The young leaves ofVernonia amygdalinaare often utilized as vegetable and for medicinal purpose compared to the old leaves. This study was designed to evaluate and compare the antidiabetic effects between ethanolic leaf extracts of old and youngV. amygdalinaon streptozotocin (STZ) induced diabetic rat for four weeks. Preliminary screening of both young and old ethanolic extracts revealed the presence of the same phytochemicals except flavonoids which was only present in the oldV. amygdalina. Difference in antioxidant power between the young and old leaf extracts was statistically significant (p<0.05). Both leaf extracts produced a significant (p<0.05) antihyperglycaemic effect. Also results from treated rats revealed increasing effect in some haematological parameters. Similarly, the higher dose (300 mg/kg) of both extracts significantly (p<0.05) reduced serum ALT, AST, and ALP levels as compared to the diabetic control rats. Results also showed significant (p<0.05) decrease in LDL-C and VLDL-C in the extract-treated rats with a corresponding increase in HDL-C, as compared to the diabetic control rats. Moreover histopathological analysis revealed ameliorative effect of pathological insults induced by the STZ in the pancreas, liver, and spleen, most significantly the regeneration of the beta cells of the islets of Langerhans in treated rats.

Aim: The study evaluated the effects of the combined extracts of Vernonia amygdalina (VA) and Gongronema latifolium (GL) on pancreatic GLUT 2 expression and caspase 3 activity in streptozotocin (STZ, 45 mg/Kg)-induced diabetic rats. Study Design: Fifteen Albino rats were used for the study and were placed in 3 groups of 5 rats each: A - normal control, B – Diabetic control and C – experimental group. Place and Duration of Study: The study was carried out in the department of Anatomy, University of Calabar. Duration:  6 months. Methodology:  Half of the diabetic rats were treated with VA+GL (400mg/kg, ratio 1:1, DE group) for 28 days, while the other half was untreated and served as diabetic control (DC). Normal control (NC) rats were untreated. After 28 days, the rats were sacrificed and their blood glucose, serum GLUT 2 and caspase 3 activity were measured.  Histochemical evaluation of the pancreas was also carried out. Results: Blood glucose concentrations for the 3 groups were 60.31±7.28, 257.00±4.43, and 116.60±10.11 mg/dl for NC, DC and DE respectively. This represented a 4-fold increase in the DC compared with NC and a significant amelioration in the extract-treated DE group compared with DC group. Serum GLUT 2 concentrations were 70 ng/ml in NC, dropped to 8 ng/ml (p<0.05) in the DC and recovered to 20 ng/ml in DE (p<0.05). Serum caspase was 3.2 ng/ml for NC, increased to 8.5 ng/ml in DC (p<0.05) and reduced to 1.8 ng/ml in DE (p<0.05). The histology of the pancreas showed distorted, degenerated and shrunken β-cells mass in DC compared with NC and DE groups. The DE group showed clear signs of regeneration of the islet cells which was corroborated by positive Feulgen’s reaction compared with the DC group. Conclusion: The data suggests that the combined VA+GL extract has the potential to effectively reverse pancreatic damage in diabetes.

Background. Increased activity of aldose reductase (AR) is one of the mechanisms involved in the development of diabetic complications. Inhibiting AR can be a target to prevent diabetes complications. This study is aimed at evaluating the effect of cyclohexane (CH) and ethanol extracts (ET) of walnut leaves on AR activity in the lens and testis of diabetic rats. Methods. Fifty-six male rats classified into seven groups as control and treatment groups and treated for 30 days. The treatment groups were treated with different concentrations of ET and CH. The diabetic control (DC) group was exposed to streptozotocin. AR activity was measured in the lens and testis. The expression of AR in the testis was evaluated by the immunohistochemistry method. Results. Both extracts significantly reduced the AR activity (ng/mg of tissue protein) in the testis (0.034±0.004, 0.038±0.010, and 0.040±0.007 in the treatment groups vs. 0.075±0.007 in the DC group) and lens (1.66±0.09, 2.70±0.47, and 1.77±0.20 in the treatment groups vs. 6.29±0.48 in the DC group) of the treatment group compared to those of the DC group (P<0.05). AR expression in the testes of the treatment groups was decreased compared with that of the DC group (P<0.0001). Conclusion. Walnut leaf extracts can reduce the activity and localization of AR in the testes and its activity in the lens of diabetic rats.

Diabetes mellitus (DM) is a serious worldwide health threat since the number of people with DM is forecasted to grow annually. Thus, effective and affordable treatment is urgently needed. Our previous studies used n-hexane and hydroethanolic root extracts of Ruellia tuberosa L. which significantly affected diabetic rats. In this study, aqueous R. tuberosa L. root extracts were used as treatments for the diabetic rat model and their effects were evaluated. Diabetes was generated by the administration of streptozotocin (STZ) at 20 mg/kg within 5 sequential days. Male Wistar rats were orally treated with the extracts and standard drug (metformin 200 mg/kg) and vehicle every day for 4 weeks. Hypoglycemic effects were assessed for normal, diabetic control, standard, and extract-treated groups. Histopathology was also carried out for the pancreatic, hepatic, and kidney tissues. The progression of diabetes was considerably diminished after extract treatment. In treated rats, the highest dose of extract induced a decline in blood glucose and serum malondialdehyde (MDA) levels at 25% and 35%, respectively. Furthermore, aqueous extract of R. tuberosa L. treatment decreased MDA levels in the pancreas by 12%. Histologic examination of the organ tissues of diabetic rats showed deteriorating alterations. After treatment, histopathological damages to the tissues and cells were reversed. The results of the experiments recommend that aqueous extract of R. tuberosa L. has antidiabetic effects on STZ-induced diabetic rats; nevertheless, a higher dose of the aqueous extracts is needed to achieve more significant results.

Objective: To assess the antioxidant activity from another part of the plant, in this study, leaf extracts in n-hexane were fractionated.Methods: Ten fractions were obtained and tested in vitro for antioxidant activity using two methods, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferricreducing antioxidant power (FRAP), to identify the most active fraction.Results: The IC50 of the most active fraction was 36.24 μg/mL using the DPPH method, and the EC50 was 39.54 μg/mL using the FRAP method. Themost active fraction was also shown to contain terpenoids.Conclusion: The most active fraction of an n-hexane extract of the leaves of Gacinia bancana Miq., which was tested by both DPPH and FRAP methodshad antioxidant activities with IC50 and EC50 values of 36.2482 μg/mL and 39.5442 μg/mL, respectively. Phytochemical screening showed that activefraction contains terpenoids.

The study aimed at evaluating the antioxidant profile of a medicinal epiphyte Viscum album L. harvested from three tree species, namely, Populus ciliata L, Ulmus villosa L., and Juglans regia L. The crude extracts were obtained with ethanol, methanol, and water and were evaluated for the total phenol content (TPC), total flavonoid content (TFC), and antioxidant activities using total reducing power (TRP), ferric reducing antioxidant power (FRAP), 1, 1-diphenyl 1-2-picryl-hydrazyl (DPPH), superoxide radical scavenging (SOR), and hydroxyl radical scavenging (•OH) assays. Our results showed that crude leaf extracts of plants harvested from the host Juglans regia L. exhibited higher yields of phytochemical constituents and noticeable antioxidative properties. The ethanolic leaf samples reported the highest phenols (13.46 ± 0.87 mg/g), flavonoids (2.38 ± 0.04 mg/g), FRAP (500.63 ± 12.58 μM Fe II/g DW), and DPPH (87.26% ± 0.30 mg/mL). Moreover, the highest values for TRP (4.24 ± 0.26 μg/mL), SOR (89.79% ± 0.73 mg/mL), and OH (67.16% ± 1.15 mg/mL) were obtained from aqueous leaf extracts. Further, Pearson correlation was used for quantifying the relationship between TPC, TFC, and antioxidant (FRAP, DPPH, SOR, OH) activities in Viscum album L. compared to their hosts. It was revealed that the epiphyte showed variation with the type of host plant and extracting solvent.

Introduction: Bridelia ferruginea is a plant known for its antidiabetic properties. However, few studies on leaf extracts have induced anti-hyperglycemic activity on normal mice subjected to carbohydrate overload. The current study was designed to assess the effect of the leaf extracts’ fraction on fructose-induced diabetic mice. Methods: The in vitro ferric-reducing antioxidant power (FRAP) assay were carried out and the condensed tannins quantified. The vanillin-HCl method was used to characterize the condensed tannins. The antidiabetic effect on fructose-induced diabetic mice was evaluated for 28 days using a fructose-enriched fat diet approach. Results: The fraction confirmed the antioxidant activity with a reducing power of 800 μg/mL comparable to ascorbic acid at 200 μg/mL. The condensed tannins were estimated at 79.6 ± 3.4 mg catechin equivalent per gram of sample. Significant decreases in blood sugar levels of 6.25% at the 7th day, 11.04% at the 14th day, 12.61% at the 21th day, and 11.35% at the 28th day were obtained in mice treated with the extract dose of 200 mg/kg of body weight, compared to the positive control group. The decreases of 37.11% of triglycerides and 40.16% of total cholesterol were also obtained. Conclusion: The investigated fraction showed notable antidiabetic activity and might be a good candidate in the treatment of diabetes.

Tea was brewed from young leaves of 4 weeks old Thai upland black waxy rice plants of the cultivar ‘Damgatondam’, which has a noticeable fragrant smell from both seeds and leaves. The plants were harvested about 1 cm above soil level, washed, and cut into pieces, about 1 cm long. These pieces were then steamed for 1 min in a steamer rack, withering for 5min by spreading them out in a thin layer, pan-roasted at 70°C for 30 min, and finally dried at 70°C for 90 min. They were either kept as pieces of leaves or ground into a powder, and both were used for brewing tea by infusion in water at 4oC, room temperature (about 25oC), for 1, 3, 5, 7, 9, 11, or 15 h and brewing at 70oC or 98oC for 1, 3, 5, 7, 9, 11 or 15 min. Directly after brewing, the tea was stored at -25oC awaiting analysis. Each sample was analyzed for total phenolic content (TPC) and for antioxidant activity, by ferric reducing antioxidant power (FRAP), 2,2 diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazo line-6-sulphonic acid) (ABTS) assays. Results showed that tea brewed from ground leaves had higher antioxidant activity than tea brewed from ungrounded leaves. The highest level of extraction was at 98oC for 15 min, where the TPC was 62.79 mg GAE/100g, antioxidant activity was 278.04 for DPPH, 195.38 for ABTS and 1150.13 mg TE/100g for FRAP. Samples extracted at 4oC or room temperature also had high levels of TPC and antioxidant activity with the maximum extraction at room temperature for 15 h; where TPC was 46.76 mg GAE/100g, and antioxidant activities were 152.72 for DPPH, 140.42 for ABTS and 950.32 mg TE/100 g for FRAP. Therefore, the level of extraction for tea brewed in all four conditions depended on both temperature and brewing time. Also, extraction of high levels of bioactive compounds depended on large surface areas of the leaves being in contact with water.

The aim of this study was to know the hypoglycaemic and hypolipidaemic activities of water extract of bungur (Lagerstroemia speciosa [L.] Pers.) leaves in alloxan-induced diabetic rat. Phytochemical studies was conducted to determinate the tannin-contents of the water extract (WE). Male Wistar rats (150- 180 9 BW) were used in this study. The rats were divided into six groups: nonnal control; diabetic control; glibenclamide; WE 0.1 g/200 9 BW; WE 0.2 g/200 9 BW; and WE 0.5 g/200 9 BW. Dried leaves of bungur extracted with boiling water (50 g/L) for 30 minutes. The extract administered orally. Fasting blood glucose, triglyceride, and total cholesterol were determined at 0, 2, 4, 6 hours after treatment. Total phenol, ellagitannin, gallotannin, and condensed-tannin were determined from the WE. The results showed that WE exhibited hypoglycaemic activity at doses 0.2 g/200 9 BW and 0.5 g/200 9 BW. The last dose showed similar hypoglycaemic activity compared with glibenclamide. All doses showed the hypolipidaemic activity higher than glibenclamide. Phytochemical studies showed that the WE containing total phenol 0.025%; ellagitannin 0.011%; gallotannin 0.0199%; and condensed-tannin 0.0167%.

Diabetes mellitus is prevalent in many countries of the world, affecting all ages both in developing and developed nations. The use of plants as remedies or preventive therapies has increased over the years. The study investigated the biochemical changes caused by  combined leaf extracts of Vernonia amygdalina (bitter leaf) and Gnetum africanum (okazi leaf) on alloxan induced diabetic wistar rats.Aqueous extracts of the leaves were prepared using the conventional method.Forty male wistar rats weighing 150-180g were  grouped into eight (five rats each). Group 1 was the normal control while diabetes was induced using alloxan (160mg/ kg)in groups 2-8.Group 2 received no treatment while groups 3-7 received varied ratios of the extracts at (BI/OK|10:90%), (BI/OK|30:70%),  (BI/OK|50:50%), (BI/OK|70:30%) and (BI/OK|90:10%). Group 8 was the diabetic control treated with the standard diabetic drug (Metformin). The animals were weighed and blood glucose was determined at 7-day intervals. They were sacrificed on the 28th day and blood samples collected for serum protein, serum electrolyte, urea, creatinine, liver enzymes and markers of oxidative stress analyses.  The results showed steady increase in the body weights (g) of the rats with (BI/OK|70:30)% treated group showing the highest increase (175.40±1.28). The fasting blood sugar (mg/dl) showed timedependent reduction in all the treated diabeti groups with (BI/OK|90:10)% having the highest (56.20±1.65) reduction. There was a significant increase (p<0.05) in total blood protein concentration (g/dl) in all the treated groups. The results of this study showed time and ratio dependent effect on the parameters measured. Since the two plants are staple vegetables in some countries, their utilization particularly in appropriate combinations should be encouraged.
