The microorganisms involved in the fermentation and spoilage of fermented cassava flour were investigated. The water samples used at the different processing sites were also investigated to determine their safety status. There was predominance ofStaphylococcus aureus,Aspergillusspp., andEscherichia coliin all samples. Coliforms were observed to be present in all of the processing water. In the fermented cassava flour, the total bacterial count ranged between4.9×106 cfu/mL from Eleso, Bakatari, and Oja Odan processing sites and8.10×106 cfu/mL in Eruku processing site. The majority of the microorganisms involved in the spoilage of “lafun” were found to beAspergillus nigerwhich ranged between4.6×105 cfu/mL in Eleso and8.1×105 cfu/mL in Kila. The control sample prepared in the laboratory had a low microbial load compared to samples collected from various sites and markets.

Background: [1, 3, 4]oxadiazolenone core containing chalcones and nucleosides were synthesized by Claisen-Schmidt condensation of a variety of benzaldehyde derivatives, obtained from oxidation of substituted 5-(3/6 substituted-4-Methylphenyl)-1, 3, 4-oxadiazole-2(3H)-one and various substituted acetophenone. The resultant chalcones were coupled with penta-O-acetylglucopyranose followed by deacetylation to get [1, 3, 4] oxadiazolenone core containing chalcones and nucleosides. Various analytical techniques viz IR, NMR, LC-MS and elemental analysis were used to confirm the structure of the synthesised compounds. The compounds were targeted against Bacillus subtilis, Staphylococcus aureus and Escherichia coli for antibacterial activity and Aspergillus flavus, Aspergillus niger and Fusarium oxysporum for antifungal activity. Methods: A mixture of Acid hydrazides (3.0 mmol) and N, Nʹ- carbonyl diimidazole (3.3 mmol) in 15 mL of dioxane was refluxed to afford substituted [1, 3, 4]-oxadiazole-2(3H)-one. The resulted [1, 3, 4]- oxadiazole-2(3H)-one (1.42 mmol) was oxidized with Chromyl chloride (1.5 mL) in 20 mL of carbon tetra chloride and condensed with acetophenones (1.42 mmol) to get chalcones 4. The equimolar ratio of obtained chalcones 4 and β -D-1,2,3,4,6- penta-O-acetylglucopyranose in presence of iodine was refluxed to get nucleosides 5. The [1, 3, 4] oxadiazolenone core containing chalcones 4 and nucleosides 5 were tested to determined minimum inhibitory concentration (MIC) value with the experimental procedure of Benson using disc-diffusion method. All compounds were tested at concentration of 5 mg/mL, 2.5 mg/mL, 1.25 mg/mL, 0.62 mg/mL, 0.31 mg/mL and 0.15 mg/mL for antifungal activity against three strains of pathogenic fungi Aspergillus flavus (A. flavus), Aspergillus niger (A. niger) and Fusarium oxysporum (F. oxysporum) and for antibacterial activity against Gram-negative bacterium: Escherichia coli (E. coli), and two Gram-positive bacteria: Staphylococcus aureus (S. aureus) and Bacillus subtilis(B. subtilis). Result: The chalcones 4 and nucleosides 5 were screened for antibacterial activity against E. coli, S. aureus and B. subtilis whereas antifungal activity against A. flavus, A. niger and F. oxysporum. Compounds 4a-t showed good antibacterial activity whereas compounds 5a-t containing glucose moiety showed better activity against fungi. The glucose moiety of compounds 5 helps to enter into the cell wall of fungi and control the cell growth. Conclusion: Chalcones 4 and nucleosides 5 incorporating [1, 3, 4] oxadiazolenone core were synthesized and characterized by various spectral techniques and elemental analysis. These compounds were evaluated for their antifungal activity against three fungi; viz. A. flavus, A. niger and F. oxysporum. In addition to this, synthesized compounds were evaluated for their antibacterial activity against gram negative bacteria E. Coli and gram positive bacteria S. aureus, B. subtilis. Compounds 4a-t showed good antibacterial activity whereas 5a-t showed better activity against fungi.

The emergence of multidrug microbial resistance is the main challenges that the modern scientists have so far been facing in the recent era. In this respect, new series of drug classes having potential to give antimicrobial effect have been synthesized. A new series of 5- substituted-1,10 b-dihydroimidazole[1,2-c]quinazoline derivatives 8a-e have been synthesized and screened for antibacterial activity and antifungal activity. Synthesized derivatives were characterized by IR, MASS and 1H-NMR spectroscopy. Synthesized compounds show good activity, which was comparable to the standard drug and it can be useful for the further clinical study. Antibacterial activity was evaluated against four different pathogenic bacterial strains like Staphylococcus aureus, Enterococcus faecalis, Escherichia coli and Pseudo-monas aeruginosa. Among the screened compounds, 8e show good antibacterial activity against Staphylococcus aureus and Escherichia coli with MIC of 50 and 100 μg/ml respectively. Antifungal activity was evaluated  against two strains of fungi. Among the synthesized derivates, compound 8c was emerged out as the potent antifungal compound against Candida albicans and Aspergillus niger with MIC of 25 μg/ml and 75μg/ml respectively. Compound 8e also shows good antifungal activity with MIC of 50 μg/ml against both Candida albicans and Aspergillus niger. The overall results of this study indicated that  synthesized quinazoline derivatives had the potential to act as an antibacterial and antifungal agent, hence further investigation is warranted.

In order to determined the antibacterial property of Apocynum venetum. Three kinds of extractants (distilled water, 75% ethanol and absolute ethanol) were used to extract the leaf, stick and skin of Apocynum venetum, respectively. Flask and plate methods were used to evaluate the antibacterial property of these extracts on four kinds of classical microbiologies, Escherichia coli, Staphylococcus aureus, Saccharomyces cerevisiae and Aspergillus niger, respectively. The results showed that the antibacterial rate of extract by 75 % ethanol was better than extracts by water or absolute ethanol on Escherichia coli and skin contained more antibacterial substance (on Escherichia coli) than others. The antibacterial rate of extract by distill water was better than extracts by 75 % ethanol or absolute ethanol on Staphylococcus aureus and stick contained more antibacterial substance (on Staphylococcus aureus) than others. Some of extracts had very weak antibacterial rate on Saccharomyces cerevisiae and all extracts had not antibacterial effect on Aspergillus niger.

The aim of this study was to determine the antibiogram of bacterial isolates from Tympanotonus fuscatus var. radula sold in markets in Nasarawa State. Nigeria. Samples of Tympanotonus fuscatus var. radula (periwinkles) were bought from soup ingredient sellers at different sale locations in Keffi, Masaka and Orange markets and were analyzed using standard bacteriological methods. The bacterial isolates were identified using morphological, cultural and biochemical techniques. The total bacteria count varied from 1.18–3.20 x 108 CFU/g for the raw samples while the total bacterial count for the boiled samples varied from 0–1.57 x 108 CFU/g. Periwinkle samples with shells from Masaka market had the highest bacterial load with a mean total bacterial count of 2.94 x 10⁸ CFU/g and mean total coliform count of 2.80 x 10⁶ CFU/g. Raw periwinkle samples with shells had a higher bacterial load than samples without shells. There was also a drastic reduction in the bacterial load in the periwinkle samples after boiling under laboratory conditions. The bacteria isolated were Bacillus spp. and Staphylococcus aureus were the Gram-positive bacteria isolated. Enterobacter spp., Escherichia coli, Salmonella spp., Pseudomonas spp., Serratia spp. and Proteus spp. The most frequently occurring gram positive bacteria was Escherichia coli with an isolation frequency of 6(24%), the least frequently occurring was Bacillus spp., 8(32)%. Antibiotic susceptibility test showed that all the gram negative organisms exhibited sensitivity to ciprofloxacin: Escherichia coli (32 mm), Enterobacter spp. (41.5 mm), Proteus spp. (40.0 mm), Salmonella spp. (37.0 mm), Serratia spp. (26.0 mm), Pseudomonas spp. (23.0 mm). All the gram negative organisms showed marked resistance to vancomycin: Escherichia coli (12.0 mm), Enterobacter spp. (10.0 mm), Proteus spp. (11.0 mm), Salmonella spp. (5.0 mm), Serratia spp. (10.0 mm) and Pseudomonas spp. (4.5 mm).
