Evaluation of yield and quality properties of Elaeagnus mollis oil produced by ultrasound-assisted solvent enzymatic extraction

Abstract To improve the extraction efficiency and quality, ultrasound-assisted solvent enzymatic extraction (UASEE) method was applied to extract oil from Elaeagnus mollis Diels. The soxhlet extraction (SE) and cold pressing (CP) were carried out to be compared with UASEE. The optimal UASEE conditions were liquid–solid ratio of 11.6 mL/g, enzyme amount of 1.1%, and ultrasound power of 583 W, which gave by Plackett–Burman and Box–Behnken design. Under optimum conditions, the oil yield of 43.35 ± 0.26% was reached, which was similar to that of SE (43.02 ± 0.77%). The oil obtained by UASEE is more desirable than that of SE, but similar with CP, especially on unsaturated fatty acids content and total tocopherol content. Overall, UASEE is a potential alternative to traditional methods for the efficient extraction of E. mollis oil for its higher oil yield and better quality.

An efficient procedure for ultrasound-assisted enzymatic extraction of crude polysaccharides fromTrichosanthes Fructus(crude TFP) using response surface methodology (RSM) was developed. The Box–Behnken design was applied to optimize the effects of pH (X1), enzyme amount (X2), extraction temperature (X3), and liquid-to-solid ratio (X4) on the extraction. The statistical analysis indicated that the independent variables (X4,X2, andX3), the quadratic coefficients (X12,X22,X32, andX42), and the interaction coefficient (X1X3) had significant impact on the yield of crude TFP. The optimal conditions were determined as follows: pH 4.5, enzyme amount 5000 u/g, extraction temperature 45°C, and liquid-to-solid ratio 30 ml/g. The experimental yield of crude TFP was 6.58%, which was very close to the predicted yield of 6.71%. TFPI was then purified and characterized with Sephadex G-100 column, UV-Vis, GPC, and FT-IR. The average molecular weight of TFPI was calculated to be 1.49 × 105 Da. TFPI exhibited strong reducing power and possessed not only remarkable scavenging activities against ABTS•+and DPPH radicals, but also high antitumor activities in C4-2, DU145, and PC3 cells. The results suggest thatTrichosanthes Fructusand TFPI could be a novel potent natural medicine with antioxidant and antitumor activities.

Background: Jicaro seeds can be considered oilseeds due to their high content of lipids, which could be used. Aqueous Enzymatic Extraction (AEE) of oils is a safe, economical and ecological technique, with the disadvantage that oil yields are lower than conventional methods. The microwaves pre-treatment of the seeds has been applied to overcome this inconvenience. Therefore, the objective of this study was to investigate the aqueous enzymatic extraction of oil from Jicaro seed pretreated in a microwave (M-AEE) compared to Soxhlet extraction, in terms of oil yield and its quality. Methods: M-AEE was evaluated varying type of enzyme, extraction time, enzyme concentration, solid/liquid ratio and particle size, according to Fractional Factorial experimental design. The obtained oil was analyzed in terms of fatty acid content and antioxidant activities. The same analyses were performed in oil obtained by Soxhlet extraction for comparison. Results: All parameters investigated in this work had a positive effect on the extraction process. The highest oil yield of 69.71% was obtained with a particle size of 0.59 mm, 3% (w/w) of Viscozyme L, solid/ liquid ratio of 1:6, and 5 h. No differences were found in fatty acid content and antioxidant activity between the oil obtained by M-AEE and that obtained by SE. Conclusion: The properties of the oil were not different in the two extraction methods evaluated; however, M-AEE has the advantage of being environmentally friendly technique. Jicaro seed oil physicochemical properties were similar to edible oils, thus could be considered as a new option of edible vegetable oil.

In this paper, ultrasonic cellulase extraction (UCE) was applied to extract polyphenols from passion fruit. The extraction conditions for total phenol content (TPC) and antioxidant activity were optimized using response surface methodology (RSM) coupled with a Box-Behnken design (BBD). The results showed that the liquid-to-solid ratio (X2) was the most significant single factor and had a positive effect on all responses. The ANOVA analysis indicated quadratic models fitted well as TPC with R2 = 0.903, DPPH scavenging activity with R2 = 0.979, and ABTS scavenging activity with R2 = 0.981. The optimal extraction parameters of passion fruit were as follows: pH value of 5 at 30 °C for extraction temperature, 50:1 (w/v) liquid-to-solid ratio with extraction time for 47 min, the experimental values were found matched with those predicted. Infrared spectroscopy suggested that the extract contained the structure of polyphenols. Furthermore, three main polyphenols were identified and quantified by HPLC. The results showed the content of phenolic compounds and antioxidant activity of the optimized UCE were 1.5~2 times higher than that determined by the single extraction method and the Soxhlet extraction method, which indicates UCE is a competitive and effective extraction technique for natural passion fruit polyphenols.

Soxhlet and microwave assisted extraction (MAE) methods were used to obtain non-polar compounds from pistachio hull. MAE parameters (liquid to solid ratio, microwave power, and extraction time) were studied to obtain maximum extraction yield. The optimal conditions were found to be liquid to solid ratio of 15:1 (v/w), microwave power of 250 W and extraction time of 12.5 min. The extraction yields were 9.81 and 9.50% for MAE and Soxhlet methods, respectively. The total phenolic content, antioxidant activity and tocopherol content of the extract obtained by MAE was found to be significantly higher than those of the Soxhlet extract (p < 0.05). The results showed that the extract contained α-tocopherols (567.65 mg/kg) and oleic acid (48.46%) as the major tocopherols and fatty acids. These findings propose that hull extracts can be considered as a good source of natural bioactive compounds and MAE can be a good alternative to the traditional Soxhlet method.
