Introduction: Bridelia ferruginea is a plant known for its antidiabetic properties. However, few studies on leaf extracts have induced anti-hyperglycemic activity on normal mice subjected to carbohydrate overload. The current study was designed to assess the effect of the leaf extracts’ fraction on fructose-induced diabetic mice. Methods: The in vitro ferric-reducing antioxidant power (FRAP) assay were carried out and the condensed tannins quantified. The vanillin-HCl method was used to characterize the condensed tannins. The antidiabetic effect on fructose-induced diabetic mice was evaluated for 28 days using a fructose-enriched fat diet approach. Results: The fraction confirmed the antioxidant activity with a reducing power of 800 μg/mL comparable to ascorbic acid at 200 μg/mL. The condensed tannins were estimated at 79.6 ± 3.4 mg catechin equivalent per gram of sample. Significant decreases in blood sugar levels of 6.25% at the 7th day, 11.04% at the 14th day, 12.61% at the 21th day, and 11.35% at the 28th day were obtained in mice treated with the extract dose of 200 mg/kg of body weight, compared to the positive control group. The decreases of 37.11% of triglycerides and 40.16% of total cholesterol were also obtained. Conclusion: The investigated fraction showed notable antidiabetic activity and might be a good candidate in the treatment of diabetes.

Like other members of the Colchicum genus, C. szovitsii subsp. szovitsii is also of medicinal importance in Turkish traditional medicine. However, its biological properties have not been fully investigated. Herein, we focused on the evaluation of the in vitro antioxidant and enzyme inhibitory effects of flower, root and leaf extracts, obtained using different extraction methods. In addition, a comprehensive (poly)-phenolic and alkaloid profiling of the different extracts was undertaken. In this regard, ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS) allowed us to putatively annotate 195 polyphenols and 87 alkaloids. The most abundant polyphenols were flavonoids (83 compounds), whilst colchicine and 2-demethylcolchicine were some of the most widespread alkaloids in each extract analyzed. However, our findings showed that C. szovitsii leaf extracts were a superior source of both total polyphenols and total alkaloids (being, on average 24.00 and 2.50 mg/g, respectively). Overall, methanolic leaf extracts showed the highest (p < 0.05) ferric reducing antioxidant power (FRAP) reducing power (on average 109.52 mgTE/g) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging (on average 90.98 mgTE/g). Interestingly, each C. szovitsii methanolic extract was more active than the water extracts when considering enzymatic inhibition such as against tyrosinase, glucosidase, and acetylcholinesterase (AChE). Strong correlations (p < 0.01) were also observed between polyphenols/alkaloids and the biological activities determined. Multivariate statistics based on supervised orthogonal projections to latent structures discriminant analysis (OPLS-DA) allowed for the detection of those compounds most affected by the different extraction methods. Therefore, this is the first detailed evidence showing that C. szovitsii subsp. szovitsii might provide beneficial effects against oxidative stress and the associated chronic diseases. Nevertheless, the detailed mechanisms of action need to be further investigated.

In this work, the phytochemical profile and the biological properties of Colchicum triphyllum (an unexplored Turkish cultivar belonging to Colchicaceae) have been comprehensively investigated for the first time. Herein, we focused on the evaluation of the in vitro antioxidant and enzyme inhibitory effects of flower, tuber, and leaf extracts, obtained using different extraction methods, namely maceration (both aqueous and methanolic), infusion, and Soxhlet. Besides, the complete phenolic and alkaloid untargeted metabolomic profiling of the different extracts was investigated. In this regard, ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS) allowed us to putatively annotate 285 compounds when considering the different matrix extracts, including mainly alkaloids, flavonoids, lignans, phenolic acids, and tyrosol equivalents. The most abundant polyphenols were flavonoids (119 compounds), while colchicine, demecolcine, and lumicolchicine isomers were some of the most widespread alkaloids in each extract analyzed. In addition, our findings showed that C. triphyllum tuber extracts were a superior source of both total alkaloids and total polyphenols, being on average 2.89 and 10.41 mg/g, respectively. Multivariate statistics following metabolomics allowed for the detection of those compounds most affected by the different extraction methods. Overall, C. triphyllum leaf extracts showed a strong in vitro antioxidant capacity, in terms of cupric reducing antioxidant power (CUPRAC; on average 96.45 mg Trolox Equivalents (TE)/g) and ferric reducing antioxidant power (FRAP) reducing power (on average 66.86 mg TE/g). Interestingly, each C. triphyllum methanolic extract analyzed (i.e., from tuber, leaf, and flower) was active against the tyrosinase in terms of inhibition, recording the higher values for methanolic macerated leaves (i.e., 125.78 mg kojic acid equivalent (KAE)/g). On the other hand, moderate inhibitory activities were observed against AChE and α-amylase. Strong correlations (p < 0.01) were also observed between the phytochemical profiles and the biological activities determined. Therefore, our findings highlighted, for the first time, the potential of C. triphhyllum extracts in food and pharmaceutical applications.

Aims: Presently the medicinal world is rapidly turning more on the therapeutic health benefits of natural product and medicinal plants in the management of major crucial disease and their complications. Medicinal plant, Stephania japonica has been studied for exploring antidiabetic potentiality as an alternative source of medicine against the global threat of Diabetes mellitus (DM). Methods: The extraction of S. japonica leaf was carried out by acetone and ethanol. Phytochemical screening and quantitative analysis of S. japonica leaf extracts were evaluated through chemically forming characterized color formation and calibration method respectively, by using standard reference substances (ascorbic acid, gallic acid, and quercetin) to assess the probable compounds present in the extract. Anti-diabetic potentiality of highest phytochemicals containing two extracts were investigated in in vitro as a ⍺-amylase inhibitors and in vivo through alloxan-induced Swiss albino diabetes mice model. Results: Alkaloids, carbohydrates, steroids, flavonoids, resins, saponins, tannins and coumarins were present in the leaf extracts. The estimated amount of total phenolics, flavonoids, flavonols and proanthrocyanidins contents of acetone and ethanol extract were 92.12±0.64 and 56.54±1.05 mg of gallic acid equivalent (GAE)/gm of dry extract, 66.02±1.42 and 46.17±0.54 mg of catechin equivalent (CAE)/gm of dry extract, 7.05±0.108 and 5.26±0.083 mg of quercetin equivalent (QUE)/gm of dry extract, 35.19±0.67 and 9.55±1.11 mg CAE/gm of dry extract, respectively. In 3, 5-dinitrosalicylic acid method, acetone and ethanol extract showed α-amylase inhibition of 51.02% and 46.62%, respectively at the concentration of 1000 µg/mL whereas in starch iodine color assay, acetone and ethanol extract showed inhibition of 57.32% and 52.12%, respectively at the concentration of 800 µg/mL. In contrast, both of the leaf extracts significantly (p<0.05 to p<0.001) improved the lipid profile parameters, blood glucose level and serum hepatic marker proteins in alloxan-induced diabetic mice. Conclusion: The present study strongly concluded that S. japonica leaf extracts process potent antidiabetic potentiality that might be significance for the management of diabetes and its complications.

The undesirable adverse effects of present available synthetic drugs endorse the modern medicine to search the superior choice for the treatment of metabolic diseases. Herbal medicine turns out to be a hopeful therapy for the effective treatment of diabetes in foreseeable future. In an effort to render a scientific evidence for the antidiabetic potential of Annona reticulata L., the present research, with the objective to evaluate the ability of A. reticulata leaf extracts on antihyperglycemic property under in vitro using yeast cell model was performed. Besides, as the diabetes and its complications are highly associated with the oxidative stress, the current study was also focussed on antioxidant property of A. reticulata leaf extracts. The scavenging ability of plant extracts on free radical 2, 2 – diphenyl- 1- picrylhydrazyl (DPPH), ferric reducing power assay and total antioxidant activity were performed to establish the antioxidant potential of A. reticulata leaf extracts. The in vitro antidiabetic ability of A. reticulata leaves was evaluated by glucose uptake method using yeast cell model. Among the four chosen solvent extracts- aqueous, methanol, ethyl acetate and n-hexane, methanol extract (MeE) exhibited high antioxidant potential. MeE exhibited 62.28% of DPPH inhibition at 200µg/ml, with total antioxidant activity 164.72 ± 2.63µg/ml and higher absorbance in reducing power assay (1.15 ± 0.03).  In vitro antidiabetic activity by glucose uptake of yeast cell assay was evaluated and observed dose dependant rise in % of glucose uptake in methanol, ethylacetate and aqueous extracts of A. reticulata. MeE showed 48.55% of glucose uptake at 500µg/ml concentration. Hence the present study could be concluded as A.reticulata leaves possess potent antioxidant activity and antidiabetic activity under in vitro conditions. With the outcome of the present initial study, research work could be extended further; thereby the exact pharmacological action of the plant compounds could be discovered.

This study explored the antioxidant and immunomodulatory potential of ethnomedicinally valuable species, namely, Arisaema jacquemontiiof north-western Himalayan region. The tubers, leaves, and fruits of this plant were subjected to extraction using different solvents.In vitroantioxidant studies were performed in terms of chelation power on ferrous ions and FRAP assay. The crude methanol extract of leaves was found to harbour better chelating capacity (58% at 100 μg/mL) and reducing power (FRAP value1085.4±0.11 μMFe3+/g dry wt.) than all the other extracts. The crude methanol extract was thus further partitioned with solvents to yield five fractions. Antioxidant study of fractions suggested that the methanol fraction possessed significant chelation capacity (49.7% at 100 μg/mL) and reducing power with FRAP value of 1435.4 μM/g dry wt. The fractions were also studied for immune modulating potential where it was observed that hexane fraction had significant suppressive effect on mitogen induced T-cell and B-cell proliferation and remarkable stimulating effect on humoral response by 141% and on DTH response by 168% in immune suppressed mice as compared to the controls. Therefore, it can be concluded thatA. jacquemontiileaves hold considerable antioxidant and immunomodulating potential and they can be explored further for the identification of their chemical composition for a better understanding of their biological activities.

Plants are well known in traditional herbal medicines for their hypoglycemic and hypolipidemic activities and are often used due to their accessibility, affordability, and corollary effects. Leptopus cordifolius has been reported to control diabetes in folkloric medicine, but no known scientific research has been conducted to assess the plausibility of this assertion. Therefore, the current study is aimed to investigate the antidiabetic and hypolipidemic effects of Leptopus cordifolius leaves in alloxan-induced diabetic mice. The antidiabetic and antihyperlipidemic evaluation was conducted in Swiss albino mice at doses of 150–250°mg/kg for 15°days. The blood glucose, total cholesterol, triglyceride, LDL, HDL, creatinine, ALP, SGPT, and SGOT levels were estimated according to standard procedures. Phytochemicals of leaves were analyzed using GC-MS analysis. Enzymatic antioxidant activity of the plant was investigated spectrophotometrically by carrying out superoxide dismutase, peroxidase, and catalase assays. The membrane stabilization potential of L. cordifolius leaf extracts was carried out using an in vitro haemolytic assay. The results revealed a dose response effect with the methanolic extract of L. cordifolius which had significant antihyperglycemic effects at 150–250°mg/kg in alloxan treated mice, although less than the positive control (glibenclamide). Hyperlipidemic activity was significant at 250 mg/kg. The biochemical parameters, such as total cholesterol, triglyceride, LDL, HDL, creatinine, ALP, SGPT, and SGOT, were significantly improved (p &lt; 0.01) by the methanolic extract of 250 mg/kg compared to the diabetic group. Treatment for 15 days showed significant elevation (p &lt; 0.01) of antioxidant enzymes. GC-MS analysis provided tentative identifications of 52 compounds in the methanolic extract of L. cordifolius, of which 12 compounds have reported antidiabetic activity. In conclusion, methanolic extract of L. cordifolius of 150 and 250°mg/kg body weight showed significant antidiabetic and antihyperlipidemic activities in alloxan-induced diabetic mice and, with further work, has the potential to be used to manage blood glucose and cholesterol levels.

Background: Quercus serrata Murray leaves have been used traditionally in the treatment of diabetes, dysmenorrhoea, inflammation and urinary tract infection. So, far no study had been reported on the toxicological profile and antioxidant properties of the plant. Objective: The present study was aimed to investigate the in-vivo toxicological profile and in-vitro antioxidant activities of the methanolic extract of standardized Quercus serrata leaves. Methods: Per-oral sub-acute toxicity study was performed in rats using three dose levels (200, 400 and 800 mg/kg b.w.) of the extract for 28-days. Control group received gum acacia suspended in water. Bodyweight was measured weekly. Biochemical parameters were analysed using the serum, the blood-cell count was done using whole blood. Pathological changes were also checked in highly perfused tissues. Further, in-vitro reducing power assay, nitric oxide scavenging assay, DPPH free-radical scavenging assay were performed to check the antioxidant activity of the extract. Results: There were no significant alterations in the blood-cell count and biochemical parameters analysed in the treatment group when compared with the normal control. Histopathology study of liver, kidney, pancreas, heart and brain revealed normal cellular architecture in the treatment groups alike the control group animals. Quercus serrata also showed a significant reduction of DPPH with IC50 4.48±0.254 µg/mL, in-vitro reducing power activity with IC50121.65±0.320 µg/mL and nitric oxide scavenging activity IC50 106.43±0.338 µg/mL. Conclusion: The above study showed that standardized methanolic extract of Quercus serrata leaves was safe after subacute oral administration in rats and has good antioxidant potential.

Objective: To assess the antioxidant activity from another part of the plant, in this study, leaf extracts in n-hexane were fractionated.Methods: Ten fractions were obtained and tested in vitro for antioxidant activity using two methods, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferricreducing antioxidant power (FRAP), to identify the most active fraction.Results: The IC50 of the most active fraction was 36.24 μg/mL using the DPPH method, and the EC50 was 39.54 μg/mL using the FRAP method. Themost active fraction was also shown to contain terpenoids.Conclusion: The most active fraction of an n-hexane extract of the leaves of Gacinia bancana Miq., which was tested by both DPPH and FRAP methodshad antioxidant activities with IC50 and EC50 values of 36.2482 μg/mL and 39.5442 μg/mL, respectively. Phytochemical screening showed that activefraction contains terpenoids.

The study aimed at evaluating the antioxidant profile of a medicinal epiphyte Viscum album L. harvested from three tree species, namely, Populus ciliata L, Ulmus villosa L., and Juglans regia L. The crude extracts were obtained with ethanol, methanol, and water and were evaluated for the total phenol content (TPC), total flavonoid content (TFC), and antioxidant activities using total reducing power (TRP), ferric reducing antioxidant power (FRAP), 1, 1-diphenyl 1-2-picryl-hydrazyl (DPPH), superoxide radical scavenging (SOR), and hydroxyl radical scavenging (•OH) assays. Our results showed that crude leaf extracts of plants harvested from the host Juglans regia L. exhibited higher yields of phytochemical constituents and noticeable antioxidative properties. The ethanolic leaf samples reported the highest phenols (13.46 ± 0.87 mg/g), flavonoids (2.38 ± 0.04 mg/g), FRAP (500.63 ± 12.58 μM Fe II/g DW), and DPPH (87.26% ± 0.30 mg/mL). Moreover, the highest values for TRP (4.24 ± 0.26 μg/mL), SOR (89.79% ± 0.73 mg/mL), and OH (67.16% ± 1.15 mg/mL) were obtained from aqueous leaf extracts. Further, Pearson correlation was used for quantifying the relationship between TPC, TFC, and antioxidant (FRAP, DPPH, SOR, OH) activities in Viscum album L. compared to their hosts. It was revealed that the epiphyte showed variation with the type of host plant and extracting solvent.

The aim of this study is to investigate in vitro antioxidant and antibacterial activities of the aqueous and hydroethanolic extracts for aerial parts of Herniaria hirsuta. Extracts were screened for their possible antioxidant activities by three tests: DPPH free radical-scavenging, reducing power, and molybdenum systems. The screening of antibacterial activity of extracts was individually evaluated against sixteen bacteria species using a disc diffusion method. Flavonoids, total phenols, and tannins content were performed for both extracts. It shows higher content in the hydroethanolic extract. The hydroethanolic extract showed a significant antioxidant activity for the three methods studies to the aqueous extract, but nonsignificant results compared to the reference (BHT). However, both extracts have negative effect on the strains studies for the antibacterial activity.
