The purpose: investigate changes in immunity parameters and cortisol level in subjects with opiate addiction during the treatment of opiate withdrawal syndrome. Methods. The study enrolled 136 subjects with opiate addiction with physical dependence receiving injections of opium extract. Patients received conventional therapy with complete opiate withdrawal. The study was performed at the following stages: at admission to the hospital (acute withdrawal syndrome (AWS); on days 5-7 of therapy (transition into post-withdrawal state - PWS); on days 25-28 of therapy (formation of therapeutic remission - FTR). Laboratory methods included determination count of lymphocytes with receptors CD3, CD4, CD8, СD16, with receptors to dopamine (D-RFC); the serum levels of IgМ, IgG, IgА, cortisol, circulating immune complexes (CIC). Results. The principal immunoendocrine pattern for all stages of withdrawal syndrome therapy is characterized in comparison to the reference normal values quantitative deficit of CD3, CD4, СD8 Т-lymphocyte subpopulations, increased count of lymphocytes with receptors to dopamine, activation of humoral immunity factors (IgM, IgG, CIC), high cortisol level. At AWS and PAS stages such changes are most pronounced; on days 25-28 of therapy positive changes in cytotoxic Т-lymphocytes (СD8) and Т-lymphocytes СD3 was revealed. CD4 count remained steadily reduced, count of lymphocytes with receptors to dopamine and cortisol level were elevated. Clinical and immunological analysis demonstrated that consumption of high opiate doses, long-term narcotization are associated with higher intensity of disorders detected. Conclusion. Dysregulation of immunoendocrine parameters was revealed in subjects with opiate addiction at all stages of withdrawal syndrome therapy within the term observed evidencing instability of therapeutic remission achieved and necessity in further rehabilitation events.

Temporomandibular joint (TMJ) is commonly involved in various immune-mediated rheumatic disorders accounting for significant disability and impaired quality of life. The aim of our study was to assess inflammatory and immune parameters in patients with TMJ arthritis related to rheumatoid arthritis (RA), juvenile idiopathic arthritis (JIA), ankylosing spondylitis (AS) and psoriatic arthritis (PsA) and to identify potential relation with severity and dysfunction of TMJ pathology. We performed a cross-sectional study in a cohort of 433 consecutive RA, 32 JIA, 258 AS, and 103 PsA. Only patients presenting with clinically significant TMJ involvement (273) related to their rheumatic condition were included in the final analysis. TMJ involvement is traditionally described in chronic inflammatory rheumatic disorders, particularly in patients with higher levels of inflammation as detected in rheumatoid arthritis and psoriatic arthritis. Disease activity and severity, as well as biological and positive serological assessments (rheumatoid factor, anti-cyclic citrullinated peptide, IL-1) remain significant determinants of the severity of TMJ arthritis.

Abstract Yeast cell wall products are common functional ingredients capable of “priming” the immune system, specifically in reference to vaccine efficacy. Twenty-four adult, female Beagles were used in a completely randomized design. Three retorted diets were used: control diet (CON), CON plus β-glucan top-dressed daily upon time of feeding (C+B), and CON plus retorted β-glucan included in diet formulation (BG). Following a 7 d adaptation to CON, dogs were fed their respective treatment diets for 42 d and were challenged with an oral Bordetella bronchiseptica vaccine on d 14 with blood collections on d 0, 21, 28, and 42. The objectives of the present study were 1) to evaluate the effects of dietary inclusion of a yeast β-1,3/1,6 glucan (150 ppm) on apparent total tract digestibility (ATTD) of macronutrients, fecal microbiota, and peripheral blood mononuclear cells (PBMC; T-cells, B-cells, and monocytes) of adult dogs and, 2) to test the effects of retorting on the efficacy of these β-glucans. All diets were well-accepted by all dogs. ATTD of both dry matter and crude protein were greater (P < 0.05) for BG than CON and with greater (P < 0.05) energy digestibility for BG than both B+G and CON. Additionally, fecal short-chain and branched-chain fatty acids, ammonia, indole and phenol concentrations did not differ among treatments. No significant treatment by time interactions among treatment groups were observed for any analyzed PBMC. These data suggest that a 150 ppm inclusion of this yeast-derived β-glucan had no detrimental effects on ATTD, fecal characteristics and metabolites, nor any analyzed PBMC; however, due to the absence of differences in immune parameters among treatments, the effect of retorting on the efficacy of this product cannot confidently be assessed with these findings. Higher doses of yeast-derived β-glucan might be needed to elicit an immunological modulation in healthy adult dogs.

Abstract Hempseed meal (HSM) is a byproduct of hemp oil production and is high in crude protein, fiber, and fat, making it a potential feedstuff for ruminants. The objective of this study was to evaluate the effect of HSM supplementation on growth performance, rumen fermentation and blood chemistry profile of growing meat goats. Forty castrated, Boer cross goats were randomly assigned to one of the four treatments (n = 10): control, 10%, 20%, and 30% HSM supplementation. Data collected over a period of a 60-day feeding trial were analyzed utilizing the mixed model analysis function of SYSTAT, version 13. The result revealed total live weight gain decreased with the increasing levels of HSM supplementation 10.75, 9.53, 8.48, and 7.80 kg, for 0, 10, 20, and 30%, respectively. Average daily gain followed the same trend 0.179, 0.159, 0.141, and 0.13 kg, with a significant difference (P < 0.05) observed between the control and 30 % supplementation. Conversely, feed to gain ratio increased with the increasing levels of supplementation 9.0, 10.2, 11.9, 12.2, likewise a significant difference was observed (P < 0.05) between the control and 30% supplementation. Acetic, propionic, butyric, valeric, iso-valeric, and iso-butyric acid concentrations as well as the total VFA concentration decreased significantly (P < 0.05) with the increasing level of supplementation. Acetic to propionic acid ratios increased with increasing level of supplementation 3.43, 4.36, 4.52, and 4.59, significant differences (P < 0.05) were observed between control-20% and 30% HSM group. Serum glucose concentration decreased with an increasing rate of HSM supplementation while BUN concentration increased with no significant differences. These findings provide new insights into the feeding value of HSM for meat goats; however, further research needs to be conducted to determine the optimal level of supplementation.

Weaning stress can elicit changes in the metabolic, hormone and immune systems of pigs and interact with prolonged disruptions stemming from maternal immune activation (MIA) during gestation. The present study advances the characterization of the combined effects of weaning stress and MIA on blood chemistry, immune and hormone indicators that inform on the health of pigs. Three-week-old female and male offspring of control gilts or gilts infected with the porcine reproductive and respiratory syndrome virus were allocated to weaned or nursed groups. The anion gap and bilirubin profiles suggest that MIA enhances tolerance to the effects of weaning stress. Interleukin 1 beta and interleukin 2 were highest among weaned MIA females, and cortisol was higher among weaned relative to nursed pigs across sexes. Canonical discriminant analysis demonstrated that weaned and nursed pigs have distinct chemistry profiles, whereas MIA and control pigs have distinct cytokine profiles. The results from this study can guide management practices that recognize the effects of the interaction between MIA and weaning stress on the performance and health of pigs.

We studied the fecal lactobacilli count and species diversity of growing pigs along with immune parameters associated with intestinal lactobacilli. Thirty pigs categorized as small (S, n = 12) or large (L, n = 18) at birth were followed from birth to slaughter in two commercial herds, H1 and H2. Herds differed in terms of their general management. We determined sow colostrum quality, colostrum intake, piglet serum immunoglobulins, and pig growth. We took individual fecal samples from pigs in the weaning and finishing units. We studied lactobacilli count and identified their diversity with 16S PCR. Total lactobacilli count increased in H1 and decreased in H2 between samplings. Lactobacilli species diversity was higher in H1 in both fecal sampling points, whereas diversity decreased over time in both herds. We identified altogether seven lactobacilli species with a maximum of five (one to five) species in one herd. However, a relatively large proportion of lactobacilli remained unidentified with the used sequencing technique. Small pigs had higher lactobacilli counts in both herds but the difference was significant only in H2 (p = 0.01). Colostrum quality was numerically better in H1 than in H2, where colostrum intake tended to be associated with total lactobacilli count (p = 0.05).
