The present study was designed to develop safer, effective, and viable cardioprotective herbal combination to control oxidative stress related cardiac ailments as new alternatives to synthetic drugs. The synergetic cardioprotective potential of herbal combination of four plantsT. arjuna(T.A.),P. nigrum(P.N),C. grandiflorus(C), andC. oxyacantha(Cr) was assessed through curative and preventive mode of treatment. In preventive mode of treatment, the cardiac injury was induced with synthetic catecholamine (salbutamol) to pretreated rabbits with the proposed herbal combination for three weeks. In curative mode of treatment, cardiotoxicity/oxidative stress was induced in rabbits with salbutamol prior to treating them with plant mixture. Cardiac marker enzymes, lipids profile, and antioxidant enzymes as biomarker of cardiotoxicity were determined in experimental animals. Rabbits administrated with mere salbutamol showed a significant increase in cardiac marker enzymes and lipid profile and decrease in antioxidant enzymes as compared to normal control indicating cardiotoxicity and myocardial cell necrosis. However, pre- and postadministration of plant mixture appreciably restored the levels of all biomarkers. Histopathological examination confirmed that the said combination was safer cardioprotective product.

Abstract Background Cisplatin (CP) is a novel drug of choice in the treatment of cancer but its major limitation is nephrotoxicity, which is dose limiting. Andrographis paniculata (AP) is a common Indian dietary component. It is well known for its medicinal properties. This present study investigated the nephroprotective effect of ethanol leaf extract of Andrographis paniculata (EEAP) on CP-induced nephrotoxicity. Methods CP was used to induce nephrotoxicity in male Wistar rats to study the effect of EEAP on renal damages using hematological parameters, biochemical parameters, histology, and immunohistochemistry studies. Results The effects of EEAP were determined by CP-induced changes in different kidney tissue on antioxidant enzymes, markers of oxidative stress, serum creatinine, and urine parameters. Administration of EEAP (200 mL/kg and 400 mg/kg orally), prior to and following a single dose CP treatment (10 mg/kg i.p), significantly mitigated the CP-induced decrease in antioxidant enzymes, and increase in markers of oxidative stress, serum creatinine, and urinary protein. On histopathological examination of the kidney tissue, there was severe glomerular degeneration and infiltration of inflammatory cells in CP only treated rats, mild glomerular degeneration, and infiltration of inflammatory cells in EEAP pre-treated rats. Furthermore, EEAP activated Nrf2 and mitigated Kim-1 pathways in CP-induced nephrotoxicity. Conclusions The results showed the protective effect of EEAP against CP-induced nephrotoxicity.

Mancozeb (MZB) is one of the fungicides used in pest control programs that might affect human health including reproductive system. The aim of this study was to demonstrate the mechanisms through which MZB induces testicular tissue damage and the probable protective effect of N-acetylcysteine (NAC), a modified amino acid, with antioxidant property, against MZB toxicity in an animal model. Male albino mice ( n = 8) were exposed to different doses of MZB (250 and 500 mg/kg/day) by oral gavage without or with NAC (200 mg/kg, twice/week) for 40 days. Sub-chronic MZB dose-dependently decreased sperm motility and count. Exposure to MZB increased lipid peroxidation and protein carbonyl, while it reduced antioxidant enzymes activities, total antioxidant capacity, and glutathione content. The histopathological examination clearly showed deleterious changes in the testicular structure. At the molecular levels, the results of quantitative real time-poly chain reaction (qRT-PCR) showed that MZB upregulated oxidative stress markers inducible nitric oxide synthase (iNOS) and NADPH oxidase 4 (NOX4) and downregulated expression of the glutathione peroxidase 1 (Gpx1) gene as one of the most important antioxidant enzymes. MZB also induced apoptosis dose-dependently in the testes as determined by the terminal dUTP nick-end labeling assay and immunoblotting. NAC administration decreased the mRNA levels of both iNOS and NOX4 with a concomitant increase in Gpx1 expression. It also significantly decreased MZB-induced oxidative stress and apoptosis. Collectively, the present study showed MZB-induced oxidative damage in testes leading to apoptosis. It revealed that antioxidants such as NAC can mitigate oxidant injury induced by the dithiocarbamate pesticides in the reproductive system.

<p>Aim: The aim of present study was to evaluate the cardioprotective efficacy of <em>Polygonum glabrum</em> on isoproterenol induced myocardial necrosis in rats. Methods: Experimental rats were treated orally with methanol extract of <em>Polygonum glabrum</em> at two doses (200 mg and 400 mg/kg) for 30 days. Isoproterenol (85 mg/kg, s.c.) was administered on 29<sup>th</sup> and 30<sup>th</sup> day to induce myocardial necrosis.  At the end of the experiment, serum cardiac marker enzymes [creatine kinase muscle brain (CK-MB), lactate dehydrogenase (LDH), serum glutamate oxaloacetate transaminase (SGOT)], serum glutamate pyruvate transaminase (SGPT) and total protein (TP) were estimated. Plasma total cholesterol (TC), triglycerides (TG), high density lipoprotein (HDL), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) levels were also recorded. Further, antioxidant parameters viz catalase (CAT), superoxide dismutase (SOD),  glutathione (GSH), glutathione peroxidase (GPx), glutathione reductase (GRD) and malondialdehyde (MDA) levels were evaluated in heart tissue homogenate. Results: The results of the study indicated that, methanol extract of <em>Polygonum glabrum</em> showed greater cardioprotection by restoring the cardiac marker enzymes and attenuated the level of plasma lipid profiles along with an increase in HDL. Additionally, level of myocardial antioxidants significantly increased along with a reduction in the content of malondialdehyde. The cardioprotective effect was compared with propranolol     (10 mg/kg, oral) which was used as the standard. Histopathological findings revealed a decrease in the degree of necrosis and inflammation following pretreatment with <em>Polygonum glabrum</em>. Conclusion: The present investigation indicates that <em>Polygonum glabrum</em> could protect myocardium from isoproterenol induced necrosis.</p>

Background. Anshen Buxin Liuwei pill (ABLP) is a Mongolian medicinal formula which has a therapeutic effect on the symptoms such as coronary heart disease, angina pectoris, arrhythmia, depression and irritability, palpitation, and short breath. However, its bioactivity against cardiac injury remains unclear. Methods. The protective effect of ABLP was evaluated using H9c2 cells. Cell viability, intracellular Ca2+, reactive oxidative indices, and mitochondrial membrane potential (∆ψ) were assessed, respectively. The mRNA levels of Ca2+ channel-related genes (DHPR, RyR2, and SCN5A) and oxidative stress-related genes (Keap1, Nrf2, and HO-1) were measured by RT-PCR. Results. 0.5–50 μg/mL ABLP could significantly decrease H2O2-induced cell injury by suppressing cell necrosis/apoptosis and excess oxidative stress, ameliorating the collapse of ∆ψ, and reducing intracellular Ca2+ concentration. Furthermore, 0.5–50 μg/mL ABLP reversed H2O2-induced imbalance in the mRNA levels of DHPR, RyR2, SCN5A, Keap1, Nrf2, and HO-1 gene in H9c2 cells, which further illustrate the mechanism. Conclusion. ABLP provided protective and therapeutic benefits against H2O2-induced H9c2 cell injury, indicating that this formula can effectively treat coronary disease. In addition, the present study also provides an in-depth understanding of the pharmacological functions of ABLP, which may lead to further successful applications of Mongolian medicine.

: Diabetes is a common metabolic disease that increases the risk of cardiovascular disease. It seems that the reduction of oxidative stress or increasing antioxidant levels improves diabetic cardiomyopathy. Antioxidant effects of immunomodulatory drug (IMODTM) and also beneficial influences of exercise on diabetic complications have been shown. The present study examined the effects of IMODTM and exercise on cardiac oxidative stress and antioxidants in diabetes. For this purpose, 64 rats were divided into 8 groups: control (C), exercise (E), IMODTM (20 mg/kg) (I), exercise plus IMODTM (E + I), diabetes (D), diabetic rats treated with exercise (D + E), diabetic rats treated with IMODTM (D + I), and diabetic rats treated with exercise plus IMODTM (D + E + I). Treatments with exercise and/or IMODTM were performed for 8 weeks. Type 1 diabetes was induced by intraperitoneal injection of 60 mg/kg streptozotocin. After the treatment period, all rats were anesthetized, and blood and heart samples were gathered for measurement of malondialdehyde (MDA) as an oxidative stress marker, lactate dehydrogenase (LDH) as a cardiac injury marker, total antioxidant capacity (TAC), and superoxide dismutase (SOD) as well as glutathione peroxidase (GPx) as antioxidant enzymes. The present study, for the first time, showed that IMODTM alone or in combination with exercise had positive effects on alleviating hyperglycemia, MDA, and LDH along with elevation of antioxidant enzymes activities in type 1 diabetic rats.

The present study was designed to investigate the cardioprotective effects of Malaysian Tualang honey against isoproterenol- (ISO-) induced myocardial infarction (MI) in rats by investigating changes in the levels of cardiac marker enzymes, cardiac troponin I (cTnI), triglycerides (TG), total cholesterol (TC), lipid peroxidation (LPO) products, and antioxidant defense system combined with histopathological examination. Male albino Wistar rats (n= 40) were pretreated orally with Tualang honey (3 g/kg/day) for 45 days. Subcutaneous injection of ISO (85 mg/kg in saline) for two consecutive days caused a significant increase in serum cardiac marker enzymes (creatine kinase-MB (CK-MB), lactate dehydrogenase (LDH), and aspartate transaminase (AST)), cTnI, serum TC, and TG levels. In addition, ISO-induced myocardial injury was confirmed by a significant increase in heart lipid peroxidation (LPO) products (TBARS) and a significant decrease in antioxidant enzymes (SOD, GPx, GRx, and GST). Pretreatment of ischemic rats with Tualang honey conferred significant protective effects on all of the investigated biochemical parameters. The biochemical findings were further confirmed by histopathological examination in both Tualang-honey-pretreated and ISO-treated hearts. The present study demonstrates that Tualang honey confers cardioprotective effects on ISO-induced oxidative stress by contributing to endogenous antioxidant enzyme activity via inhibition of lipid peroxidation.

<p>The present study was executed to evaluate the myocardial protective effect of methanol extract of <em>Bougainvillea glabra </em>against isoproterenol induced myocardial necrosis in rats. Myocardial necrosis was induced by subcutaneous injection of isoproterenol (85mg/kg body weight) on 29<sup>th</sup> and 30<sup>th</sup> day at an interval of 24 hours. Myocardial necrosis was evident from the changes of marker enzymes in serum, plasma and heart tissue. The activities of serum cardiac marker enzymes such as lactate dehydrogenase (LDH), creatine kinase myoglobin (CK-MB), serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), triglycerides (TG), total cholesterol (TC), high density lipoproteins (HDL), low density lipoproteins (LDL), very low density lipoproteins (VLDL) and total protein (TP) were estimated. In addition, plasma TBARS and plasma LDH levels were also recorded. Antioxidant parameters viz catalase (CAT), superoxide dismutase (SOD), glutathione (GSH), glutathione peroxidase (GPx) and malondialdehyde (MDA) levels were performed in heart tissue homogenate. The outcome of the study indicated that, pretreatment with methanol extract of <em>Bougainvillea glabra</em> to isoproterenol induced rats significantly prevented the altered serum cardiac marker enzymes, plasma levels and antioxidant parameters to near normal status. The cardioprotective effect was compared with propranolol (10 mg/kg, oral) which was used as the standard. Histopathological findings exposed a reduced degree of necrosis and inflammation succeeding pretreatment with <em>Bougainvillea glabra.</em>Based on these results, it was suggested that methanol extract of <em>Bougainvillea glabra</em> prevents myocardial necrosis and oxidative stress induced by isoproterenol.</p>

Curcumin has anti-inflammatory, antioxidative, anticarcinogenic, and cardiovascular protective effects. Our study is aimed at evaluating the effects of pretreatment with curcumin nanoparticles (CCNP) compared to conventional curcumin (CC) on isoproterenol (ISO) induced myocardial infarction (MI) in rats. Fifty-six Wistar-Bratislava white rats were randomly divided into eight groups of seven rats each. Curcumin and curcumin nanoparticles were given by gavage in three different doses (100 mg/kg body weight (bw), 150 mg/kg bw, and 200 mg/kg bw) for 15 days. The MI was induced on day 13 using 100 mg/kg bw ISO administered twice, with the second dose 24 h after the initial dose. The blood samples were taken 24 h after the last dose of ISO. The antioxidant, anti-inflammatory, and cardioprotective effects were evaluated in all groups. All doses of CC and CCNP offered a cardioprotective effect by preventing creatine kinase-MB leakage from cardiomyocytes, with the best result for CCNP. All the oxidative stress parameters were significantly improved after CCNP compared to CC pretreatment. CCNP was more efficient than CC in limiting the increase in inflammatory cytokine levels (such as TNF-α, IL-6, IL-1α, IL-1β, MCP-1, and RANTES) after MI. MMP-2 and MMP-9 levels decreased more after pretreatment with CCNP than with CC. CCNP better prevented myocardial necrosis and reduced interstitial edema and neutrophil infiltration than CC, on histopathological examination. Therefore, improving the bioactivity of curcumin by nanotechnology may help limit cardiac injury after myocardial infarction.

Endometriosis is described as a gynecological disorder characterized by the presence of endometrial tissue outside the uterus; extensively explored because of its increasing incidency, with an indubitable diagnostic only after invasive surgery, with no efficient treatment, it has still many aspects to be elucidated. A growing body of facts sustain oxidative stress as a crucial factor between the numerous incriminated factors implicated in endometriosis ethiopathogeny. Reactive oxygen species(ROS) act to decline reproductive function. Our study intends to determine if an experimental model of endometriosis may be useful to assess the impact of oxidative stress on endometrial cells; we have used a murine model of 18 adult Wistar female rats. A fragment from their left uterine horn was implanted in the abdominal wall. After 4 weeks, a laparatomy was performed, 5 endometrial implants were removed, followed by biochemical tissue assay of superoxide dismutase(SOD) and catalase(CAT). At the end of the experiment, the rats were sacrificed, the implants were removed for histopathological exam and biochemical assay of antioxidant enzymes. The results revealed decreased levels of antioxidant enzymes, pointing on significant oxidative stress involvement.
