Extracts from raw materials from different plant parts, tissue-cultured plants, and callus cultures of Oroxylum indicum were analyzed for in vitro antioxidant activities determined by DPPH radical scavenging assay and evaluated for phytochemical profiles by TLC and LC-MS methods. The results were analyzed by principal component analysis (PCA) to evaluate the similarity. Stalk, pedicel, flower, seed, and whole fruit and callus extracts promoted strong antioxidant activity with high total phenolic and total flavonoid contents. The main phytochemicals found in extracts were baicalin, baicalein, and chrysin. Baicalein and baicalin promoted strong antioxidant effects and existed in most extracts while chrysin, which promoted very low antioxidant activity, was a major flavonoid in the leaves and tissue-cultured plants. From PCA analysis by total phenolic and total flavonoid contents, four main clusters including callus and tissue-cultured plant groups from different growth stages, flower group, and whole fruit and leaf group could be organized. When the results were analyzed by PCA using antioxidant activity with total phenolic or total flavonoid contents, all O. indicum samples could be grouped together except the extracts from the root of tissue-cultured plants which separated from the rest due to their low phytochemical contents and weak antioxidant activities.

In the present study the extracts ofin vivoandin vitrogrown plants as well as callus tissue of red clover were tested for their antioxidant activities, using different extraction solvent and different antioxidant assays. The total flavonoid and phenolic contents as well as extraction yield of the extracts were also investigated to determine their correlation with the antioxidant activity of the extracts. Among all the tested extracts the highest amounts of total phenolic and total flavonoids content were found in methanol extract ofin vivogrown plants. The antioxidant activity of tested samples followed the orderin vivoplant extract > callus extract >in vitroextract. The highest reducing power, 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging, and chelating power were found in methanol extracts ofin vivogrown red clover, while the chloroform fraction ofin vivogrown plants showed the highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, superoxide anion radical scavenging and hydrogen peroxide scavenging compared to the other tested extracts. A significant correlation was found between the antioxidant activity of extracts and their total phenolic and total flavonoid content. According to the findings, the extract ofin vitroculture of red clover especially the callus tissue possesses a comparable antioxidant activity to thein vivocultured plants’ extract.

The aim of this study is to evaluate in vitro antioxidant activities of Ruta montana L.  extracts. This activity was evaluated by three methods : DPPH (2, 2'-diphenyl- 1- picrylhydrazy), bleaching of β-carotene and chelation of ferrous iron. Results showed that ethyl acetate extract (EAE) represents the highest amount of total polyphenols, tannins and flavonoids with 257,1 ± 0,703µg gallic acid equivalent/mg of extract,  251 ± 1.41 µg tannic acid equivalent /mg of extract,117,4 ± 3,451 µg quercetin equivalents/mg of extract, 139,5 ± 4,107 µg rutin equivalents/mg of extract, respectively. In the DPPH assay, ethyl acetate extract showed the higher scavenging capacity (IC50 = 0.044 ± 0.001 mg/ml) followed by methanol, aqueous and chloroform extract. Whereas, AqE showed the best chelating effect and the best inhibitory capacity of the coupled oxidation of linoleic acid/ β-carotene. Summary Introduction: Chrysopogon aucheri is a species native to the South of Iran. It is used for the treatment of some oxidative-based disorders. Objective: Due to lack of biological research on C. aucheri, we were prompted to investigate the antioxidant activities of leaf total extract and different sub-fractions for the first time. The phenolic and flavonoid contents were also determined in the leaves as the interaction between these components and the antioxidant activity. Methods: The antioxidant effects of total extract and sub-fractions were evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method. Total phenolic and flavonoid contents were also determined by Folin-Ciocalteu and aluminium chloride colorimetric methods, respectively. Results: Total extract of C. aucheri leaves was found to possess desirable antioxidant activity in vitro in comparison with standard antioxidant BHT (butylated hydroxytoluene). The highest phenolic and flavonoid contents were observed in the methanol sub-fraction. Results showed weak and moderate correlations with phenolics and flavonoids, respectively, and therefore other metabolites of C. aucheri leaves might be responsible for antioxidant activities. Conclusion: The results suggested that C. aucheri leaves possess desirable antioxidant activity when compared with BHT and support the ethnomedicinal claims of the use of the leaves in the management of some oxidative-based diseases.

The in vitro antiproliferative and antioxidant activities of the aqueous, chloroform and methanol extracts of Muntingia calabura leaves were determined in the present study. Assessed using the 3,(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) (MTT) assay, the aqueous and methanol extracts of M. calabura inhibited the proliferation of MCF-7, HeLa, HT-29, HL-60 and K-562 cancer cells while the chloroform extract only inhibited the proliferation of MCF-7, HeLa, HL-60 and K-562 cancer cells. Interestingly, all extracts of M. calabura, which failed to inhibit the MDA-MB-231 cells proliferation, did not inhibit the proliferation of 3T3 (normal) cells, indicating its safety. All extracts (20, 100 and 500 μg/ml) were found to possess antioxidant activity when tested using the DPPH radical scavenging and superoxide scavenging assays with the methanol, followed by the aqueous and chloroform, extract exhibiting the highest antioxidant activity in both assays. The total phenolic content for the aqueous, methanol and chloroform extracts were 2970.4 ± 6.6, 1279.9 ± 6.1 and 2978.1 ± 4.3 mg/100 g gallic acid, respectively. In conclusion, the M. calabura leaves possess potential antiproliferative and antioxidant activities that could be attributed to its high content of phenolic compounds, and thus, needs to be further explored.

Second-grade dates (Phoenix dactyliferaL.), with hard texture, from three selected Tunisian cultivars (Allig, Deglet Nour, and Bejo) were analysed from their antioxidant activities using DPPH radical scavenging activity, FRAP assay, H2O2scavenging activity, and metal chelating activity. Date extracts showed strong and concentration-dependant activity in all tested methods. The results showed that the best antioxidant activity was obtained in Allig, followed by Bejo and Deglet Nour. Total phenolics, total flavonoids, carotenoids, and tannins were determined spectrophotometrically in three date extracts. Results indicated that date contained significantly different amounts of these compounds. In fact, Allig presented the highest antioxidant compounds, followed by Bejo and Deglet Nour. Correlation analyses indicated a positive linear relationship between antioxidant activities and total phenolic and flavonoid content of date. This study demonstrates the potential antioxidant activity with Tunisian date, where we can use these natural extracts as food additives in replacement of synthetic compounds.

Cordia macleodii, an important medicinal plant traditionally used for the treatment of various disorders, including wound healing, jaundice, gastric ulcer, hepatoprotective activity and antivenom potential, thus the plant showed the high antioxidant properties. Successive extraction of leaf and barks powder with methanol and water was tested for quantitative determination of total phenolics, total flavonoids and various in vitro antioxidant activities. Phytochemical screening of crude plant extracts revealed the presence of sterols, alkaloid, flavonoid, phenolic, Reducing sugar, glycosides, tannins and saponins. All extracts showed the significant total phenolic and flavonoid contents and as well as radical scavenging and iron-chelating activities. Among all, methanol leaf extract was observed to be higher antioxidant activity than that of other extract. The methanol extracts showed greater antioxidant activity by DPPH scavenging the free radical with IC50 values of 7.63 ± 0.38 μg gm-1 along with scavenged free radical in a concentration depended manner, which is compare to the standard ascorbic acid.  Our results revealed that the presence of grater antioxidant activity of C. macleodii extract may be due to the high level of flavonoid and phenolic content in the plant. These results clearly indicated that C. macleodii is an effective natural antioxidant. Key words: Cordia macleodii, antioxidant, radical scavenging, phenolics, flavonoid, phytochemical.

Oroxylum indicum extracts from the seeds collected from Lampang and Pattani provinces in Thailand, and young fruits and flowers exhibited in vitro display antioxidant and antibacterial activities against clinically isolated zoonotic bacteria including Staphylococcus intermedius, Streptococcus suis, Pseudomonas aeruginosa, β-hemolytic Escherichia coli and Staphylococcus aureus. The orange crystals and yellow precipitates were obtained from the preparation processes of the seed extracts. The orange-red crystals from the seeds collected from Lampang province exhibited strong in vitro 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging effects (EC50 value = 25.99 ± 3.30 μg/mL) and antibacterial effects on S. intermedius and β-hemolytic E. coli while the yellow precipitate from the same source exhibited only antioxidant activity. Quantitative analysis of phytochemicals in O. indicum samples by spectrophotometric and HPLC techniques showed that they contained different amounts of total phenolic, total flavonoid and three major flavones; baicalin, baicalein and chrysin contents. Young fruit extract, which contained low amounts of flavone contents, still promoted antibacterial effects against the tested bacteria with IC50 values lower than 1 mg/mL and MIC values between 4 to 10 mg/mL in S. intermedius, S. aureus and S suis while higher IC50 and MIC values against P. aeruginosa and β-hemolytic E. coli were found. From scanning electron microscopy, the extract of the young fruit of O. indicum promoted morphological changes in the bacterial cells by disrupting the bacterial cell walls, inducing leakage of the cellular content, and generating the abnormal accumulation of cells. The mechanism of action of the extract for this antibacterial effect may be the disruption of the cell membrane and abnormal cell aggregations. Regression analysis of the results suggests the correlation between total phenolic and total flavonoid contents and antioxidant and antibacterial effects. Baicalin was found to have a high correlation with an inhibitory effect against β-hemolytic E. coli while three unidentified peaks, which could be flavones, showed high correlations with an inhibitory effect against S. intermedius, S. suis, P. aeruginosa and S. aureus.

Nonalcoholic fatty liver disease (NAFLD) is a major cause of morbidity and mortality worldwide. Additional therapies using functional foods and dietary supplements have been investigated and used in clinical practice, showing them to be beneficial. Honeybee pollen from Chile has shown a large concentration of phenolic compounds and high antioxidant activity. In this work, we characterized twenty-eight bee pollen extracts from the central zone of Chile according to botanical origin, phenolic profile, quercetin concentration, and antioxidant activity (FRAP and ORAC-FL). Our results show a statistically significant positive correlation between total phenolic content and antioxidant capacity. Selected samples were evaluated on the ability to reverse the steatosis in an in vitro cell model using Hepa1-6 cells. The pollen extracts protected Hepa1-6 cells against oxidative damage triggered by 2,2′-azo-bis(2-amidinopropane) dihydrochloride (AAPH)derived free radicals. This effect can be credited to the ability of the phenolic compounds present in the extract to protect the liver cells from chemical-induced injury, which might be correlated to their free radical scavenging potential. Additionally, bee pollen extracts reduce lipid accumulation in a cellular model of steatosis. In summary, our results support the antioxidant, hepatoprotective, and anti-steatosis effect of bee pollen in an in vitro model.

Antioxidant activities of acetone and ethyl acetate extracts from Metaplexis japonica Makino, one of famous medicine plants in the eastnorth region of China, named luomo in Chinese, were examined by a DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical-scavenging assay and a β-carotene-linoleic acid test. In DPPH, the antioxidant activity of the acetone extracts, ethyl acetate extracts and derivative were IC50 were 313.21, 266.92 and 118.78μg/mL, respectively. In the β-carotene-linoleic acid test, IC50 were 285.09, 351.57 and 123.89μg/mL. It was concluded that Metaplexis japonica Makino and its derivatives might be a potential natural source of antioxidants .

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.
