Sleep is a vital physiological state, and it is essential for optimal daytime function. Sleep patterns undergo tremendous quantitative and structural changes throughout the maturational process from infancy to late adolescence. These universal alterations in sleep and sleep patterns reflect the essential need for optimal sleep during normal healthy development. Unfortunately, sleep loss has become omnipresent in modern adult culture and has also become a widespread phenomenon among children, particularly with respect to adolescents. Specifically, sleep loss in adolescence is a function of multiple normative endogenous changes often amplified by exogenous influences. Substance abuse has also achieved epidemic proportions among today’s adolescents, with well-documented negative functional outcomes. This chapter summarizes objective and subjective studies examining the bidirectional links between alcohol and substance use and inadequate sleep and/or sleep patterns in adolescents.

Chromatin structure at the fundamental level of the nucleosome is important in vital cellular processes. Recent biochemical and genetic analyses show that nucleosome structure and structural changes are very active participants in gene expression, facilitating or inhibiting transcription and reflecting the physiological state of the cell. Structural states and transitions for this macromolecular complex, composed of DNA wound about a heterotypic octamer of variously modified histone proteins, have been measured by physico-chemical techniques and by enzyme-accessibility and are recognized to occur with various post-translational modifications, gene activation, transformation and with ionic-environment. In spite of studies which indicate various forms of nucleosome structure, all current x-ray and neutron diffraction studies have consistently resulted in only one structure, suggestive of a static conformation. In contrast, two-dimensional electron microscopy studies and three-dimensional reconstruction techniques have yielded different structures. These fundamental differences between EM and other ultrastructural studies have created a long standing quandary, which I have addressed and resolved using spectroscopic electron microscopy and statistical analyses of nucleosome images in a study of nucleosome structure with ionic environment.

AbstractSleep loss has emerged as a risk factor for the development of impaired glucose tolerance. The mechanisms underpinning this observation are unknown; however, both mitochondrial dysfunction and circadian misalignment have been proposed. Given that exercise improves glucose tolerance, mitochondrial function, and alters circadian rhythms, we investigated whether exercise may counteract the effects induced by inadequate sleep. We report that sleeping 4 hours per night, for five nights, reduced glucose tolerance, with novel observations of associated reductions in mitochondrial function, sarcoplasmic protein synthesis, and measures of circadian rhythmicity; however, incorporating three sessions of high-intensity interval exercise (HIIE) during this period mitigates these effects. These data demonstrate, for the first time, a sleep loss-induced concomitant reduction in a range of physiological processes linked to metabolic function. These same effects are not observed when exercise is performed during a period of inadequate sleep, supporting the use of HIIE as an intervention to mitigate the detrimental physiological effects of sleep loss.

Natural autofluorescence is a widespread phenomenon observed in different types of tissues and organisms. Depending on the origin of the autofluorescence, its intensity can provide insights on the physiological state of an organism. Fungal autofluorescence has been reported in terrestrial and human-derived fungal samples. Yet, despite the recently reported ubiquitous presence and importance of marine fungi in the ocean, the autofluorescence of pelagic fungi has never been examined. Here, we investigated the existence and intensity of autofluorescence in five different pelagic fungal isolates. Preliminary experiments of fungal autofluorescence at different growth stages and nutrient conditions were conducted, reflecting contrasting physiological states of the fungi. In addition, we analysed the effect of natural autofluorescence on co-staining with DAPI. We found that all the marine pelagic fungi that were studied exhibited autofluorescence. The intensity of fungal autofluorescence changed depending on the species and the excitation wavelength used. Furthermore, fungal autofluorescence varied depending on the growth stage and on the concentration of available nutrients. Collectively, our results indicate that marine fungi can be auto-fluorescent, although its intensity depends on the species and growth condition. Hence, oceanic fungal autofluorescence should be considered in future studies when fungal samples are stained with fluorescent probes (i.e., fluorescence in situ hybridization) since this could lead to misinterpretation of results.

The purpose of this study was to describe the sleep patterns and fatigue of both mothers and fathers before and after childbirth. The authors used wrist actigraphy and questionnaires to estimate sleep and fatigue in 72 couples during their last month of pregnancy and 1st month postpartum. Both parents experienced more sleep disruption at night during the postpartum period as compared to the last month of pregnancy. Compared to fathers, with their stable 24-h sleep patterns over time, mothers had less sleep at night and more sleep during the day after the baby was born. Sleep patterns were also related to parents’work status and type of infant feeding. Both parents self-reported more sleep disturbance and fatigue during the 1st month postpartum than during pregnancy. Mothers reported more sleep disturbance than fathers, but there was no gender difference in ratings of fatigue. At both time points, fathers obtained less total sleep than mothers when sleep was objectively measured throughout the entire 24-h day. Further research is needed to determine the duration of sleep loss for both mothers and fathers, to evaluate the effect of disrupted sleep and sleep loss on psychosocial functioning and job performance, and to develop interventions for improving sleep patterns of new parents.

The article considers changes in biochemical blood parameters depending on the degree of adaptation process manifesting on the plantar surface of hind limbs of different sex rabbits and physiological state under cage husbandry technology. Groups of stud bucks (1st series), pregnant and lactating does (2nd and 3rd series of experiments) were formed according to the stages of structural changes depending on the stress of physiological regulatory processes of adaptation (the systematics proposed by R.M. Baevsky). The authors studied the following factors in serum: total protein, protein fractions (albumin, α-, β-, γ-globulin), creatinine, total bilirubin, urea, uric acid, alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase, glucose triacylglycerol and cholesterol. At the stressed stage of regulatory systems the group of bucks showed a reduction of total bilirubin on the background of decrease of ALT, AST and alkaline phosphatase, which can testify to activation of liver function at this stage, and increase in bilirubin, ALT, AST with a simultaneous decrease in alkaline phosphatase, noted at the overstress stage of physiological adaptation regulatory processes, can testify to decrease in the functional activity of hepatocytes.Pregnant rabbits have the most pronounced compensatory reactions in the third stage of the adaptation process: the increase in absolute and relative γ-globulin values indicates the activation of immune factors. An increase in creatinine and urea levels during the gestation period, against a background of developing adaptive changes on the plantar surface of the feet, indicates a decrease in the filtration capacity of the renal tubules.Lactating animals experience an increased physiological load on the liver, which is expressed by an increase in triacylglycerols throughout the adaptation.

Abstract Introduction An abundance of cross-sectional research links inadequate sleep with poor emotional health, but experimental studies in children are rare. Further, the impact of sleep loss is not uniform across individuals, and pre-existing anxiety might potentiate the effects of poor sleep on children’s emotional functioning. Methods N=53 children (mean age 9.0 years; 56% female) completed multi-modal, emotional assessments in the lab when rested and after two nights of sleep restriction (7h and 6h in bed, respectively). Sleep was monitored with polysomnography and actigraphy. Subjective reports of affect and arousal, psychophysiological reactivity, and objective emotional expression were examined during two emotional processing tasks, including one where children were asked to suppress their emotional responses. Results After sleep restriction, deleterious alterations were observed in children’s affect and their emotional reactivity, expression, and regulation. These effects were primarily limited to positive emotional stimuli. The presence of anxiety symptoms moderated most of the alterations in emotional processing observed after sleep restriction. Conclusion Results suggest inadequate sleep preferentially impacts positive compared to negative emotion in pre-pubertal children and that pre-existing anxiety symptoms amplify these effects. Tim23p is an essential channel-forming component of the multisubunit TIM23 complex of the mitochondrial inner membrane that mediates protein import. Radiolabeled Tim23p monocysteine mutants were imported in vitro, incorporated into functional TIM23 complexes, and subjected to chemical cross-linking. Three regions of proximity between Tim23p and other subunits of the TIM23 complex were identified: Tim17p and the first transmembrane segment of Tim23p; Tim50p and the C-terminal end of the Tim23p hydrophilic region; and the entire hydrophilic domains of Tim23p molecules. These regions of proximity reversibly change in response to changes in membrane potential across the inner membrane and also when a translocating substrate is trapped in the TIM23 complex. These structural changes reveal that the macromolecular arrangement within the TIM23 complex is dynamic and varies with the physiological state of the mitochondrion.

Aim:This study examines the relationship between the personality traits of Neuroticism and Extroversion and sleep patterns and others sleep behaviours in medical students.Method:Neuroticism and Extroversion were assessed using the Portuguese version of the Eysenck Personality Inventory. Sleep was assessed with a self-response questionnaire covering a wide range of sleep-wake habits and difficulties. A factor analysis with varimax rotation was performed with seventeen items of the questionnaire and four factors were extracted: Sleep Disturbance, Sleep Timing, Insufficient Sleep and Dreams/Nightmares. A total of 203 third year medical students (127 females, 62.6%) mean age 20.01 (SD=1.09; Range=18-24) participated in the study.Results:There were no significant gender differences in Neuroticism and Extroversion mean scores (Neuroticism: p=.454; Extroversion: p=.891). Extroversion was significantly correlated with Sleep Timing (p=.000), whereasthere was a trend for Neuroticism to be associated with Sleep Disturbance (p=.095). Concerning the relationship of Neuroticism and Extroversion mean scores with other Sleep problems, Neuroticism was found associated with concentration difficulties in the morning (p=.006), inadequate sleep duration (p=.001), sleep problems (p=.012), and use of sleep medication (p=.052). No significant associations were found with Extroversion.Conclusion:Neuroticism was significantly associated with sleep problems, use of sleep medication, inadequate sleep duration, and concentration difficulties in the morning. Extroversion was correlated only with later Sleep Timing.

AbstractDifferent current ideas on the origin of life are critically examined. Comparison of the now fashionable FeS/H2S pyrite-based autotrophic theory of the origin of life with the heterotrophic viewpoint suggest that the later is still the most fertile explanation for the emergence of life. However, the theory of chemical evolution and heterotrophic origins of life requires major updating, which should include the abandonment of the idea that the appearance of life was a slow process involving billions of years. Stability of organic compounds and the genetics of bacteria suggest that the origin and early diversification of life took place in a time period of the order of 10 million years. Current evidence suggest that the abiotic synthesis of organic compounds may be a widespread phenomenon in the Galaxy and may have a deterministic nature. However, the history of the biosphere does not exhibits any obvious trend towards greater complexity or «higher» forms of life. Therefore, the role of contingency in biological evolution should not be understimated in the discussions of the possibilities of life in the Universe.
