Țaga cheese is a traditional Romanian smear-ripened cheese made from bovine milk and identified with the name of the village and caves where it is produced. As no previously reported microbiological and chemical studies have been undertaken on this product, this research aimed to investigate the microbiological and biochemical characteristics which ensure the uniqueness of Țaga cheese during the ripening process, to inform producers as to key quality determinants. Cheese samples, consisting of retail blocks, were collected on days 2, 5, 12, 18, and 25 of the ripening process. The evolution of lactic microbiota during the production and maturation of traditional cheeses involves isolating lactic acid microorganisms present in cheese. Cheese samples were analyzed for pH, fat, NaCl, fatty acids, and volatile compounds. The microbial ecosystem naturally changes during the maturation process, leading to variation in the microorganisms involved during ripening. Our results show that specific bacteria were identified in high levels during the entire ripening process and may be responsible for milk fat lipolysis contributing directly to cheese flavor by imparting detailed fatty acid flavor notes, or indirectly as precursors formation of other flavor compounds.

Abstract A published liquid chromatographic (LC) method was modified for quantitation of phenylbutazone (PBZ) residues in the range of 25-300 ng/mL in bovine milk. Milk samples (1 m└) were diluted with absolute ethanol and 25% NH4OH. Diethyl ether and petroleum ether were added sequentially to the milk extract and the mixture was agitated on a Vortex mixer to partition out milk fat. The organic phase was removed and discarded. Tetrahydrofuran- hexane (1 + 4) was added to the aqueous phase and the extract was acidified with 3M HCI. The samples were mixed on a Vortex mixer for 30 min, and centrifuged. The organic layer, containing PBZ, was transferred to a clean test tube. The organic solvents were evaporated to dryness under a stream of N2 at room temperature. The resulting extract was dissolved in 1 m└ mobile phase and filtered before injection. The chromatographic system was a C18 reversed-phase column connected to a UV detector set at 264 nm. Recoveries of PBZ from raw bovine milk fortified at 25- 300 ng/mL ranged from 79 to 84%; relative standard deviations (RSDs) ranged from 6 to 7%. The RSDs for incurred PBZ quantitated from 34 to 229 ng/mL ranged from 1 to 4%.

This study is motivated the identification of policies issued by the regional Governmentof Rokan Hulu in the form of Regulatory region number 1 by 2015 on the determination of thevillage and Indigenous Village. Political dynamics based on various interests against themanufacture of, and decision-making in the process of formation of the corresponding localregulations determination of Indigenous Villages in the Rokan Hulu is impacted to a verysignificantamount of changes from the initial draft of the number i.e. 21 (twenty one) the villagebecame Customary 89 (eighty-nine) the Indigenous Villages who have passed. Type of thisresearch is a qualitative descriptive data analysis techniques. The research aims to describe theState of the real situation in a systematic and accurate fact analysis unit or related research, aswell as observations of the field based on the data (information). Method of data collectionwas done with interviews, documentation, and observations through fieldwork (field research).The results of the research on the process of discussion of the draft local regulations andmutual agreement about Designation of Indigenous Villages in the Rokan Hulu is, showed thatthe political dynamics that occur due to the presence of various political interests, rejectionorally by Villagers who were judged to have met the requirements of Draft Regulations to beformulated and the area for the set to be Indigenous Villages, and also there is a desire fromsome villages in the yet to Draft local regulations in order to set the Indigenous village , there isa wide range of interests of these aspects influenced the agreement to assign the entire localVillage which is in the Rokan Hulu become Indigenous village, and the village of Transmigrationinto administrative Villages where the initiator of the changes in the number of IndigenousVillages in the Rokan Hulu it is the desire of the local Government of its own.
