Deer antler velvet is one of the important ingredients used in traditional medicine by many Asean countries including Malaysia. However, there is doubt regarding the halalness of the medicine derived from deer antler velvet taken alive due to the conflict of fatwa. The objective of this paper is to analyse the position of medicine derives from deer antler velvet from Shariah perspectives. The qualitative methode used through documents analysis, interview and observation. The finding shows that the conflict exists because of the different of ijtihad when the deer antler velvet taken from the animal alive. It is due to the issue, whether it hurt the animal or vice versa. Based on the scientific observation, it is proven that deer antler velvet is part of the animal organ and the nerve connected can make the animal suffering if it is taken alive. It is totally different from halal animal fur that remove while they still alive. Thus, any medication derived from deer antler velvet is not permissible and unclean (najs). Alternatively, to make it halal, the animal should be slaughtered before removing the deer antler valvet.   Keywords: deer antler velvet, medicine, fatwa, halal, haram   ABSTRAK: Tanduk rusa baldu merupakan antara bahan penting yang dimanfaatkan dalam bidang perubatan herba tradisional di kebanyakan negara asia, termasuk di Malaysia. Bagaimanapun, timbul ketidakpastian hukum terhadap ubat tanduk rusa baldu yang dipotong ketika masih hidup, iaitu sama ada ia boleh dikategorikan sebagai produk yang halal atau haram untuk dimanfaatkan, lantaran wujud konflik fatwa di Malaysia mengenainya. Objektif kajian ini adalah bagi menganalisis hukum ubat daripada tanduk rusa baldu menurut perpektif halal dan haram. Bagi mencapai objektif tersebut, kaedah kualitatif digunakan menerusi pendekatan analisis dokumen, temubual dan pemerhatian. Hasil kajian mendapati antara punca perbezaan pendapat fuqaha dalam menentukan hukum tanduk yang dipotong semasa hidup adalah disebabkan ijtihad mereka yang berbeza terhadap tanduk haiwan, iaitu sama ada ia termasuk dalam kategori anggota yang menyakitkan jika dipotong atau tidak menyakitkan. Berdasarkan maklumat saintifik dan pemerhatian, terbukti bahawa tanduk rusa baldu adalah sejenis anggota yang bersaraf dan menyakitkan jika dipotong ketika hidup, dan tidak boleh disamakan dengan kes bulu haiwan halal. Justeru, produk perubatan berasaskan tanduk rusa yang diperolehi secara kaedah pemotongan tersebut adalah haram dan najis. Sebagai alternatif, pengambilan tanduk rusa menerusi kaedah penyembelihan secara syarak boleh menukar statusnya menjadi produk yang halal.   Kata kunci: tanduk rusa, baldu, perubatan, fatwa, halal, haram

Abstract Objectives Glucocorticoid-induced osteoporosis (dexamethasone) is a primary cause of secondary osteoporosis by the decreasing formation and increasing resorption activities. Previously, the in vitro study showed that 70% ethanol and aqueous extract of deer antler have increased alkaline phosphatase in osteoblast cell that known as marker of bone formation. The mind of this study is to analyze the effect of deer antlers in increasing the bone trabecular density of osteoporosis-induced male mice. Methods This study used a post-test control group design. A total of 54 healthy male mice were randomly divided to nine groups, i.e., healthy control, osteoporotic, positive control, 70% ethanol (4, 8, and 12 mg/kg BW), and aqueous extracts (4, 8, and 12 mg/kg BW) of deer antler groups. All of the interventions were given 1 mL of test sample for 4 weeks orally. The bone densities were determined using histomorphometry by Image J and Adobe Photoshop. The statistical data were performed using SPSS 23 and statistical significance was set at p<0.05. Results The results showed that alendronate group, 70% ethanol, and aqueous extract groups increased bone density and calcium levels in serum (p<0.05) compared to osteoporotic group in dose dependent manner. It indicated that 70% ethanol and aqueous extract of deer antler stimulating bone turnover and aqueous extract showed the highest. Conclusions Dexamethasone induction for 4 weeks caused osteoporotic mice and the administration of 70% ethanol and aqueous extracts of deer antler from East Kalimantan increased trabecular bone density and calcium levels in dose dependent manner.

Deer antlers are the only mammalian appendage capable of regeneration. We aimed to investigate the effect of red deer antler extract in regulating hair growth, using a mouse model. The backs of male mice were shaved at eight weeks of age. Crude aqueous extracts of deer antler were prepared at either 4°C or 100°C and injected subcutaneously to two separate groups of mice (n=9) at 1 mL/day for 10 consecutive days, with water as a vehicle control group. The mice skin quantitative hair growth parameters were measured and 5-bromo-2-deoxyuridine was used to identify label-retaining cells. We found that, in both the 4°C and the 100°C deer antler aqueous extract-injection groups, the anagen phase was extended, while the number of BrdU-incorporated cells was dramatically increased. These results indicate that deer antler aqueous extract promotes hair growth by extending the anagen phase and regulating cell proliferation in the hair follicle region.

Deer antlers are a rare example of mammalian epimorphic regeneration. Each year, the antlers re-grow by a modified endochondral ossification process that involves extensive remodelling of cartilage by osteoclasts. This study identified regenerating antler cartilage as a site of osteoclastogenesis in vivo. An in vitro model was then developed to study antler osteoclast differentiation. Cultured as a high-density micromass, cells from non-mineralised cartilage supported the differentiation of large numbers of osteoclast-like multinucleated cells (MNCs) in the absence of factors normally required for osteoclastogenesis. After 48 h of culture, tartrate-resistant acid phosphatase (TRAP)-positive mononuclear cells (osteoclast precursors) were visible, and by day 14 a large number of TRAP-positive MNCs had formed (783+/−200 per well, mean +/− s.e.m., N=4). Reverse transcriptase/polymerase chain reaction (RT-PCR) showed that receptor activator of NF κ B ligand (RANKL) and macrophage colony stimulating factor (M-CSF) mRNAs were expressed in micromass cultures. Antler MNCs have the phenotype of osteoclasts from mammalian bone; they expressed TRAP, vitronectin and calcitonin receptors and, when cultured on dentine, formed F-actin rings and large resorption pits. When cultured on glass, antler MNCs appeared to digest the matrix of the micromass and endocytose type I collagen. Matrix metalloproteinase-9 (MMP-9) may play a role in the resorption of this non-mineralised matrix since it is highly expressed in 100 % of MNCs. In contrast, cathepsin K, another enzyme expressed in osteoclasts from bone, is only highly expressed in resorbing MNCs cultured on dentine. This study identifies the deer antler as a valuable model that can be used to study the differentiation and function of osteoclasts in adult regenerating mineralised tissues.

Background/Aims: Sika deer (Cervus nippon Temminck) antler is traditional animal medicine of renal protection in East Asia. This study measured the effect of sika deer antler protein (SDAPR) on gentamicin (GM)-induced cytotoxicity in HEK293 cells, and investigated the effect of SDAPR against GM-induced nephrotoxicity in mice. Methods: HEK293 cells viability and oxidative stress were measured in HEK293 cells while flow cytometry was used for apoptosis analysis. The acute kidney injury biomarkers, kidney injury molecule-1 (KIM-1), neutrophil gelatinase-associated lipocalin (NGAL) and cystatin c (Cys-C), were repeatedly measured by ELISA assay. ICR male mice were randomly assigned six groups: Control, GM with vehicle, single SDAPR, GM with SDAPR at three concentrations 50, 100, 200 mg/kg/d, p.o., 10 d. GM was injected for 8 consecutive days (100 mg/kg/d, i.p.). Renal function, oxidative stress and levels of inflammatory factors were measured in vivo. Renal tissues were stained with H&E to observe pathological changes. Results: Pretreatment with SDAPR (0.5-4.0 mg/mL) significantly improved cell viability. Treatment with SDAPR could reduce KIM-1, NGAL and Cys-C activity. SDAPR could improve antioxidant defense and attenuated apoptosis on HEK293 cells. SDAPR also ameliorated GM-induced histopathologic changes, and decreased blood urea nitrogen (BUN) and serum creatinine (Cr). Additionally, SDAPR significantly regulated oxidative stress marker and interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) inflammatory cytokines. Conclusion: These results show that SDAPR could be an effective dietary supplement to relieve GM-induced nephrotoxicity by improved antioxidase activity, suppressed inflammation, and inhibited apoptosis in vitro and vivo.
