Interspecific variability of 1,8-cineole content, phenolics and bioactivity among nine Eucalyptus taxa growing under the sub-humid bioclimate stage

AbstractBackgroundEucalyptus essential oils and extracts are used since the ancient times in alternative medicine. The 1,8-cineole is the most significant constituent of Eucalyptus essential oil, while phenolic contents define the value of eucalyptus extracts. Based on the last considerations, interspecific variability of 1,8-cineole content, phenolics and antioxidant potentials among nine Eucalyptus taxa growing under the sub-humid bioclimate stage of Tunisia was investigated.MethodsEssential oil profiling was assessed based on gas chromatography-mass spectrometry analysis. Total phenolic and flavonoid contents were assessed using Folin–Ciocalteau and aluminium chloride colorimetric methods, respectively. The antioxidant ability of Eucalyptus volatile metabolites and extracts was achieved based on two test systems namely DPPH and FRAP assays.ResultsQualitative and quantitative variations in the composition of essential oils according to the studied taxon were shown. The total phenolic and flavonoid contents varied also significantly among the investigated samples. Based on the obtained results, the species Eucalyptus sideroxylon exhibits the highest 1,8-cineole content (76.24 ± 0.86), total phenolics and flavonoids contents (38.5 ± 1.4 mg GAE /g DW and 18.6 ± 0.3 mg RE /g DW, respectively). Moreover this species highlighted the highest free radical-scavenging ability and ferric reducing power for both essential oil and methanolic extracts. Chemometric multivariate analysis showed the classification of the nine studied taxa to three clusters.ConclusionsThe pattern of 1,8-cineole concentration, phenolic and flavonoid contents for the studied nine Eucalyptus species and hybrid showed E. sideroxylon species as the potential candidate for further improvement strategies regarding the production of eucalyptus essential oils and phenolics with high quality for pharmaceutical industry.

 The efficiencies of three extraction methods (percolation, Soxhlet and ultrasonically assisted extraction) were evaluated for the extraction of antioxidants from Vicia faba L. bean and hulls. Antioxidant activities of extracts were evaluated using four different tests. Total phenolic and flavonoid contents of the extracts were determined using the Folin–Ciocalteu and aluminium chloride methods, respectively. Although the yield of extractions for ultrasonically assisted extraction was about half to one-fourth of that of other methods, the extraction ratio of total phenol was higher. Hull extracts had higher total phenolic and flavonoid contents and antioxidant activities than bean extracts. The hull ultrasonic extract showed the best DPPH (IC50= 56.9 ± 2.5 g ml-1) and NO radical scavenging (11.3 ± 0.5 gml-1). The hull percolation extract showed the best iron chelating ability (171.8 ± 6.8 g ml-1) and reducing power. The results indicated that all extraction methods could effectively extract antioxidants from medicinal plants

Objective: In this study, the leaf powder of Aegle marmelos (L.) Correa. was subjected to preliminary phytochemical and in vitro antioxidant analysis. Methods: The freshly prepared plant leaf extract was subjected to preliminary phytochemical screening, which revealed the presence of alkaloids, tannins, saponins, flavonoids, glycosides, phenolic compounds, terpenoids, and steroids. Reducing power, superoxide (SO) anion radical, nitric oxide (NO) radical, and hydroxyl radical scavenging assays were carried out to evaluate the antioxidant potential of the methanolic leaf extract of this plant. The amounts of total phenolic and flavonoid compounds were also determined. Results: This study has revealed that the A. marmelos (L.) Correa. leaf extract showed considerably high amounts of most of the phytochemicals, total antioxidant capacity, total phenolic, and flavonoid content. The study also indicated that the A. marmelos (L.) Correa. showed comparatively good scavenging activity, i.e., inhibition of hydroxyl radical, NO and SO anion scavenging and reducing power activities when compared with the respective standards. Conclusion: The leaf powder of A. marmelos (L.) Correa. can be used as easily accessible source of natural antioxidant and as a possible food supplement or in pharmaceutical industry.

Pterygota alata is a large deciduous tree of Malvaceae family. The present study was designed to evaluate in vitro antioxidant activity of stem bark of the plant. Ferric reducing power (FRP) test, 1, 1- diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging test and total antioxidant capacity (TAC) assay were used to detect the antioxidant activity. Total phenolic and flavonoid contents were also determined as they are well known phytochemicals with antioxidant property. The bark of the plant showed efficient reducing power as well as free radical scavenging property (IC50 values 52.25 - 172.05). The bark also found rich in total phenolic and flavonoid content. The highest amount of total phenolic content was found in chloroform soluble fraction (29.898 µg/mL) followed by ethyl acetate soluble fraction (15.88 µg/mL). The highest content of total flavonoid also detected in chloroform soluble fraction (107.56 µg/mL) but followed by crude ethanol extract (98.66 µg/mL). Overall, the bark of the plant possesses significant antioxidant activity, therefore can be used as a good natural source of antioxidant. © 2014 JSR Publications. Origanum vulgare L., the most complex and variable species of the genus Origanum, is used to cure various diseases in the traditional medicinal system. In the current investigation, O. vulgare was collected from its natural origin and further multiplied for two years at the experimental farm of DIBER field station, Auli, and afterwards, it was grown at three different altitudes, i.e., DIBER field station of Auli (2744 m), Pithoragarh (1524 m), and Haldwani (412 m), under natural field conditions. The essential oil was extracted using the Clevenger apparatus and analyzed by GC/MS and GC/FID. The total phenolic, flavonoid, and tannin content in the methanolic extracts of all the cultivations were estimated by using Folin–Ciocalteu reagent, aluminium chloride reagent, and Folin–Denis methods, respectively. The antioxidant activity of the essential oils and extracts of all the cultivations was also estimated by DPPH, ABTS, and reducing power methods. The highest amount of phenolics, flavonoids, and tannins was obtained in the Pithoragarh cultivation. Thymol was the major component in all the essential oils contributing 38.81% (Auli), 52.83% (Pithoragarh), and 31.86% (Haldwani) of the total oil. The highest antioxidant activity was exhibited by oregano oil of Pithoragarh cultivation, followed by Auli and Haldwani, respectively. The alcoholic extract of Pithoragarh cultivation had the highest antioxidant activity. The study infers that the plant demonstrated good results in terms of fresh weight, oil yield, antioxidant potential, and EO composition when grown at higher altitudes.

Abstract Soy whey, a liquid nutritional by-product of soybean manufacture, is rich in proteins, oligosaccharides and isoflavones. Soy whey can be used to produce functional beverages, instead of discarding it as a waste. In this study, unfermented soy whey (USW) and Cordyceps militaris SN-18-fermented soy whey (FSW) were investigated and compared for their physicochemical and functional properties by high performance liquid chromatography (HPLC) and DNA damage assay. Results show that C. militaris SN-18 fermentation could increase the contents of essential amino acids, total phenolic and flavonoid and isoflavone aglycones and eliminate the oligosaccharides in soy whey. Furthermore, C. militaris SN-18 could significantly enhance the ABTS radical scavenging ability, reducing power and ferric reducing power of soy whey, and its fermented products could prominently attenuate Fenton reaction-induced DNA damage. These findings indicate that soy whey can potentially be converted into a novel soy functional beverage by C. militaris SN-18 fermentation. Graphical abstract

Abstract Twenty-eight populations of ajowan (Trachyspermum ammi L.) were evaluated for morphological traits and oil yield in two consecutive years. Then, selected ajowan populations that revealed medium and high oil yield and higher seed weight in two years were selected for further studies. These were assessed for physiological traits, total phenolic and flavonoid contents and antioxidant capacity under four salt treatments control, 60, 90, and 120 mM NaCl. The essential oil composition was analyzed by gas chromatography-mass spectrometry (GC-MS) Thymol (32.7-54.29%), γ -terpinene (21.71–32.81%), and p-cymene (18.74–26.16%) were major components. The highest and lowest thymol were recorded for Qazvin (control) and Qazvin (Low salt concentration), respectively. Salt stress caused an increase in essential oil content of Esfahfo and Qazvin populations. The highest phenolic and flavonoid contents were found in Arak population grown in 60 mM NaCl (183.83 mg TAE g− 1 DW) and Yazd population grown in 90 mM NaCl (5.94 mg QE g− 1 DW). Moreover, Yazd population exhibited the strongest antioxidant activity based on DPPH (IC50 = 1566µg/mL) under 60 mM NaCl and the highest reducing power (0.69 nm) under 120 mM NaCl. Overall, the results revealed that low and moderate salt stress improves the phytochemicals of ajowan, being especially useful for pharmaceutical and food applications.

Background: Cinnamomum zeylanicum Blume bark and leaf essential oil possesses eugenol as the major component. Both the essential oil have the potential antioxidant, anti-inflammatory, antimicrobial and negligible genotoxicity. Objective: A comparative chemical composition and biological evaluation of Cinnamomum zeylanicum Blume, bark and leaf essential oils, originated from North-East India has been investigated in the present study. Methods: Gas chromatography/mass spectroscopy analysis was used to investigate the chemical composition of the essential oil samples. DPPH free radical scavenging activity, reducing power assay was used for antioxidant activity evaluation. Similarly, egg albumin denaturation assay, protease inhibitory activities were used for anti-inflammatory activity, while genotoxicity was evaluated by using Allium cepa assay. Results: The results showed that eugenol is the major component of C. zeylanicum bark (84.48%) and leaf (48.78 %) essential oil. Significant antioxidant activity were observed for both essential oils in the DPPH free radical scavenging assay (IC50= 103.2 μg/mL, bark; IC50= 234.7 μg/mL, leaf), and reducing power assay (absorbance of 1.802 nm, 0.907 nm in 48 µg/mL for bark and leaf essential oil respectively). In-vitro anti-inflammatory activity revealed strong potential of leaf essential oil (protein denaturation assay, IC50= 0.05279 μg/mL; protease inhibitory activity IC50= 3.607 μg/mL) in comparison to bark essential oil (protein denaturation assay IC50= 0.1491, protease inhibitory activity IC50= 61.06). Allium cepa root growth test, mitotic index, and chromosomal aberration test were performed to investigate the genotoxicity of C. zeylanicum bark and leaf essential oil, which resulted no toxic nature of both the essential oils. Antimicrobial activity against eight bacterial and fungal strains revealed good antimicrobial properties with the MIC value of Staphylococcus aureus at 5 μL/mL (bark), Aspergillus niger at 2.5 μL/mL (bark); Bacillus cereus at 5 μL/mL (leaf), Aspergillus niger at 2.5 μL/mL (leaf). Conclusion: Collectively, these findings indicated the strong biological potential of both essential oils, although bark essential possesses better biologically activities than the leaf essential oil and could be used in the food and beverages industries.

An alternating synergetic ultrasound/microwave method was applied to the simultaneous extraction of essential oils and polysaccharides with deep eutectic solvent (DES) from Schisandra chinensis. Under the optimal conditions, extract in the selected choline chloride-ethylene glycol 1:3 solvent yielded 12.2 mL/kg and 8.56 g/100g of essential oils and polysaccharides, respectively. The free radical scavenging and immunological activities of the polysaccharides and the antioxidant activity of the essential oils have also been investigated. The lymphocyte proliferation capacity was substantially improved by adding concanavalin A or lipopolysaccharides to polysaccharides (0.20 mg/mL). The IC50 values of the essential oils for scavenging DPPH obtained by hydro-distillation and DES ultrasound/microwave-assisted hydro-distillation (DES UMHD) were 52.34 µg/mL and 29.82 µg/mL, respectively. The essential oil obtained by DES UMHD had the highest reducing power (856.05 (TE)/g) at 150 g/mL and had the strongest inhibitory capacity (SC% = 18.12%). S. chinensis has the potential to be developed as a natural antioxidant.

Present study reports a comparative screening of three ecotypes of Ziziphora tenuior L., sampled from three region with different elevation in middle and northeastern Turkey, based on their essential oil composition, total phenolic content and antioxidant capacities. Z. tenuior ecotypes showed twenty compounds, representing 89.18-91.44% of the essential oil (EOs) identified by GC-MS. The major components of EOs in ecotypes were pulegone (37.23-49.12%), and followed by 1,8-Cineole (2.26-7.78%), limonene (4.20-5.44%), ?-Caryophyllene (3.88-5.11%) and thymol (2.68-4.11%), respectively. Ecotypes showed variable pulegone content and it was increased with elevation increase. The total phenolic contents varied among ecotypes ranged from 21.13 to 27.50 mg of gallic acid equivalent/g EOs. Antioxidant capacity was determined in vitro using DPPH radical-scavenging and FRAP (Ferric reducing power) assays and expressed as concentration of each extract required to inhibit radical by 50% (IC50 and EC50) values that ranged from 0.80 to 0.97 mg/ml in DPPH and 0.42 to 0.55 mg/ml in FRAP assays, respectively. Our results indicated that antioxidant capacity changed among ecotypes. Taking into account the essential oil content, it seems that to select ecotype was the most appropriate to obtain more pulegone for this plant from its wild habitat.
