The present study demonstrated the antioxidant activity of 2-bromo-3-hydroxy-2-nitroproylcinnamte versus alcohol induced oxidative damage in albino wistar rats. In this study, 30 % alcohol exposed rats were found to be more prone to peroxidative risk as they are calculated by species of thiobarbituric acid. It was observed that after the rats induction with 30% alcohol, concentration of lipid peroxidation has been obtained expressively (p ≤ 0.001) high in a liver and serum, beside with concomitant substantial (p ≤ 0.001) reduced in enzymatic and non-enzymatic antioxidants levels like catalase (CAT), superoxide dismutase (SOD), glutathione reductase (GR), glutathione peroxidase (GPx), vitamin E (tocopherol), reduced glutathione (GSH), glutathione-s-transferase (GST), vitamin C (ascorbic acid), β-carotene as well as ceruloplasmin in serum along with liver, only 30% alcohol was treated. If rats obtained 2- bromo-3-hydroxy-2-nitroproylcinnamte at the dose level of 20 mg/kg bw/day, PO, for 30 days, the peroxidative damage has been marginal in serum along with liver, alongside efficiently encouraging the potential of antioxidant in the rats treated by alcohol. This study revealed that in liver the raised peroxidative risk is probably allied with alcohol induction pathology that can be decreased by increasing the antioxidant potential by free radical scavenging activity, therefore promising as artificial antioxidants for 2-bromo-3-hydroxy-2-nitroproylcinnamte.

The pathogenesis of cervical cancer is related to oxidative damage caused by persistent infection by one of the oncogenic types of human papillomavirus (HPV). This damage comes from oxidative stress, which is the imbalance caused by the increase in reactive oxygen and nitrogen species and impaired antioxidant mechanisms, promoting tumor progression through metabolic processes. The incorporation of HPV into the cellular genome leads to the expression of oncoproteins, which are associated with chronic inflammation and increased production of reactive oxygen species, oxidizing proteins, lipids and DNA. The increase in these parameters is related, in general, to the reduction of circulating levels of enzymatic antioxidants—superoxide dismutase, catalase, glutathione peroxidase and glutathione-S-transferase; and non-enzymatic antioxidants—reduced glutathione, coenzyme Q10 and vitamins A, C and E, according to tumor staging. In contrast, some enzymatic antioxidants suffer upregulation in the tumor tissue as a way of adapting to the oxidative environment generated by themselves, such as glutathione-S-transferase, reduced glutathione, glutathione peroxidase, superoxide dismutase 2, induced nitric oxide synthase, peroxiredoxins 1, 3 and 6, and thioredoxin reductase 2. The decrease in the expression and activity of certain circulatory antioxidants and increasing the redox status of the tumor cells are thus key to cervical carcinoma prognosis. In addition, vitamin deficit is considered a possible modifiable risk factor by supplementation, since the cellular functions can have a protective effect on the development of cervical cancer. In this review, we will discuss the impact of oxidative damage on cervical cancer progression, as well as the main oxidative markers and therapeutic potentialities of antioxidants.

An experiment was conducted to evaluate the potential of honey, propolis, and bee pollen for the reversal of lipid peroxidation induced by tebuconazole (TEB) in South American catfish (Rhamdia quelen), in which the concentration of thiobarbituric acid reactive substances (TBARS), the activity of the antioxidant enzyme glutathione-S-transferase (GST) and the concentrations of non-enzymatic antioxidants, reduced glutathione (GSH), ascorbic acid, and non-protein thiols were assessed. Honey (0.125 g L-1) and bee pollen (0.05 g L-1) added to the water reverse the production of TBARS induced by TEB, while propolis demonstrated a pro-oxidant effect, inducing an increase in TBARS production. The data presented herein suggest that the addition of water to honey and bee pollen potentially protects against the oxidative stress caused by agrichemicals.

The antioxidant activity and bioactive compounds of different blackberry fruit genotypes from the major Brazilian producer region (three cultivars and four selections) were evaluated and compared to the Cherokee cultivar. Phenolic and anthocyanic extracts were obtained and evaluated for each fruit genotype. The phenolic extracts of selections 02/96 and 07/001 presented higher antioxidant activity than those of cultivars in most assays. This activity was partially correlated to the higher amount of total phenolics in these samples. Thus, the phenolic compounds are probably the major responsible for the antioxidant activity in the diphenyl-2-picrylhydrazyl radical scavenging assay (DPPH), ferric reducing antioxidant power (FRAP) assay and thiobarbituric acid reactive substances (TBARS) assay. Quercetin seems to be responsible for the antioxidant activity of blackberry phenolic extracts in the β-carotene bleaching assay. Concerning anthocyanic extracts, the selection 02/96 and Cherokee cultivar from harvest 2007 had higher antioxidant activity than the other genotypes in most assays. Anthocyanins appear to be the major responsible for the antioxidant activity of anthocyanic extracts in the DPPH and FRAP assays, although ascorbic acid also contributed to the DPPH antioxidant activity. Selection 02/96 appears to have higher antioxidant activity than the commercial cultivars cultivated in the southern Brazil and appears to be promising for nutritional and health purposes.

AbstractThe antioxidant potential of fruit pulp and seeds of extracts of the Annona coriacea, and A. sylvatica(Annonaceae) were investigated, as well contents total phenolics, flavonoids, condensed tannins and ascorbic acid. Was used to determine the antioxidant activity the 1,1-diphenyl-1-picrylhydrazyl free radical (DPPH), β-carotene bleaching and ABTS radical cation method. The total phenol, total flavonoid, condensed tannin, and ascorbic acid contents were measured spectrophotometrically. In this study, the pulp and seeds of the fruits were extracted using methanol/water (8:2) for maceration. The seed extracts of A. coriacea demonstrated a moderate antioxidant effect with free radical scavenging activity of 31.53%, by the DPPH test, 51.59% by the β-carotene bleaching test and 159.50 µM trolx/g of extract in the ABTS assay. We found that the hydromethanolic seed extract of A. coriacea had high total phenol (147.08 ± 4.20 mg of GAE/g of extract) and flavonoid (131.18 ± 2.31 mg of QE/g of extract) content. This indicated that the antioxidant activity of the extracts was related to the contents of these constituents.

In the present study, Okara, a soybean by-product from the production of tofu and soymilk, was processed by solid state fermentation (SSF) with Neurospora crassa Mai 1-1, a filamentous and edible fungus which was firstly isolated in our group. The proximate composition, dietary fiber content, water-soluble polysaccharides and antioxidant activity of Okara was investigated. The crude fat and protein, soluble dietary fiber content and ergosterol content of Okara was significantly increased by SSF with N.crassa . And the antioxidant activity (evaluated by the ABTS radical cation scavenging activity and DPPH radical scavenging activity) of Okara was significantly enhanced by SSF with N. crassa . In addition, emulsion-type sausage was taken as a model system for farther investigation, sausage with fermented Okara power had reduced values of thiobarbituric acid reactive substances (TBARS) ( p < 0.05 ), peroxide value. Therefore, this study demonstrated that fermented Okara showed higher nutrition value and antioxidant activity compared to the non-fermented Okara counterpart, and as an additive has good effect on antioxidant of emulsion-type sausage.

ABSTRACT: Plants that contain antioxidant compounds have attracted increasing interest for their vital role in the attenuation of oxidative damage caused by free radicals and in the treatment of various diseases. The present study investigated the β-ecdysone content and the antioxidant activity of Brazilian ginseng (Pfaffia glomerata) extracts obtained from inflorescences, stems, and roots. The P. glomerata extracts were tested for antioxidant activity using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method, β-carotene bleaching test, and phosphomolybdenum method. The β-ecdysone content of P. glomerata extracts was measured by high-performance liquid chromatography (HPLC). The P. glomerata inflorescences showed the strongest DPPH radical scavenging activity and the strongest antioxidant activity in the β-carotene bleaching assay and phosphomolybdenum test. The roots showed the lowest antioxidant capacity in all of the assays. The concentration of β-ecdysone in the plant organs followed the following decreasing order: inflorescences > stems > roots. The present study showed that P. glomerata inflorescence extract had high antioxidant capacity that could be attributed to the presence of β-ecdysone.

AbstractThe aim of the present study was to examine the antioxidant activity of three Veronica species (Plantaginaceae). The antioxidant potential of various extracts obtained from aerial flowering parts was evaluated by DPPH-free (1,1-diphenyl-2-picrylhydrazyl-free) radical scavenging activity and ferric-reducing antioxidant power assays. Considerable antioxidant activity was observed in the plant samples (FRAP values ranged from 0.97 to 4.85 mmol Fe2+/g, and DPPH IC50 values from 12.58 to 66.34 µg/ml); however, these levels were lower than the activity of the control compound butylated hydroxytoluene (BHT) (FRAP: 10.58 mmol Fe2+/g; DPPH IC50: 9.57 µg/ml). Also, the in vivo antioxidant effects were evaluated in several hepatic antioxidant systems in rats (activities of glutathione peroxidase, glutathione reductase, peroxidase, catalase, xanthine oxidase, glutathione content and level of thiobarbituric acid reactive substances) after treatment with different Veronica extracts, or in combination with carbon tetrachloride (CCl4). Pretreatment with 100 mg/kg b.w. of Veronica extracts inhibited CCl4-induced liver injury by decreasing TBA-RS level, increasing GSH content, and bringing the activities of CAT and Px to control levels. The present study suggests that the extracts analyzed could protect the liver cells from CCl4-induced liver damage by their antioxidative effect on hepatocytes.

Porcupine bezoars (PBs) are masses of undigested calcareous concretions formed within the gastrointestinal tract. There are undocumented claims that PBs have antioxidant activity and can treat cancers. However, limited scientific study has been carried out to verify these traditional claims. Hence, this study was conducted to characterize the chemical profile and validate the antioxidant and anticancer activity against melanoma cells (A375). PB extract was initially subjected to Fourier-transform infrared spectroscopy (FTIR), gas chromatography–mass spectrometry (GCMS), total phenolic content (TPC), and total flavonoid content (TFC) analyses. The bioautography of antioxidant assays, namely 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS), 2,2-diphenyl-1-picrylhydrazy (DPPH), and β-carotene was performed. An in vitro A375 cell viability assay, apoptosis assay, cell cycle arrest assay, and gene expression assay were carried out as well. The experimental finding revealed 5,10-diethoxy-2,3,7,8-tetrahydro-1H,6H-dipyrrolo[1,2-a:1’,2’-d]pyrazine, ursodeoxycholic acid, and cholest-5-en-3-ol (3 beta)-, carbonochloridate are major compounds detected in PB extract. PB extract has low phenolic content, viz. 698.7 ± 0.93 (µg GAE/5 mg dry weight). The bioautography antioxidant assays revealed a potent antioxidant effect (ABTS > DPPH > β-carotene), with free radical scavenging activity. Furthermore, PB extract exhibited dose- and time-dependent inhibition of cancer activity on A375 cells due to the exhibition of apoptosis via an intrinsic pathway.

Gracilariopsis lemaneiformis polysaccharides (GLP) were degraded using pectinase, glucoamylase, cellulase, xylanase, and β-dextranase into low-molecular-weight polysaccharides, namely, GPP, GGP, GCP, GXP, and GDP, respectively, and their antioxidant capacities were investigated. The degraded GLPs showed higher antioxidant activities than natural GLP, and GDP exhibited the highest antioxidant activity. After the optimization of degradation conditions through single-factor and orthogonal optimization experiments, four polysaccharide fractions (GDP1, GDP2, GDP3, and GDP4) with high antioxidant abilities (hydroxyl radical scavenging activity, DPPH radical scavenging activity, reduction capacity, and total antioxidant capacity) were obtained. Their cytoprotective activities against H2O2-induced oxidative damage in human fetal lung fibroblast 1 (HFL1) cells were examined. Results suggested that GDP pretreatment can significantly improve cell viability, reduce reactive oxygen species and malonaldehyde levels, improve antioxidant enzyme activity and mitochondria membrane potential, and alleviate oxidative damage in HFL1 cells. Thus, the enzyme degradation of GLP with β-dextranase can significantly improve its antioxidant activity, and GDP might be a suitable source of natural antioxidants.

Traditional medicines (TM) used to treat symptoms of diabetes by two Cree communities were assessed for their free radical scavenging activity using the stable 1,1-diphenyl-2-picrylhydrazyl radical, and their ability to protect human low-density lipoprotein from Cu2+-mediated oxidation by measuring lag time before the appearance of conjugated dienes and formation of thiobarbituric acid-reactive substances. Water-soluble phenolic content was also measured. A total of 20 medicinal plants from Whapmagoostui and 16 from Mistissini were compared with 16 extracts of plants that were not used medicinally. Medicinal plant extracts, particularly those from Larix laricina, displayed high antioxidant activity, comparable with ascorbic acid, Trolox, and the known antioxidant flavonoids quercetin, epicatechin, catechin. Extracts of Pinaceae and Ericaceae contained the highest levels of phenolics. Factors such as season and area of plant collection, as well as the plant part from which the extract was derived, affected antioxidant activity. Positive correlations were found between the established traditional knowledge of Cree Elders and Healers and the antioxidant activity for medicinal plants used in Mistissini (r = 0.3134; p = 0.058) and in Whapmagoostui (r = 0.5165; p = 0.001). Significant correlations between phenolic content were also seen with the existing ethnobotanical data (r = 0.5015; p = 0.003) and bioassays (r = 0.4178; p = 0.003). These results indicate that a clear majority of plants used by the Cree are excellent sources of antioxidants.
