Laboratory-dried specimens yield distorted information about the state of hydromorphic soil. Changes in the gas medium, humidity, temperature, aggregation capacity and illumination have an irreversible impact on the dried and powdered soil specimen. Properties of the dried hydromorphic soil can be altered significantly. Most altered are (1) the colour of the hydromorphic soils, (2) the рН value, and (3) the content of mobile trace metal (TM) compounds after drying the wet soil. Colour change (browning) of the hydromorphic soil is related to the oxidation of Fe(II). In a highly ferruginous soil, the browning is rapid and visible even to the naked eye. In the weakly ferruginous gley, the colour change is invisible to the naked eye, but instrumental field measurements reveal a slow and slight yellowing of the dried gley. In dried soils, laboratory рН values determined in soil suspension differ from the values determined in soil solution at the same time. The process of drying provokes a significant decrease in the content of mobile TM compounds, resulting in the illusion of less soil contamination than the real situation during hydromorphic soil humidification. When studying hydromorphic soils, it is desirable to accomplish the maximum possible field measurements. Laboratory analyses should only be carried out for specimens stored under dark conditions since they retain the initial humidity, temperature and gas composition.

Staining of wood with various substances and processes is an important part of surface finishing of wood. Colour differences as a result of staining and of exposure of coloured wood during its utilisation are usually evaluated by instrumental measurements. However, the measurement results can show something else compared to what our naked eye can see. Due to inhomogeneity of wood, this discrepancy can be even greater in the case of finished surfaces. The aim of our research was to evaluate distinctions between visual perception and numerically determined colour differences on differently finished wooden surfaces, to get information at which starting point the colour difference becomes visible, and to establish whether it is related to the nature of the surface. We established that the visual assessment is influenced by many factors and that there is a correlation between visual and instrumental assessments. The colour difference ΔE* of 0.5 should be considered as a value when it starts to become visible, and at the value of 2.0, observers already considered the colour difference as a different colour. It was stated that we have some tolerance in perceiving the colour change. This tolerance is more expressed in the case of transparent coating systems.

Poly– diallyldimethylammonium chloride (poly-DADMAC) is an established coagulant in the treatment of drinking water. Reports have indicated that poly-DADMAC can degrade into a suspected carcinogenic form which is N-nitrosodimethylamine (NDMA). Consequently, water treatment plant operators are required to know the residual concentration of polyelectrolytes at various stages in the treatment process and the eventual quality of the treated water. Historical research has proven that, over the years, a number of methods such as extraction-spectrophotometry, fluorometry and tannic acid have been developed and implemented for the analysis of polymers in drinking water. However, they produced poor linearity, sensitivity and precision, high detection limits or produced false positives due to matrix effects. The laboratory method that has proven to be simple, affordable and accurate is the colloidal titration method. However, this method cannot be used at the plant for quick and accurate monitoring of poly-DADMAC. In this study, the aim was to fabricate a Lovibond portable colorimetric comparator device based on gold nanoparticles for colorimetric quantification and detection of poly-DADMAC in raw and treated potable water. The colorimetric disk and comparator was fabricated from 14 nm gold nanoparticles with the concentration of poly-DADMAC varying from 1 to 10 mg L-1. The addition of higher concentrations of poly-DADMAC resulted in the aggregation of gold nanoparticles with the colour changing from red to blue. The gold nanoparticles were prepared via the citrate reduction method. Characterisation of the gold nanoparticles was done by ultraviolet- visible (UV/VIS) spectrophotometry and transmission electron microscopy (TEM). The Lovibond comparator was fabricated with a colour filter disk for the screening of residual poly-DADMAC in raw and potable water. The colorimetric disk was printed on the plastic slide and inserted in the plastic compartment of the comparator. The Lovibond comparator was verified with raw and potable water samples from different sampling points in and around the Mhlathuze river area located in the province of KwaZulu-Natal, South Africa. Preliminary results showed that the developed colorimetric comparator device can visually detect poly-DADMAC concentrations lower than 1 mg L-1. The colour development was first developed on normal paper and then optimised by UV/VIS spectrophotometry. The method developed has a linear range from zero to 10 mg L-1 with the correlation coefficient of R=0.9954. The effectiveness of the device was investigated by doing a recovery study on a potable water sample. Potable water is water that is suitable for drinking. In this research, potable water refers to tap water. The potable water sample was spiked with 1 mg L-1 poly-DADMAC. This exercise was done three times. The acceptance criterion for recovery is 80 to 120%. The 3 recoveries that were obtained are 107.95, 91.26 and 100.3%. The average recovery was 99.84%. This shows that the proposed method can detect poly-DADMAC with the acceptable level of accuracy. One of the important parameters that a quality method must have is selectivity. This parameter shows that the method can accurately detect the analyte of interest in the midst of different matrices. This was done by analysing the raw water samples together with their treated samples. Physical-chemical parameters were also analysed to show the broader state of the samples. The poly-DADMAC results obtained from the UV/VIS spectrophotometer compared quite well with those obtained from using the Lovibond colorimetric filter. The limited observation of colours using our eyes is a major contributor of systematic errors during the application of colorimetric devices. Thus, such a limitation can be reduced by using CIELAB system. A gold nanoparticle-based colorimetric CIELAB system for detection of poly-DADMAC in potable and raw water was also demonstrated. The method is based on the application of a paper-based analytical device which is printed on the normal A4 white printing paper. Fully enclosed 6 X 9 hydrophobic wells were fabricated on this paper. This work provides a clear evidence of the application of CIELAB colour system, and thus, replacing the conventional spectrophotometric technique to quantify polymers. Results of this work showed that the intensity of the fabricated well is proportional to the concentration of the detected polymer. The change in colour (ΔE) was calculated for each fabricated well and clear evidence of the colour change was observed upon the variation of the polymer. Moreover to the application of ΔE, the chromaticity using CIEYxy was used to verify colour change, it was observed that they followed the expected shift from red to blue, symbolising aggregation due to Van Der Waal inter-particle attractions as a result of the addition of poly-DADMAC. The results of this experiment were validated using the spectrophotometric technique which further emphasised the appearance of the new surface Plasmon resonance peak formed at 610nm symbolising aggregation. Importantly, the intensity of the new Surface Plasmon Resonance (SPR) peak at 610 nm increased by increasing the concentration of poly-DADMAC. Comparison of the Lovibond and UV/VIS results showed that there was no significant difference between the two methods. This proved that the fabricated Lovibond colour comparator is capable of the detection of residual poly-DADMAC in water treatment. This therefore implies that plant operators can be able to detect poly-DADMAC at any stage during the water treatment process by using a rapid, user-friendly portable device.

Drinking water contamination of lead from various environmental sources, leaching consumer products and intrinsic water-pipe infrastructure is still today a matter of great concern. Therefore, new highly sensitive and convenient Pb2+ measurement schemes are necessary, especially for in-situ measurements at a low-cost. Within this work dye/ionophore/Pb2+ co-extraction and effective water phase de-colorization was utilized for highly sensitive lead measurements and sub-ppb naked-eye detection. A low-cost ionophore Benzo-18-Crown-6-ether was used, and a simple test-tube mix and separate procedure was developed. Instrumental detection limits were in the low ppt region (LOD=3, LOQ=10), and naked-eye detection was 500 ppt. Note, however, that this sensing scheme still has improvement potential as concentrations of fluorophore and ionophore were not optimized. Artificial tap-water samples, leached by a standardized method, demonstrated drinking water application. Implications for this method are convenient in-situ lead ion measurements.

A simple and rapid positive–negative colorimetric approach to determine the presence of antimony ions based on the use of gold nanoparticles conjugated with oligonucleotide (poly-A sequence) is developed. Colorimetric measurements reveal that the aggregates of modified gold nanoparticles were afforded after adding antimony ions, thus changing the solution color from pink to blue. The results of aptamer’s interaction on the gold nanoparticle surface with the target analyte can be detected either by photometry or by the naked eye. The realized assay provides rapid (2 min), sensitive (detection limit 10 ng/mL), specific, and precise (variation coefficient less than 3.8%) detection of antimony (III) in drinking water.

A new hydrazone-based colorimetric Cu2+ chemosensor is synthesized. Its structure was confirmed by single-crystal X-ray diffraction. Its binding properties towards various metal ions are examined through absorption spectroscopy. In aqueous THF solution, the chemosensor exhibits selective recognition towards Cu2+ over other metal ions with a colour change from colourless to pink. The complex formed between the chemosensor and Cu2+ ions forms a 2:1 stoichiometry with an association constant of 2.46 × 108M−2. The analytical detection limit for Cu2+ by the naked eye is as low as 10.0 μM.

Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis. Detection and control of infectious diseases is a major problem, especially in developing countries. Lateral flow immunoassay (LFIA) has been introduced as a handheld immunoassay-based point-of-care platform for an automated detection of TB. The CFP10-ESAT6 antigen of M. tuberculosis was used as the target in early detection of TB using LFIA strip-based POC strategy. An interesting platform based on optical signals is implemented as a colour change in the detection area that is visible to the naked eye. The gold nanoparticles (AuNPs) were used as the colour probe for the detection of a target of interest. The high-resolution transmission electron microscopy (HRTEM) image and ultraviolet-visible spectrophotometer (UV-Vis) analysis confirmed that the synthesized AuNPs were appropriate for the immunoassay designed. The platform consists of AuNPs conjugated with specific antibodies (Ab) to capture the antigen of M. tuberculosis. Under the capillary effect, sandwich immunoreactions of AuNP-Ab-antigen were performed on the test pad of the immunostrip, which can be observed by the colour signal on the test line of the strip with a short assay time. Furthermore, the newly developed biosensor was utilized in CFP10-ESAT6 antigen detection in human sputum specimens with satisfactory results. The characteristic coloured bands enable visual detection (naked eye) of target analyte without instrumentation. This noninvasive diagnose system which is sputum-based detection could provide user-friendly and affordable diagnostic tests in developing countries.
