The effect of temperature on symptom expression and detection of banana streak badnavirus (BSV) by immunosorbent electronmicroscopy (ISEM) and enzyme-linked immunosorbent assay of 12 in vitro-propagated plantain hybrids (genome AAB × AA), 3 ABB cooking banana, and 3 AAB plantain landraces was studied. Experiments were done for 2 years under two temperature regimes, 28 to 35°C in a screenhouse and 22°C in a temperature-controlled room. Most BSV-infected plants of plantain hybrids expressed symptoms under both conditions. Symptom expression was enhanced when plants were continuously grown at 22°C, but later became indiscernible when plants were continuously grown at 28 to 35°C. Plants grown at 22°C and showing severe symptoms contained significantly higher virus titer than plants grown at 28 to 35°C. When asymptomatic plants with very low virus titer at 28 to 35°C were transferred back to 22°C, there was a significant increase in both symptom severity and concentration of virus (greater than 3 to 5 times) in leaf tissues after 9 months. In contrast, the concentration of virus and symptom severity decreased in plants after transfer from 22°C to 28 to 35°C. Micropropagated plants of AAB plantain landrace cv. Mimi Abue and ABB cooking bananas (cvs. Bluggoe, Cardaba, and Pelipita) did not express visible symptoms under either temperature regime, but BSV was detected by ISEM in 23% of the plants. After 2 years at 22°C, virus was detected in 64% of the plants, but the concentration of virus remained low. Implications of these results on quarantine screening of in vitro plants and virus diagnosis are discussed.

Summary. An offtype has been identified from micropropagated Lady Finger bananas (Musa spp., AAB group, Pome subgroup) that is characterised by its slow growth and poor bunch size. Bunch weights were approximately 25% those of normal Lady Finger plants and all of the fruit produced was unmarketable. This particular offtype is the most commonly encountered from micropropagated Lady Finger plants and, in 2 instances, blocks of 3000 and 1500 plants were entirely comprised of this single offtype. Detection of offtype plants was possible during establishment and growth of plants in the glasshouse by the presence of chlorotic streaks in the leaves. In more severe cases the streaks coalesced into chlorotic patches that developed thin, necrotic areas that eventually produced holes or splits in the leaves. Symptom expression was not ameliorated by the addition of fertiliser and even though symptoms were similar to severe Ca and B deficiency, both normal and offtype plants had similar levels of these elements in the leaves. The offtype plants were also slow growing in the glasshouse and produced significantly (P<0.05) smaller pseudostems and leaves than normal plants. Offtype plants could be readily detected after 4 weeks deflasking using the presence of chlorotic streaks in the leaves as the main selection criterion. Maximum discrimination was possible between weeks 5–7 and at the 6-leaf stage when all of the offtypes could be detected.

Symptoms resembling those of viral leaf streak disease, caused by banana streak badnavirus (BSV), were observed in May 1998 on two banana (Musa spp.) landraces grown from farmer-collected propagules in a farmer's field at Kiboje Uchukuni, Zanzibar. Those showing symptoms were “French plantain” cv. Mzuzu and “Cavendish” banana cv. Mtwike. Leaf symptoms were expressed as chlorotic streaks and blotches. Leaf samples were indexed by immunosorbent electron microscopy with BSV and cucumber mosaic cucumovirus (CMV) antibodies, using partially purified preparations (2). The two landraces tested positive for BSV, corroborating the occurrence of BSV in Zanzibar. In addition, cv. Mtwike was found to be coinfected with CMV. No other viruslike particles were seen by electron microscopy. Although BSV has been reported in Zanzibar (1), it was only from symptoms in the Musa field genebank at Kizimbani Research Station. BSV has been found in many Musa collections worldwide, particularly in the widely grown cv. Mysore. Banana streak badnavirus (BSV) has been reported from Musa spp. in many parts of West Africa, including Benin, Côte d'Ivoire, Ghana, and Nigeria (1). Symptoms of BSV infection in Musa spp. are sometimes similar to and confused with those caused by cucumber mosaic virus (CMV). BSV is prevalent in areas of southern Nigeria bordering Cameroon, and the disease may also be present in other Central African countries. In June 1996, six leaf samples with viruslike yellow/chlorotic streak symptoms were collected from plantain in the four villages, Awae, M'Balmayo, Nkolfep, and Nkolfoulou, within a 60-km radius of Yaoundé, Cameroon's capital. The samples were indexed for BSV and CMV by both triple antibody sandwich indirect enzyme-linked immunosorbent assay (TAS-ELISA) and immunosorbent electron microscopy (ISEM) to ascertain the presence of these two viruses. The TAS-ELISA was performed with rabbit polyclonal antiserum (obtained from B. E. L. Lockhart, University of Minnesota) for trapping and mouse polyclonal antiserum (obtained from G. Thottappilly, IITA) for detection. Out of the six samples, one tested strongly positive (>×2 A405 of the healthy control) and four were weakly positive (<×2 but >×1.5 A405 of the healthy control) for BSV by TAS-ELISA. However, all six samples contained BSV particles when examined by ISEM with rabbit polyclonal antiserum (from B. E. L. Lockhart) for trapping. None of the samples tested positive for CMV. The origin of mica polytypes has long been a challenging problem for crystal- lographers, mineralogists and petrologists. From the petrological point of view, interest in this field arose from the potential use of layer stacking data to furnish further informations about equilibrium and/or kinetic conditions prevailing during the crystallization of the widespread mica-bearing rocks. From the compilation of previous experimental works dealing with the occurrence domains of the various mica "polymorphs" (1Mr, 1M, 2M1, 2M2 and 3T) within water-pressure vs temperature fields, it became clear that most of these modifications should be considered as metastable for a fixed mica species. Furthermore, the natural occurrence of long-period (or complex) polytypes could not be accounted for by phase considerations. This highlighted the need of a more detailed kinetic approach of the problem and, in particular, of the role growth mechanisms of basal faces could play in this crystallographic phenomenon.

Excessive presence of polysaccharides, polyphenol and secondary metabolites in banana plant affects the quality of DNA and it leads to difficult in isolating good quality of DNA. An optimized modified CTAB protocol for the isolation of high quality and quantity of DNA obtained from banana leaf tissues has been developed. In this protocol a slight increased salt (NaCl) concentration (2.0M) was used in the extraction buffer. Polyvinylpyrrolidone (PVP) and Octanol were used for the removal of polyphenols and polymerase chain reaction (PCR) inhibitors. Proteins like various enzymes were degraded by Proteinase K and removed by centrifugation from plant extract during the isolation process resulting in pure genomic DNA, ready to use in downstream applications including PCR, quantitative polymerase chain reaction (qPCR), ligation, restriction and sequencing. This protocol yielded a high molecular weight DNA isolated from polyphenols rich leaves of Musa spp which was free from contamination and colour. The average yields of total DNA from leaf ranged from 917.4 to 1860.9 ng/ìL. This modified CTAB protocol reported here is less time consuming 4-5h, reproducible and can be used for a broad spectrum of plant species which have polyphenol and polysaccharide compounds.

The water purification procedure aims to obtain a product appropriate for human consumption, minimizing the presence of contaminants and toxic substances present in the water. Among these contaminants, some radionuclides of natural origin, such as uranium, thorium and their descendants, have been identified. Studies have shown that the stages of purification are quite effective in removing the radionuclides contained in water. The removal is due to co-precipitation of the radionuclides with the suspended materials and the precipitated material is accumulated and characterized as a Technologically Concentrated Natural Occurrence Radioactive Material (TENORM) by the United States Environmental Protection Agency (USEPA). This residue can present significant levels of radioactivity and, when discarded in the environment without any treatment, can generate a problem of environmental impact and a risk to the health of the population. In this way, some gamma emitters of the series of U, Th and the K-40 were determined in the residues generated at the Potable Water Treatment Plants – PWTPs in six municipalities of Pernambuco. The results obtain corroborate the classification of the residues generated in the PWTPs as concentrators of the radioactive components contained in the water supplied to the system and reinforce the need for the release to the environment, which is the usual way of disposal of this waste, to be carried out only after considering the radiological protection standards established.
