Effects of xylene and formaldehyde inhalations on renal oxidative stress and some serum biochemical parameters in rats

In this study, it was aimed to demonstrate the possible renal oxidative stress and some serum biochemical parameters and their alterations caused by the exposure to xylene and formaldehyde (HCHO) in rats. Weighing 150—200 g, 12-week-old, 24 female Sprague-Dawley rats were used. The rats were randomly divided into four groups: Group 1 (control), Group 2 (300-ppm technical xylene), Group 3 (6-ppm HCHO) and Group 4 (150-ppm technical xylene + 3-ppm HCHO). The animals were exposed to gases eight hours per day for six weeks. Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities, glutathione (GSH) and malondialdehyde (MDA) levels were measured. In addition, serum total protein, albumin, urea and creatinine levels were evaluated. Compared with the control animals, urea levels increased significantly in all groups ( P < 0.001). GSH activities and MDA levels increased in xylene and xylene + HCHO groups ( P < 0.05). No statistically considerable differences were found in SOD, CAT and GSH-Px activities, total protein, albumin and creatinine levels among all groups ( P > 0.05). The present study indicates but not statistically confirms the renal toxicity of the exposures to xylene, HCHO and a mixture of them.

This study aimed to assess potential feeding effect of camel milk curd mass and its mixes to experimental rat’s blood serum biochemical parameters, enzymatic activity and the peptide toxicity. Fifty healthy male Sprague-Dawley rats were divided into five groups (n = 10 each). Each group was fed with camel milk pure curd mass and its mixes for 16 days. At the end of the experiment, rats were sacrificed to collect the samples from the blood serum. Blood serum biochemical parameters total protein, cholesterol, glucose, albumin, triglycerides; the enzymatic activities of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase were determined on the A25 automatic analyser, and peptide toxicity analysed by the reference method. The statistical data have shown no significant differences in body weight gain in all groups. Total protein decreased in group II, IV, and V; however, it increased in group III compared to the control group. Cholesterol grew up in group II and it slightly increased in group V, dropped in groups III and IV compared to group I result. Glucose increased in groups II, III, IV compared to group I; still, group V results show a slight decrease. Albumin decreased in group IV, yet in group V it increased than the group I result. Simultaneously, groups II and III results were changed with less percentage. Triglyceride grew up in groups II, V, and it dropped significantly in groups III, IV compared to the control group. De Ritis ratio of enzymes in groups II, III, and IV fluctuated between 1.31 and 0.98 IU/L; however, group V demonstrated significant data versus group I. Diets peptide toxicity in all groups was lower than control group data. The experimental results indicated that curd mass from camel milk could be used as a pure or with additives and it did not discover the observed side effects.

The response of activated skeletal muscle to a ramp stretch is complex. Force rises rapidly above the isometric plateau during the initial phase of stretch. However, after a strain of ∼1–2%, force yields and continues to rise but with a slower slope. The resistance to stretch during the initial phase can be characterized by the stiffness of the muscle and/or the preyield modulus ( Epre). Similarly, a measure of modulus also can be used to characterize the postyield modulus response ( Epost). This study examined the effects of muscle atrophy and altered myosin heavy chain (MyHC) isoform composition on both Epre and Epost. Female Sprague-Dawley rats were assigned to 1) control group, 2) a hypothyroid group, 3) a hyperthyroid group, 4) a hindlimb suspension group, and 5) a hindlimb suspension + hyperthyroid group. These interventions were used either to alter the MyHC isoform composition of the muscle or to induce atrophy. Soleus muscles were stretched at strain rates that ranged from ∼0.15 to 1.25 muscle length/s. The findings of this study demonstrate that 4 wk of hindlimb suspension can produce a large (i.e., 40–60%) reduction in Epre. Hindlimb suspension did not produce a proportional change in Epost. Analyses of the Epre-strain rate relationship demonstrated that there was little dependence on MyHC isoform composition. In summary, the disproportionate decrease in Epre of atrophied muscle has important implications with respect to issues related to joint stability, especially under dynamic conditions and conditions where the static joint stabilizers (i.e., ligaments) have been compromised by injury.

Summary The aim of this study was to evaluate dichlorvos toxicity in terms of nitro-oxidative stress by determining 3-nitrotyrosine (3-NT) levels in the fore, mid, and hindbrain regions in acutely exposed rats. Male Sprague- Dawley rats were randomly allocated to three groups of eight. Group 1 was administered a single intraperitoneal dichlorvos dose of 1.8 mg kg-1 (0.1xLD50) and group 2 a dose of 9 mg kg-1 (0.5xLD50). The control group received 0.5 mL saline solution via the same route. 3-NT and tyrosine (TYR) levels were measured using high performance liquid chromatography with a photodiode array detector (HPLC-PDA) and expressed as a ratio of 3-NT to TYR. The 3-NT/1000 TYR ratios increased significantly in the fore-, mid- and hindbrains of the exposed groups compared to control (p<0.01). In the forebrain, the increase was also significant between the treated groups. Our study has confirmed that acute exposure to dichlorvos leads to nitro-oxidative stress in the brain and that 3-NT may play a role in the mechanism of dichlorvos neurotoxicity.

The present experiment was conducted to study the effects of different levels of oligosaccharide(FOS) and complex CPP preparation(CPP) in diets on growth performance and serum biochemical parameters for weaned piglets. 240 crossbred DLY( Duroc× Landrace× Yorkshire) 21-day-old weaning piglets, about 7.2 kg in initial body weight were selected and randomly divided into 6 groups, with 2 repeats for each group, 20 each repeat(half male and half female.). Group 1, as the control group, was fed base diet(BD), group 2 was fed BD adding 0.10% aureomycin, group 3, 4, 5, 6, respectively, was fed BD adding 0.05% CPP, 0.05% FOS, 0.20% FOS and 0.10% FOS + 0.05% of CPP. Feeding trial was conducted for 35 days. Results: (1) The combination of 0.10%FOS and 0.05%CPP(group 6) were added, the average daily gain of weaned pigs increased by 13.71 % (P < 0.05) and the feed conversion ratio was reduced by 7.41 % (P < 0.05) compared with control. The diarrhea rate of piglets decreased by 94.63 % (P < 0.01) compared with the control (p < 0.05), and decreased by 62.86 % (P <0.05) compared with group 2.(2)There was no significant difference in the apparent digestibility of nutrient, such as ether extract and crude protein, by adding FOS and CPP individually or jointly in diet of weaned piglets, but apparent digestibility of crude fiber increased , to acertain extent, and there was no significant difference with diet adding 0.10 % of antibiotics (group 2)(P >0.05).(3) There was no significant difference in serum biochemical indexes, such as serum total protein, albumin, urea nitrogen, cholesterol, triglycerides, alanine aminotransferase, aspartate aminotransferase and sugar by adding FOS and CPP individually or jointly in diet of weaned piglets (P > 0.05);there was no significant effect in serum immunoglobulin IgG and IgM (P> 0.05), but the serum IgE content was increased in different degrees. Conclusion: Adding FOS and CPP in diet for weaned piglets can improve the growth performance, reduce diarrhea rate of piglets, and promote digestion and absorption of feed nutrients, without significant difference between them and diet adding antibiotics. Among them, the effect of adding 0.10%FOS+0.05%CPP was the best.

The influence of age on serum biochemical components in the domestic rabbit was investigated. The serum total protein (g/dL), albumin (g/dL), globulin (g/dL), creatinine (mg/dL), urea mg/dL, cholesterol (mg/dL), Alkaline phosphatase (ALP; iu/l), Aspartate aminotranferase (AST; iu/l) and Alanine amimotransferase (ALT; iu/l) were assessed in 24 male rabbits at 7, 14 and 21 weeks of age to represent weaners, growers and pubertal stages. Data were analysed using descriptive statistics and analysis of variance at á . Results of 0.05 serum biochemistry revealed a steady significant increase in total protein (4.2±0.3; 5.3±0.5; 6.9±0.2), albumin (2.2±0.7; 2.8±0.4; 4.0±0.2), globulin (1.9±0.3; 2.5±0.4, 2.9±0.2), cholesterol (49.5±1.1; 56.4±4.2; 62.4±5.4) and creatinine (0.8±0.1; 1.2±0.1; 1.9±0.3) at 7, 14 and 21 weeks respectively. Serum urea levels were similar across the ages. The values recorded for ALP and AST at 7 weeks (39.6±7.5; 101.1±6.7) and 14 weeks (38.7±6.4; 104.5±8.7) were significantly higher than the 21 weeks (33.5±2.7; 95.8±5.9) while ALT at 7 weeks (27.33±5.6) was significantly lower than 14 (38.2±3.9) and 21 weeks (37.4±3.0). The age of the rabbits had a significant effect on their serum biochemical parameters. They should therefore be grouped according to age when setting serum reference values for them.

Objectives: Nephrotoxicity is a renal-specific situation in which the excretion of toxic metabolites is reduced due to toxic agents and drugs. Gentamicin is an antibiotic belongs to aminoglycoside group which may induce nephrotoxicity due to induction of oxidative stress. Curcumin is a component of traditional medicine with significant nephroprotective effect. Therefore, the objective of the present study was to evaluate the nephroprotective effect of curcumin on gentamicin-induced nephrotoxicity. Methods: A total of 30 male Sprague-Dawley rats were used which divided into Group 1 (n=10): Rats treated with distilled water 5 ml/kg plus normal saline 5 ml/kg for 12 days, Group 2 (n=10): Rats treated with distilled water 5 ml/kg plus gentamicin 100 mg/kg for 12 days, and Group 3 (n=10): Rats treated with curcumin 100 mg/kg plus gentamicin 100 mg/kg for 12 days. Blood urea, serum creatinine, malondialdehyde (MDA), kidney injury molecule (KIM-1), and cystatin-C were measured in both control and experimental groups. Results: Rats treated with gentamicin showed nephrotoxicity as evident by significant elevation in blood urea, serum creatinine, KIM-1, MDA, and cystatin-C sera levels. Curcumin leads to significant reduction of blood urea and serum creatinine compared to gentamicin group, p<0.05. Curcumin also reduced MDA, KIM-1, and cystatin-C sera levels significantly compared to gentamicin group, p<0.01. Conclusion: Curcumin produced significant nephroprotective effect on gentamicin-induced nephrotoxicity through modulation of oxidative stress and inflammatory biomarkers.

Background: Sulodexide is a powerful antithrombin agent with reno-protective property. However, whether it has beneficial effects on Contrast-Induced Nephropathy (CIN) remained elusive. In the current study, we evaluated the therapeutic effects of Sulodexide on CIN and investigated the potential mechanisms. Methods: CIN model was induced by intravenous injection of indomethacin, followed by Ioversol and L-NAME. Sprague-Dawley rats were divided into 4 groups: control group, CIN group, CIN+vehicle group (CIN rats pretreated with vehicle) and CIN+ Sulodexide (CIN rats pretreated with Sulodexide). Sulodexide or an equivalent volume of vehicle was intravenously delivered 30 min before the induction of CIN. All the animals were sacrificed at 24h after CIN and tissues were harvested to evaluate renal injury, kidney oxidative stress and apoptosis levels. Plasma antithrombin III (ATIII) activities were also measured. Results: Compared to the untreated CIN group, improved renal function, reduced tubular injury, decreased levels of oxidative stress and apoptosis were observed in CIN rats receiving Sulodexide injection. In addition, we also found that ATIII activity was significantly higher in Sulodexide-administered group than that in vehicle-injected CIN rats. For in vitro studies, HK2 cells were exposed to Ioversol and the cyto-protective effects of Sulodexide were also determined. Sulodexide pretreatment protected HK2 cells against the cytotoxicity of Ioversol via inhibiting caspase-3 activity. Preincubation with Sulodexide could also attenuate H2O2-induced increases in ROS, apoptosis and caspase-3 levels. Conclusions: Taken together, Sulodexide could protect against CIN through activating ATIII, and inhibiting oxidative stress, inflammation and apoptosis.

Our aim was to evaluate effect of etanercept on oxidative stress parameters in rats with experimental peritonitis and investigate the availability of etanercept usage in the treatment of peritonitis in the future. Twenty-eight rats were divided into four groups as control (group 1), peritonitis (group 2), peritonitis + cefazolin sodium (group 3), and peritonitis + cefazolin sodium + etanercept (group 4). Peritoneal tissue and blood samples were taken from all of the rats for histopathological and biochemical examination. The oxidative stress parameters were examined in blood and tissue samples. It was observed that rats with peritonitis benefit from cefazolin sodium treatment. Evaluating the effectiveness of etanercept was our main objective for this study. In this perspective, we compared group 3 and group 4 and found statistically significant decreases in oxidative parameters and statistically significant increases in antioxidants in serum and tissue samples in group 4. It is observed that there was a significant contribution of etanercept on biochemical and also histopathological results. As a result, the TNF-αinhibitor, etanercept, in addition to antibiotics given in the early treatment of peritonitis results in more significant improvement of histopathological and oxidative parameters as compared to antibiotics alone.

The aim of the study was to investigate the effects of different doses of acetylsalicylic acid (ASA) and sodium salicylate (SS) administration on weight gains and serum biochemical parameters in broiler chickens. This study was performed to investigate the safety of ASA and SS in chickens in conditions of moderate overdose. Seventy five broiler chickens (Sanhuangji breed) were divided into control and experimental groups. The chickens were administered orally with ASA or SS in the following daily doses: 50, 100 and 200 mg/kg for 20 days. In the course of the experiment weight gains were recorded. On day 14 and day 20 the chickens were sampled for blood biochemical analysis. Investigated parameters included serum total protein, albumin, uric acid, creatinine, potassium, sodium and calcium contents as well as alanine transaminase activity. After the last samplings the chickens were dissected and liver and kidney to body weight ratios were recorded. It was demonstrated that SS at a dose of 200 mg/kg increased serum total protein level. ASA increased serum proteins at a dose of 100 mg/kg but not at a higher dose. At the dose of 200 mg/kg ASA increased the kidney to body weight ratio. In all salicylate treated groups the liver weights were lower as compared to the control. No clinical manifestations of intolerance were observed. It is assumed that chickens tolerate ASA and SS well, even in doses producing distinct side effects in several mammalian species.
