1. Intravenous infusions of the brain/gut hormone, cholecystokinin, have been shown to reduce food intake in a subsequent test meal. However, in previous studies the doses administered were large and likely to have produced plasma concentrations far in excess of the normal post-prandial range. 2. In this study cholecystokinin-8 was infused intravenously to six healthy subjects in doses that reproduced physiological post-prandial concentrations. Plasma concentrations of cholecystokinin were measured using a novel sensitive and specific radioimmunoassay. The effect of cholecystokinin-8 infusion on subsequent food intake in a standard test meal was compared with the effect of saline infusion in the same subjects. 3. Food intake (mean ± SEM) was significantly less during cholecystokinin (5092 ± 665 kJ) than during saline infusion (6418 ± 723 kJ, P = 0.03). During cholecystokinin infusion, plasma concentrations increased from 0.45 ± 0.06 pmol/l to 7.28 ± 2.43 pmol/l immediately before the meal. With saline infusion there was no premeal increase in plasma cholecystokinin concentration. 4. This paper describes a novel radioimmunoassay for measurement of plasma concentrations of cholecystokinin. Using this assay we have demonstrated that cholecystokinin is important in control of satiety in humans.

AbstractThe reduced fertility that is becoming more evident in high yielding dairy cows may be related to many factors including changes in milk production, food intake and fluctuations in body condition. Metabolic and production markers have been studied as a way of predicting success to a particular artificial insemination. Successful conception to a particular service was not associated with milk production, body condition or plasma concentrations of several indicators of metabolic state around the time if insemination. This highlights the importance of time of information collection in fertility management programmes. Increased food intake may reduce systemic progesterone concentrations. This is more evident in sheep than cattle, but a positive relationship between systemic progesterone early post mating and establishment of pregnancy in cattle has been reported. However, progesterone concentrations in the ovarian vein and endometrium are not strongly correlated with systemic progesterone. Thus, the significance of modest changes in systemic progesterone in affecting oocyte and embryo development must be questioned. Blood urea concentrations can be altered by diet, and reduced pregnancy rates have been reported in cows with high urea concentrations. However, in other recent studies, no difference was reported in serum urea in cows that conceived and those that failed to conceive. Pregnancy rate was equally high in heifers when in-vitro produced embryos were transferred to heifers on high and low urea diets. When embryos were produced in sheep on high and low dietary urea, the effects on embryo development appear to occur early in the developmental process, suggesting a substantial effect on the development of the oocyte. The developmental capacity of oocytes and quality of embryos is reduced in cattle maintained on extremely high dietary intakes. Oocyte developmental capacity is reduced in cows of higher genetic merit and embryo quality can be substantially reduced in the early postpartum period. Collectively, these results suggest that high dietary intake or high metabolic load is deleterious to normal oocyte development and establishment of pregnancy. This highlights the importance of further studies on the effect of dietary intake on metabolic state and follicle, oocyte and embryo development. In a practical context, these results highlight the importance of nutritional management and avoiding changes in the amount or type of diet around the time of mating in high-production dairy cows.

AbstractRecently, we reported that, in mice, hunger causes the autophagy-dependent release of a protein called “acyl-CoA-binding protein” or “diazepam binding inhibitor” (ACBP/DBI) from cells, resulting in an increase in plasma ACBP concentrations. Administration of extra ACBP is orexigenic and obesogenic, while its neutralization is anorexigenic in mice, suggesting that ACBP is a major stimulator of appetite and lipo-anabolism. Accordingly, obese persons have higher circulating ACBP levels than lean individuals, and anorexia nervosa is associated with subnormal ACBP plasma concentrations. Here, we investigated whether ACBP might play a phylogenetically conserved role in appetite stimulation. We found that extracellular ACBP favors sporulation in Saccharomyces cerevisiae, knowing that sporulation is a strategy for yeast to seek new food sources. Moreover, in the nematode Caenorhabditis elegans, ACBP increased the ingestion of bacteria as well as the frequency pharyngeal pumping. These observations indicate that ACBP has a phylogenetically ancient role as a ‘hunger factor’ that favors food intake.

Abstract Background and Aims Vascular calcification (VC) is an important contributor to the high rate of cardiovascular mortality associated to chronic kidney disease. The inability to eliminate phosphorus (P) and the subsequent P retention promotes CV. P metabolism and uremic VC are influenced by obesity and by the caloric content of the diet. Caloric restriction (CR) has been shown to have multiple beneficial effects on health, for example, CR has been reported to improve vascular health and retard vascular ageing. However, to our knowledge the effect of CR on the development of uremic VC has not been explored. We hypothesize that CR may be beneficial to prevent the development of uremic VCs. Thus, the objective of the present study was to determine if rats subjected to CR were protected against VC. Method 48 Wistar rats were divided in four groups. The control diet provided Metabolizable Energy = 3.528 kcal/g and contained 0.6% Calcium (Ca) and 0.6% P. Additional diets of identical composition to the control diet but containing varying levels of Ca and P: 0.9% Ca, 0.9% P; 0.6% Ca, 1.2% P; and 0.9% Ca, 1.8% P, were also used in the experiments. Rats in Group 1 and 3 were fed 15 g/day of the control diet. Rats in Group 2 and 4 were calorie restricted and fed 10 g/day of diet with Ca/P = 0.9%/0.9%. Thus the daily P intake should be identical in the four groups. Uremia was induced by 5/6 nephrectomy (Nx). After Nx rats in Group 1 and 2 were fed ad libitum a diet with 0.6% Ca and 1.2% P. While rats in Group 3 and 4 were fed ad libitum a diet with 0.9% Ca and 1.8% P. Rats were supplemented with calcitriol. At the end of the experiment, rats were sacrificed to obtain blood samples and tissue samples (thoracic and abdominal aortas). After blood collection, plasma was separated by centrifugation and stored at –20° C until assayed. Plasma creatinine, Ca and P were measured by spectrophotometry. Energy intake was calculated based on food intake. VC was studied by histology and by measuring the tissue Ca content. Values are expressed as mean ± standard error (SE), the difference between groups was assessed by ANOVA. Fisher LSD test was used as a post-hoc procedure. p<0.05 was considered significant. Results Before Nx, caloric intake was significantly lower in calorie restricted rats (35.4 ± 0.1 and 35.8 ± 0.1 kcal/day) than in rats eating normal calories (52.7 ± 0.1 and 52.8 ± 0.2 kcal/day); however, P intake was almost identical in the four groups and ranged between 89.8 and 91.6 mg/day. After Nx, rats in all groups reduced food intake and, consequently, caloric intake. Thus, although the P content of the diet was increased after Nx, daily P intake was not increased in Groups 1 and 2; however, P intake was significantly increased in Groups 3 and 4 (120.9 ± 4.6 and 122.2 ± 6.2 mg/day, respectively). In all groups, rats had high plasma concentrations of creatinine and P, and low plasma concentrations of Ca. Also, all rats had elevated Ca content in the aorta. No significant differences between the study groups were found in any of these parameters (Table 1). Von Kossa staining of the aortas showed abundant mineral deposition in the four groups. Conclusion This study shows that, contrary to what was expected, CR did not prevent or ameliorate uremic calcifications.

The quantification of medical or toxic substances in vitreous humour (VH) could be very useful in forensic toxicology when blood sample determinations are impossible due to absence or deterioration. However, few studies have been made in this area and even fewer have tried to find a relationship between drug levels in both samples. To determine a correlation ratio between blood and VH diazepam (DZ) levels, we performed an experimental study using rabbits administered with a sub-toxic dose of DZ under known and controlled conditions. Blood and VH samples were collected 0.5, 1, 2, 3 and 6 hours after the drug administration in order to determine DZ and its main active metabolite, desmethyldiazepam (DMD). In addition, we have studied an animal group sacrificed 2 hours after intramuscular (i.m.) drug administration with blood and VH collection 24 hours later, to evaluate the existence of possible post-mortem changes. After DZ administration, a fast absorption phase was observed with a plasma Cmax value 1 hour after, followed by a rapid concentration decrease, with a half-life of 1 hour, indicating that, besides elimination, a fast distribution to other organs and tissues and/or hepatic metabolism occurred. Diazepam Cmax value in VH was achieved between 1 and 2 hours, when plasma concentrations had already decreased to half the value. The plasma/VH DZ ratio calculated at this time was 10. In the post-mortem study, while plasma DZ concentration at 24 hours was smaller, DMD levels were higher than those at the time of death. In the VH, both DZ and DMD concentrations at 24 hours were higher than those obtained at the time of death. That is, in both fluids DZ and DMD concentrations were different from those at the time of death and post-mortem distribution and redistribution phenomena occurred. The combination of antemortem and post-mortem studies has allowed the determination of a correlation ratio for DZ in the rabbit of 6×, comparing the concentrations in VH collected 24 hours after death with the concentrations detected in plasma at the time of death. This study opens new perspectives for the use of VH as a complementary sample to blood for DZ detection and confirmation. The putative relevance of the correlation ratio obtained, for forensic toxicology practice with medical substances, namely benzodiazepines, recommends further studies in humans.

AbstractThe rôle of protein in food intake regulation is complex in ruminants. Previous research has shown that a deficiency in degradable nitrogen (N) could affect microbial activity and decrease intake. On the other hand, an increase in metabolizable protein content of the diet seems to stimulate food intake in lactating dairy cows. The aim of this experiment was to determine whether metabolizable protein supply plays a direct rôle in the stimulation of food intake. Treatments comprised two infusions of soya protein isolate (800 g/day) either into the rumen (RP) or into the duodenum (DP), which were compared with two iso-energy infusions of glucose (880 g/day) either into the rumen (RG) or into the duodenum (DG). Four ruminally and duodenally cannulated cows producing 36·5 kg/day of milk were assigned to a 4 ✕ 4 Latin-square design with periods of 4 weeks. Duodenal infusions of protein (DP) significantly increased (P < 0·05) dry-matter intake (DMI) ( +1·9 kg/day), rate of intake ( + 8·2 g DMI per min), milk yield ( + 4 kg/day), protein content ( + 2·3 g/kg) and protein yield ( +191 g/day) compared with the glucose infusion in the duodenum (DG). No significant effect was observed with ruminal infusion of protein (RP) compared with the glucose infusion in the rumen (RG). The protein infusions had no effect (P > 0·05) on the apparent digestibility of dry matter, organic matter, neutral-detergent fibre or acid-detergent fibre and also no or only small effects on ruminal fermentation variables. Plasma concentrations of most of the essential amino acids increased significantly with the duodenal infusion of protein, whereas ruminal infusion of protein had no significant effect. It is concluded that direct supply of metabolizable protein stimulates intake independently of ruminal digestion effects.

AbstractAn experiment was made to examine the effects of food intake and an orally active progestagen on plasma steroid concentrations and subsequent reproductive performance of multiparous sows. Thirty-five multiparous Landrace × Large White sows were assigned to one of four treatments immediately after parturition. Treatment H-AT sows were offered a high level of food intake throughout lactation: 3·5 kg of a diet containing 160 g/kg dry matter (DM) of crude protein and 13 MJ digestible energy per kg DM given twice daily. Treatment H+AT sows were offered the same food intake as H-AT sows and in addition they were given 20 mglday of allyl trenbolone (AT) mixed with the morning feed. Treatment L-AT sows were given 1·5 kg of the same diet offered twice daily during lactation and treatment L+AT sows were also offered this lower level of food intake and given AT. The respective levels of food intake were offered to sows from the 1st day of lactation onwards until weaning at 21 days post partum. Litter weights at weaning were influenced significantly by feeding level (P < 0·001) and also by AT administration (P <0·05). Sows on the high level of feeding had the heaviest litters and food-restricted sows had the lightest litters. AT treatment depressed litter weight at weaning. There was no effect of food level or AT on the plasma concentrations of progesterone or oestrogens during lactation. There was a highly significant (P <0·001) difference in mean plasma oestrogen concentration between high (H-AT and H+AT) and low (L-AT and L+AT) fed groups during early pregnancy in the subsequent cycle. Sows given a combination of high energy in lactation and AT (H+AT) exhibited shorter intervals from weaning to oestrus than both groups of food-restricted (L-AT and L+AT) sows fP <0·01) but treatment had no significant effect on either the farrowing rate or on the subsequent litter size. It is concluded that despite significant changes in the live weight and condition of sows and changes in steroid hormone concentrations due to food intake and the administration of allyl trenbolone, there were no significant effects on reproductive performance.
