Background: Emerging resistant Candida species become a severe problem for public health. The study aims to examine the efficacy of Terminalia subspathulata bark and fruit extracts as an alternative source of antifungals for resistant strains of Candida species. Methodology: Candida albicans was isolated between 4–5th interdigital foot space and C. parapsilosis were isolated from the left-hand third finger of male patients. Morphological and molecular methods identified the isolates. Antifungal susceptibility testing (AFST) was determined using azole derivatives, amphotericin B (AmpB) and echinocandins based on E-test and broth microdilution (BMD) methods. The AFST activity of the 10 μL methanolic extracts with concentrations between 1.25–10 mg/mL was assessed by disk diffusion method according to CLSI guidelines. Results: The AFST results showed total resistance to all azoles (FLU, MIC ≥ 256 μg/ml and VOR, ITR and POS each with MIC ≥ 32 μg/ml). The BMD method agreed with E-test MICs. The T. subspathulata bark extract tested indicated activity on the C. albicans lawn with the zone of inhibition between 9–13 mm. The fruit extract indicated zone between 8–10 mm. Candida parapsilosis indicated susceptibility to bark extract with the zone of inhibition between 10–15 mm, while the fruit extract showed zone between 8–13 mm. Conclusion: Despite azole resistance, the extracts showed good activity against the isolates. Findings showed alternatives to AmpB and echinocandins because of toxicity and parenteral administration, respectively. Based on our findings, our study first reports the medicinal value of T. subspathulata.

The present study targeted to evaluate the phytoconstituents of Nyctanthes arbortristis Linn., and its biological activity. By solvent extraction procedure, a total of four extracts were designated from the leaf and bark of the plant using two solvents (leaf extract in ethyl acetate and methanol: NALE & NALM; bark extract in ethyl acetate and methanol: NABE & NABM). Phytochemical screening was conducted by qualitative analysis and thin-layer chromatography. Parallelly, antioxidant property (by DPPH free radical scavenging method) and antimicrobial activity (by disk diffusion method) were also investigated to determine bioactivity. The presence of alkaloids and glycosides was detected by qualitative phytochemical assay of the extracts. Furthermore, TLC successfully observed the versatility of the compound's presence, such as phenolic compounds, different alkaloids, and glycosides. In DPPH assay, methanolic extracts are highly capable of scavenging the radicals than the ethyl acetate extracts. Surprisingly, IC50 for NABM (1.69 µg/ml) is less than ascorbic acid (3.58µg/ml), which is an exploration of the excellent antioxidant potential of the plant. But any extracts showed no positive result in the antimicrobial test against gram-positive or gram-negative bacteria, even against yeast.

Objective. The study aims to speciate clinical Candida isolates and detect their biofilm-forming ability and antifungal resistance. Methods. All the Candida spp. isolated from different clinical samples like pus, urine, blood, and body fluid were included in the study. Biofilm production was tested by the microtiter plate method. Antifungal susceptibility was studied by the disk diffusion method. Patient’s demographic details such as age, sex, and clinical information were collected. Presence of other risk factors such as diabetes mellitus, history of antibiotic use, and any urinary tract instrumentations was also recorded. Results. Among 90 Candida species isolated, most predominant species was found to be C. albicans (45.5%) followed by C. tropicalis (28.88%), C. krusei (20%), C. glabrata (3.33%), and C. parapsilosis (2.22%). Candida spp. were isolated from urine (43%), BAL/sputum (18.88%), high vaginal swab (8.88%), suction tips (7.77%), blood and wound swabs (6.66%), pus (3.33%), bile aspirate (2.22%), and deep tissue (1.11%). A larger number of females were affected than males, and the age group of 51 to 60 years was more susceptible to candidiasis. A higher number of C. albicans isolates produced biofilm followed by C. parapsilosis, C. tropicalis, and C. krusei. However, C. glabrata showed no biofilm production in our study. All Candida isolates were 100% sensitive to amphotericin B. Voriconazole was the next effective drug with 81.11% susceptibility. 24.44% of strains were resistant to fluconazole. Conclusion. Speciation of Candida isolates, detection of ability to form the biofilm, and monitoring of antifungal susceptibility testing are necessary for appropriate treatment.

Dillenia serrata Thunb.is a member of the Dillenia clan which is endemic to Sulawesi. D. serrata fruit is a seasonal fruit and can be used as a cooking ingredient as a sour taste, raw material for several food products such as candied fruit and syrup, and has the potential to be processed into chips and jam. Traditionally, the leaves and bark of D. serrata are also used by the local community as a sprue medicine, fever, wound medicine, treating swelling or inflammation and treating vomiting of blood. It is believed that there are still many untapped benefits from this plant. This study was conducted to determine the effectiveness of D. serrata fruit extract as an antimicrobial that causes disease in humans, and to determine its effectiveness as an antioxidant. The antioxidant test was carried out using the DPPH method while the antimicrobial test was carried out by the agar diffusion method (Kirby-Bauer). The ripe Dillenia fruit was extracted using methanol, the extract was then diluted in various concentrations for the DPPH test The results showed that the D. serrata extract had the ability as an antioxidant. The antioxidant activity of fresh fruit extracts was higher than that of dried fruit extracts. D. serrata extract is effective in inhibiting the growth of Pseudomonas aeruginosa, Staphylococcus aureus, and Streptococcus mutans bacteria.

ABSTRACT Aim The present study was conducted to evaluate the efficacy of commercially available herbal toothpastes against the different periodontopathogens. Materials and methods Six herbal toothpastes that were commonly commercially available were included in the study. Colgate herbal, Babool, Meswak, Neem active, Dabur red toothpastes were tested for the study whereas sterile normal saline was used as control. Antimicrobial efficacies of dentifrices were evaluated against Streptococcus mutans and Actinobacillus actinomycetemcomitans. The antimicrobial properties of dentifrices were tested by measuring the maximum zone of inhibition at 24 hours on the Mueller Hinton Agar media inoculated with microbial strain using disk diffusion method. Each dentifrice was tested at 100% concentration (full strength). Results The study showed that all dentifrices selected for the study were effective against the entire test organism but to varying degree. Neem active tooth paste gave a reading of 25.4 mm as the zone of inhibition which was highest amongst all of the test dentifrices. Colgate Herbal and Meswak dentifrices recorded a larger maximum zone of inhibition, measuring 23 and 22.6 mm respectively, compared to other toothpastes. All other dentifrices showed the zone of inhibition to be between 17 and 19 mm respectively. Conclusion The antibacterial properties of six dentifrices were studied in vitro and concluded that almost all of the dentifrices available commercially had antibacterial properties to some extent to benefit dental health or antiplaque action. Seven out of eight methanolic extracts from five plants native to Mexico were inactive against ten bacterial strains of clinical interest. The fruit extract of Chenopodium ambrosioides inhibited the bacteria Enterococcus faecalis (MIC = 4375 μg/ml), Escherichia coli (MIC = 1094 μg/ml), and Salmonella typhimurium (MIC = 137 μg/ml). The fruit extract of C. ambrosioides was with CC50 = 45 μg/ml most cytotoxic against the cell-line Caco-2, followed by the leaf extract from Pithecellobium dulce (CC50 = 126 μg/ml); interestingly, leaves of C. ambrosioides (CC50 = 563 μg/ml) and bark of P. dulce (CC50 = 347 μg/ml) extracts were much less cytotoxic. We describe for the first time the cytotoxic effect from extracts of the aerial parts and the flowers of Cirsium mexicanum (CC50 = 323 μg/ml and CC50 = 250 μg/ml, resp.). Phytochemical analysis demonstrated for both extracts high tannin and saponin and low flavonoid content, while terpenoids were found in the flowers. For the first time we report a cytotoxicological study on an extract of Eryngium carlinae (CC50 = 356 μg/ml) and likewise the bark extract from Amphipterygium adstringens (CC50 = 342 μg/ml). In conclusion the fruit extract of C. ambrosioides is a potential candidate for further biological studies.

Today, the tendency to use of natural preservatives to increase food security has expanded. In the present study, antibacterial effects of Areca Nut fruit extracts were evaluated against Staphylococcus aureus, Escherichia coli, Salmonella enterica, and Enterobacter aerogenes bacteria using agar disc diffusion technique. Methanol, ethanol, and water were used as solvents for extraction by maceration method, and extracts were analyzed by GC-MS. The antibacterial activity was evaluated using microtiter broth dilution method to determine minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). Results revealed that all ATCC strains were significantly inhibited by ethanolic and methanolic extracts. Escherichia coli produced a significantly larger zone of inhibition for Gentamicin ( 35 ± 0.65   mm ) and Penicillin ( 25   mm ± 0.56 ), while Enterobacter aerogenes produced smaller zone of inhibition for Gentamicin (20 ± 0.87 mm) and Penicillin ( 15 ± 0.87   mm ). Also, methanolic extract had considerable antibacterial activity with MIC value of 1.56 mg/mL against Escherichia coli. All of extracts were used to evaluate antibacterial effects in prepared cake, and as a result, all pathogenies were the most sensitive by methanolic extract in 100 mg/L of concentration except Escherichia coli that were more sensitive by ethanolic extract. In conclusion, the Areca Nut fruit extracts may be used as a natural preservative in food industries. Future studies should focus on the effect of Areca Nut fruit extracts in bakery and drinking industries.

Abstract The genus Malassezia belongs to Basidiomycota and includes 16 species, from which M. pachydermatis is the most common in dogs. M. pachydermatis is a member of the normal mycobiota of the skin and mucosal sites of dogs. Under certain conditions, these yeasts can be opportunistic pathogens and involved skin and ear canal infections of these animals. Topical and oral antifungal agents are used for the therapy of Malassezia dermatitis and otitis. With the expanding use of antifungal agents, resistant strains of Malassezia are increasingly detected. In this study, the susceptibility of 40 M. pachydermatis isolates to fluconazole, itraconazole, ketoconazole, clotrimazole and nystatin were evaluated in vitro based on the modified standard disk diffusion method M44-2A.

In this study, we are presenting the effect of three ripening stages of air-dried bitter gourd fruit extracts on phenolic acid composition, antioxidant, antibacterial, and anticancer activities. The results showed mature bitter gourd fruit extract in 100% methanol showing 78% DPPHº scavenging activity. Immature dried fruit extract in 80% and 100% methanol showed promising antibacterial activities, i.e., >18.5 ± 0.21 mm zone-of-inhibition against Staphylococcus aureus, while mature dried fruit extract in 80% methanol showed 18.4 ± 0.17 mm zone-of-inhibition against Escherichia coli. Anticancer activity results of 100% methanol extracts of ripened fruit possess showed 6.72 ± 1.81 and 3.55 ± 0.51 mg/mL IC50 values with HeLa and MDBK cancer cell lines, respectively. The overall results indicate that the immature and ripen fruits of BG could be extracted in pure methanol as an antibacterial and anticancer phytomedicine.

Background: Algeria, by its vast terrestrial extent and its climatic variation, has an abundant, rich and varied flora in which it was counted many aromatic and medicinal species that provide bioactive compounds characterized by their broad biological activities. In this context, this work is based on the evaluation of the antimicrobial activity of Quercus robur L. leaves extracts (Family of Fagaceae). Methods: Firstly, the collected plant material was defatted; then, the extraction of tannins and saponins was carried out according to a standard protocol where the extracts obtained were tested on some uropathogenic microbial strains by disk diffusion method with the determination of the Minimum Inhibitory Concentrations (MICs) by broth macro-dilution method. Results: The extraction yield of the selective extracts was 7.93 and 16.94% for tannins and saponins, respectively. The antibiotic resistance profile of the tested strains showed a resistance relatively important to several antibiotics, namely amoxicillin +clavulanic acid and ampicillin for Escherichia coli, while Pseudomonas aeruginosa showed resistance to amoxicillin+clavulanic acid, amikacin, cefotaxime and ceftazidime. However, Staphylococcus aureus was susceptible to penicillin, gentamicin, ofloxacin and chloramphenicol. Antifungal susceptibility testing has been shown that Candida albicans was susceptible to amphotericin B, econazole and it was clinically categorized as intermediate to miconazole drug. For antimicrobial tests, the tannins and saponins extracts exhibited a low to strong inhibitory effect at tested concentrations lower than 30 mg/mL (ranged from no inhibition to an inhibition zone diameter of 17.5 mm), depending on dose levels and tested microbial strains. Conclusion: This activity is proportional to the tested concentrations, knowing that tannins extract was more active compared to saponins extract. For this, Q. robur could constitute an important source for drug discovery.

Introduction: Plants have been used for thousands of years to treat health disorders, to prevent diseases including epidemics, to flavor and conserve food. It is estimated that 250 to 500 thousand plant species are present on the earth, out of which only 10% is used as a source of food by humans and animals Objective: In the present work, antibacterial activity of five different Indian spices: turmeric, clove, pepper, cinnamon and garlic were investigated against pathogens isolated from wound samples. Method: The unknown bacteria were identified by different types of morphological and biochemical techniques such as serial dilutions, spread plate methods, morphological studies and biochemical tests. The swabs of the patients were inoculated in 10 mL of sterile nutrient broth and incubated at 37oC for 24 h. The antibacterial activities of these Indian spices were evaluated using the disk diffusion method. A suspension of the microorganisms to be tested was spread on nutrient agar and MacConkey agar medium. The filter paper discs were placed on the agar plates, which were saturated with extract of spice. The plates were then incubated at 37oC for 24 h.. After incubation, the zone of inhibition was determined by evaluating the diameter of the zone of inhibition. Results: The antibacterial activities of these Indian spices were evaluated using the disk diffusion method and the inhibitory zones were recorded. It was clear from the result that cinnamon had a larger inhibition zone against P1NA3, P1MAC1 and least with P2NA2. Garlic showed a larger zone of inhibition against P2NA2, P3NA3 and least for P1MAC1. Clove had maximum zone of inhibition against P1MAC2 and least with P3NA3. These spices showed zone of inhibition somewhat close to the control antibiotic drug ampicilline. Conclusion: P1MNC2 showed a maximum zone of inhibition with clove (3 cm) at 100%. P2NA2 showed the highest antibacterial activity with garlic (3.6 cm) at 100% and minimum with cinnamon (1.1cm) at 20%. P3NA3 showed maximum antibacterial activity result with garlic (4.2 cm) at 100% and minimum with clove (1.2cm) at 20%. Spices are economical, more accessible to most of the population in the world. So, medicinal plants should be encouraged to use as potential sources of new drugs.
