ANTIBIOTIC XG AS A SEED TREATMENT FOR THE CONTROL OF LEAF AND POD SPOT OF PEAS CAUSED BY ASCOCHYTA PISI

Antibiotic XG has been studied for its possible application as a pea seed treatment in the control of Ascochyta leaf and pod spot. Fungistatic properties of Antibiotic XG against Ascochyta Pisi at various concentrations in relation to pH and temperature in vitro were studied. At antibiotic concentrations up to 1 p.p.m. no fungistatic effect was observed at pH values from 3.5 to 6.5, but, at concentrations above 1 p.p.m., complete inhibition of growth was found at all pH values tested. In the temperature range of 22 °C. to 30 °C. antibiotic concentrations of 1 p.p.m. had no effect on growth. Fungicidal activity to spores of A. Pisi has also been tested. A spore suspension of 10,000 spores per ml. required an exposure of 12 hr. with an antibiotic concentration of 100 p.p.m. for complete lethal dosage. The soaking of diseased pea seed samples in Antibiotic XG at a concentration of 25 p.p.m. for a period of 18 hr. effectively controls A. Pisi infection as determined by laboratory test and does not seriously impair germination. This treatment is compatible with Spergon applied after the seed has been air-dried. Storage of treated seed for a period of three months did not affect the degree of control but reduced the germinative capacity in laboratory plate tests.

Atherosclerosis is associated with the increased trans-sialidase activity, which can be detected in the blood plasma of atherosclerosis patients. The likely involvement in the disease pathogenesis made this activity an interesting research subject and the enzyme that may perform such activity was isolated and characterized in terms of substrate specificity and enzymatic properties. It was found that the enzyme has distinct optimum pH values, and its activity was enhanced by the presence of Ca2+ ions. Most importantly, the enzyme was able to cause atherogenic modification of lowdensity lipoprotein (LDL) particles in vitro. However, the identity of the discovered enzyme remained to be defined. Currently, sialyltransferases, mainly ST6Gal I, are regarded as major contributors to sialic acid metabolism in human blood. In this mini-review, we discuss the possibility that atherosclerosis- associated trans-sialidase does, in fact, belong to the sialyltransferases family.

The MEIC (multicentre evaluation of in vitro cytotoxicity) programme is a five-year programme to validate in vitro tests for general toxicity, and is organised by the Scandinavian Society for Cell Toxicology. Interested laboratories are invited, on an international basis, to test 50 published reference chemicals in their respective assays. Submitted results will then be evaluated yearly by the MEIC Committee for their relevance to various types of human toxicity, including an evaluation for the same chemicals of the prediction by animal tests of human toxicity. To establish the validation methods, a preliminary validation cycle is being performed in 1989/90 with submitted results for the first ten MEIC chemicals. The present paper is the very first step of this preliminary validation process. The prediction of human toxicity by five cytotoxicity assays (altogether 14 different cell systems/endpoints) has been evaluated, and also compared with the predictive value of rodent LD50 tests. Mouse LD50 prediction of human lethal dosage for these substances was good, while rat LD50 prediction was less satisfactory. The collective predictions by all 14 cell systems/endpoints of human toxicity in the form of a multivariate PLS (partial least squares) model of human acute lethal blood concentrations, as well as the corresponding prediction by a HeLa cell assay, were comparable to the efficiency of mouse LD50 prediction of human lethal dosage. When combined with simple toxicokinetic data (absorption of chemicals in the intestine and distribution volumes), the PLS model and the HeLa assay were able to predict human lethal dosage of the ten chemicals as accurately as the mouse LD50 value. The small number of chemicals studied to date means that general conclusions cannot be drawn from these results. Further validation of more chemicals with the in vitro methods is essential and promises to be worthwhile.

Abstract Objectives The study aimed to evaluate and compare various commercially available local anesthetic solutions. Materials and Methods A total of 150 commercially available local anesthetic cartridges of similar composition (2% lidocaine with epinephrine 1:100,000) were randomly collected and divided into 3 groups. The designations of groups were selected from their product names such that each group consisted of 60 cartridges. Group S (Septodont, France) Group M (Medicaine, Korea) and Group H (HD-Caine, Pakistan). The samples were divided into five sub-groups, each consisting of 10 cartridges from each group to investigate each parameter. Results The acquired data was statistically analyzed and compared (using SPSS version 12). Compositional analysis revealed a non-significant (P>0.05) difference when the three Groups were compared with standard lidocaine and epinephrine solutions. The mean pH values of samples from group S, M and H respectively fell within the range of pH values of commercially available solutions. Non-significant difference in EPT values of Group S and H was found when efficacy was compared (p = 0.3), however a significant difference (p < 0.01) was observed in contrast to Group M. Anti-bacterial activity was observed in all the group and a non-significant difference in cell viability values of Group S and M was found (p = 0.6), while the difference was significant in comparison to Group H. Conclusion Within the limitations of these investigations, it appears that the properties of different manufacturers fall within the recommended ranges as mentioned in literature and do not appear to be statistically different in the variables we have tested.

The objective of this study was to evaluate the efficacy of the active ingredient abamectin, either in isolation or in combination with fungicides and insecticides formulated for the industrial seed treatment, on the population of Meloidogyne javanica, tested under greenhouse and in vitro conditions. In both tests, the combination of the following treatments was assessed: abamectin; thiamethoxam; fludioxonil + metalaxyl-M + thiabendazole. Water was used as control for the in vitro assay, whereas under greenhouse conditions, controls were inoculated and not inoculated with M. javanica. The tests were conducted in a completely randomized design with six (in vitro) and ten (greenhouse) replicates. For in vitro studies the effect on hatching, motility, and mortality of juveniles of M. javanica was evaluated. Under greenhouse conditions, the soybean ‘BRS 133’ seeds were treated, and at 15, 30, and 60 days after inoculation (DAI) with M. javanica, plant measurements were recorded. The penetration of second stage juveniles (J2) was also evaluated at 15 DAI. At 30 DAI, galls, egg masses, nematodes/g of root, and final population were evaluated. At 60 DAI, the final population of nematodes in the roots was quantified. The treatments containing abamectin were the most effective in diminishing the hatching of juveniles. All treatments had an effect on nematode motility when compared to the control, and in the treatment containing only abamectin, total juvenile mortality was observed. In greenhouse conditions, at 15 DAI, the treated soybean plants had the highest root mass and shoot length, differing statistically from the inoculated control. All treatments reduced the number of nematodes per gram of root, differing from the control. At 30 DAI, treatment efficiency was observed in reducing the final population of M. javanica, particularly the treatments using abamectin, and abamectin + thiamethoxam + fludioxonil + metalaxyl-M + thiabendazole. However, at 60 DAI, the effect of the treatments on the population of M. javanica did not persist.

Yeasts can adapt to a wide range of pH fluctuations (2 to 10), while Helicobacter pylori, a facultative intracellular bacterium, can adapt to a range from pH 6 to 8. This work analyzed if H. pylori J99 can protect itself from acidic pH by entering into Candida albicans ATCC 90028. Growth curves were determined for H. pylori and C. albicans at pH 3, 4, and 7. Both microorganisms were co-incubated at the same pH values, and the presence of intra-yeast bacteria was evaluated. Intra-yeast bacteria-like bodies were detected using wet mounting, and intra-yeast binding of anti-H. pylori antibodies was detected using immunofluorescence. The presence of the H. pylori rDNA 16S gene in total DNA from yeasts was demonstrated after PCR amplification. H. pylori showed larger death percentages at pH 3 and 4 than at pH 7. On the contrary, the viability of the yeast was not affected by any of the pHs evaluated. H. pylori entered into C. albicans at all the pH values assayed but to a greater extent at unfavorable pH values (pH 3 or 4, p = 0.014 and p = 0.001, respectively). In conclusion, it is possible to suggest that H. pylori can shelter itself within C. albicans under unfavorable pH conditions.

Endophytes were isolated from 16.7% of surface-disinfested seeds and 100% of stems and roots of field-growth plants. Strains from Israel with broad-spectrum in vitro antibiosis were mainly Bacillus spp., and some were chitinolytic. Following dipping of cut cotton roots into suspensions of these strains, endophytes were detected up to 72 days later by isolation and by autoradiograms of 14C-labelled bacteria. Selected endophytes exhibited biological control potential based on significant reductions in disease severity on cotton inoculated with Rhizoctonia solani or Fusarium oxysporum f. sp. vasinfectum as well as control of Sclerotium rolfsii on bean. Neither salicylic acid nor chitinase levels increased in plants as a result of endophytic colonization, suggesting that the observed biocontrol was not accounted for by PR protein production. Some biocontrol endophytes secreted chitinolytic enzymes. Model endophytic strains inoculated into cotton stems via stem injection showed only limited movement within the stem. When introduced into stems at low concentrations, endophytes increased in population density at the injection site. After examining several experimental and semi-practical inoculation systems, seed treatment was selected as an efficient way to reintroduce most endophytes into plants.

Abstract To find an in vitro predictor of in vivoM/em&gt; semen motility prompted this study. Our objective was to evaluate semen motility for an 8-hour period immediately following a breeding soundness exam. Ejaculates from 52 Angus and 56 Charolais bulls were evaluated. Motility, morphology, scrotal circumference and pH of ejaculate were evaluated at the time of collection. Ejaculates were then extended using a one to one ratio and incubated in a water bath held at 37 degrees Celsius and evaluated hourly. Motility was evaluated hourly for 8 hours, or until motility of the sample reached zero. Data were analyzed for breed and hourly effects using the GLIMMIX procedure of SAS. There was statistical evidence for difference (P &lt; 0.0001) between breeds for motility over time. Angus ejaculates had higher pH values than Charolais ejaculates showing an association between breed and pH (6.82 vs 6.76, respectively). Primary spermatozoa abnormalities were greater (P &lt; 0.0001) for Angus bulls compared to Charolais bulls (13.33% vs. 10.91%, respectively). Scrotal circumference between breeds tended to be different (P &lt; 0.07), with Charolais bulls having a larger scrotal circumference compared to Angus bulls (38.29 vs. 38.03 centimeters, respectively). There was no difference (P &gt; 0.05) between breeds for secondary abnormalities. There was a significant interaction (P &lt; 0.01) between breed and time of motility measurement. Angus bull’s motility decreased drastically until hour 4, it then had a more gradual decrease until hour 8. Charolais bulls had a more gradual decrease in the percentage of motile sperm over time. In conclusion, there was evidence for difference between breeds for pH, primary spermatozoa abnormalities, and long-term motility, and a scrotal tendency. Understanding the effects of breed and individual biological factors may help producers adjust BSE expectations and lead to future research in long term semen motility.
