statistically superior to those obtained with non-retained ART restorations (p=0.014). No statistical differences were detected between materials (p=0.761). Conclusion: Retentive grooves improved fracture resistance of Class II GIC ART restorations. KeywordsDental materials; ART; Glass ionomer cements.

The aim of the study was to investigate the impact of soil chemistry on the concentrations of Co, Ni, Zn, Mn, Cu and Fe in oats (Avena sativa L. cv. Fiia) grown on Finnish acid sulphate (AS) soils with varying geochemical characteristics. Twenty two soil profiles, which were sampled to a depth of 1 m (five 20 cm section splits), and 26 composite oat grain samples were collected on a total of five fields. The concentrations of Co, Ni, Zn and Mn in the grains were correlated with the NH4Ac-EDTA-extractable concentrations in the soils. However, as these four chalcophilic metals are in general easily lost to drains and not retained as a large pool in the soil in easily-extractable form, also the concentrations in the oats were not in general elevated as compared with average values on other soils. On one of the fields, however, the Co and Ni concentrations in the soil, and thus also in the oats, were clearly elevated. Copper and Fe displayed no correlation between the soil and oat concentrations, indicating that the plant-uptake mechanisms are much more important than variations in geochemistry. It was suggested that the NH4Ac-EDTA solution was not efficient in extracting Fe and Cu, which shows that these metals are bound in relatively immobile oxyhydroxides.;

Polyvinyl alcohol (PVA) was grafted on graphene nanosheets (GN) in the reduction of graphene oxide with hydrazine hydrate. The obtained GN-PVA (GP) suspension was treated with the freezing–thawing cycle to fabricate 3D porous monolithic GP materials, which were modified with carbon disulfide to introduce xanthan groups on the wall of porous materials, marked as GPCs. The characterization of GPCs confirmed that PVA was attached on the surface of GNs, and xanthan groups were effectively functionalized on the porous structures, which were composed of randomly oriented GNs. The Pb[Formula: see text] adsorption pattern for GPC materials was investigated. The kinetic adsorption and isotherm data fit the pseudo second-order kinetic and the Langmuir isotherm models, respectively. The maximum adsorption capacity of Pb[Formula: see text] reached 242.7[Formula: see text]mg/g. And GPCs for Pb[Formula: see text] adsorption could be regenerated with ethylenediamine tetracetic acid (EDTA) solution for repetitious adsorption.

The nonlethal procedure of incubation in EDTA solution makes the peripheral regions of ascites sarcoma 37 cells more easily deformable, as reflected in measurements of the decreased amount of negative pressure required to suck out standard hemispherical bulges from the cells into micropipettes. The facilitation of deformability was abolished after reincubation of cells in calcium-containing saline, and this mechanical parameter was partially restored to normal after reincubation in magnesium-containing saline; the mechanical effect of EDTA treatment is, therefore, thought to be due mainly to the removal of calcium from the cell periphery. As EDTA treatment produces no detectable change in cellular electrophoretic mobility, it is concluded that peripheral calcium must be bound to anionic sites deeper than about 10 A from the cellular hydrodynamic slip plane. The data are discussed with emphasis on the view that they should not be extrapolated freely to other cell types.

Cetaceans greatly depend on their hearing system to perform many vital activities. The spiral ganglion is an essential component of the auditory pathway and can even be associated with injuries caused by anthropogenic noise. However, its anatomical location, characterized by surrounding bony structures, makes the anatomical and anatomopathological study of the spiral ganglion a difficult task. In order to obtain high-quality tissue samples, a perfect balance between decalcification and the preservation of neural components must be achieved. In this study, different methodologies for spiral ganglion sample preparation and preservation were evaluated. Hydrochloric acid had the shortest decalcification time but damaged the tissue extensively. Both formic acid and EDTA decalcification solutions had a longer decalcification time but exhibited better preservation of the neurons. However, improved cell morphology and staining were observed on ears pretreated with EDTA solution. Therefore, we suggest that decalcifying methodologies based on EDTA solutions should be used to obtain the highest quality samples for studying cell morphology and antigenicity in cetacean spiral ganglion neurons.
