Origanum vulgare L., the most complex and variable species of the genus Origanum, is used to cure various diseases in the traditional medicinal system. In the current investigation, O. vulgare was collected from its natural origin and further multiplied for two years at the experimental farm of DIBER field station, Auli, and afterwards, it was grown at three different altitudes, i.e., DIBER field station of Auli (2744 m), Pithoragarh (1524 m), and Haldwani (412 m), under natural field conditions. The essential oil was extracted using the Clevenger apparatus and analyzed by GC/MS and GC/FID. The total phenolic, flavonoid, and tannin content in the methanolic extracts of all the cultivations were estimated by using Folin–Ciocalteu reagent, aluminium chloride reagent, and Folin–Denis methods, respectively. The antioxidant activity of the essential oils and extracts of all the cultivations was also estimated by DPPH, ABTS, and reducing power methods. The highest amount of phenolics, flavonoids, and tannins was obtained in the Pithoragarh cultivation. Thymol was the major component in all the essential oils contributing 38.81% (Auli), 52.83% (Pithoragarh), and 31.86% (Haldwani) of the total oil. The highest antioxidant activity was exhibited by oregano oil of Pithoragarh cultivation, followed by Auli and Haldwani, respectively. The alcoholic extract of Pithoragarh cultivation had the highest antioxidant activity. The study infers that the plant demonstrated good results in terms of fresh weight, oil yield, antioxidant potential, and EO composition when grown at higher altitudes.

Abstract Origanum vulgare L. var. aureum is an aromatic medicinal plant belonging to the Lamiaceae family, rich in essential oils and antioxidants. Its specific chemical composition represents an important source of biologically active principles with diverse applications in food products and nutraceuticals. The aim of this study was to determine the differences between chemical composition of volatile oils, phenolic content and antioxidant activity of different plants parts: leaves, flowers, and stems. Essential oils from leaves and flowers had similar compositions with only few differences between γ-terpinene and trans-ß-ocimene. Total phenolic content and antioxidant activity were determined for the hydro-alcoholic extract obtained from post-distillation oregano waste material. The total phenolic content was determined using Folin-Ciocalteu reagent and varied from 3173 to 6522 mg GAE/L. The major compounds identified by HPLC were syringic acid, pyrocatechol, and catechin. Furthermore, all extracts showed high antioxidant activity, ranging from 31.3 to 44.5 mg GAE/L, with an inhibition percent varying from 63.1 to 88.6%.

The antioxidant activity of aqueous and alcoholic extracts of different plant parts viz, leaves, flower, stem and root of Ajuga bracteosa was investigated against various in-vitro antioxidant assays. The total phenolic, flavonoid and tannin contents also estimated. The results revealed the significant antioxidant potential and variation in the IC50, EC50 and phytochemical contents among all the plant parts. The aqueous extract of leaves exhibited significantly (P<0.05) highest antioxidant activity on 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acixd), 2,2-diphenyl-1-picryl hydrazyl, and potassium ferricyanide reducing power assay, with IC50 values of 0.2707±0.0008, 0.4409±0.0020, and EC50 value 0.3413±0.0030 mg/mL, respectively, followed by the other parts of the plant. The leaves extract also possess the highest total phenolics, flavonoid and tannin contents among all the parts. Similarly, the aqueous extract is better than the alcoholic extract of different parts as far as phytochemical contents and antioxidant activity concerned. The present study revealed that the aqueous extract of leaves had the highest antioxidant potential, which correlated with the high level of total phenolic fl flavonoid, and tannin contents. Therefore, higher the phytochemical contents, higher will be the antioxidant potential.

Objective: This present study is planned to reveal about the pharmacognostical details, phytochemistry, antimicrobial activity, and antioxidant activity of aqueous and alcoholic extract of the berries of Solanum torvum Swartz. belonging to the family Solanaceae, which is a very good anti-inflammatory medicine in traditional system of therapies.Methods: The physicochemical parameters such as ash values, extractive values, and total fiber content, qualitative preliminary phytochemical studies, quantitative estimation for total phenolic contents and flavonoid content, anjd inorganic minerals such as sodium, calcium, and potassium were estimated. Flavonoids present in the methanolic extract were isolated and its Rf values were calculated by high-performance thin-layer chromatography method and compared with that of the standard epicatechin. The antioxidant activity was evaluated by Reducing power ability, 1, 1-diphenyl-2-picrylhydrazyl method, and hydrogen peroxide methods. The antibacterial and antifungal activities of the aqueous and methanolic extracts are evaluated by disc diffusion method comparing the zone of inhibition with that of the standards such as amikacin and Ketoconazole.Results: It contains a number of potentially pharmacologically active chemicals including the sapogenin steroid, chlorogenin, alkaloids, flavonoids, phenols, and tannins. Both the extracts showed good activity, but the alcohol extract is more active toward all bacterial organisms and also toward fungi organism. Methanol extract shows good antioxidant activity. Antimicrobial activity and antioxidant activity of the berries are due to the presence of phytoconstituents present in it.Conclusion: These berries can be used for new formulation in nutraceuticals and polyherbal natural formulations for reducing the oxidative stress-related diseases and different therapies.

The total phenolics, flavonoids and in vitro antioxidant activity of petroleum ether, benzene, ethyl acetate, methanol and ethanol extracts of stem of Drypetes sepiaria were determined using various antioxidant model systems viz, DPPH, hydroxyl, superoxide, ABTS and reducing power. Total phenolic content was estimated by Folin-Ciocalteau method. Flavonoids were determined by Aluminium chloride method. The total phenolics and flavonoids contents were found to be 0.81 g 100 g-1 and 1.12g 100 g-1 respectively in the methanol extract. Among the solvent tested, methanol and ethanol extracts of D. sepiaria stem showed potent in vitro antioxidant activities. This investigation explored that D. sepiaria stem is a potential source of natural antioxidant.

Abstract Background Wedelia chinensis has been reported as a folk medicine for the treatment of different diseases including neurodegenerative disease. Although the plant has been studied well for diverse biological activities, the effect of this plant in neurological disorder is largely unknown. The present study was undertaken to evaluate the cholinesterase inhibitory and antioxidant potential of W. chinensis. Methods The extract and fractions of the plant were evaluated for acetylcholinesterase and butyrylcholinesterase inhibitory activity by modified Ellman method. The antioxidant activity was assessed in several in vitro models/assays such as reducing power, total antioxidant capacity, total phenolic and flavonoid content, scavenging of 2,2’-diphenyl-1-picrylhydrazyl (DPPH) free radical and hydroxyl radical, and inhibition of brain lipid peroxidation. Chromatographic and spectroscopic methods were used to isolate and identify the active compound from the extract. Results Among the extract and functions, aqueous fraction (AQF) and ethylacetate fraction (EAF) exhibited high inhibition against acetylcholinesterase (IC50: 40.02 ± 0.164 µg/ml and 57.76 ± 0.370 µg/ml) and butyrylcholinesterase (IC50: 31.79 ± 0.182 µg/ml and 48.41 ± 0.053 µg/ml). Similarly, the EAF and AQF had high content of phenolics and flavonoids and possess strong antioxidant activity in several antioxidant assays including DPPH and hydroxyl radical scavenging, reducing power and total antioxidant activity. They effectively inhibited the peroxidation of brain lipid in vitro with IC50 values of 45.20 ± 0.995 µg/ml and 25.53 ± 0.042 µg/ml, respectively. A significant correlation was observed between total flavonoids and antioxidant and cholinesterase inhibitory activity. Activity guided chromatographic separation led to the isolation of a major active compound from the EAF and its structure was elucidated as apigenin by spectral analysis. Conclusions The potential ability of W. chinensis to inhibit the cholinesterase activity and peroxidation of lipids suggest that the plant might be useful for the management of AD.

The influence of various gamma-radiation dose absorptions on oregano (<i>Origanum vulgare</i> L.) solid samples was monitored by means of electron paramagnetic resonance (EPR) spectroscopy. Further, the antioxidant activity of oregano methanol/water extracts was characterised using DPPH (1,1-diphenyl-2-picrylhydrazyl free radical); thiobarbituric acid reactive substances (TBARS); ferric reducing power (FRP); and total content of phenolic compounds (TPC) assays. EPR spectroscopy was used for the investigation of the influence of the absorbed dose on the character of the paramagnetic structures formed, as well as for their thermal stability and life-time characterisation. EPR spectrum of the reference (non-irradiated) sample represents a broad singlet line with unresolved hyperfine splitting, attributable to Mn(II) ions, upon which the additional sharp EPR signal (g = 2.0022, &Delta;B<sub>pp</sub> &asymp; 1 mT) is superimposed, assigned to stable semiquinone radicals produced by the oxidation of polyphenolic compounds present in plants. The additional paramagnetic structures of different origin (mostly of cellulose and carbohydrate), possessing diverse thermal stability and life-time, were identified in the gamma-irradiated samples. Immediately after irradiation, a statistically significant increase was observed of the TBARS values and the total content of phenolic compounds in methanol/water oregano extract. The alterations of the antioxidant properties of oregano extracts with the time after the radiation treatment were also monitored. A substantial time-dependent decrease of antioxidant activity was observed, probably as a result of storage, with both irradiated and non-irradiated oregano samples, as obvious from the ferric reducing power test and the content of total phenolic substances. The influence of irradiation and subsequent storage of oregano samples on the DPPH radical-scavenging ability was only negligible.

Abstract The aim of the present study is to evaluate the total phenolic contents, antioxidant and anti-proliferative activities of the n-BuOH extract of Centaurea maroccana (BECM). The total phenolic and flavonoid of the butanolic extract of the plant were assessed by using Folin-Ciocalteau and aluminium chloride colorimetric assays, respectively, and the quantitative estimation of total flavonoids and phenols revealed the richness of the extract in these compounds. Antioxidant activity was evaluated using standard lab colorimetric methods, while the anti-proliferative activity was evaluated using sulforhodamine B (SRB) assay against C6 (Rattus norvegicus brain glioma) and HT29 (Homo sapiens colorectal adenocarcinoma) cell lines. The n-butanolic extract of Centaurea maroccana showed a strong antioxidant activity through DPPH, ABTS•+, DMSO alkalin, Reducing power, β-Carotene-linoleic acid and CUPRAC assays. Furthermore, the anti-proliferative activity against C6 and HT29 of Centaurea maroccana exhibited a high effect by the decrease in viability of both cancer cell lines. Our results suggest a possible use of Centaurea maroccana as a source of natural antioxidant and chemo-preventive agents against cancer.

Oregano ( Thymbra capitata and Origanum vulgare) essential oils (EOs), cultivated and extracted in the South-East of Spain, were analysed by GC/MS to determine their composition. ( E)-β-Caryophyllene (0.5–4.9%), thymol (0.2–5.8%), p-cymene (3.8–8.2%), γ-terpinene (2.1–10.7%) and carvacrol (58.7–77.4%) were determined as the main molecules. This characterisation was completed with enantioselective gas chromatography, where (-)-( E)-β-caryophyllene, (+)-α-pinene and (+)-β-pinene were determined as the main enantiomers. Antioxidant activity was evaluated positively by several methods, accounting for activity against free radicals and reducing power. Important inhibitory activity on lipoxygenase (LOX) and acetylcholinesterase (AChE) was observed supporting potential anti-inflammatory, anti-Alzheimer and insecticidal activities, mainly due to carvacrol. These properties support the potential use of oregano EOs as natural cosmetic and natural pharmaceutical ingredients.

AbstractBackgroundEucalyptus essential oils and extracts are used since the ancient times in alternative medicine. The 1,8-cineole is the most significant constituent of Eucalyptus essential oil, while phenolic contents define the value of eucalyptus extracts. Based on the last considerations, interspecific variability of 1,8-cineole content, phenolics and antioxidant potentials among nine Eucalyptus taxa growing under the sub-humid bioclimate stage of Tunisia was investigated.MethodsEssential oil profiling was assessed based on gas chromatography-mass spectrometry analysis. Total phenolic and flavonoid contents were assessed using Folin–Ciocalteau and aluminium chloride colorimetric methods, respectively. The antioxidant ability of Eucalyptus volatile metabolites and extracts was achieved based on two test systems namely DPPH and FRAP assays.ResultsQualitative and quantitative variations in the composition of essential oils according to the studied taxon were shown. The total phenolic and flavonoid contents varied also significantly among the investigated samples. Based on the obtained results, the species Eucalyptus sideroxylon exhibits the highest 1,8-cineole content (76.24 ± 0.86), total phenolics and flavonoids contents (38.5 ± 1.4 mg GAE /g DW and 18.6 ± 0.3 mg RE /g DW, respectively). Moreover this species highlighted the highest free radical-scavenging ability and ferric reducing power for both essential oil and methanolic extracts. Chemometric multivariate analysis showed the classification of the nine studied taxa to three clusters.ConclusionsThe pattern of 1,8-cineole concentration, phenolic and flavonoid contents for the studied nine Eucalyptus species and hybrid showed E. sideroxylon species as the potential candidate for further improvement strategies regarding the production of eucalyptus essential oils and phenolics with high quality for pharmaceutical industry.

AbstractA number of herbal plants from Romania widely used as natural food additives or for health promotion in traditional medicine were investigated for their antioxidant activity. Methanol extracts were obtained from plants belonging to the Lamiaceae family (lavender Lavandula angustifolia L.; lemon balm Melissa officinalis; sage Salvia officinalis; oregano Origanum vulgare L.; rosemary Rosmarinus officinalis L.; thyme Thymus vulgaris L.; mullein Verbascum phlomoides; mint Mentha longifolia), Clusiaceae family (St John’s wort Hypericum perforatum L.), and Compositae family (elecampane Inula helenium). Total phenolic concentration was determined using the Folin-Ciocalteu phenol reagent method, while total flavonoids were measured using the aluminium chloride colorimetric method. Relationships between total antioxidant activity and composition of plant extracts were evaluated. Origanum vulgare extract showed the highest antioxidant activity and total phenolic content compared to the other plants extracts. A positive correlation was observed between total antioxidant activity and total phenolic content of the analyzed extracts.
