Lignocellulose degradation by Fusarium species

Eighteen strains of fungi in the genus Fusarium, including varieties of F. episphaeria, F. lateritium, F. moniliforme, F. nivale, F. oxysporum, F. rigidiusculum, F. roseum, F. solani, and F. tricinctum, slowly degraded lignocelluloses from blue spruce (Picea pungens) and wheat (Triticum aestivum). When grown with [lignin-14C]lignocellulose from blue spruce, 15 of the Fusarium strains converted 2.2 to 4.3% of the [14C]lignin in 60 days to 14CO2 and 3.9 to 8.4% to labeled water-soluble products. When grown with unlabeled lignocellulose from wheat straw, the strains caused total weight losses in 60 days of 7 to 25%, acid-insoluble (Klason) lignin losses of 2 to 17%, and carbohydrate losses of 3 to 33%. Crude protein contents of degraded wheat-straw lignocellulose samples were 3.2 to 5.1%. Among the aromatic degradation products from wheat-straw lignocellulose degraded by different strains, as shown by gas chromatography, were p-coumaric acid, vanillic acid, vanillin, syringaldehyde, and p-hydroxybenzaldehyde.

Abstract Background Water-soluble vitamins are often included simultaneously in pharmaceutical formulations as food complements or in parenteral nutrition mixtures. Given their sensitivity to heat, light or pH variations, it is important to study their stability using validated stability indicating methods. We thus aimed to validate a liquid chromatography (LC) stability-indicating method for the simultaneous quantification of 5 water-soluble vitamins. Methods We analyzed four water-soluble B vitamins (nicotinamide, pyridoxine, folic acid, cyanocobalamin) and ascorbic acid using a LC method with diode array detector. They were separated on a C18 stationary phase under gradient elution of solvent A [0.2 % of metaphosphoric acid in water and acetonitrile 98:2] and solvent B (100 % acetonitrile). All vitamins were subjected to forced degradation conditions and we showed that the obtained degradation products didn’t interfere with the vitamins. Results The method allows the separation of the 5 water-soluble vitamins in a 30 minute run without any interference from the breakdown products obtained with acid/alkaline solutions, hydrogen peroxide, temperature and light. It meets all the qualitative and quantitative criteria for validation with an acceptable accuracy and good linearity. Conclusions This stability-indicating method can be used for carrying out stability studies of water-soluble vitamins in pharmaceutical preparations.

Water-soluble carboxymethylated lignin (CML) was synthesized using wheat straw alkali lignin (WAL) in aqueous medium. The process of carboxymethylation was optimized with respect to the NaOH concentration, monochloroacetic acid concentration, reaction temperature and time. The optimized product has a yield of 80.47% and a carboxyl group content of 2.8231 mmol•g-1, respectively. The optimum conditions for carboxymethylation are NaOH concentration of 20.0% (wt%), monochloroacetic acid concentration of 37.5% (wt%), temperature of 70 °C and time of 90 min. The optimized CML was characterized by FTIR spectroscopy, 1H NMR spectroscopy and interfacial tension apparatus. The result shows that the substitution reaction of carboxymethylation occurs simultaneously in the phenolic hydroxyl group and aliphatic hydroxyl group in WAL. CML has the surface activity in water for industrial application as dispersant.

Two major components of insensitive munition formulations, nitroguanidine (NQ) and 3-nitro-1,2,4-triazol-5-one (NTO), are highly water soluble and therefore likely to photo-transform while in solution in the environment. The ecotoxicities of NQ and NTO solutions are known to increase with UV exposure, but a detailed accounting of aqueous degradation rates, products, and pathways under different exposure wavelengths is currently lacking. We irradiated aqueous solutions of NQ and NTO over a 32-h period at three ultraviolet wavelengths and analyzed their degradation rates and transformation products. NQ was completely degraded by 30 min at 254 nm and by 4 h at 300 nm, but it was only 10% degraded after 32 h at 350 nm. Mass recoveries of NQ and its transformation products were >80% for all three wavelengths. NTO degradation was greatest at 300 nm with 3% remaining after 32 h, followed by 254 nm (7% remaining) and 350 nm (20% remaining). Mass recoveries of NTO and its transformation products were high for the first 8 h but decreased to 22–48% by 32 h. Environmental half-lives of NQ and NTO in pure water were estimated as 4 and 6 days, respectively. We propose photo-degradation pathways for NQ and NTO supported by observed and quantified degradation products and changes in solution pH.

A survey was carried out in 2013–2019 on the life cycle of the bud blight Gemmamyces piceae (Borthw.) Casagrande and the trend assessment in the infestation of the Colorado blue spruce Picea pungens Engelm. stands. Four ecologically different plots were chosen in the Fláje region in the Eastern part of the Krušné hory Mountains. The pycnidia were formed at the beginning of June on buds infested the previous year. Later, in the second half of June, they formed on the spring attack buds. Conidia occurred in the first week of July on the buds of the old infestation, or later in the case of the spring attack buds, in the middle of July. The production of conidia lasted till the end of September. Ascospores occurred in the middle of July and were produced until the end of September. The ability of host trees to sprout became continually weaker, along with the defoliation progress. A distinct decline in the sprouting ability was noticed on stands with a dense crown canopy and on stands with competition of broad-leaved trees – with an admixture of birch.

1. Native DNA from two strains of Bacillus subtilis was chromatographed by stepwise elution from MAK (methylated albumin on kieselguhr). 2. Transforming activity was confined to two out of the three main fractions, activity being distributed between the two peaks differently for DNA from the different strains. 3. Fractionation of DNA from both strains on 2% agarose gel gave two components. Approx. 75% of the material was eluted within the void volume of the column. Approx. 25% of the material consisted of degradation products of lower molecular weight. 4. Chromatography on MAK of the material of high molecular weight eluted from agarose gel gave a number of peaks differing in molecular weight, indicating that degradation of the DNA takes place during chromatography on MAK. 5. The distribution of transforming activity among the fractions from MAK suggests that degradation occurs preferentially in certain regions of the DNA.

Selected anticorrosion preserving agents were subjected to a comparison on the basis of tests made under operating conditions. Resistance of materials to corrosion markedly varies with operating conditions and impact of outer environment with the most important factors influencing the corrosion rate being SO<sub>2</sub> content, temperature and humidity. Relative protective efficiency of preserving agents was assessed on the basis of weight losses of protected and unprotected samples. It was found out that the material&rsquo;s resistance to corrosion is significantly depending on warehousing conditions (SO<sub>2</sub>, temperature, humidity, etc.) and on the quality of the applied preservant. Depending on various environments, the values of relative protective efficiency of water-soluble preservant and preserving agent based on vegetable oils were ranging from 38&ndash;87% and 47&ndash;75%, respectively.

Eleven actinomycetes, isolated from the gut of worker termites (Macrotermes, Armitermes, Microcerotermes, Odontotermes), were identified as Streptomyces chromofuscus, S. chromogenus, S. diastaticus, and S. rochei. Their ability to grow on natural lignocellulosic substrates was tested in solid state fermentation experiments using wheat straw (C/N = 49.8) as a sole carbon source. Weight loss was 4.7–20.9% of the initial substrate, after 5 weeks at 30 °C; lignin and cellulose content decreased 2.0–16.1 and 3.5–32.9%, respectively. When the 11 Streptomyces were grown on wheat straw pretreated with (NH4)HCO3 (C/N = 28.2), weight loss was 9.3–29.9% of the initial substrate, indicating an overall enhancement of lignocellulose degradation. Weight, lignin, and cellulose losses were enhanced when S. chromofuscus (strain A2 and A11) and S. rochei A4 were grown on pretreated wheat straw instead of the untreated substrate. With S. rochei A10 the weight loss and lignin degradation were enhanced, while cellulolysis was slightly depressed. Weight loss and cellulose degradation were both enhanced when the remaining strains were grown on pretreated wheat straw. In this case, lignin degradation was depressed (S. chromofuscus A6 and A8, S. diastaticus A12, S. rochei A14) or remained essentially the same (S. diastaticus A3 and S. chromogenus A7).
