Abstract Marine sponges are associated with marine fungi. The associated fungi produce secondary metabolites for sponge survival in extreme habitats. Despite the important role of the associated fungi on their host, the research on marine fungi however has not been studied well. This study aimed to isolate sponge-associated marine fungi and analyze the potency of fungal secondary metabolites against Candida albicans. The sponge used was Clathria sp. collected from Pramuka Island, Indonesia. Fungal isolation used the direct inoculation method. Fungal identification was done by morphological and molecular characteristics of ITS rDNA region. Detection of anti-Candida used the well diffusion method. The isolate has typical morphological characteristics of the genus Gymnoascus with noduled chlamydospore and arthroconidia. The isolated fungus was identified as Gymnoascus udagawae based on morphological and molecular analysis. This is the first record of marine fungi G. udagawae from Indonesia. The ethyl acetate extract of fungal filtrate showed 1.4 cm inhibition diameter of 500 mg/μL extract. The inhibition is moderate category compared to that of clotrimazole a drug commonly used for candidiasis as the positive control with showed 2.8 cm inhibition diameter at 100 mg/μL. The fungus is a potential source of the secondary metabolite active against C. albicans.

Saluang Belum (Luvunga sarmentosa Kurz) adalah tumbuhan yang dikenal sebagai obat tradisional di Kalimantan Tengah. Batang L. sarmentosa Kurz dipercayai memiliki manfaat secara empiris sebagai antioksidan yang dimanfaatkan sebagai anti-aging (anti penuaan dini). Batang L. sarmentosa Kurz mengandung metabolit sekunder yaitu tanin, saponin, steroid dan flavonoid yang diduga memiliki aktivitas sebagai antibakteri. Berdasarkan khasiat empiris dan kandungan metabolit sekunder yang ada dalam tumbuhan L. sarmentosa Kurz, maka dilakukan penelitian tentang aktivitas daya hambat ekstrak etanol tumbuhan ini terhadap bakteri P. acnes yang merupakan salah satu bakteri penyebab jerawat. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas daya hambat ekstrak etanol batang L. sarmentosa Kurz terhadap bakteri P.acnes. Metode penelitian yang dilakukan pada penelitian ini dimulai dari pengambilan simplisia, pembuatan ekstrak etanol dengan metode perkolasi dan uji daya hambat dengan menggunakan metode Disk Difusion yang menggunakan kontrol positif antibiotik klindamicin. Analisis data dilakukan dengan membandingkan lebar zona hambat dari ekstrak etanol batang L. sarmentosa Kurz  dengan kategori penghambatan antimikroba klindamisin berdasarkan diameter zona hambat dari Clinical and Laboratory Standards Institute.  Hasil uji daya hambat didapatkan zona hambat ekstrak etanol batang Saluang Belum pada semua konsentrasi 0,5%, 1%, 5%, 10% dan 15% secara berturut-turut yaitu  Intermediate, Resistant, Intermediate, Susceptible dan Susceptible dan hasil dari zona hambat klindamicin pada semua konsentrasi 0,5%,1%,5%,10%,dan 15% adalah susceptible. Simpulan dari penelitian ini adalah ekstrak etanol batang L. sarmentosa Kurz mempunyai kemampuan dalam menghambat pertumbuhan bakteri P.acnes.           Kata Kunci: Obat Tradisional, Jerawat, Uji Daya Hambat, Propionibacterium acnes, Batang Saluang Belum Saluang belum (Luvunga sarmentosa Kurz) is one of traditional medicine in Central Kalimantan. The stem of L. sarmentosa Kurz is believed to have empirical benefits as an antioxidant which is used as anti-ageing (anti-ageing). The stem of L. sarmentosa Kurz contains secondary metabolites, namely tannins, saponins, steroids and flavonoids which are thought to have antibacterial activity. Based on the empirical properties and the content of secondary metabolites in L. sarmentosa Kurz, a study was conducted on the inhibitory activity of this plant's ethanol extract against P. acnes, which is one of the bacteria that causes acne. The purpose of this study was to determine the inhibitory activity of the ethanol extract of L. sarmentosa Kurz stem against P.acnes bacteria. The research method carried out in this study started from taking simplicia, making ethanol extract using the percolation method and the inhibition test using the disk diffusion method which used a positive control of the clindamycin antibiotic. Data analysis was performed by comparing the inhibition zone width of the ethanol extract of L. sarmentosa Kurz stem with the clindamycin antimicrobial inhibition category based on the inhibition zone diameter of the Clinical and Laboratory Standards Institute. The results of the inhibition test obtained the inhibition zone of Saluang Belum stem ethanol extract at all concentrations of 0.5%, 1%, 5%, 10% and 15% respectively, Intermediate, Resistant, Intermediate, Susceptible and Susceptible and the results of the inhibition zone. Clindamycin at all concentrations of 0.5%, 1%, 5%, 10%, and 15% are susceptible. This research concludes that the ethanol extract of L. sarmentosa Kurz stem can inhibit the growth of P.acnes bacteria.

The biosynthesis of compounds with antibiotic activity produced by marine fungi, strongly depends on their growth conditions. A good understanding of the role of culture conditions in the biosynthesis of metabolites may lead to better exploitation of microbial metabolites. In this study, the influence of culture conditions including incubation period, initial pH and salinity on antimicrobial activity and secondary metabolites production of marine fungus 01NT.1.1.5 was investigated. This isolate, obtained from sponge Stylissa sp. in Nha Trang Bay, exhibited a broad spectrum of in vitro antimicrobial activity to Bacillus cereus ATCC 11778, Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Listeria monocytogenes ATCC 19111, Streptococcus faecalis ATCC 19433 and Candida albicans ATCC 10231. According to morphological characteristics and sequence analysis of 28S rDNA, the fungus was identified as Aspergillus flocculosus. The results indicated that antimicrobial activity and metabolite amount were highest when the fungus was cultivated in rice medium with incubation period of 20 days. The optimum salinity of 35 g/L and initial pH of 6.0 were found for the maximum antibiotic production. The colony growth, antimicrobial activity and production of secondary metabolites of the strain A. flocculosus 01NT.1.1.5 varied depending on salt concentrations and initial pH of medium. Particularly, extract of this fungus only showed activity against C. albicans when it was cultured in medium with 30-35 g/L salinity and initial pH 4.0-8.0. The results  indicate that salinity and initial pH along with cultivation period are important factors influencing antimicrobial activity and secondary metabolites of A. flocculosus 01NT.1.1.5, and might be for other marine fungi.

Abstract. Handayani D, Artasasta MA, Safira N, Ayuni DF, Tallei TE, Hertiani T. 2020. Fungal isolates from marine sponge Chelonaplysilla sp.: Diversity, antimicrobial and cytotoxic activities. Biodiversitas 21: 1954-1960. The purpose of this research was to study the diversity of fungi associated with marine sponges Chelonaplysilla sp. and their bioactivities. Fungal isolation was carried out by the multilevel dilution method in Saboraud Dextrose Agar (SDA). Twelve fungal isolates were successfully purified, then cultivated using rice for 4-6 weeks at room temperature and subsequently extracted using ethyl acetate. Antimicrobial activities of the fungal extracts were tested against Staphylococcus aureus, Escherichia coli, and Candida albicans by using the agar diffusion method. The extracts of isolates Ch05 and Ch12 showed a significant antagonistic effect against S. aureus and E. coli with the diameter that ranged from 15 to 17 mm. Using the brine shrimp lethality test (BSLT), six fungal extracts revealed cytotoxic activity with LC50 <100 µg/mL. Isolate Ch10 was the most potential fungus with the strong cytotoxic activity of LC50 of 0.90 µg/mL. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was conducted also for six potential fungal extracts against breast cancer cell (T47D). The isolate Ch05 showed moderate cytotoxic activity with IC50 of 83.69 µg/mL. The molecular identification was carried out for potential fungi using the ITS marker. The results showed that Ch02 was Aspergillus oryzae, Ch05 was Phomompsis sp., Ch06 was Penicillium simplicissimum, Ch10 was B. bassiana and Ch12 was Aspergillus mellinus. This study concluded that fungal isolates from marine sponge Chelonaplysilla sp. can be explored further for new sources of antimicrobial and anticancer compounds.

Medicinal plants are a wealthy source of natural medicinal properties and remain as base for new drug discoveries. Endophyte from the specific medicinal plants produce the analogous metabolites as that of the host plant. The metabolites from the endophytes comprise maximum therapeutic properties and have been extensively applied in treating various diseases and disorders. This study was focused on identification of the endophytic fungi from the medicinal plant Blumea axillaris and investigates the diversity of endophytic fungi from various explants of the same plant. The explants were cultured on potato dextrose agar and 6 endophytic fungi were successfully isolated from Blumea axillaris. They were identified morphologically and confirmed with molecular analysis as Xylaria arbuscula, Paraphoma radicina, Phomopsis phaseoli, Sordaria fimicola, Aspergillus amstelodami, Diaporthe eucalyptorum. The DNA sequences were analyzed by BLAST and the phylogenetic tree was constructed with neighbor joining method. The six isolates were subjected to antagonistic activity for the selection of potential strain and the bioactive strain Xylaria arbuscula was selected for the production of secondary metabolites by optimization. The parameters like pH, temperature, incubation period, carbon and nitrogen (organic and inorganic source) were optimized for secondary metabolite production. The fungal metabolite was extracted by solvent extraction method using polar and non-polar solvents like propanol, methanol, chloroform, acetone and ethyl acetate. To investigate the bioactivities of the fungal crude extract was subjected first for its antioxidant activity using DPPH radical scavenging method, followed by antimicrobial activity of methanolic (MeOH) extract of Xylaria arbuscula, that were also analyzed by the agar well-diffusion method against the clinical pathogens Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pnuemoniae, Proteus mirabilis, Aspergillus niger and Candida albicans.

AbstractEndophytic fungi associated with Nigerian plants have recently generated significant interest in drug discovery programmes due to their immense potential to contribute to the discovery of new bioactive compounds. This study was carried out to investigate the secondary metabolites of endophytic fungi isolated from leaves ofNewbouldia laevis, Ocimum gratissimum, andCarica papayaThe plants were collected from Agulu, Anambra State, South-East Nigeria. Endophytic fungal isolation, fungal fermentation; and extraction of secondary metabolites were carried out using standard methods. The crude extracts were screened for antimicrobial activities using the agar well diffusion method, and were also subjected to high performance liquid chromatography (HPLC) analysis to identify their constituents. A total of five endophytic fungi was isolated, two fromN. laevis(NL-L1 and NL-L2), one fromO. gratissimum(SL-L1), and two fromC. papaya(PPL-LAC and PPL-LE2). In the antimicrobial assay, the extracts of NL-L2, SL-L1, and PPL-LE2 displayed mild antibacterial activity against both Gram negative and Gram positive test bacteria. PPL-LAC extract showed mild activity only againstS. aureus, while no antimicrobial activity was recorded for NL-L1 extract. All the endophytic fungal extracts showed no activity against the test fungiC. albicansandA. fumigatusHPLC analysis of the fungal extracts revealed the presence of ethyl 4-hydroxyphenyl acetate and ferulic acid in NL-L1; ruspolinone in NL-L2; protocatechuic acid, scytalone, and cladosporin in SL-L1; indole-3-acetic acid and indole-3-carbaldehyde in PPL-LE2; and indole-3-acetic acid in PPL-LAC. The findings of this study revealed the potentials possessed by these plants as source of endophytes that express biological active compounds. These endophytes hold key of possibilities to the discovery of novel molecules for pharmaceutical, agricultural and industrial applications.

Frogs of the family Centrolenidae are adapted to live at high altitudes, and so it is not surprising that their greatest diversity is in the Andes mountain range. Here we extend the known geographic distribution of Vitreorana ritae by more than 550 km towards southern Amazonia. The species was identified by morphological, acoustic and molecular characteristics (mitochondrial marker 16S rRNA). The records reported here are the first for the genus Vitreorana in the state of Mato Grosso, Brazil, as well as for the transition zone between the Amazonia and Cerrado biomes. These records are consistent with the hypothesis that the low number of species of centrolenids reported in the lowlands of the Amazon region may be the result of limited sampling. Even though V. ritae is distributed throughout the Amazon, most of its diagnostic morphological characteristics are conserved. Thus, the great rivers of the Amazon Basin do not seem to act as geographical barriers for this species; however, due to the limited sample size, further acoustic and molecular studies are needed to confirm this hypothesis.

Abstract Parthenitae and cercariae of Plagiorchis. multiglandularis Semenov, 1927 are recorded in Lymnaea stagnalis (Linnaeus, 1758) for the fi rst time in Ukraine; their morphological characteristics are specifi ed. Diagnostic characters of P. multiglandularis parthenitae and cercariae found in Ukrainian Polissia are compared with those from other regions. To confi rm the validity of the species, a comparison of the morphometric data of this trematode larvae with the cercariae of Plagiorchis elegans (Rudolphi, 1802) Braun, 1902, found in molluscs L. stagnalis, L. ralustris and L. corvuses, was performed. It was determined that P. multiglandularis cercariae diff er from those of P. elegans in size and position of the penetration glands.

Diseases mediated by the immune system are difficult problems to treat such as human immunodeficiency virus (HIV) and other lethal viruses. Infections that occur in normal people are generally brief and rarely leave permanent damage. Treatment of this disease requires an aggressive and innovative approach to the development of new treatments so that it requires the role of immunomodulators to improve the immune system. A substance that acts as an enhancer or immune enhancer can be obtained by using herbs that are efficacious as immunostimulants. One of the herbs used is herbal binara (Artemisia vulgaris L) which has been studied as a potential immunomodulator with high antioxidant activity. Previous research also stated that red shoots (Syzygium oleana) were studied as potential immunomodulators with high antioxidant activity. Several other species such as Syzygium samarangense have 16 flavonoida compounds which show pharmacological immunological activity. The purpose of this study was to determine the content of secondary metabolites of ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) and to determine the best dose of extract from the ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) can reduce the volume of swelling of mouse feet. Examination of the chemical content of secondary metabolites from the ethanol extract of herbal binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) is carried out by chemical screening and characterization of simplicia and extract. The method used is the slow type hypersensitivity method. In this test the independent variable is the secondary metabolite of ethanol extract of herb binara (Artemisia vulgaris L.) with red shoots (Syzygium oleana) with four concentrations (50, 100, 200 and 400 mg / kgBB). The positive control used by Stimuno dose is 32.5 mg / kgBB

AbstractDifficult-to-treat infections caused by methicillin-resistant Staphylococcus aureus (MRSA) are of concern in people living with HIV infection as they are more vulnerable to infection. We aimed to identify molecular characteristics of MRSA colonizing newly diagnosed HIV-infected adults in Tanzania. Individuals newly diagnosed with HIV infection were recruited in Dar es Salaam, Tanzania, from April 2017 to May 2018, as part of the randomized clinical trial CoTrimResist (ClinicalTrials.gov identifier: NCT03087890). Nasal/nasopharyngeal isolates of Staphylococcus aureus were susceptibility tested by disk diffusion method, and cefoxitin-resistant isolates were characterized by short-reads whole genome sequencing. Four percent (22/537) of patients carried MRSA in the nose/nasopharynx. MRSA isolates were frequently resistant towards gentamicin (95%), ciprofloxacin (91%), and erythromycin (82%) but less often towards trimethoprim-sulfamethoxazole (9%). Seventy-three percent had inducible clindamycin resistance. Erythromycin-resistant isolates harbored ermC (15/18) and LmrS (3/18) resistance genes. Ciprofloxacin resistance was mediated by mutations of the quinolone resistance-determining region (QRDR) sequence in the gyrA (S84L) and parC (S80Y) genes. All isolates belonged to the CC8 and ST8-SCCmecIV MRSA clone. Ninety-five percent of the MRSA isolates were spa-type t1476, and one exhibited spa-type t064. All isolates were negative for Panton-Valentine leucocidin (PVL) and arginine catabolic mobile element (ACME) type 1. All ST8-SCCmecIV-spa-t1476 MRSA clones from Tanzania were unrelated to the globally successful USA300 clone. Carriage of ST8 MRSA (non-USA300) was common among newly diagnosed HIV-infected adults in Tanzania. Frequent co-resistance to non-beta lactam antibiotics limits therapeutic options when infection occurs.

BACKGROUND: The Mexican serviceberry, Malacomeles denticulata, have been used as a successful oral therapy by Mexican communities without enough scientific support. OBJECTIVE: To evaluate the M. denticulata extracts with selective antibacterial properties over dental biofilm bacteria. METHODS: Fruit, Leaf, and Stem of M. denticulata extracts were evaluated with micro-broth dilution method using ATCC bacteria. OD600 values had compared against each positive control (T-student-test). Anaerobically viability had confirmed by Colony-Forming-Units. Thin-Layer-Chromatography was used to identify the number of compounds and phytochemicals to identify secondary metabolites of the selected extracts. RESULTS: Streptococcus mutans showed Minimum-Bactericidal-Concentrations_(MBC) at 30 mg/mL to Fruit, Leaf, and Stem extracts. Periodontal-pathogens Aggregatibacter actinomycetemcomitans serotype b_(MBC = 30 mg/mL_p <  0.01); Fusobacterium nucleatum subsp. nucleatum_(MBC = 30 mg/mL_p<0.001); Parvimonas micra_(MBC = 15 mg/mL_NS); Porphyromonas gingivalis_(MBC = 30 mg/mL_NS); and Prevotella intermedia_(MBC = 3.75 mg/mL_NS) presented higher sensitivity to Leaf-Methanol, than the primary colonizers. Phytochemicals showed positive results to anthraquinones, coumarins, flavonoids, saponins, saponins steroids/triterpenoids, steroids/triterpenes, and tannins/phenols. CONCLUSION: We suggest the natural extracts of fruit and leaf of the Mexican serviceberry for the preventive use over the oral cariogenic or periodontal biofilm species, by their selective antibacterial properties against pathogenic species evaluated in-vitro, and due to the presence of antibacterial secondary metabolites identified as flavonoids and saponins of M. denticulata leaf extracts.
