Germination response of Arabian desert species to gibberellic acid and potassium nitrate seed treatment

Poor early growth and uneven crop establishment are reported as the major bottlenecks in wide-scale adoption and optimal yield realization of dry direct-seeded rice (DSR). Seed priming can potentially help overcome these problems in DSR. Therefore, laboratory and field studies were conducted at Punjab Agricultural University, Ludhiana, India, during kharif/wet-season 2018 and 2019 to evaluate the effect of different priming techniques on germination, establishment, growth, and grain yield of rice under DSR conditions. The following priming treatments were evaluated: dry non-primed seed (control), hydropriming with distilled water, halopriming with 2.0% potassium nitrate, hormopriming with 50 ppm gibberellic acid (GA3), and osmopriming with polyethylene glycol (PEG)(−0.6 MPa), each with 12 and 24 h priming duration. In 2019, priming treatments were tested under two DSR establishment methods—conventional DSR (sowing in dry soil followed by irrigation) and soil mulch DSR (locally known as vattar DSR) (sowing in moist soil after pre-sowing irrigation), whereas in 2018, priming treatments were evaluated under conventional DSR only. In both years, halopriming and hormopriming resulted in a 7–11% increase in rice yields compared to non-primed dry seed (control). Osmopriming resulted in a 4% yield increase compared to control in 2018 but not in 2019. The higher yields in halopriming and hormopriming were attributed to higher and rapid germination/crop emergence, better root growth, and improvement in yield attributes. Priming effect on crop emergence, growth, and yield did not differ by DSR establishment methods and duration of priming. Conventional DSR and soil mulch DSR did not differ in grain yield, whereas they differed in crop emergence, growth, and yield attributes. These results suggest that halopriming with 2.0% potassium nitrate and hormopriming with 50 ppm GA3 has good potential to improve crop establishment and yield of rice in both conventional and soil mulch DSR systems.

To break dormancy and increase the germination of Prunus avium L. (mazzard cherry) seeds, various methods were tested including the removal of the seed coat after cold moist stratification and treatment with GA<sub>3</sub>, KNO<sub>3</sub>, or thiourea. Treatments with 7,500 ppm KNO<sub>3</sub> after 120 days of stratification were more effective, yielding 64.54% germination of seeds with coat. In seeds without coat, 500 ppm GA<sub>3</sub> treatment after 120 days of stratification gave 79.74% germination; a value increased about 29% compared to control. &nbsp;

Effects of brief red and far-red light on germination of seeds from dormant lines of wild oat (Avena fatua L.) were studied in combination with mechanical injury to the seed coat, application of gibberellin A3, or changes in relative humidity during afterripening. Aberrant germination responses to phytochrome action were observed in the mechanically injured seeds in some of the lines, i.e., brief red light inhibited or delayed germination induced by injury, and immediately following far-red light cancelled the negative effects. Phytochrome action influenced germination of the gibberellin-treated seeds in a normal fashion, although effects of the gibberellic acid and brief red light on germination were not additive. Brief red light inhibited germination of seeds afterripened in zero relative humidity; the same light promoted germination of those in 30 and 60% relative humidity. Germination response to phytochrome in wild oat depends on specific seed dormancy states, illustrated by genetic origins, dormancy-breaking methods and afterripening conditions. The Passiflora L. genus covers a diversity of wild species with ornamental potential, especially due to the intrinsic beauty of its exotic flowers, flowering more than once a year and the lush foliage. However, Passiflora seeds present dormancy complicating seed germination and the establishment of commercial plant production with species with high ornamental potential. This study was conducted to determine the best pre-germination treatments to overcome seed dormancy for Passiflora quadrangularis, P. nitida, P. foetida, P. eichleriana, P. alata, P. cincinnata, P. mucronata, P. micropetala, P. suberosa, P. morifolia and P. tenuifila. The experimental design was completely randomized, with five treatments and four replicates, with 25 seeds per plot. Pre-germination treatments were: seeds soaked in 1,000 mg L- 1 GA3 (gibberellic acid) for 6 hours, seeds soaked in 0.2 % KNO3 (potassium nitrate) for 24 hours, seeds soaked in 1 % KNO3 for 24 hours, partial seedcoat scarification with sandpaper number 120 and control (seeds untreated). Percentage of germination, germination velocity index and radicle length were evaluated for all species. The results showed that GA3 was effective to overcome seed dormancy in P. suberosa (86%), P. morifolia (68 %) and P. tenuifila (54%). KNO3 1% had significant effect on overcoming dormancy in seeds of P. eichleriana (66%) and scarification with sandpaper increased seed germination of P. micropetala (38%).

The effect of seed treatment with solutions of a purified amount of triterpene glycosides (PATG) containing, as major components, oleanolic acid glycosides – sylphiosides B, C, E, G, and extract (E) enriched with sylphiosides, from Silphium perfoliatum L. (Asteraceae) leaves on growth parameters and nitrate reductase activity (NR, EC 1.6.6.1) of 7-day winter wheat plants (Tritium aestivum L.) was studied. It was shown that, seed treatment with PATG solutions in concentrations of 0.0005 and 0.001% and E in concentrations of 0.2 and 0.4% caused an increase in the length of roots, shoots, wet and dry weight of seedlings compared to the control. The stimulating effect of these concentrations of PATG and E on the total nitrate reductase activity of the roots and leaves of seedlings has been established, and an increase in the stimulating effect of preparations on the activity NR оf against the background of substrate activation of the enzyme potassium nitrate (KNO3) was observed. The greatest increase in the total NR activity of roots and leaves of winter wheat plants was observed when PATG acted at a concentration of 0.001% and E – at a concentration of 0.4%, which amounted respectively 122 and 116%, when adding 1 ml of 50 mM KNO3 solution into the growing medium of plants – 141 and 137% relative to the control. The stimulating effect of exogenous triterpene glycosides on NR activity has been established for the first time. The obtained data allow to theoretically substantiate the possibility of practical use of triterpene glycosides and preparations based on them for the regulation of growth and nitrogen metabolism of plants.
