Fetal myocardial hypertrophy in an experimental model of gestational diabetes

Introduction: The frequent occurrence of prenatal hypertrophy of the muscular ventricular septum has been widely reported in fetuses of diabetic mothers. Objectives: This experimental study was carried out to test the hypothesis that the weight of the heart, the ratio of the cardiac weight to that of the body, the thickness of the muscular ventricular septum, and the myocytic profile within the ventricular septum are all increased in fetuses of diabetic rats in comparison to fetuses of normal rats. Methods: Diabetes was induced in 5 pregnant Wistar rats, bearing 30 fetuses, on the eighth day after conception, by intraperitoneal injection of 50mg/kg of streptozotocin. Five normal pregnant Wistar rats, bearing 20 fetuses, made up the control group. Morphometric data were obtained by a computer-assisted method applied to the measurements of the thickness of the ventricular septum, and myocytic nuclear area. Statistical analysis utilized Student's t-test and Kruskal-Wallis test. Results: The mean thickness of the septum was 675.56 μm (± 159) in the control fetuses, and 904.39 μm (± 262) in the fetuses carried by diabetic mothers (p < 0.001). The cardiac weight was 0.016 g (± 0.004) in the control group, and 0.023 g (± 0.005) in the group of diabetic fetuses (p < 0.001). The ratio of cardiac to body weight was 0.294% (± 0.079) in the control group, and 0.514% (± 0.073) in the diabetic group (p < 0.001). The myocytic nuclear area was 14.70 μm2 in the control group, and 21.43 μm2 in the diabetic group (p < 0.001). Conclusions: The presence of cellular and morphologic cardiac hypertrophy in fetuses of diabetic rats was demonstrated by the significant difference between the two groups for each analyzed feature.

PURPOSE: To compare body weight and length, heart weight and length, heart-to-body weight ratio, glycemia, and morphometric cellular data of offspring of diabetic rats (ODR) and of normal rats (control). METHODS: Diabetes was induced in 3 pregnant Wistar rats, bearing 30 rats, on the 11th day after conception by intraperitoneal injection of 50 mg/kg of streptozotocin. Six normal pregnant Wistar rats, bearing 50 rats, made up the control group. Morphometric data were obtained using a scale for the weight, length, heart and body measurements. Morphometric cellular data were obtained by a computer assisted method applied to the measurements of myocytes. Statistical analysis utilized Student's t-test, ANOVA and Levene test. RESULTS: Control offspring had greater mean body weight and length than offspring of diabetic rats (p < 0.001). Heart weight and length and heart-to-body ratios of newborn rats differed between groups at birth (p < 0.001), but showed no difference at 21 days. Mean nuclei area and perimetric value of the myocytes decrees throughout the first 21 days of life (p < 0.01) in the diabetic group. CONCLUSIONS: Heart hypertrophy on the offspring of diabetic rats at birth was demonstrated by the significant difference between the groups. After the eleventh day, no difference was found, which confirmed regression of cardiomegaly. The significant difference between the first and the 21th day of life, for nuclei area feature, demonstrate regression of cardiac hypertrophy in the offspring of diabetic rats.

The aim of this research was to investigate the effects of extract ethanol of pegagan (Centella asiatica) after treated with restraint stress by measuring the eight arms radial maze performance of rats. Pre-test and post-test controlled groups design was applied in this research. Male Wistar rats (Rattus novergicus) with the mean age of 8 weeks and the body weight of 150-200 grams which used for sample were divided randomly into 4 groups, each group consisted of 6 rats, i.e. (i) control group (without any treatment), (ii) stress group (it was given by restraint stress for 2 hours/day for each rat), (iii) pegagan group (it was given by 0.3 mg/g BW/day/rat extract ethanol of pegagan), and (iv) pegagan and stress group (it was given by 0.3 mg/g BW/day/rat extract ethanol of pegagan and restraint stress for 2 hours/day for each rat). The treatments were given for 21 days. Within 12 days for each pre-treatment and post-treatment, a test on the eight arm radial maze was conducted on individual rat to observe its performance. The assessment of rat performance in the eight arms radial maze test was conducted based on error type B. Kruskal-Wallis and Mann-Whitney tests with SPSS for Windows 16 version were applied to analyze statistically the difference between four groups. Kruskal-Wallis test was used to show the significant performance level difference between four groups of rats with p=0.001, while Mann-Whitney test was used to determine the significant difference between stress group and pegagan group (p=0.001), also stress group and pegagan and stress group (p=0.001). The result of research showed that there was no significant difference between control group and stress group (p=0.051), control group and pegagan group (p=0.143), control group and pegagan and stress group (p=0.143), also pegagan group and pegagan and stress group (p=0.952). It was concluded that extract ethanol of pegagan improved the performances of rats on the eight arms radial maze after treated with restraint stress.

Introduction Type 1 diabetes is an autoimmune disorder characterized by lack of insulin production by the β cells of the pancreas. This lack of insulin causes a variety of systemic effects on the metabolism of the body, one of which is reproductive dysfunction. The present study investigates the effects of diabetes on the male reproductive system of streptozotocin (STZ)-induced diabetic rats. Material and Methods A total of 18 adult male Wistar rats weighing ∼ between 250 and 300 g were included in the present study. The animals were divided into normal and diabetic groups. The diabetic group was further subdivided into 2 subgroups with durations of 24 and 48 days. A single dose of STZ (40 mg/kg body weight) was administrated intraperitoneally to the animals of the diabetic group. After the planned duration, the testes and epididymides were dissected, and their gross weight was measured. The tissues were then processed for histological study. Results The gross weight of the testes and epididymides in diabetic rats at 24 and 48 days showed a decrease in comparison to the control. (p < 0.01 for testes and epididymides).Diabetic animals presented a significant decrease in the diameter of the seminiferous tubules compared with the control group (p < 0.01). The epididymides in the diabetic groups showed a considerable reduction in the tubular surface area compared with the control group (p < 0.01). There was also a reduction in the mean diameter, which was measured using the maximum and minimum diameter of the tubules (p < 0.01). Conclusion The present study is an insight into the adverse effects that diabetes can have on the tissue structure of the testes, of the epididymides, and ultimately on the process of spermatogenesis.

The study evaluated the effects of moringa oleifeira on the histoarchitecture of the cerebral cortex, the body weight and brain weight of young wistar rats. Fifteen (15) young wistar rats of both sexes weighing 20-30g were used for this study. They were randomly assigned into three groups (A, B and C) of five rats each. Group A served as control and received distilled water, group B and C received 100 mg/kg and 200mg/kg of moringa oleifera respectively. Treatment lasted for a period of 6 weeks (orally). Rats were weighed and sacrificed under ketamine (30 mg/kg) anaesthesia. The cerebrum was harvested and fixed immediately in 10% formolcalcium, for further histological processing. One-way ANOVA was used to analyze data, followed by Newan-Keuls (SNK) for multiple comparisons. The results showed that there was significant increase in the feed intake of animals in groups B and C starting from the 4th week of administration. There was no significant difference in the relative brain weight and the mean weight of the rats in group B and C when compared with group A. Histological findings revealed that there was slight distortion in group B and more distortion in group C when compared with the normal histoarchitecture in control group A. The results obtained from this study showed that high doses of Moringa oleifera caused damage of some parts of histoarchitecture of the frontal cortex of developing wistarKey words: MORINGA OLEIFERA; Cerebrum; Frontal Cortex

Aim and Objectives: Demonstrating the exact quantity of Moringa oleifera (MO) that will cure any hepatocyte diseases and the one that can harm the hepatocytes of the Adult Wistar rats. Methods: Twenty (20) adult Wistar rats (both sex) were used for the study (caged based on same sex to avoid mating and pregnancy) and were randomly assigned into four groups (n=5); A-Control, B-250 mg/body weight (BW) of MO, C-500 mg/BW of MO, and D-750 mg/BW of MO orally. Result: The crude aqueous extract of MO Lam, shows insignificant increased in BW at the 1st week of administration which latter dropped little by little doing the weeks of the administration in groups (B, C, and D) rats, by comparing (p<0.05) to the control group after MO administration, the organ (Kidneys) shows a significant difference between the kidneys (left and right kidneys) in relation to the control group rats. MO increases the weight of the animals morphologically by comparing the weight of the animals before and after administration. Histological sections shows a normal Glomerulli, Peri-Glomerular Space, Convoluted Tubules, and Interstitium, after administration of the Crude Aqueous Extract of MO lame in terms with the control group rats. Conclusion: MO is a good herb that has no damage effect on the body and hepatocytes but of more beneficial.

Objectives. This study aimed to explore the expression of GLP-1 receptor in hypothalamus and gastrointestinal tissues after Roux-en-Y gastric bypass (RYGB) surgery in diabetic rats.Methods. Male 12-week-old Wistar rats (control) and Goto-Kakizaki rats (diabetic) were randomly divided into two groups, respectively: control sham surgery group (C), control RYGB group (C + R), diabetic sham surgery group (D), and diabetic RYGB group (D + R). Body weight and blood glucose were monitored before and after surgery every week. Eight weeks after surgery, all rats were sacrificed and the serum fasting GLP-1 concentrations were measured by ELISA. GLP-1R and DPP-4 expression in hypothalamus and ileum were measured by RT-PCR.Results. The body weight and fasting/random blood glucose in the D + R group decreased significantly compared with the D group (P<0.05). Serum GLP-1 levels in diabetic rats treated with RYGB were higher than the corresponding sham surgery rats. The expression of GLP-1R of hypothalamus in RYGB-treated diabetic rats was significantly higher than those of the sham surgery diabetic rats and both control group rats (P<0.05). We found a negative correlation between hypothalamus GLP-1R mRNA and blood glucose level. No significant difference was seen in ileum GLP-1R and DPP-4 expression among all groups.Conclusions. RYGB efficiently promoted serum GLP-1 levels and the expression of GLP-1 receptor in the hypothalamus in diabetic rats. These data suggest that the hypothalamus GLP-1R may play an important role in the GLP-1 system for improving glucose homeostasis after reconstruction of the gastrointestinal tract.

Objective: The present study was designed to evaluate the effects of Atorvastatin (ATO) plus Melatonin (MEL) on streptozocin-induced Diabetic Retinopathy (DR) in rats. Methods: Diabetes was induced in Wistar rats with an intraperitoneal injection of streptozocin (50 mg/kg). Animals were randomly assigned to one of the following groups (8 rats/group): Control group, Diabetic group, Diabetic + MEL group (20 mg/kg/day), Diabetic + ATO group (10 mg/kg/day), Diabetic + MEL + ATO group (as above). Treatments were started one week after induction of diabetes and continued for 7 weeks. At the end of the experiment, angiography was performed and the rats were killed and retinas were harvested for pathological and molecular examinations. Results: Administration of MEL reduced the fluorescein leakage, MDA and ROS levels compared to diabetic group. Treatment with ATO only reduced ROS levels compared to diabetic group. In addition, administration of ATO plus MEL decreased these indices compared to the diabetic and ATO groups. Histologically, retinal vascular congestion was not observed in the combined ATO and MEL group as compared to the diabetic, ATO, and MEL groups. Conclusion: These data provide evidence for the therapeutic value of MEL in combination with ATO in clinical practice for prevention of DR.

In this study the antidiabetic effect of a polyherbal formulation- ZPC was investigated in Wistar rats. Diabetes was induced by intraperitoneal injection of streptozotocin at a dose of 50 mg/kg. Rats having Fasting Blood Sugar (FBS) level above 250 mg/dl after 72 hrs were considered diabetic and used for the studies. Five rats served as non- diabetic control (Group 1) while twenty diabetic rats were randomized into 4 groups of 5 rats each. The four groups (Groups 2,3,4 and 5) received 1ml (diabetic control), 250 mg/kg chlorpropamide and ZPC at doses of 250 and 500mg/kg respectively for 28 days. During the treatment period, the FBS and bodyweight of rats were monitored weekly and on day 28, the rats were euthanized and blood samples collected for serum lipid profile analysis. Results obtained indicated that following administration of streptozotocin, there was a significant (p<0.05) increase in the FBS, total cholesterol, triglycerides and LDL concentration with a corresponding significant (p<0.05) decrease in HDL concentration compared to non- diabetic control. However, following the treatment with the polyherbal formulation, there was a significant (p<0.05) reduction in the FBS level and a significant (p<0.05) increase in the body weight of rats compared to the diabetic control. The polyherbal formulation also produced a significant (p<0.05) reduction in total cholesterol, triglycerides and LDL concentration with a corresponding significant (p<0.05) increase in HDL concentration compared to diabetic control. It was concluded that, ZPC might serve as a good alternative or as an adjunct to the oral hypoglycaemic agents in the management of diabetes.

Someone who dies will experience a decrease in body temperature from body temperature at the beginning of death, both within normal and abnormal limits to room temperature. Decrease in body temperature according to sex may differ in duration. Coupled with exposure to methanol, it is also possible to influence a decrease in body temperature of the corpse. This study aimed to analyze the differences in body temperature reduction in male and female Wistar rats  induced with methanol.This research is an experimental study with a pre and post test control group design approach. The design of this study was to observe the body temperature of male and female Wistar rats while still alive and after death and the duration of decrease in body temperature of male and female Wistar mice after being induced with methanol. The population studied was male and female Wistar rat. Based on the results from analysis test, it can be concluded that there is no significant difference between the decrease in body temperature of male and female Wistar rats that died induced by methanol, but there is a significant difference between the decrease in body temperature of male and female Wistar rats who died induced by methanol and without methanol.

We studied lungs of spontaneously diabetic Bio-Breeding/Worcester (BB/W) Wistar rats which resemble human insulin-dependent diabetes mellitus. Compared with the age-matched control group, the body weight of the diabetic rats tended to be smaller and lung wet and dry weight were similar, but lung dry weight, relative to body weight and to lung wet weight, was significantly larger. Both air and saline lung volumes were reduced in the diabetic rats, and volume-pressure (V-P) curves expressed as a percent of maximal lung volume were significantly shifted downward and to the right of those in the control group over the midportion. Total DNA and RNA contents were similar in both groups, whereas protein content and concentration and protein/DNA and RNA/DNA ratios were significantly reduced in the diabetic rats. In contrast, content and concentration of 4-hydroxy-L-proline, elastin, and crude connective tissue were significantly higher in the diabetic group. We conclude that the increase in connective tissue proteins in the BB/W rats is most likely responsible for the shift in the V-P curves.
