Abstract Trinder's method for glucose has nearly all the attributes of an ideal automated colorimetric glucose oxidase procedure. The chemicals used in the color reaction with peroxidase are readily available, the solutions are stable and can be prepared by the user, the method is highly specific and largely free of interferences, the sensitivity can be adjusted by the user to cover a wide range of glucose concentrations, and the reagents are not hazardous. We found very good agreement between results by this method and by the hexokinase and Beckman Glucose Analyzer methods. The method has been modified and adapted to the AutoAnalyzer I and SMA 6/60 (Technicon) with manifolds that give very little interaction between specimens. A study of the method by the simplex technique revealed that the glucose oxidase activity in the reagent is the most critical variable.

Romania is known among the main European honey producers, due to the variety of landforms as well as the diversity of the flora. Thirty-four honey samples of the acacia, linden and multifloral types, produced in eastern Romania and collected during 2013–2018, were physico-chemically analyzed using methods provided by the national and EU standards. The results of water-insoluble solids, color and refractive index were found to be 0.023–0.131%, 0.3–76.4 mm Pfund and 1.485–1.499, respectively. The moisture content ranged between 15.20% and 20.77%, solid substances content ranged between 79.23% and 84.80% and total soluble substances content ranged between 77.83 °Brix and 83.26 °Brix. The obtained values of specific gravity were from 1.414 to 1.450 g/cm3, pH ranged from 3.673 to 5.503 and free acidity ranged between 2.4 meq and 50 meq kg−1. The ash content and the electrical conductivity varied between 0.030 and 0.543% and 130 and 679 µS cm−1, respectively. Pearsonʹs correlation analysis showed an intense association of the ash content with electrical conductivity (r = 0.81). Our findings reveal the qualitative level of Romanian honey and the variation in quality parameters due to factors such as geographical region, climatic conditions, botanical origin and handling or storage conditions.

Currently, American Foulbrood (AFB) represents one of the most important problems for beekeepers, due to economic losses and to the absence of an effective therapeutic treatment. The aim of this work was to characterize fifteen Italian honeys in order to assess their inhibitory activity against Paenibacillus larvae ATCC 9545. Each honey was analyzed for the activity of the following enzymes: glucose oxidase and catalase. Moreover, melissopalynological analysis and other biochemical parameters, namely gluconic acid, total phenolic and total flavonoid contents were determined. For each honey, the Minimum Inhibitory Concentration (M.I.C.) and the Minimum Bactericidal Concentration (M.B.C.) against P. larvae were determined. All tested honey samples had an inhibitory activity on P. larvae. In particular, the lowest M.I.C. and M.B.C. values (53.8 mg/mL and 107.5 mg/mL, respectively) were recorded for an Arbutus honey sample. Arbutus honeys also had the highest gluconic acid and total phenolic contents (12.6 ± 1.7 g/kg and 243.2 ± 25.1 mg/kg, respectively) and the highest glucose oxidase activity (13.0 ± 1.9 nM H2O2/min). Dark honeys, including Arbutus, seem to have a higher gluconic acid content and a higher antimicrobial activity. Thus, honey characterization, including colour and physico-chemical characteristics (e.g. gluconic acid concentration, total phenolic and total flavonoid contents, glucose oxidase activity), could be crucial for the assessment of its employment against P. larvae.
