1. A meningococcus vaccine suspended in salt solution has been given subcutaneously as a prophylactic to about 3,700 volunteers in three injections of 2,000 million, 4,000 million, and 4,000 or 8,000 million cocci at weekly intervals. 2. These doses rarely caused more than the mildest local and general reactions. Exceptionally a more severe reaction emphasized the presence of an unusual individual susceptibility to the vaccine. In such instances the symptoms were in part those of meningeal irritation and sometimes simulated the onset of meningitis. 3. Specific meningococcus agglutinins have been demonstrated in the blood serum of vaccinated men as compared with normal controls. 4. Moreover, agglutinins have been demonstrated in the blood serum of chronic carriers of the meningococcus. Evidence is thus brought forward that the relative immunity of chronic carriers to epidemic meningitis may be due to the presence of specific antibodies in the blood stream.

Specific antibodies against estradiol (Es) and progesterone (Pg) are known to modulate blood serum concentrations of these hormones and their biological effects after immunization of animals. It was suggested that specific IgA-Es and IgA-Pg could influence on Es and Pg levels in human blood serum. The purpose of this study was to identify the suggested correlations between serum Es and Pg and specific IgA-Es and IgA-Pg in postmenopausal healthy women (HW) and breast cancer patients (BCP). The serum levels of Es, Pg, IgA-Es and IgA-Pg were studied in 226 HW and 633 BCP by means of solid-phase immunoassay. The following results were obtained. The levels of Es in BCP (0.25 nmol/l) were higher than in HW (0.16; р < 0.0001). The levels of Pg were lower (0.79 vs 0.87; р < 0.0001), and individual Pg/Es ratios were lower (3.19 vs 6.64; р < 0.0001). Individual IgA-Pg/IgA-Es ratios correlated with decrease of Es (rs = -0.15; p = 0.029), with increase in Pg (rs = 0.38; р < 0.0001), and with increased Pg/Es ratio (rs = 0.29; р < 0.0001) in healthy women. Similar correlations were determined in BCP (correspondingly: rs = -0.14, р < 0.001; rs = 0.1, р = 0.009; rs = 0.15, р < 0.0001). The decrease of Es and increase of Pg and Pg/Es in BCP were less significant than in HW: the a quotients in regression у = ах+b (y = hormones levels and x = antibodies levels) in BCP were 3 to 4-fold lower than in HW. These peciliarities of interrelations between hormones and specific antibody levels were revealed only in ER+/PR+ BCP but not in ER+/PR- and ER-/PR- BCP. In conclusion, we have confirmed a suggestion about participation of specific antibodies in regulation of steroids levels in human blood serum. The immune regulation of hormonal status was weakened in BCP.

1. The filtered blood serum of rabbits infected with pneumococci is not toxic. 2. Extracts of pneumococci prepared by keeping emulsions of the bacteria in salt solution at 37° C. for varying periods of time may be toxic, and when injected intravenously into guinea pigs, may produce a train of symptoms followed by acute death resembling that seen in acute anaphylaxis. Such extracts, however, are not uniformly toxic and it has been impossible to discover the exact conditions under which such extracts become toxic. 3. When the centrifugalized peritoneal washings of guinea pigs infected with pneumococci are injected into the circulation of normal guinea pigs, these animals very frequently exhibit symptoms like those seen in acute anaphylaxis, and a considerable proportion of the animals die acutely. 4. When pneumococci are dissolved in dilute solutions of bile salts and the solution resulting is injected intravenously into rabbits and guinea pigs, these animals show with great constancy the same symptoms that are seen in acute anaphylaxis. The solution of pneumococci in bile may occur in ten minutes at 37° C. or in half an hour on ice. This is considered evidence that the toxicity of the solution does not result from digestion of the bacterial protein, but is due to substances preformed in the bacterial cells and set free on their solution. The toxicity of the solution is diminished or destroyed by heating to 55° C. or over.

Metabolic disorders and dysfunctional state of the digestive organs lead to the development of distal extremities diseases. The greatest danger is neсrobacrillosis. Vaccination is one of the ways to prevent and control this disease. The article reflects the data on the effect of vaccination (“Nekovak” and formol-emulsion neсrobacrillosis vaccine of Federal Centre for toxicology, radiation and biological safety) on immunological and serological parameters of blood and blood serum of cattle in the Udmurt Republic. After immunization with “Nekovak” vaccine and formol-emulsion neсrobacrillosis vaccine, we determined an increase of the number of leukocytes to 7th day, and also an increase of T-lymphocytes to 14th day. The number of B-lymphocytes reaches a maximum on 21st day. The content of gamma globulins, including specific antibodies against the neсrobacrillosis pathogen, exceeded the control and reached a maximum on the 21st day after vaccination.

Metal-on-metal (MoM) hip prostheses are known to release chromium and cobalt (Co), which negatively affect the health status, leading to prosthesis explant. Albumin (ALB) is the main serum protein-binding divalent transition metals. Its binding capacity can be affected by gene mutations or modification of the protein N-terminal region, giving the ischaemia-modified albumin (IMA). This study evaluated ALB, at gene and protein level, as marker of individual susceptibility to Co in MoM patients, to understand whether it could be responsible for the different management of this ion. Co was measured in whole blood, serum and urine of 40 MoM patients. A mutational screening of ALB was performed to detect links between mutations and metal binding. Finally, serum concentration of total ALB and IMA were measured. Serum total ALB concentration was in the normal range for all patients. None of the subjects presented mutations in the investigated gene. Whole blood, serum and urine Co did not correlate with serum total ALB or IMA, although IMA was above the normal limit in most subjects. The individual susceptibility is very important for patients’ health status. Despite the limited results of this study, we provide indications on possible future investigations on the toxicological response to Co.

Specific antibodies against environmental chemical gene toxicants and endogenous steroid hormones are shown to modulate concentrations of these compounds in blood serum and their biological effects in experimental models. However, probable hazards of such antibodies in human teratogenesis are still unknown. In particular, potential correlations between specific serum antibodies, sex hormone levels in pregnant women, and congenital malformations in newborns are not clear. The aim of this study was to identify possible associations between occurrence of antibodies to benzo[a]pyrene, estradiol and progesterone (Bp, Es and Pg, respectively), and congenital malformations, and effects of these antibodies upon Es and Pg concentrations in blood serum of pregnant women. We have included into the study 182 women with normal pregnancy and 101 females with congenital malformations of fetus. A non-competitive solid phase immunoassay was performed using Bp, Es and Pg conjugated to bovine serum albumin as antigens. Es and Pg serum concentrations were measured using immunoassay test-systems of “Immunotech” (Moscow). Results: strong positive correlations were revealed between the levels of studied antibodies in the both groups. High IgA-Bp/IgA-Es (> 3) and IgA-Bp/IgA-Pg ratios (> 3) were associated with congenital malformations (OR = 2.2, p = 0.013 and OR = 6.8, p < 0.0001). Positive correlations were revealed between Pg/Es and IgA-Bp/IgA-Es (rS = 0.62, p < 0.0001), and IgA-Bp/IgA-Pg ratios (rS = 0.77, p < 0.0001) in cases with inborn malformations. Similar correlations were found for the women who had normal pregnancy (rS = 0.4, p = 0.0001, and rS = 0.23, p = 0.026, respectively). The Pg/Es proportion correlated with IgG-Bp/IgG-Es (rS = 0.46, p = 0.002), and with IgG-Bp/IgG-Pg ratio (rS = 0.5, p = 0.0009) in cases of malformations, but not in women with normal pregnancy. Conclusion: we have revealed novel associations between congenital malformations of fetus and ratios of IgA-Bp/IgA-Es, as well as IgA-Bp/IgA-Pg, like as positive correlations between hormonal Pg/Es proportions, and ratios of specific antibodies in pregnant women.

The aim of the study was to determine the prevalence of SRLV infection in sheep breeds farmed in mid-Eastern Poland. Out of 6,470 mother ewes kept in 98 nucleus flocks of various sizes, 2,924 belonging to 15 breeds and lines were selected for serum sample collection. The selection of the animals analyzed was carried out using stratification which enabled the determination of infection prevalence with an error margin no greater than 8%. The assay of SRLV-specific antibodies in blood serum was performed using the ELISA test. The analyses revealed a large diversity of prevalence between the sheep breeds studied. The lowest prevalence was observed in Uhruska sheep, in which only 5.07% had a positive serological reaction, despite the fact that the number of animals of this breed included in the analysis was large (1675 mother ewes). The prevalence for this variety was over 5-7 times lower in comparison to Wielkopolska sheep (35%), Polish Pogorze sheep (22.73%), Podhale Zackel sheep (29.03%) and over 10 times lower than Świniarka sheep (57.04%). The results should be used for developing breeding strategies and establishing anti-SRLV programs..

Comparative investigations of the sensitivity and specificity of the indirect immunoenzyme probe - iELISA and the hemagglutination inhibition test (HI test) in serological diagnostics of the Newcastle disease in poultry were carried out using samples of blood serum taken from non-vaccinated and vaccinated poultry. A total of 14 samples of blood serum from non-vaccinated poultry were examined using the immunoenzyme probe - iELISA, and nine of these were found to be positive to the presence of specific antigen against the Newcastle disease virus, while two samples were suspect, and no presence of specific antibodies was established in three samples. Examinations of 82 samples of blood serum from vaccinated poultry for the presence of specific antibodies against the Newcastle disease virus established their presence in 80 serum samples, while one sample was suspect and one sample was negative. The values of the titer of specific antibodies in blood serum samples of vaccinated and non-vaccinated poultry established using the hemagglutination inhibition test (HI test) ranged from 1:2 to 1:32.


In a previous communication on precipitin reactions, we brought forward observations which led to the conclusion that the precipitum is derived mainly from the antiserum and not from the homologous protein. In a recent review of our paper the suggestion was made that gravimetric evidence might be more conclusive. Acting on this suggestion, we have carried out experiments in which a considerable amount of antiserum was allowed to interact with a known small amount of homologous protein. As soon as the interaction was completed, the deposits were collected, washed, and weighed. In every case the amount of dried precipitum exceeded the amount of dried homologous protein. The smallest precipitum obtained weighed more than twice, the largest more than 25 times, the homologous protein employed in the interaction. Former observations had revealed that antisera require very different amounts of protein to ensure the formation of maximal deposits. It was, therefore, to be expected that considerable variations would be obtained in the weight of deposits produced by small amounts of protein in different antisera. Method of Experiment . The antiserum was obtained from rabbits which were bled on the day of the experiment. One milligramme or 2·5 milligrammes dried egg-white or blood serum was dissolved in 10 c. c. salt solution and added to the homologous antiserum, together with about 90 c. c. salt solution. The whole of the available clear antiserum, with the exception of less than 1 c. c. reserved for control observations, was employed. The dried protein was prepared under aseptic conditions, and very precaution was taken to exclude micro-organisms during the experiment. After 48 hours the clear superfluids were removed, and were usually subjected to further experiment, either by addition of more homologous protein, or by addition of fresh antiserum from another immunised animal. Since it had been shown that different homologous antisera might interact to yield a precipitate, appropriate controls were carried out which showed that no trace of deposit occurred as the result of the interactions with one another of the antisera used in these experiments.

Serological investigations were performed on 24,208 samples of cattle blood serum, representing 31.18% of the reproductive cattle fund in the territory of nine municipalities in the Southern Backa region over a period of five years. The samples were examined using the method of microscopic agglutination, with live antigens of nine Leptospira serotypes: L. pomona, L. icterohaemorrhagiae, L. grippotyphosa, L. sejroe, L. tarassovi, L. australis, L. bataviae, L. canicola, and L. hardjo. Specific antibodies against leptospira were established in 286 or 1.18% samples of cattle serum. The level of seroprevalence of leptospira infection at an annual level ranged from 2.56% to 0.05%. The biggest number of seropositive cattle were registered in the municipality of Novi Sad-151 cattle, or 3.54%. In the course of the observation period, the presence of four serotypes of ieptospira were diagnozed in catlt: L. pomona, L. icterohaemorrhagiae, L. grippotyphosa, L. hardjo. The most represented serotype was L. hardjo, 41.02%, while the serotypes L. grippotyphosa (34.61%), L. icterohaemorrhagiae (14.74%) and L. pomona (9.61%) were diagnosed in smaller percentages. An increasing tendency was observed in the number of cattle infected with leptospira at an annual level of 0.195%. On the basis of the presented analysis, it can be predicted with a probability of 95%, that leptospirosis in cattle will appear within the range of 0.09% to 3.50% in the epizootiological region of Southern Backa in the coming period.

Introduction. The objective of our research was to point to the significance of serological diagnostic methods, that is, competitive immunoenzyme test - cELISA as well as the method of classical agglutination of O and H salmonella antigen, for specific antibodies against Salmonella enterica subspecies enterica serovar Enteritidis presence and titre detection in blood serum samples of bothvaccinated and unvaccinated poultry. Material and methods. In our work, we have used commercial competitive immunoenzyme test - cELISA and classical agglutination method with O and H antigens of Salmonella enterica subspecies enterica serovar Enteritidis. Comparative testing included 177 blood serum samples of poultry, out of which 137 was from unvaccinated and 40 from vaccinated individuals originating from majority of poutry farms. Results. In 74 blood serum samples, that is 54,01% out of the total of tested samples originating from unvaccinated flocks, by the use of cELISA test, there were found specific antibodies against S. Enteritidis, while by the method of classical agglutination specific antibodies against O antigen 1,9 and 12 were found in 58 samples, that is in 42,34% , and specific antibodies against H antigen g and m were found in 61 samples,what was 44,53% . In all the tested blood serum samples of vaccinated poultry, there was determined the presence of of specific antibodies against S. Enteritidis, both by the use of competitive cELISA method and classical agglutination with somatic O and flagellar H antigens. By the statistical analysis of the results obtained by the method of classical agglutination and cELISA use of kappa test, there was found out a very good compliance (kappa=0,813). Conclusion. Based on the compared results of blood serum testing on the presence of specific antibodies against S. Enteritidis, it can be concluded that cELISA and classic agglutination with O and H antigen methods have a significant place in serological diagnostics of poultry samonelosis, because their application enables detection of the titre of specific antibodies against S. Enteritidis presence in the population of unvaccinated poultry sensitized with antigens of the mentioned causative agent, as well as in unvaccinated animals.
