AbstractBackgroundFish is considered an important part of a healthy diet and is frequently recommended as a main course at least twice a week.ObjectiveTo study the frequency of fish consumption among young overweight European adults and their compliance to varying seafood consumption in weight loss intervention diets.DesignAfter meeting the inclusion criteria, the subject’s seafood intake was evaluated. Subjects were randomly assigned into four groups and were advised energy-restricted diets for 8 weeks, including no seafood (control), cod, salmon or fish oil. A validated FFQ was used to evaluate the consumption of seafood at baseline, midpoint and endpoint, and long-chain n-3 fatty acids in blood erythrocytes were measured.SettingIceland, Ireland and Spain.SubjectsThe sample (n 324); 20–40-year-olds with BMI = 27·5–32·5 kg/m2; 85 % participated.ResultsAt baseline, 34 % of the participants reported eating fish at least twice a week as the main course. During the intervention, six participants reported that they did not finish their fish portions, 15 % of the participants consumed small amount of fish additional to the study protocol in weeks 1–4 and 23 % in weeks 5–8 (P = 0·010). Changes in erythrocyte long-chain n-3 fatty acids confirmed good compliance, with increases in the salmon (P < 0·001) and fish oil (P < 0·001) groups, smaller increase in the cod group (P = 0·037) and decrease in the control group (P = 0·030).ConclusionFrequency of fish consumption among 66 % of young European overweight adults is lower than frequently recommended. Compliance to varying seafood consumption was good. Therefore, including more fish in the diet of this group should be encouraged.

AbstractEight mid-lactation Holstein-Friesian dairy cows were used in an experiment to investigate whether poor transfer to milk of (n-3) long chain (≥C20) poly-unsaturated fatty acids (n-3 LCPUFA), from diets containing fish oil, is related to their mode of transport in plasma lipoproteins. The cows were split into two groups of four, each of which was housed with 24 other cows which did not provide blood data in this experiment. All cows received a basal diet ad libitum consisting of (kg/t dry matter) silage (539), grain distillers’ grains (148), rolled barley (248) and soya (65). All cows also received a mid-day meal of 2·76 kg per cow per day of molassed sugar-beet shreds. The control group (C) received no oil supplement but those in the treatment group (F) received 300 g per cow per day of fish oil absorbed onto the sugar-beet shreds. Fish oil inclusion progressively depressed milk fat concentration of the four cows from each group over the 3 weeks of the experiment so that levels were approximately 20 g/kg lower for cows in treatment F than for those in C (P < 0·05). Mean concentrations over all cows of the long-chain (≥C20, n-3) poly-unsaturated fatty acids in milk fat (g/100 g) were 0·12 and 0·54 for treatments C and F respectively, whilst corresponding output in milk (g/day) increased from 116 to 3·91, an average efficiency of transfer of 4·6%. The fatty acid compositions of the lipoproteins were measured in plasma samples from the four cows from each group after 3 weeks on the experimental diets The proportions (g/100 g fatty acids) of C20: 5 (n-3) in cholesteryl (CE) and phospholipid (PL) of high-density lipoprotein (HDL) in the plasma were respectively increased from 0·97 to 4·39 (P < 0·05) and from 1·15 to 4·74 (P < 0·01) by the fish-oil supplementation. For C22: 6 (n-3), the proportions in CE and PL of HDL respectively increased (P < 0·001) from 0·02 to 0·23 and from 0·16 to 2·18 as a result of the supplementation. The mean proportions (g/100 g fatty acids) of C20: 5 (n-3) and C22: 6 (n-3) in the triglyceride fraction of very low-density lipoprotein were 1·05 and 2·3 respectively and were not significantly altered by fish-oil supplementation (P > 0·05). It is suggested that the low transfer efficiency of (n-3) LCPUFA to the milk is partly due to their transport in the plasma largely as components of HDL lipids which are unavailable to the mammary gland.

This study assessed the potential of omega-3 long chain polyunsaturated fatty acids (n-3 LC-PUFA) enrichment in red earthworm (Eisenia fetida, REW), via the addition of fish oil (FO, anchovy oil) to the culture-compost. FO addition to compost was tested at graded inclusion doses (0, 2.5, 5 and 10 g/100 g compost) and for different timeperiods, up to 96h. Dose, time, as well as dose and time interactions of FO compost enrichment had significant effects on all fatty acids of earthworm bodies. The enrichment with 10 g FO/100 g compost sharply increased DHA levels of the worms (14.01 mol%) within just 24 h, compared to the control group (0.92 mol%). Similar increases (ranging between 10.99 and 15.55 mol%) occurred only after 48 and 96 h in lower FO enrichment levels (2.5 and 5.0 g/100 g compost, respectively). Therefore, it was concluded that, to obtain maximum n-3 LC-PUFA enrichment efficiency in REW, an enrichment period of 24 h is ideal and recommended for the 10 g FO/ 100 g compost, whereas longer enrichment periods (48-96 h) are more suitable for lower levels of FO inclusions (2.5 or 5 g/100 g compost).

Dietary polyphenols promote cardiometabolic health and are linked with long-chain polyunsaturated fatty acids in plasma phospholipids (LC-PUFA). The FADS2 polymorphisms are associated with LC-PUFA metabolism and overweight/obesity. This 4-week study examined the link between polyphenol intake, FADS2 variants (rs174593, rs174616, rs174576) and obesity in 62 overweight adults (BMI ≥ 25), allocated to consume 100 mL daily of either: Aronia juice, a rich source of polyphenols, with 1177.11 mg polyphenols (expressed as gallic acid equivalents)/100 mL (AJ, n = 22), Aronia juice with 294.28 mg polyphenols/100 mL (MJ, n = 20), or nutritionally matched polyphenol-lacking placebo as a control (PLB, n = 20). We analyzed LC-PUFA (% of total pool) by gas chromatography and FADS2 variants by real-time PCR. Four-week changes in LC-PUFA, BMI, and body weight were included in statistical models, controlling for gender and PUFA intake. Only upon AJ and MJ, the presence of FADS2 variant alleles affected changes in linoleic, arachidonic, and eicosapentaenoic acid (EPA). Upon MJ treatment, changes in EPA were inversely linked with changes in BMI (β= −0.73, p = 0.029) and weight gain (β= −2.17, p = 0.024). Only in subjects drinking AJ, the link between changes in EPA and anthropometric indices was modified by the rs174576 variant allele. Our results indicate the interaction between FADS2, fatty acid metabolism, and polyphenol intake in overweight subjects.

Based on the potential benefits to human health, there is interest in developing sustainable nutritional strategies to enhance the concentration of long-chainn-3 fatty acids in ruminant-derived foods. Four Aberdeen Angus steers fitted with rumen and duodenal cannulae were used in a 4 × 4 Latin square experiment with 21 d experimental periods to examine the potential of fish oil (FO) in the diet to enhance the supply of 20 : 5n-3 and 22 : 6n-3 available for absorption in growing cattle. Treatments consisted of total mixed rations based on maize silage fed at a rate of 85 g DM/kg live weight0·75/d containing 0, 8, 16 and 24 g FO/kg diet DM. Supplements of FO reduced linearly (P < 0·01) DM intake and shifted (P < 0·01) rumen fermentation towards propionate at the expense of acetate and butyrate. FO in the diet enhanced linearly (P < 0·05) the flow oftrans-16 : 1,trans-18 : 1,trans-18 : 2, 20 : 5n-3 and 22 : 6n-3, and decreased linearly (P < 0·05) 18 : 0 and 18 : 3n-3 at the duodenum. Increases in the flow oftrans-18 : 1 were isomer dependent and were determined primarily by higher amounts oftrans-11 reaching the duodenum. In conclusion, FO alters ruminal lipid metabolism of growing cattle in a dose-dependent manner consistent with an inhibition of ruminal biohydrogenation, and enhances the amount of long-chainn-3 fatty acids at the duodenum, but the increases are marginal due to extensive biohydrogenation in the rumen.

To improve the parameters of the transesterification process by the basic route for the production of biodiesel, crude fish oil (AP), obtained from the viscera of fish, and residual vegetable oils (AR) blenders were used. Because the AP contains high percentages of long chain polyunsaturated fatty acids that are responsible for its instability is that it was mixed with increasing percentages of AR. This translated into a higher conversion performance to biodiesel by the basic route, also depending on the level of free fatty acids. The index of methyl esters obtained from the blends and the original oils was analyzed. The calculation of the performance of each reaction is made based on the produced esters. The analysis is complemented by measuring the kinematic viscosity of the blends and the pure oils, as well as the biodiesel produced. The results showed yields higher than 90% when using the process of transesterification of the blends by the basic route. The standardization of blends by its acidity index is of great importance reaching a maximum value of 3.5 mg KOH/g. On the other hand, the viscosity of the biodiesel produced, from the blends, is kept within international standards; In addition, the process conditions are improved.

Introduction: Dyslipidemia is a prevalent condition in obesity and type 2 diabetes. Although fish oil rich in omega-3 fatty acids (ω-3) is a widely used hypolipidemic agent, it is often required at high doses. At high doses, these fatty acids can induce oxidative stress or endothelial activation and therefore, strategies to improve their beneficial effects are needed. We previously reported that fish oil in combination with cyclooxygenase (COX) inhibitors exerts enhanced hypolipidemic and anti-inflammatory effects in low density lipoprotein receptor knock-out mice. Here, we sought to determine the effects of ω-3 fatty acids in combination with naproxen (NX), a COX inhibitor, on dyslipidemia and gene expression in subcutaneous adipose tissue (scAT) in humans. Methods: Obese dyslipidemic patients were randomly assigned to receive one of these interventions (n=8/group) for 12 wk: 1) Standard nutrition counseling (control), 2) ω-3 (2 g twice daily), 3) NX (220 mg twice daily), and 4) ω-3 (2 g twice daily) + NX (220 mg twice daily). Results: The body mass index, HOMA-IR, and plasma total, LDL, and HDL cholesterol levels were not altered significantly in any of the groups. The percent change in plasma triglycerides (TG) from baseline was 75% ( P <0.1) and 68% ( P <0.05) in ω-3 and ω3 + NX-treated subjects, respectively. Notably, 25% of subjects who received ω-3s alone did not show a reduction in TG whereas all the patients that received ω-3 + NX showed a reduction in TG. Realtime PCR analysis of scAT showed that the expression of glucose transporter 4 (GLUT-4), a marker of glucose uptake and a key regulator of glucose homeostasis was significantly reduced in ω-3 compared to control group ( P <0.01). However, combining NX with ω-3 abolished this effect. Moreover, the expression of MCP-1 and VCAM-1, markers of inflammatory response or endothelial activation, was significantly increased in ω-3 but not in ω-3 + NX group. The plasma levels of MCP-1 and E-selectin did not vary significantly in any of the groups. Conclusions: Our data reveal previously unrecognized effects of fish oil in scAT. Our data suggest that combining NX with ω-3 fatty acids will increase their effectiveness in reducing plasma TG and improve the benefits of ω-3 supplements by favorably altering gene expression in scAT.

Background: Obesity affects young adults, leading to future morbidity and mortality. Early behavioral intervention may promote long-term weight control. Mobile technology-based (mHealth) interventions may be particularly effective in young adults. We compared both an mHealth behavioral weight loss intervention and a personal coaching weight loss intervention to no intervention (and to each other) in overweight/obese young adults. Methods: We randomized 365 generally healthy adults age 18-35 years with BMI > 25 kg/m2 (overweight or obese) to 24-months of intervention delivered primarily via investigator-designed cell phone (CP) or intervention delivered primarily via in-person (6 weekly) and by phone (23 monthly) coaching (PC), compared to usual care control group (Control). Primary outcome was weight change from baseline to 24 months. This study was conducted as part of the Early Adult Reduction of weight through LifestYle (EARLY) cooperative trials. Results: Randomized participants (N=365) had mean BMI 35 kg/m2, mean age 29yrs, were 70% women, 36% African American, 6% Latino. Final weight was obtained in 86%; missing weight was multiply imputed. At 24 months, weight loss was not different in either PC or CP vs Control (see Figure). Weight loss in PC was significantly greater than Control at 6 months. From baseline to 24 months, clinically significant weight loss (> 3% per national guidelines) occurred in 40% of PC, 34% of CP, and 30% of Control. Conclusions: mHealth alone may not be sufficient for weight loss in young adults but mHealth-enhanced contact with an interventionist has a modest short-term effect. Future interventions should maximize the complementarity of mHealth and personal contact to achieve larger and more sustained effect.

Background and Aims: Obesity is an increasing problem in patients after total knee replacement. The aim of this study was to investigate whether a weight loss intervention before primary total knee replacement would improve quality of life, knee function, mobility, and body composition 1 year after surgery. Material and Methods: Patients scheduled for total knee replacement due to osteoarthritis of the knee and obesity were randomized to a control group receiving standard care or to an intervention group receiving 8-week low-energy diet before total knee replacement. Patient-reported quality of life, 6-Min Walk Test, and body composition by dual-energy X-ray absorptiometry were assessed before intervention for the diet group, and within 1 week preoperatively for both groups, and the changes in outcome from baseline to 1 year after total knee replacement were compared between groups. The number of participants was lower than planned, which might introduce a type-2 error and underestimate the trend for a better outcome after weight loss. Results: The analyses are based on a total of 76 patients, 38 in each group. This study showed major improvement in both study groups in quality of life and knee function, though no statistically significant differences between the groups were observed 1 year after total knee replacement. The average weight loss after 8-week preoperative intervention was 10.7 kg and consisted of a 6.7 kg reduction in fat mass. One year after total knee replacement, the participants in the diet group managed to maintain the weight reduction, whereas there was no change in the control group. Conclusion: The results suggest that it is feasible and safe to implement an intensive weight loss program shortly before total knee replacement. The preoperative intervention resulted in a 10% body weight loss, improved body composition, lower cardiovascular risk factors, and sustained s-leptin.
