Phytochemical investigation and cytotoxic characterization of bioactive constituents from Conyza dioscoridis

Understanding the bioactive constituents and physicochemical components in cereals can provide insights into their potential health benefits and food applications. This study evaluated some bioactive constituents, carbohydrate profiles and pasting properties of 16 Striga-resistant hybrids, with yellow-orange kernel color and semi-flint to flint kernel texture, grown in two replications at two field locations in Nigeria. Carotenoids were quantified using HPLC, while other analyses were carried out using standard laboratory methods. The ranges of major carotenoids (μg/g) across the two locations varied from 2.6 to 9.6 for lutein, from 2.1 to 9.7 for zeaxanthin, from 0.8 to 2.9 for β-cryptoxanthin, from 1.4 to 4.1 for β-carotene; with total xanthophylls and provitamin A carotenoids (pVAC) ranging from 5.4 to 17.1 and 1.4 to 4.1 μg/g, respectively. Tannins content ranged from 2.1 to 7.3 mg/g, while phytate ranged from 0.4 to 7.1%. Starch, free sugar, amylose and amylopectin ranged from 40.1 to 88.9%, 1.09 to 6.5%, 15.0 to 34.1%, and 65.9 to 85.0%, respectively. Peak and final viscosities ranged from 57.8 to 114.9 and 120.3 to 261.6 Rapid Visco Units (RVU), respectively. Total xanthophylls, β-carotene, tannins, phytate, sugar, amylose and amylopectin levels, as well as peak and final viscosities, varied significantly (p < 0.05) across the hybrids. Amylose was significantly correlated (p < 0.05) with total xanthophylls, β-carotene, pVAC, phytate and pasting temperature (r = 0.3, 0.3, 0.4, 0.3, 0.3, respectively), but starch significantly correlated with tannins (r = 0.3). Hence, the Striga-resistant yellow-orange maize hybrids have a good combination of bioactive constituents, carbohydrate profile and pasting properties, which are partly influenced by hybrid.

Phytochemical investigation of the Australian lichen, Ramalina glaucescens resulted in the isolation of a new halogenated depside, 5-chlorosekikaic acid 5, together with (+)-usnic acid 1, sekikaic acid 2, atranorin 6 and parietin 7, the latter of which was isolated from the associated (co-occurring) lichen, X. parietina. Compound 5 is suspected to be an artifact of the isolation procedure. All structures were assigned using spectroscopic methods and mass spectrometry. In addition to the full characterization of 5, this report represents the first application of 2D NMR spectroscopy to complete the unequivocal chemical shift assignment for compounds 2 and 7. Compounds 1-2 and 5-7 all displayed varying degrees of antitumor activity (ranging from an IC50 of 15 μM to >44 μM) with compounds 1, 2 and 5 also displaying antibacterial properties. Of these, (+)-usnic acid 1 displayed the most significant antitumor and antibacterial activities.

In the present research work the phytochemical and Chromatographic separation of bio active compounds Berberis lycium occurred. Phytochemical investigation of the CH2 Cl2 /CH3 OH (1:1) extract ethanol of root shown the presence of terpenoids, flavonoids, saponins, anthraquinones and alkaloids, steroids and tannins was absent. Chromatographic separation of CH2 Cl2 /CH3 OH (1:1) root ethanol extract yielded methyl 9, 10-dihydro-3, 8-hydroxy-6-methyl-9, 10-dioxoanthracene-2- carboxylate, isolated for the first time from the species of Berberis lycium. With the help of spectroscopic methods (IR, 1 H NMR, 13C NMR, and DEPT-135 and UV-Vis,) Partial characterization of the isolated compound was done.

Abstract A phytochemical investigation of the EtOH extract of the flowers of Lagerstroemia indica L. led to the isolation and characterization of a new pyrrole alkaloid, named lagerindicine (1), along with four known compounds (2–5). Their structures were elucidated by the detailed spectroscopic analysis and comparison with literature data, whereas the structure, in particularly, the absolute configuration (AC) of 1, was firmly determined by total synthesis. All the isolates were evaluated for their cytotoxic effects against human colon cancer cell (HCT-116), and compound 3 exhibited weak cytotoxicity with IC50 value of 28.4 μM. Graphic Abstract

The herbal medicine combination of notoginseng-safflower has been commonly used clinically for the prevention and treatment of cardiovascular diseases. A reliable liquid chromatography-tandem mass spectrometry (LC–MS/MS) method was developed for simultaneous determination of six bioactive components (hydroxysafflor yellow A, notoginsenoide R1, ginsenoside Rb1, Re, Rd, and Rg1) in rat urine and feces after oral administration of notoginseng total saponins (NS), safflower total flavonoids (SF), and the combination of NS and SF (CNS). The chromatographic separation was achieved on a Waters HSS T3 column under gradient elution with acetonitrile and water containing formic acid as the mobile phase. The calibration curves were linear, with correlation coefficient (r) > 0.99 for six components. The intra- and interday precision (RSD) and accuracy (RE) of QC samples were within −14.9% and 14.9%, respectively. The method was successfully applied to study of the urinary and fecal excretion of six bioactive constituents following oral administration of NS, SF, and CNS in rats. Compared to the single herb, the cumulative excretion ratios of six constituents were decreased in the herbal combination. The study indicated that the combination of notoginseng and safflower could reduce the renal and fecal excretion of the major bioactive constituents and promote their absorption in rats.

AbstractIn this paper, examination of the aerial parts of seven Senecio species from Serbia and Montenegro is reported. Phytochemical investigation of Senecio erucifolius led to the isolation and characterization of semiquinol butyl 2-(1-hydroxy-4-oxocyclohexa-2,5-dienyl)acetate (I), along with methyl 2-(1-hydroxy-4-oxocyclohexa-2,5-dienyl)acetate (jacaranone, II), and methyl 2-(4-hidroxyphenyl)acetate (III). The structure of I was established based on spectroscopic studies (1H- and 13C-NMR, IR, and CI-MS). Compound II was also isolated from S. carpathicus and S. subalpinus. The presence of jacaranone in the methanol extracts of S. wagneri, S. othonnae, and S. paludosus was confirmed by LC/ESI-TOF MS.
