Serum activity of matrix metalloproteinase-2 and -9 is increased in chronic post-stroke individuals: a cross-sectional exploratory study

Background. Encapsulating peritoneal sclerosis (EPS) is a rare but serious complication of peritoneal dialysis (PD). So far, there is no biomarker-based prediction tool available for EPS. Matrix metalloproteinase-2 (MMP-2) is a protein involved in the breakdown of the extracellular matrix, and the effluent MMP-2 can be a potential biomarker of EPS. This study is aimed at developing a nomogram for EPS based on effluent MMP-2 levels. Patients and Methods. We enrolled 18 EPS patients and 90 gender-matched PD patients without EPS in this cross-sectional case-controlled study. The effluent MMP-2 levels and possible risk factors for EPS were analyzed using multivariable logistic regression, and a nomogram was developed. The nomogram was validated using 200 bootstrap resamples to reduce overfit bias. Results. The effluent MMP-2 levels in EPS patients were significantly higher than those in normal PD patients ( p < 0.001 , Manny-Whitney U test). Effluent MMP-2 levels and PD duration were independently associated with EPS risks ( p < 0.001 and p = 0.001 ) in multivariate logistic regression. A nomogram based on MMP-2 levels and PD duration was proposed. The AUC of MMP-2 was 0.824, and the AUC of the nomogram was 0.907 ( p = 0.05 ). Conclusion. A nomogram based on effluent MMP-2 levels and PD duration may predict EPS with high accuracy.

BACKGROUND: Left ventricular end-diastolic volume (LVEDV) on echocardiography is one of the tests performed on heart failure. This refers to the volume of the left ventricle at the end of the diastolic phase, which would be increased when there is a disturbance in preload, afterload, and contractility factors. Matrix metalloproteinase-2 (MMP2) is a marker of congestive heart failure that can be examined through laboratory examinations. AIM: The objective of the study was to provide evidence of the association between MMP and inflammatory process as well as its correlation with LVEDV in children with heart failure. METHODS: This was a cross-sectional study conducted on children aged 3 months–12 years old with heart failure, who visited Sanglah Hospital, Denpasar, Indonesia from May 2017 to March 2018. Echocardiographic examination (LVEDV) and blood samples were taken to measure the serum level of MMP2 on day 1 after the subjects were diagnosed with heart failure. RESULTS: A total of 32 subjects with heart failure were analyzed in this study. Acyanotic congenital heart defect (CHD) was the most common cause of heart failure, as observed in 23 subjects (71.9%). Characteristics data revealed that 24 subjects (75%) were underweight, 23 (71.9%) had cardiomegaly, and 22 (68.8%) had mild heart failure. Data analysis showed a moderate positive correlation between MMP2 levels with LVEDV after controlling for the influence of age (p = 0.02; r = 0.425). CONCLUSION: There was a moderate positive correlation between MMP2 level and LVEDV after controlling for the age factor.

Ketuban pecah dini adalah masalah penting dalam obstetri dengan insiden 8 % pada kehamilan. Kejadian KPD disebabkan oleh muktifaktorial, diantaranya adalah faktor infeksi dan nutrisi ibu hamil yang mempengaruhi pembentukan selaput ketuban. Beberapa peneliti menemukan bahwa peningkatan matrix metalloproteinase-2  akan mendegradasi matrix extraseluler, menjadi lemah sehingga memicu terjadinya KPD. Tujuan penelitian untuk mengetahui hubungan MMP-2 serum dengan ketuban pecah dini dan kehamilan normal. Penelitian dilakukan dengan menggunakan desain cross sectional comparative study. Penelitian dilakukan pada empat rumah sakit di Padang pada bulan September – Oktober 2018. Pengambilan sampel dengan consecutive sampling. Populasi pada penelitian ini adalah ibu hamil dengan diagnosa ketuban pecah dini (n=29) dan seluruh wanita dengan kehamilan normal(n=29). Pemeriksaan MMP-2 dilakukan dengan metode ELISA. Uji statistik menggunakan uji t tidak berpasangan dan korelasi Pearson. Hasil penelitian menunjukkan rerata kadar MMP-2 pada KPD yaitu 35,17 ± 2,23 ng/ml, dan pada kehamilan normal 33,27 ± 0,82 ng/ml (p<0,05). Terdapat perbedaan kadar MMP-2 pada ketuban pecah dini dan kehamilan normal. Kesimpulan penelitian terdapat perbedaan rerata kadar MMP-2 pada ketuban pecah dini dan kehamilan normal.

Objective: To determine the role of matrix metalloproteinase-2(MMP-2) in serum and peritoneal fluid of endometrioticpatients.Methods: Research’s design using cross-sectional method in Dr.Wahidin Sudirohusodo hospital and several other hospitals inMakassar within May 2015 until May 2016. Subjects were chosenusing consecutive sampling technique. The examination usingthe ELISA method. The data were analysed using Fisher exact,t-independent, Mann-Whitney, and Spearman association.Results: A total of 50 subjects were recruited in this study. Mostlythe value of MMP-2 serum and peritoneal fluid in endometriosisgroup was higher compare to study control. There was significantdifferent between the total of MMP-2 serum and peritoneal fluid.There was also a significant association between the value ofMMP-2 serum and peritoneal fluid with endometriosis.Conclusion: The value of MMP-2 serum and peritoneal fluid werehigher in endometriotic patients compared to those withoutendometriosis. Purpose This exploratory study examined the language skills and the type and frequency of disfluencies in the spoken narrative production of Spanish–English bilingual children who do not stutter. Method A cross-sectional sample of 29 bilingual students (16 boys and 13 girls) enrolled in grades prekindergarten through Grade 4 produced a total of 58 narrative retell language samples in English and Spanish. Key outcome measures in each language included the percentage of normal (%ND) and stuttering-like (%SLD) disfluencies, percentage of words in mazes (%MzWds), number of total words, number of different words, and mean length of utterance in words. Results Cross-linguistic, pairwise comparisons revealed significant differences with medium effect sizes for %ND and %MzWds (both lower for English) as well as for number of different words (lower for Spanish). On average, the total percentage of mazed words was higher than 10% in both languages, a pattern driven primarily by %ND; %SLDs were below 1% in both languages. Multiple linear regression models for %ND and %SLD in each language indicated that %MzWds was the primary predictor across languages beyond other language measures and demographic variables. Conclusions The findings extend the evidence base with regard to the frequency and type of disfluencies that can be expected in bilingual children who do not stutter in grades prekindergarten to Grade 4. The data indicate that %MzWds and %ND can similarly index the normal disfluencies of bilingual children during narrative production. The potential clinical implications of the findings from this study are discussed.
