Introduction: The prevalence of obesity is high and increasing worldwide. Obesity is generally associated with an increased risk of chronic disease and mortality. The objective of the study was to test the effect of a lifestyle intervention on body weight and other chronic disease risk markers. Methods: A non-randomized controlled trial was conducted, including mostly middle-aged and elderly participants recruited from the general population in rural northwest Germany (intervention: n = 114; control: n = 87). The intervention consisted of a 1-year lifestyle programme focusing on four key areas: a largely plant-based diet (strongest emphasis), physical activity, stress management, and community support. Parameters were assessed at baseline, 10 weeks, 6 months, and 1 year. The control group received no intervention. Results: Compared to control, in the intervention group significantly lower 1-year trajectories were observed for body weight, body mass index (BMI), waist circumference, total cholesterol, calculated LDL cholesterol, non-HDL cholesterol, remnant cholesterol (REM-C), glucose, HbA1c, and resting heart rate (RHR). However, between-group differences at 1 year were small for glucose, HbA1c, and cholesterol (apart from REM-C). No significant between-group differences were found for 1-year trajectories of measured LDL cholesterol, HDL cholesterol, triglycerides, insulin, blood pressure, and pulse pressure. Conclusion: The intervention successfully reduced body weight, BMI, waist circumference, REM-C, and RHR. However, at 1 year, effectiveness of the intervention regarding other risk markers was either very modest or could not be shown.

Introduction: A systematic review and meta-analysis of clinical trials was undertaken to evaluate the effect of diacerein intake on cardiometabolic profiles in patients with type 2 diabetes mellitus (T2DM). Methods: Electronic databases such as PubMed, EMBASE, Scopus, Web of Science, Google Scholar, and Cochrane Central Register of Controlled Trials were searched from inception to 31 July 2019. Statistical heterogeneity was evaluated using Cochran’s Q test and I-square (I2 ) statistic. Data were pooled using random-effect models and weighted mean difference (WMD). Results: From 1,733 citations, seven clinical trials were eligible for inclusion and meta-analysis. A significant reduction in hemoglobin A1c (HbA1c) (WMD -0.73; 95%CI -1.25 to -0.21; P= 0.006; I2 = 72.2%) and body mass index (BMI) (WMD -0.55; 95%CI -1.03 to -0.07; P= 0.026; I2 = 9.5%) were identified. However, no significant effect of diacerein intake was identified on fasting blood sugar (FBS) (WMD - 9.00; 95%CI -22.57 to 4.57; P= 0.194; I2 = 60.5%), homeostatic model assessment for insulin resistance (HOMA-IR) (WMD 0.39; 95%CI 0.95 to 1.73; P= 0.569; I2 = 2.2%), body weight (WMD -0.54; 95%CI -1.10 to 0.02; P= 0.059), triglycerides (WMD -0.56; 95%CI -24.16 to 23.03; P= 0.963; I2 = 0.0%), total-cholesterol (WMD -0.21; 95%CI -12.19 to 11.78; P= 0.973; I2 = 0.0%), HDL-cholesterol (WMD -0.96; 95%CI -2.85 to 0.93; P= 0.321; I2 = 0.0%), and LDL-cholesterol levels (WMD -0.09; 95%CI -8.43 to 8.25; P= 0.983; I2 = 37.8%). Conclusion: Diacerein intake may reduce HbA1c and BMI; however, no evidence of effect was observed for FBS, HOMA-IR, body weight, triglycerides, total-cholesterol, HDL-cholesterol or LDL-cholesterol.

Background: It is unclear whether vitamin D deficiency is associated with a higher risk of cardiovascular disease, and through what biochemical pathways this could occur. We investigated the relationship between serum 25-OH vitamin D and typical cardiovascular risk markers as well as incident myocardial infarction (MI) in a large group of high-risk individuals from the community of Calgary, Alberta, Canada. Methods: Calgary Laboratory Services databases were queried for age, sex, body mass index (BMI), personal healthcare number (PHN) and first available serum 25-OH vitamin D measure from patients who received an electrocardiogram or urine creatinine clearance test from 2010-2013. Data was linked by PHN to first available laboratory results for total cholesterol, HDL cholesterol, triglycerides, LDL cholesterol, fasting glucose and HbA1c as well as Alberta Health Services hospital discharge data for first myocardial infarction (ICD-10: I21.1-9) occurring after 25-OH vitamin D measurement. Multiple linear and logistic regression were used to examine all associations. Results: There were 36 000-50 000 complete patient records for analysis of each of the risk markers, with a median follow-up of 8-11 months. A 30 mmol/L increase in serum 25-OH vitamin D was associated with significantly (p<0.001) lower total cholesterol (-0.07 mmol/L), LDL cholesterol (-0.06 mmol/L), triglycerides (-0.14 mmol/L), fasting glucose (-0.12 mmol/L), and HbA1c (-0.13% mmol/L), but higher HDL cholesterol (+0.06 mmol/L) after adjusting for age, sex, BMI, monthly hours of sun-exposure and time between measures. Among these individuals, there were 458 cases of MI occurring after 25-OH vitamin D measurement, with a median follow-up of 1 year. In a case-cohort analysis that included 2500 controls, a 30 mmol/L increase in 25-OH vitamin D was associated with a 21% (p<0.001) lower odds of MI after multivariate adjustment. This association was strongly attenuated after adjusting LDL, HDL, fasting glucose and HbA1c. Conclusion: In a high-risk group of community patients from Calgary, Alberta, Canada, higher serum 25-OH vitamin D was associated with a lower risk of MI, which was explained by changes in commonly measured cardiovascular risk markers. Further study is needed to determine whether changes in cardiovascular risk markers are causally related to changes in 25-OH vitamin D.

Abstract Objectives The study was aimed at assessing the effects of Lactobacillus rhamnosus yoba on the serum lipid profile and body weight of male Wistar rats fed a high fat diet. Methods 20 seven week old male rats weighing between 120 g and 180 g were used for the study and divided into 4 groups of 5 rats each. The control group was fed normal mice pellets and distilled water, while the other groups were fed mice pellets supplemented with 3% cholesterol and 2% saturated fat in addition to any of distilled water, Lactobacillus rhamnosus yoba or Norvastatin respectively. The body weight was measured at the start of the study and after 2 weeks while serum parameters were measured after 8 weeks. Data was analyzed using SPSS Version 20. ANOVA and Tukey's tests determined significant differences in means at 95% confidence interval. Results Lactobacillus rhamnosus yoba significantly (P < 0.005) modulated weight gain, serum total cholesterol and triglycerides when compared to the control. Similarly, LDL-cholesterol was significantly modulated (P < 0.005) while HDL-cholesterol was significantly enhanced (P < 0.005) when compared to the control. Conclusions The ability of Lactobacillus rhamnosus yoba to elevate HDL cholesterol and modulate LDL-cholesterol without the side effects of statins makes it a potential functional food. In line with the findings, the present study justifies the use of Lactobacillus rhamnosus yoba as a probiotic in fermented foods. Funding Sources NA.

This study investigated the hypoglycaemic and hypolipidaemic effects of Treculia africana plant used in Nigeria as medicinal plant. Diabetes mellitus was induced by a single dose intraperitoneal injection of alloxan 150 mg/kg body weight. Twenty five (25) male albino rats were divided into five groups, five (5) rats per group; normal control, diabetic control and diabetic groups treated with aqueous leaves extract of 200,400 and 800 mg/Kg body weight respectively for 21 days orally. The effects of the extract on some biochemical parameters were evaluated; fasting blood glucose level was assayed using glucose oxidase method, total cholesterol and HDL –cholesterol were assayed using enzymatic method while LDL- cholesterol was determined by Friedewald equation. The results showed that, extract significantly (p<0.05) decrease the elevated fasting blood glucose levels, total cholesterol, triglyceride and LDL- cholesterol when compared with the diabetic control rats. The extract also caused significant (p<0.05) increased in HDL –cholesterol and body weight when compared with diabetic control rats. Aqueous leave extract of Treculia africana possess hypoglycemic effect and the most effective dose was 800 mg/Kg body weight in amelioration of hyperglycaemia and most all toxicity effects of alloxan on lipid profile.

AbstractCholine is an essential nutrient involved in several processes, including the export of lipids from the liver. Recent studies have underlined that low choline intake may be linked to greater body weight and liver dysfunction. The aim of this study was thus to determine whether choline intake is associated with body weight, body mass index (BMI), body composition, lipid profile, or liver steatosis indices.407 healthy subjects aged 20–40 were enrolled in Poznań, Poland from 2016 to 2018. Food intake was assessed using three-day food records. Choline intake was analyzed using the USDA Database for the Choline Content of Common Foods, which summarizes the levels of choline found in a range of food items. Weight to 0.1 kg and height to 0.01 m were measured using an electronic scale and a stadiometer, respectively. BMI was calculated as body weight in kilograms divided by height in meters squared. Fat mass and lean body mass were determined using whole-body air-displacement plethysmography. Waist and hip circumferences were measured to 0.5 cm using nonelastic tape. Total cholesterol, LDL cholesterol, HDL cholesterol, and triglyceride levels in serum were determined using a biochemical analyzer. The following biomarkers of liver steatosis were calculated: NAFLD liver fat score (NAFLD-LFS), fatty liver index (FLI), and hepatic steatosis index (HSI). To analyze associations between choline intake and these parameters, we used multiple regression with adjustments for age, sex, and energy intake.The mean BMI was 25.9 ± 5.28 kg/m2, the mean body weight was 78.39 ± 18.03 kg, the mean body fat percentage was 29.1 ± 10.79%, and the mean choline intake was 447.93 ± 235 mg/day. Choline intake was negatively associated with body weight, body fat percentage, waist circumference, and FLI index (p < 0.05 for all associations) and positively associated with HDL cholesterol (p < 0.05). There were no associations between choline intake and BMI, hip circumference, total cholesterol, LDL cholesterol, and the following fatty liver indexes: HSI and NAFLD-LFS.Our study suggests that higher choline intake is associated with favorable body composition and may have a protective role on liver status. Background: Identifying predictive factors that contribute to changes in body weight may well be an interesting approach to the management of obesity. Objective: This study was firstly aimed at examining the effect of a one-year lifestyle program based on improvements in the habitual diet and increased levels of physical activity on weight loss. Secondly, it was focused on identifying anthropometric, and serum hormonal, metabolic and haematochemical factors which can be associated with the degree of weight loss in Kg. Methods: 488 overweight or obese subjects, 383 women and 105 men, aged 18-67 years, were enrolled in the study. Body mass index, waist circumference, serum blood glucose, lipids, uric acid, creatinine, insulin, TSH, FT3, FT4, and 24-h urine catecholamines were measured. Results: Weight loss was positively associated with BMI (P < 0.01), waist circumference (P < 0.01), uric acid (P < 0.01), creatinine (P < 0.05), smoking (P < 0.01), and negatively correlated with age (P < 0.01), total cholesterol (P < 0.05), LDL-cholesterol (P < 0.01), HDL cholesterol (P < 0.05). In a multiple regression model considering weight loss as a dependent variable, and smoking, age, BMI, uric acid, creatinine, total cholesterol, LDL-cholesterol and HDL cholesterol as independent variables, weight loss maintained a direct independent relationship with BMI (P < 0.001), uric acid (P < 0.05), LDL-cholesterol (P < 0.05), and HDL-cholesterol (P < 0.05), and an inverse independent association with cholesterol (P < 0.01). Conclusions: This study suggests that higher BMI and uric acid levels, and lower total cholesterol concentrations are associated with a greater potential to lose weight.

The objective of the present study was to investigate the antihyperlipidaemic efficacy of Trachyspermum ammi (L) Sprague, seed (Ajowan) powder in albino rabbits. Hyperlipidaemia was induced with butter ad libitum and oral intubation of cholesterol 400 mg/kg body weight. Simvastatin, (Tablet survive®) was used as a synthetic cholesterol lowering drug. The results suggest that 2 g/kg T. ammi seed powder produced hypolipidaemic activity, rendering 49%, 53%, 71% and 63% reduction in total lipids, triglycerides, total cholesterol and LDL-cholesterol, respectively. However, at this dosage level 62% increase in the value of HDL-cholesterol was induced by T. ammi seed powder. Nonetheless, 0.5 g/kg and 1 g/kg T. ammi seed powder did not show antihyperlipidaemic activity. T. ammi seed powder at the rate of 2 g/kg and simvastatin (0.6 mg/kg body weight) were equally effective in treating hyperlipidaemia in albino rabbits. Moreover, this dosage level also significantly reduced the cholesterol content of liver tissue. It can be anticipated that the lipid lowering mechanism may involve enhanced removal or catabolism of lipoproteins, inhibition of HMG COA reductase, and/or inhibition of lysosomal lipid hydrolytic enzymes secreted by the liver.

Time constraints and lack of awareness of risk factors for future chronic disease development prevent many young adults from accessing lifestyle programs offered by local health services. This study aimed to determine the effectiveness of a rural pharmacy-based multidisciplinary healthy lifestyle pilot program on reducing risk factors for chronic disease development among young adults. Individuals under the age of 50 with chronic disease risk factors were referred to the program. All subjects were provided with free after-hours nutritional counselling from a dietitian at the local community pharmacy, a comprehensive medication review conducted by the pharmacist, gym membership and access to cooking classes and supermarket tours. Selected participants also received bulk-billed GP appointments and assistance with establishing a home vegetable garden. Body weight, waist circumference, fruit and vegetable consumption and physical activity were assessed at baseline and after program conclusion. Forty participants regularly attended the program between March 2009 and March 2010. At program conclusion, mean body weight was reduced by 3.8 ± 6.7 kg (P < 0.001) and waist circumference reduced by 3.9 ± 6.5 cm (P < 0.001). Fruit consumption increased by 1.2 ± 0.2 serves/day (P < 0.001) and vegetable intake increased by 1.6 ± 1.0 serves/day (P < 0.001). Participants also spent an average of 88.0 ± 47.7 more min/week (P < 0.001) engaged in physical activity on completion of the program. The community pharmacy provided an accessible location for the delivery of a successful chronic disease risk reduction program targeting young adults in a rural area.

Background: Abdominal adiposity, insulin resistance dyslipidemia, and endothelial dysfunction emerge during menopause. Objectives: To assess the relationship between lifestyle, eating behavior, and cardiometabolic biomarkers in Algerian postmenopausal women. Subjects and Methods: A prospective cross-sectional survey was conducted among 228 postmenopausal women (57.65±6.42 years) in Oran (Algeria). Women were divided into quartiles according to their waist circumference (WC); Q1 (69-89cm), Q2 (90-98.5cm), Q3 (99-108cm), and Q4 (108-125cm). We assessed for 3 days, food consumption by the 24h recall and record method, and daily energy expenditure (DEE). In serum, we analyzed the lipid profile, inflammation markers, and oxidative status. Results: DEE and total energy intake were similar in all groups. A decrease in metabolism equivalent tasks (Mets) was observed according to WC increase (<1.5). The Mets was negatively correlated with LDL-cholesterol, triacylglycerols (TG), lipid accumulation products (LAP), CRP, thiobarbituric acid reactive substances (TBARS), TBARS-LDL, and carbonyls and positively correlated with the activity of lecithin cholesterol acyltransferase (LCAT), superoxide dismutase (SOD) and catalase. An inverse relationship was noted between the intake of meats, poultry, eggs, fish, and antioxidant enzymatic activities. Fat intake was positively correlated with lipid accumulation products (r=0.293, p<0.001) and negatively with HDL-cholesterol (r=-0.396, p<0.001), LCAT activity (r= -0.275, p<0.001) and C-Reactive Protein (CRP) (r= -0.315, p<0.001). Fruits and vegetables intake was negatively correlated with LDL-Cholesterol (r=- 0.279, p<0.001) and LDL-TBARS (r= -0.284, p<0.001). Conclusion: Unhealthy diet and sedentary lifestyle were associated with high cardiometabolic risk factors in postmenopausal women and exposed them to cardiovascular diseases.
