Near Infrared Spectroscopy Estimation of Feeding Value of Forage Perennial Grasses in Breeding Programmes by Global and Specific Calibrations. Estimation of Chemical Composition and Digestibility

Near infrared (NIR) spectral analysis with a NIRSystems 6500 monochromator was applied to evaluate accuracy of predictive models for forage quality in clone breeding processes of the original Bulgarian varieties over different cuts and years. The varieties were the perennial grasses: Dactylis glomerata L., Festuca arundinacea Schreb. and Bromus inermis Leyss. Global calibrations for the 418 perennial grass samples and specific calibrations for each single grass species and internal cross-validations were performed by the PLS regression method. The effect of different spectral data pre-treatments was investigated on the residual standard errors of the NIR predictive models. Among 60 calibration equations, the model with the lowest SECV value was retained for each parameter in each database. No particular data pre-treatment was really better than the other ones. Generally, the best results of the global calibrations were obtained with SNVD and MSC. For the specific calibrations, SNVD and WMSC were the best treatments. In both cases, the first or second derivatives were needed after the first pre-treatment. Chemical composition and in vitro enzymatic digestibility of clones were predicted with accuracy similar to that of classical laboratory methods. For the cell wall component contents, the standard errors of cross-validation SECV(%DM) ranged from 0.49 for ADL ( Festuca) to 2.02 for NDF ( Dactylis). The digestibilities of dry and organic matter, IVDMD and IVOMD, were estimated with SECVs from 2.6 to 3.0%, the relative intake, from 0.06 to 0.09 rel% body weight and the relative feeding value, from 4.39 to 5.64 rel%. The global calibration models offer an acceptable accuracy for the estimation of the cell wall nutrient contents, the digestibility and the nutritive value. The standard errors of prediction of specific single species calibrations with smaller numbers of terms were lower in 60% of the cases than those obtained from the best global calibrations with higher numbers of terms. On average, SECVs from specific calibrations are better than those from global calibrations, but the differences are quite small, and for the prediction of totally new samples (new crops, another year), the global calibrations will detect less outlier samples. Even with very high variability between cuts and years, NIR spectroscopy is able via ANOVA GL Models to sort clones on their feeding value and to provide relevant information for the breeding programmes.

Bracken cut in June was ensiled in various ways, and some of the products were examined for chemical composition, digestibility and palatability. The main findings were:1. Overheated silage was moderately palatable to sheep and cattle, but its digestibility was low and it approximated to wheat or barley straw in feeding value.2. Silage subjected to less heat was unpalatable, and in some cases was absolutely refused by sheep. The ensiled stems were preferred to the leaves. The digestibility was low but slightly better than that of the overheated silage.3. Ensiling seriously affects the digestibility of bracken protein. In two silages none of the true protein was digestible, and in one the digestibility of this constituent was only 12·9%.It is concluded that the ensiling of bracken is unlikely to prove profitable.

Metabolisable energy (ME) is frequently used as the sole indicator of forage quality by researchers, rural professionals and farmers, but it is hardly ever measured and is not always a good predictor of feeding value. Forage ME is usually calculated from chemical composition and digestibility, often by near infrared reflectance spectroscopy (NIRS). Although ME is superior to dry matter (DM) as a measure of feeding value and can indicate forage quality, it should not be used to predict animal production. The ME content of DM may imply a potential for production, but other components of the diet, especially protein, structural fibre and feed availability will provide more information than ME alone. Researchers, rural professionals and farmers should understand the basis for measuring ME, exercise discretion when using ME, and include fibre and protein concentrations in their criteria for feed appraisal. In the present study, the potential of application of near infrared reflectance (NIR) spectroscopy for the estimation of the chemical composition of traditional (village style) sausages was examined. The chemical composition (moisture, ash, protein and, fat) was determined by standard reference methods. For the development of the calibration model, 39 samples of traditional fresh sausages were used, while for external validation, 10 samples of sausages were used. The correlation coefficients of calibration (RMSEC) and standard errors (SEC) were 0.92 and 1.58 (moisture), 0.77 and 0.18 (ash), 0.87 and 0.89 (protein) and 0.93 and 1.73 (fat). The cross-validation correlation coefficients (RMSECV) and standard errors (SECV) were 0.86 and 2.13 (moisture), 0.56 and 0.26 (ash), 0.78 and 1.17 (protein), and 0.88 and 2.17 (fat). The results of the calibration model showed that NIR spectroscopy can be applied to estimate with very good precision the fat content of traditional village-style sausages, whereas moisture and protein content can be estimated with good accuracy. The external validation confirmed the ability of NIR spectroscopy to predict the chemical composition of sausages.

The purpose of this study was to evaluate the anti-fatigue activity of maca hydroalcoholic extract (ME), which mainly contains macamides and polysaccharides. ME was prepared by circumfluence extraction with enzymatic pre-treatment. Anti-fatigue activity of ME was investigated in weight-loaded forced swimming mice, with pure macamides and commercially available maca tablet as positive control. Compared with normal group, pure macamides treatment group could prolong the swimming time to exhaustion, but there was no statistically significant difference (P > 0.05); while ME (middle-dose and high-dose groups) could effectively prolong the swimming durations (P < 0.05). Supplementation with pure macamides significantly decreased blood lactic acid (BLA), whereas ME significantly increased hepatic glycogen (HG), decreased BLA, and blood urea nitrogen (BUN) compared with those in normal control (P < 0.05). The results suggested that the anti-fatigue effect of ME was better than that of pure macamides, which can be explained by the increase of glycogen storage and the reduction of metabolites accumulation.

Background: Cell lytic enzyme is a kind of highly evolved protein, which can destroy the cell structure and kill the bacteria. Compared with antibiotics, cell lytic enzyme will not cause serious problem of drug resistance of pathogenic bacteria. Thus, the study of cell wall lytic enzymes aims at finding an efficient way for curing bacteria infectious. Compared with using antibiotics, the problem of drug resistance becomes more serious. Therefore, it is a good choice for curing bacterial infections by using cell lytic enzymes. Cell lytic enzyme includes endolysin and autolysin and the difference between them is the purpose of the break of cell wall. The identification of the type of cell lytic enzymes is meaningful for the study of cell wall enzymes. Objective: In this article, our motivation is to predict the type of cell lytic enzyme. Cell lytic enzyme is helpful for killing bacteria, so it is meaningful for study the type of cell lytic enzyme. However, it is time consuming to detect the type of cell lytic enzyme by experimental methods. Thus, an efficient computational method for the type of cell lytic enzyme prediction is proposed in our work. Method: We propose a computational method for the prediction of endolysin and autolysin. First, a data set containing 27 endolysins and 41 autolysins is built. Then the protein is represented by tripeptides composition. The features are selected with larger confidence degree. At last, the classifier is trained by the labeled vectors based on support vector machine. The learned classifier is used to predict the type of cell lytic enzyme. Results: Following the proposed method, the experimental results show that the overall accuracy can attain 97.06%, when 44 features are selected. Compared with Ding's method, our method improves the overall accuracy by nearly 4.5% ((97.06-92.9)/92.9%). The performance of our proposed method is stable, when the selected feature number is from 40 to 70. The overall accuracy of tripeptides optimal feature set is 94.12%, and the overall accuracy of Chou's amphiphilic PseAAC method is 76.2%. The experimental results also demonstrate that the overall accuracy is improved by nearly 18% when using the tripeptides optimal feature set. Conclusion: The paper proposed an efficient method for identifying endolysin and autolysin. In this paper, support vector machine is used to predict the type of cell lytic enzyme. The experimental results show that the overall accuracy of the proposed method is 94.12%, which is better than some existing methods. In conclusion, the selected 44 features can improve the overall accuracy for identification of the type of cell lytic enzyme. Support vector machine performs better than other classifiers when using the selected feature set on the benchmark data set.

Abstract Background The current first-line treatments for uncomplicated malaria recommended by the National Malaria Control Programme in Mali are artemether–lumefantrine (AL) and artesunate–amodiaquine (ASAQ). From 2015 to 2016, an in vivo study was carried out to assess the clinical and parasitological responses to AL and ASAQ in Sélingué, Mali. Methods Children between 6 and 59 months of age with uncomplicated Plasmodium falciparum infection and 2000–200,000 asexual parasites/μL of blood were enrolled, randomly assigned to either AL or ASAQ, and followed up for 42 days. Uncorrected and PCR-corrected efficacy results at days 28 and 42. were calculated. Known markers of resistance in the Pfk13, Pfmdr1, and Pfcrt genes were assessed using Sanger sequencing. Results A total of 449 patients were enrolled: 225 in the AL group and 224 in the ASAQ group. Uncorrected efficacy at day 28 was 83.4% (95% CI 78.5–88.4%) in the AL arm and 93.1% (95% CI 89.7–96.5%) in the ASAQ arm. The per protocol PCR-corrected efficacy at day 28 was 91.0% (86.0–95.9%) in the AL arm and 97.1% (93.6–100%) in the ASAQ arm. ASAQ was significantly (p < 0.05) better than AL for each of the aforementioned efficacy outcomes. No mutations associated with artemisinin resistance were identified in the Pfk13 gene. Overall, for Pfmdr1, the N86 allele and the NFD haplotype were the most common. The NFD haplotype was significantly more prevalent in the post-treatment than in the pre-treatment isolates in the AL arm (p < 0.01) but not in the ASAQ arm. For Pfcrt, the CVIET haplotype was the most common. Conclusions The findings indicate that both AL and ASAQ remain effective for the treatment of uncomplicated malaria in Sélingué, Mali.

Abstract Background Product quality and production efficiency of Atlantic salmon are, to a large extent, influenced by the deposition and depletion of lipid reserves. Fillet lipid content is a heritable trait and is unfavourably correlated with growth, thus genetic management of fillet lipid content is needed for sustained genetic progress in these two traits. The laboratory-based reference method for recording fillet lipid content is highly accurate and precise but, at the same time, expensive, time-consuming, and destructive. Here, we test the use of rapid and cheaper vibrational spectroscopy methods, namely near-infrared (NIR) and Raman spectroscopy both as individual phenotypes and phenotypic predictors of lipid content in Atlantic salmon. Results Remarkably, 827 of the 1500 individual Raman variables (i.e. Raman shifts) of the Raman spectrum were significantly heritable (heritability (h2) ranging from 0.15 to 0.65). Similarly, 407 of the 2696 NIR spectral landscape variables (i.e. wavelengths) were significantly heritable (h2 = 0.27–0.40). Both Raman and NIR spectral landscapes had significantly heritable regions, which are also informative in spectroscopic predictions of lipid content. Partial least square predicted lipid content using Raman and NIR spectra were highly concordant and highly genetically correlated with the lipid content values ($${r}_{\text{g}}$$ r g = 0.91–0.98) obtained with the reference method using Lin’s concordance correlation coefficient (CCC = 0.63–0.90), and were significantly heritable ($${h}^{2}$$ h 2 = 0.52–0.67). Conclusions Both NIR and Raman spectral landscapes show substantial additive genetic variation and are highly genetically correlated with the reference method. These findings lay down the foundation for rapid spectroscopic measurement of lipid content in salmonid breeding programmes.
