Abstract Background Pleurotus ostreatus is an edible mushroom popularly cultivated worldwide. Distilled grain waste (DGW) is a potential substrate for P. ostreatus cultivation. However, components in DGW restrict P. ostreatus mycelial growth. Therefore, a cost-effective approach to facilitate rapid P. ostreatus colonization on DGW substrate will benefit P. ostreatus cultivation and DGW recycling. Results Five dominant indigenous bacteria, Sphingobacterium sp. X1, Ureibacillus sp. X2, Pseudoxanthomonas sp. X3, Geobacillus sp. X4, and Aeribacillus sp. X5, were isolated from DGW and selected to develop a consortium-based microbial agent to compost DGW for P. ostreatus cultivation. Microbial agent inoculation led to faster carbohydrate metabolism, a higher temperature (73.2 vs. 71.2 °C), a longer thermophilic phase (5 vs. 3 days), and significant dynamic changes in microbial community composition and diversity in composts than those of the controls. Metagenomic analysis showed the enhanced microbial metabolisms, such as xenobiotic biodegradation and metabolism and terpenoid and polyketide metabolism, during the mesophilic phase after microbial agent inoculation, which may facilitate the fungal colonization on the substrate. In accordance with the bioinformatic analysis, a faster colonization of P. ostreatus was observed in the composts with microbial inoculation than in control after composting for 48 h, as indicated from substantially higher fungal ergosterol content, faster lignocellulose degradation, and higher lignocellulase activities in the former than in the latter. The final mushroom yield shared no significant difference between composts with microbial inoculation and control, with 0.67 ± 0.05 and 0.60 ± 0.04 kg fresh mushroom/kg DGW, respectively (p > 0.05). Conclusion The consortium-based microbial agent comprised indigenous microorganisms showing application potential in composting DGW for providing substrate for P. ostreatus cultivation and will provide an alternative to facilitate DGW recycling.

Pleurotus ostreatus, an edible mushroom is an essential food product. Recognised as one of the cheapest sources of proteins, their high nutritional and anti-nutritional properties are of immense importance to humans. The present study evaluated the effect of sawdust and dried plantain leaves on the growth and yield of Pleurotus ostreatus. Sawdust and dried plantain leaves were composted into seven substrates; as single substrates and at different combinations. The composted substrates include sawdust 100 % (M0), Dry plantain leaves 100% (M1), Dry plantain leaves + sawdust at the ratio of 1:4 (M2), 2:3 (M3), 3:2 (M4), 1:1 (M5) and 4:1(M6). Growth parameters of interest assessed flush include mycelia running rate (MRR), area of pileus, length of the stipe, the girth of the stipe, fresh weight and dry weight, number of fruit body, total yield and biological efficiency. At the end of the three weeks spawn running period, M0 was overall best in supporting mycelia running rate with a mean MRR of 16.00 cm. M1 substrates produced mushroom with longer and bigger stipes (7.17 cm) at the first flush. A higher number of fruit bodies (82.66), total yield (130.35 g), and biological efficiency (43.45 g) were all seen in M0 substrates. Sawdust at 100 % (M0) proved to be the best substrate for the cultivation of Pleurotus ostreatus.

Penambahan nutrisi perlu dilakukan untuk meningkatkan produktivitas pertumbuhan jamur tiram putih (Pleurotus ostreatus). Salah satu bahan yang dapat digunakan adalah serbuk jagung. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan serbuk jagung pada komposisi media tanam terhadap pertumbuhan miselium bibit F1 jamur tiram putih (Pleurotus ostreatus) dan mengetahui konsentrasi serbuk jagung yang paling baik digunakan pada pertumbuhan  miselium bibit F1 jamur tiram putih (Pleurotus ostreatus). Penelitian ini mengunakan Rancangan Acak Lengkap (RAL) 4 perlakuan dan 3 ulangan sehingga diperoleh 12 satuan percobaan; P0 (perlakuan kontrol), P1 (25%), P2 (50%), dan P3 (100%). Variabel yang diukur adalah persentase pertumbuhan miselium yang diamati pada hari ke-6, 12 dan 18 dan waktu penyebaran miselium Hari Setelah Inokulasi (HSI). Data hasil penelitian dianalisis menggunakan analysis of variance (ANOVA) dengan uji lanjut menggunakan uji Beda Nyata Terkecil (BNT). Hasil penelitian menunjukkan bahwa ada pengaruh pada pertumbuhan miselium bibit F1 jamur tiram putih (Pleurotus ostreatus) dengan penambahan serbuk jagung pada komposisi media tanam. Selain Itu, konsentrasi serbuk jagung yang memberikan hasil yang paling baik terhadap pertumbuhan miselium adalah P2 dengan konsentrasi 50%. Kata kunci : miselium F1; Pleurotus ostreatus; serbuk jagung  ABSTRACTAdditional nutrition needs to be done to increase the productivity of oyster mushroom (Pleurotus ostreatus) growth. One of the materials that can be used is corn powder. This study aimed to determine the effect of corn powder on the planting medium composition towards mycelium growth of oyster mushroom F1 seeds (Pleurotus ostreatus) and to find the best concentration of corn powder to grow mycelium seeds F1 oyster mushroom (Pleurotus ostreatus). This research was an experimental study using a Completely Randomized Design (CRD) with 4 treatments namely; P0 (control treatment), P1 (25%), P2 (50%), and P3 (100%), and 3 repetitions so that obtained 12 experimental units. The variables measured were the mycelium growth percentage on days 6, 12, and 18, and the mycelium widespread time after the day of inoculation. The data were analyzed using ANOVA then continued on the Least Significant Difference (LSD) test. The results showed that the addition of corn powder on oyster mushroom growing media affected mycelium growth of the oyster mushroom F1 seeds (Pleurotus ostreatus). It also found that the P2 with a 50% concentration of corn powder gave the widest mycelium growth.  Keywords: corn powder; mycelium; Pleurotus ostreatus

The batik waste industry is one source of water pollution. It must undergo treatment before discharge into the environment. Batik waste treatment technologies can be done biologically, chemically, physically, or combination of three processes. This research applied a combination of biology and physic technologies, namely used spent mushroom of Pleurotus ostreatus as the decolorization agent. Mycelium of P. ostreatus was used as biological agent, and cellulose from spent mushroom was used as physical agent. The aims of this research were to determine the effect of spent mushroom with variation incubation time on batik waste decolorization, and to know the optimum incubation time with highest decolorization batik waste percentage using spent mushroom of P. ostreatus. This research used experimental method with completely randomized design (CRD). The data obtained were statistically analyzed using Varian Test (ANOVA) on confidence level of 95% and 99% and continued with Honest Significant Difference Test (BNJ) with 95% confidence level. The result showed that variation incubation time treatment of batik waste decolorization used spent mushroom of P. ostreatus provide a highly signification effect on batik waste decolorization. The optimum incubation time was 60 hour (LW5), with decolorization percentage was 85,64%.

Pleurotus ostreatus is a well-known edible mushroom species which shows fast growth. The fungus can be used for medical, nutritional, filter, or packaging purposes. In this study, cultivation experiments were carried out with Pleurotus ostreatus growing on polyacrylonitrile (PAN) nanofiber mats in the presence of saccharose and Lutrol F68. The aim of this study was to find out whether modified PAN nanofiber mats are well suited for the growth of fungal mycelium, to increase growth rates and to affect mycelium fiber morphologies. Our results show that Pleurotus ostreatus mycelium grows on nanofiber mats in different morphologies, depending on the specific substrate, and can be used to produce a composite from fungal mycelium and nanofiber mats for biomedical and biotechnological applications.

Background: Glioblastoma (GB) is one of the most common (~30%) and lethal cancers of the central nervous system. Although new therapies are emerging, chemoresistance to treatment is one of the major challenges in cancer treatment. Brain cytoplasmic 200 (BC200) RNA, also known as BCYRN1, is a long noncoding RNA (lncRNA) that has recently emerged as one of the crucial members of the lncRNA family. BC200 atypical expression is observed in many human cancers. BC200 expression is higher in invasive cancers than in benign tumors. However, the clinical significance of BC200 and its effect on GB multiforme is still unexplored and remains unclear. Methods: BC200 expression in GB patients and cell lines were investigated through RT-qPCR, immunoblotting, and immunohistochemistry analysis. The biological importance of BC200 was investigated in vitro and in vivo through knockdown and overexpression. Bioinformatic analysis was performed to determine miRNAs associated with BC200 RNA. Results: Our findings revealed that in GB patients, BC200 RNA expression was higher in blood and tumor tissues than in normal tissues. BC200 RNA expression have a statistically significant difference between the IDH1 and P53 status. Moreover, the BC200 RNA expression was higher than both p53, a prognostic marker of glioma, and Ki-67, a reliable indicator of tumor cell proliferation activity. Overexpression and silencing of BC200 RNA both in vitro and in vivo significantly modulated the proliferation, self-renewal, pluripotency, and temozolomide (TMZ) chemo-resistance of GB cells. It was found that the expressions of BC200 were up-regulated and that of miR-218-5p were down-regulated in GB tissues and cells. miR-218-5p inhibited the expression of BC200. Conclusions: This study is the first to show that the molecular mechanism of BC200 promotes GB oncogenicity and TMZ resistance through miR-218-5p expression modulation. Thus, the noncoding RNA BC200/miR-218-5p signaling circuit is a potential clinical biomarker or therapeutic target for GB.
