AbstractBackgroundProgranulin (PGRN) is implicated in obesity and insulin resistance (IR). The aim of this study was to evaluate the effects of 8 weeks of circuit resistance training (CRT) on plasma PGRN, IR and body composition in obese men.Materials and methodsTwenty-eight healthy obese men [age: 36 ± 7.7 years, body weight (BW): 96.4 ± 15.6 kg, body mass index (BMI): 32.4 ± 4.5 kg/m2] completed the study. Subjects were randomly assigned to two groups of control and training. Subjects in the training group underwent training for 8 weeks, 3 times a week. Blood samples and anthropometric characteristics were taken before the commencement of the exercise protocol and 72 h after the last training session. The homeostatic model assessment of insulin resistance (HOMA-IR) was used to measure IR.ResultsBW, BF%, BMI, waist-hip ratio (WHR), HOMA-IR and plasma PGRN levels except lean body mass (LBM) were significantly reduced in the training group (p < 0.05). Additionally, except for LBM, subjects in the training group had significantly decreased BW, BF%, BMI, WHR, HOMA-IR and plasma PGRN levels compared to changes in those in the control group (p < 0.05). Significant correlations were found between the changes in plasma PGRN and the changes in insulin, HOMA-IR and BMI (p < 0.05).ConclusionsThe findings showed that 8 weeks of CRT improved body composition and IR which were accompanied by reduced plasma PGRN levels. This study suggests that CRT has the potential for obese individuals to counteract obesity-associated health impairments.

Objective: The overweight-related health problems among adolescents are obvious. Resistance training is recognized as a safe and efficacious exercise modality to have health-promoting effects in overweight adolescents. This study aimed to assess the efficacy of circuit resistance training (CRT) in improving inflammatory status, insulin resistance and body composition in overweight male adolescents. Materials and Methods: Twenty overweight adolescent boys (aged: 18.5 (±1), weight: 81.1 (±4.5), body mass index: 27.7 (±0.7)) completed this study. The participants were randomly and equally divided into two groups of control (CG; n=10) and circuit resistance training (CRT; n=10). The CG did their daily routine activities and the CRT group performed its training protocol 3 days a week for 6 weeks. Body composition components and serum variables were measured a day before and after the study. Insulin resistance index was measured by HOMA-IR. Results: The subjects in the CG showed significantly increased changes compared with pre-training values. Moreover, significant changes were found fort the changes of BW ( P -value= 0.005), BMI ( P -value= 0.001) and BF% ( P -value= 0.003) between groups. Conclusion: This study suggested that although six weeks of CRT failed to induce meaningful anti-inflammatory cytokine responses, and to improve body composition and HOMA-IR in overweight adolescent boys, but had protective effects on inflammatory status, HOMA-IR and body composition and prevented them from being deteriorated

Few studies have examined the improving effects of exercise on the association between metabolites of impaired protein metabolism and insulin resistance in obese children. Therefore, this study aims to investigate the effect of circuit resistance training (CRT) on plasma levels of amino acids, alpha-hydroxybutyrate (α-HB), mannose, and urinary levels of glycine conjugated adducts in obese adolescent boys. Forty obese adolescent boys (BMI above the 95th percentile) with an age range of 14-17 were randomly divided into CRT group (n= 20) and control group (n= 20). The CRT program (3 times/week, 70-80% of 1 repetition maximum) was performed for 8 weeks. The results indicated that the body composition and plasma levels of glucose, insulin resistance, valine, mannose, lysine and the sum of branched-chain amino acids (BCAAs) were decreased due to CRT. The plasma levels of asparagine, glycine, serine, and urinary levels of glycine conjugated adduct also increased in the CRT group. Although α-HB level decreased during CRT, it had no significant difference from that of the control group. It can be concluded that the improvement in obesity complications including insulin resistance in obese adolescent boys after CRT may be due to decrease in plasma levels of mannose and BCAAs and increase urinary metabolites. A systematic review with meta-analysis was conducted to evaluate the effects of circuit resistance training (CRT) on cardiorespiratory fitness, strength, and body composition in middle-aged and older women. Sixteen studies were included in the meta-analysis. The CRT interventions led to a significant decrease in weight, body mass index, and fat mass along with an increase in muscle mass. Significant differences were found in the fat mass and a trend to develop muscle mass when compared with the control group. CRT led to a significant increase in VO2max, walking endurance, and time to exhaustion; likewise, significant differences were observed when compared with the control group. CRT had a moderate and large favorable effect on arm, trunk, and lower limb strength. Furthermore, the increases in strength observed in the CRT were significantly greater than the changes observed in the control group. In middle-aged and older women, CRT improved cardiorespiratory fitness and strength and optimized body composition.

Exercise can generate alterations in body composition and modulate the immune system. The objective of this study was to verify whether a circuit resistance training (CRT) protocol can increase lean body mass (LM), and reduce fat body mass (FM) and the percent of FM (%FM) of sedentary women, without inducing inflammatory responses, indicated by serum cytokine levels. The initial hypothesis was that CRT would improve body composition, without changing serum cytokine levels. The study consisted of 14 healthy, sedentary women, aged 33–45 years (mean ± SD, 40.23 ± 3.98 years), with a normal body mass index. They participated in 3 sessions per week of CRT, which included 2 rounds in 9 stations with 1 set of 8–12 repetition maximum at each station, for 10 weeks. During the 10-week CRT period, participants maintained their pretraining nutritional standard. Body composition was analysed with dual-energy X-ray absorptiometry both pre- and post-training. Blood samples were collected after 96 h of rest pre- and post-training, and 5 min, 24 h, and 48 h after the second and last training sessions to measure serum cytokine levels by flow cytometry. The nutritional standard was accompanied throughout the study period with 24-h dietary recall. Increases in LM (35.937 ± 4.926 to 39.130 ± 4.950 kg) and decreases in FM (21.911 ± 8.150 to 17.824 ± 4.235 kg) and %FM (37.10 ± 10.84 to 31.19 ± 6.06), without concurrent changes in serum cytokine levels, and in the nutritional standard (α = 0.05). The proposed CRT improved body composition and did not induce any changes in serum cytokine levels characteristic of the inflammatory response in women.

Introdution: Non-alcoholic fatty liver disease (NFALD) is a disease associated with metabolic syndrome and lifestyle. The purpose of the present study was to investigate the effect of 12 weeks of circuit resistance training course on interleukin-17 (IL-17) levels, 70 kDa thermal shock protein (HSP70), insulin resistance and serum levels of alanine aminotransferase (ALT), as well as aspartate aminotransferase (AST). Method: In this clinical trial study, 30 males with NFALD were selected through targeted sampling and were randomly assigned to one of two groups of practice and control intervention. The training program included 12 weeks of resistive training and 3 training sessions per week, which was performed as periodic circuit resistance training. Before eating breakfast, blood sampling was done one day before the start of training and 3 days after the last training session. For statistical analysis, paired t-test was used to compare intra-group data. Independent t-test was used for comparison between groups. Data were used via the SPSS v 22 software. The significance was defined as P≤0.05. Results: After the training course, compared to the control group a significant decrease was observed in weight (P = 0.008), BMI (P = 0.006), body fat percentage (P <0.001), insulin resistance (P <0.001), HSP70 (P <0.001), IL-17 (P <0.001), serum levels of ALT (P <0.001) and AST (P <0.001) enzymes. Conclusion: Regarding the results of this study, circuit resistance training has an effective role in improving the liver function in NFALD patients by reducing insulin resistance and levels of inflammatory cytokines, this method of resistance training can be useful in the treatment of these patients.

Although weight training can enhance muscular strength, power, and body composition, outdated beliefs about muscular adaptations have limited its use in dancers. The purpose of this study was to investigate the effect of an 8-week progressive overload resistance-training (RT) program on muscular strength, power and body composition in collegiate female dancers. Sixteen subjects were randomised into a dance-only control group (CON) or an RT + dance training group (EXP). EXP met 3×/week for RT. Body fat (%BF), lean body mass (LBM), girth measurements, vertical jump (VJ), muscular strength (1-repetition max (RM) squat (SQT) and 1-RM bench press (BP)), maximal oxygen consumption (V̇O2max, and ventilatory threshold (VT)) were assessed pre- and post-study. Baseline performance characteristics across all subjects were: %BF=28.2±5.7%, VJ=33.4±5.4 cm, SQT=57.5±12.1 kg, BP=30.1±7.6 kg, (V̇O2max)=40.6±3.4 ml/kg/min, and VT=71.2±3.4%VO2max. Strength improvements were significantly greater for EXP than CON (P<0.05). No significant differences were found between groups for %BF, LBM, girth measurements, VJheight, (V̇O2max), or VT (P>0.05). However, a trend was observed for VJpower favouring EXP (P=0.07). EXP significantly improved strength, while no significant changes were observed in body composition. However, EXP improved their power-to-weight ratio, which may be a positive performance adaptation. RT for dancers can improve strength and power, allowing enhanced muscular loading and fatigue-resistance, which may optimise performance and decrease injury risk.

Insulin resistance and hyperandrogenemia observed in polycystic ovary syndrome (PCOS) are associated with metabolic disturbances and could be connected with body composition pattern. To date, several studies defining the parameters of body composition using dual energy X-ray absorptiometry (DXA) method in the group of PCOS patients have been published, however, without the analysis in different phenotypes. The aim of the present study was to investigate the relationships between serum androgens concentration, insulin resistance and distribution of fat mass using DXA method in various PCOS phenotypes according to the Rotterdam criteria. We examined 146 women: 34 (38%) had PCOS phenotype A, 20 (23%) phenotype B, 20 (23%) phenotype C and 15 (16%) phenotype D (with mean age of each phenotype 25 years), and 57 control subjects (mean age of 25.5 years). Homeostasis model assessment of insulin resistance (HOMA-IR) was calculated. Serum concentrations of testosterone, androstenedione and dehydroepiandrosterone sulfate (DHEA-S) were assessed and free androgen index (FAI) was calculated. In phenotypes A, B and C, we observed higher FAI in comparison to the control group (all p < 0.01). Serum concentrations of androstenedione and DHEA-S were higher in phenotypes A and C in comparison to the control group (all p < 0.01). However, only in phenotype A we found higher visceral adipose tissue (VAT) mass and android/gynoid ratio (A/G ratio) in comparison to the control group (all p < 0.01). In phenotype A, we observed connection of VAT with FAI (r = 0.58, p < 0.01). Accordingly, A/G ratio was related with FAI in all phenotypes (all p < 0.05). Additionally, in phenotype C, A/G ratio was related to serum concentrations of DHEA-S and androstenedione (r = 0.46, p = 0.03; r = 0.53, p = 0.01, respectively). We also found connections of HOMA-IR with VAT and A/G ratio in all phenotypes (all p < 0.05). Women with phenotype A had higher amount of VAT and A/G ratio in comparison to the control group. Serum concentration of androgens and insulin resistance are connected with VAT and A/G ratio in normoandrogenic and hyperandrogenic PCOS phenotypes.

Abstract Introduction: Back pain is a serious social problem which frequently affects women. So far, the effectiveness and safety of physical activities recommended to them have not been confirmed scientifically. The aim of the work was to determine the influence of circuit resistance training on the quality of life and disability of women with back pain. Materials and methods: The study included 25 women (mean age 52.81 ± 13.83 years) with back pain in the course of degenerative changes in the lumbar spine. Circuit resistance training was done on the machines with hydraulic resistance. Prior to the training, each participant was diagnosed on the basis of the McKenzie functional assessment. The quality of life was evaluated with the use of SF-36 survey, while pain intensity was assessed with visual analogue scale (VAS). The level of disability was examined with Roland-Morris disability questionnaire (RMDQ). The examinations were repeated after a three-month period of training and compared with a control group including 31 women (mean age 43.43 ± 14.64 years) who did not take up any physical activity in this period. Results: In the research group, 73.68% of the subjects improved their quality of life assessed with SF-36 survey, mainly in the category of physical functioning (p=0.006). Moreover, their disability decreased significantly (p=0.01) and pain subsided (p=0.003) compared to the control group. Conclusions: Circuit resistance training decreased low back pain, reduced disability and improved the quality of life of the subjects. This type of training may be recommended to individuals with non-specific low back pain.

Context: Zinc-α2 glycoprotein (ZAG) has been proposed as a new candidate in the pathogenesis of obesity, but most of the information stems from studies performed in rodents and in vitro assays. Objective: The main aim of the study was to compare serum levels of ZAG and its expression (mRNA levels and protein) in adipose tissue and the liver between obese and nonobese subjects. The relationship between ZAG and insulin resistance was also explored. Design: This was a case-control study. Setting: The study was conducted at a university referral center. Patients and Methods: Samples of serum, sc adipose tissue (SAT), visceral adipose tissue (VAT), and liver were obtained from 20 obese subjects during bariatric surgery. Samples from 10 nonobese patients matched by age and gender were used as a control group. Serum ZAG levels were determined by ELISA. ZAG mRNA levels were measured by real-time PCR and protein content by Western blot. The effect of insulin on liver production of ZAG was assessed using HepG2 cultures. Results: Serum concentration of ZAG (micrograms per milliliter) was significantly lower in obese subjects (40.87 ± 10.45 vs. 63.26 ± 16.40; P = 0.002). ZAG expression was significantly lower in the adipose tissue (SAT and VAT) and liver of obese patients than in control subjects. Significant negative correlations between body mass index and circulating ZAG (r = −0.65, P &lt; 0.001) as well as between body mass index and mRNA ZAG levels in SAT (r = −0.68, P &lt; 0.001) and VAT were detected (r = −0.64, P &lt; 0.001). No relationship was found between ZAG and homeostasis model assessment for insulin resistance and insulin had no effect on ZAG production in vitro. Conclusion: A down-regulation of ZAG in SAT, VAT, and liver exists in obese patients but seems unrelated to insulin resistance. A downregulation of zinc-α2 glycoprotein in adipose tissue and liver exists in obese patients, and it is unrelated to insulin resistance.
