Recycling of by-products from the food industry has become a central part of research to help create a more sustainable future. Brewers’ spent grain is one of the main side-streams of the brewing industry, rich in protein and fibre. Its inclusion in bread, however, has been challenging and requires additional processing. Fermentation represents a promising tool to elevate ingredient functionality and improve bread quality. Wheat bread was fortified with spray-dried brewers’ spent grain (BSG) and fermented brewers’ spent grain (FBSG) at two addition levels to achieve “source of fibre” and “high in fibre” claims according to EU regulations. The impact of BSG and FBSG on bread dough, final bread quality and nutritional value was investigated and compared to baker’s flour (BF) and wholemeal flour (WMF) breads. The inclusion of BSG and FBSG resulted in a stronger and faster gluten development; reduced starch pasting capacity; and increased dough resistance/stiffness. However, fermentation improved bread characteristics resulting in increased specific volume, reduced crumb hardness and restricted microbial growth rate over time. Additionally, the inclusion of FBSG slowed the release in reducing sugars over time during in vitro starch digestion. Thus, fermentation of BSG can ameliorate bread techno-functional properties and improve nutritional quality of breads.

For thousands of years, truffles have been used as essential foods in different cultures around the world because of their rich nutritional value and their pleasant and characteristic smell. We have studied the effect of truffles (Tirmania Nivea and Tirmania Pinoyi) extracts on the antioxidant stress properties issued from the Moroccan desert. Antioxidant and anti-free radical activities were studied using three analytical methods: trapping capacity of 1,1-diphenyl-2-picrylhydrazyl, phosphomolybdate, and reducing ferric antioxidant capacity; in addition, phenol and flavonoid levels were measured. The results of the FRAP, DPPH and PPM tests of T. Nivea were respectively 4.112±0.217, 0.142±0.006, 2.235±0.110 mg/mL for methanols and 3.424±0.034, 0.137±0.025, 0.858±0.010 mg/mL for ethyl acetate extracts. The results of the tests of T. pinoyi were respectively 3.670±0.572, 0.102±0.004, 0.907±0.014 mg/mL for methanols and 3.404±0.096, 0.080±0.003, 0.693±0.057 mg/mL for ethyl acetate extracts. For this work, we propose a valorization of the Moroccan truffle in the prevention of oxidative stress.

Corpuscles of Stannius tissue of Atlantic cod (Gadus morhua L.) transformed [4-14C] pregnenolone to progesterone and [4-14C] progesterone to 11-desoxycorticosterone in vitro. These results establish the presence of 3β-hydroxysteroid dehydrogenase, Δ5-3-ketoisomerase and 21-hydroxylase. Transformation of the 14C-steroid precursors to several as yet unidentified substances also occurred.

Variations in the occurrence of Oikopleura spp. were strongly linked to the wind field and ensuing water temperatures in an inshore region of eastern Newfoundland during June–August of 1979 and 1981–83. Oikopleura foul inshore fishing gear with their discarded houses ("slub") and fluctuate in abundance from day to day as a function of wind-driven upwelling. Densities of Oikopleura in cold upwelling water during four years ranged between 20 and 800/m3. Oikopleura densities < 1/m3 occurred when upwelling ceased and warmer water occupied the inshore region. A simple model incorporating a daily averaged wind vector, Julian day, and growing degree-day explained 62% of the daily variation in Oikopleura abundance in 1979. The model yielded density estimates that were significantly (p < 0.05) correlated with observed densities in each of the other three years. We provide evidence that net fouling by Oikopleura in the inshore and maximum Atlantic cod (Gadus morhua) catch is coincident and suggest that an annual hindcast index of slub intensity may help determine the impact of slub conditions on the inshore fishery.

Abstract Background Liver tumor-initiating cells (T-ICs) contribute to tumorigenesis, progression, recurrence and drug resistance of hepatocellular carcinoma (HCC). However, the underlying mechanism for the propagation of liver T-ICs remains unclear. Methods Real-time PCR was used to detect the expression of miR-96 in liver tumor-initiating cells (T-ICs). The impact of miR-96 on liver T-ICs expansion was investigated both in vivo and in vitro . The correlation between miR-96 expression and sorafenib benefits in HCC was further evaluated in patient-derived xenografts (PDXs). Results Our finding shows that miR-96 is upregulated in liver T-ICs. Functional studies revealed that forced miR-96 promotes liver T-ICs self-renewal and tumorigenesis. Conversely, knockdown miR-96 inhibits liver T-ICs self-renewal and tumorigenesis. Mechanistically, miR-96 downregulates SOX6 via its mRNA 3’UTR in liver T-ICs. Furthermore, the miR-96 expression determines the responses of hepatoma cells to sorafenib treatment. Analysis of patient-derived xenografts (PDXs) further demonstrated that the miR-96 may predict sorafenib benefits in HCC patients. Conclusion Our findings revealed the crucial role of the miR-96 in liver T-ICs expansion and sorafenib response, rendering miR-96 as an optimal target for the prevention and intervention of HCC.

Abstract Telencephalic radial glial progenitors (RGPs) are retained in the ventricular zone (VZ), the niche for neural stem cells during cortical development. However, the underlying mechanism is not well understood. To study whether protein phosphatase 2A (PP2A) may regulate the above process, we generate Ppp2cα conditional knockout (cKO) mice, in which PP2A catalytic subunit α (PP2Acα) is inactivated in neural progenitor cells in the dorsal telencephalon. We show that RGPs are ectopically distributed in cortical areas outside of the VZ in Ppp2cα cKO embryos. Whereas deletion of PP2Acα does not affect the proliferation of RGPs, it significantly impairs the generation of late-born neurons. We find complete loss of apical adherens junctions (AJs) in the ventricular membrane in Ppp2cα cKO cortices. We observe abundant colocalization for N-cadherin and PP2Acα in control AJs. Moreover, in vitro analysis reveals direct interactions of N-cadherin to PP2Acα and to β-catenin. Overall, this study not only uncovers a novel function of PP2Acα in retaining RGPs into the VZ but also demonstrates the impact of PP2A-dependent retention of RGPs on the generation for late-born neurons.

In todays competitive world, corporate companies all around the world are trying to maximize the wealth of their shareholders in order to gain market value as well as satisfy their stakeholders. With the gaining popularity of value based performance measures like Economic Value Added (EVA), Total Shareholder Return (TSR), Cash Value Added (CVA) etc., many corporate companies in India have started assessing their value in terms of these measures. This paper investigates the relationship between EVA and share prices of select companies in BSE-SENSEX for a period of six years from 2008 to 2013. The study focuses on the explanatory power of EVA with respect to share prices of the selected companies. In turn, the performance of the selected companies belonging to different sectors in BSE-SENSEX was analyzed using EVA. The volatile nature of the capital markets characterized by various speculative activities have a greater influence on share prices, eventually undermining the impact of performance metrics on them. Thus, the findings of the study enumerates that EVA does not have a considerable explanatory power on share prices.

AbstractMatrix metallopeptidases, commonly known as matrix metalloproteinases (MMPs), are a group of proteolytic enzymes whose main function is the remodeling of the extracellular matrix. Changes in the activity of these enzymes are observed in many pathological states, including cancer metastases. An increasing body of evidence indicates that nanoparticles (NPs) can lead to the deregulation of MMP expression and/or activity both in vitro and in vivo. In this work, we summarized the current state of knowledge on the impact of NPs on MMPs. The literature analysis showed that the impact of NPs on MMP expression and/or activity is inconclusive. NPs exhibit both stimulating and inhibitory effects, which might be dependent on multiple factors, such as NP size and coating or a cellular model used in the research.

Depending on the stage of gonad maturation, as well as other factors, gonadal steroids can exert either a positive or negative feedback at the brain and pituitary level. While this has been demonstrated in many teleost species, little is known about the nature of steroid feedback in Gadiform fish. Using an optimized in vitro model system of the Atlantic cod pituitary, the present study investigated the potential effects of two physiologically relevant doses of estradiol, testosterone (TS) or dihydrotestosterone (DHTS) on cell viability and gene expression of gonadotropin subunits (fshb/lhb) and two suggested reproduction-relevant gonadotropin-releasing hormone receptors (gnrhr1b/gnrhr2a) during three stages of sexual maturity. In general, all steroids stimulated cell viability in terms of metabolic activity and membrane integrity. Furthermore, all steroids affected fshb expression, with the effect depending on both the specific steroid, dose and maturity status. Conversely, only DHTS exposure affected lhb levels, and this occurred only during the spawning season. Using single-cell qPCR, co-transcription of gnrhr1b and gnrhr2a was confirmed to both fshb- and lhb- expressing gonadotropes, with gnrhr2a being the most prominently expressed isoform. While steroid exposure had no effect on gnrhr1b expression, all steroids affected gnrhr2a transcript levels in at least one maturity stage. These and previous results from our group point to Gnrhr2a as the main modulator of gonadotropin regulation in cod and that regulation of its gene expression level might function as a direct mechanism for steroid feedback at the pituitary level.
