The aerial parts of the Lamiaceae Hedeoma drummondii (Benth.) are used in Mexico to prepare a herbal tea and by North American Amerindians as a spice. The methanolic extract of the aerial parts exhibited a strong antioxidant effect measured by the scavenging of the free diphenyl picrylhydrazyl (DPPH) radical. Assay-guided fractionation of the crude methanolic extract allowed the identification of three major active constituents, chlorogenic, caffeic and rosmarinic acid, as well as sideritoflavone derivatives and simple phenolics. The TEAC, FRAP, total phenolic and flavonoid content were determined. The high content of caffeic acid and rosmarinic acid relates to the antioxidant activity of H. drummondii.

<p><strong>Objective: </strong>The current study was to deal the isolation and identification of secondary metabolites from <em>Polygonum equisetiforme</em> and evaluation of antioxidant activity of its extract.</p><p><strong>Methods: </strong>The methanol-water extract (7:3) of the air-dried aerial parts of <em>Polygonum equisetiforme</em> was fractionated and separated to obtain the isolated compounds by different chromatographic techniques. Structures of these compounds were elucidated by UV and 1D⁄2D H⁄ C NMR spectroscopy and compared with the literature data. The crude extract was evaluated for <em>in vitro</em> antioxidant activity using the 2,2 diphenyl dipicryl hydrazine (DPPH) method.</p><p><strong>Results: </strong>Ten secondary metabolites were isolated from <em>Polygonum equisetiforme</em> in this study. Of which three new flavonoids named as 3,5,7,2’,5’ pentahydroxyflavone 3-<em>O</em>-b-D-glucopyranoside (1), 3,5,7,2’,5’ pentahydroxyflavone 3-<em>O</em>-b-D-glucopyranoside 8 C-sulphated (2) and quercetin 3-<em>O-β</em>-D-glucucorinde 6''-methyl ester 8-sulphated (3) as well as quercetin 3-<em>O-β</em>-D-glucucorinde methyl ester (4), quercetin 3-<em>O-</em>β-D-glucopyranoside (5), quercetin 7-<em>O-β-</em>D-glucopyranoside (6),<em> </em>quercetin(7)<sub>, </sub>myricetin (8), <em>P</em><sub>-</sub>methoxy gallic acid methyl ester (9) and gallic acid (10). The antioxidant potential of <em>P. equisetiforme</em> extract was evaluated by investigating it's total phenolic and flavonoid content and DPPH radical scavenging activity whereby the extract showed significant antioxidant activity (IC<sub>50 </sub>= 37.45 μg/ml). The total phenolic and flavonoid content was found to be 130.79±5.502 and 45.8±1.63 μg/ml, respectively.</p><p><strong>Conclusion: </strong><em>Polygonum equisetiforme</em> is a promising medicinal plant, and our study tends to support the therapeutic value of this plant as an antioxidant drug.</p>

To evaluate the antioxidant potential of less valued green leafy vegetables of Bangladesh, the methanolic extracts of Alternanthera sessilis, A. philoxeroides, A. paronychioides, Glinus oppositifolius and Portulaca oleracea were subjected to assay for total phenolic content, total flavonoid content and DPPH free radicle scavenging activity. Among the studied materials, A. sessilis showed the highest phenolic and flavonoid content. However, G. oppositifolius presented the lowest phenolic content (14.53 ± 0.48 mg of GAE/g of sample) and P. oleracea exhibited minimum (7.005 ± 0.25 mg of RUE/g of sample) flavonoid content. DPPH radical scavenging activity was found to increase with increase in the concentration of each individual material. A. philoxiroides (IC50 = 6.07 ± 0.27 μg/ml) was found as the most active in DPPH radical percentage inhibition, which was followed by A. sessilis (IC50 = 7.17 ± 0.67 μg/ml). Interestingly, A. paronychioides, in spite of having good amount of total phenolic and flavonoid content, showed less DPPH scavenging activity. No linear relationship was found to exist in case of the total phenolic, flavonoid content and DPPH scavenging activity among G. oppositifolius, P. oleracea and A. paronychioides. The results from different methods may differ substantially because each complex chemical reaction generates unique values. However, all the five plants exhibited creditable antioxidant activity and thus consumption of these vegetables may play important role in combating oxidative stresses and thereby help in retaining good health.

Basil (Ocimum spp.) is a traditional herbal medicine abundant in antioxidants such as phenolic compounds. As part of a diet, this herb is proved to have some roles in decreasing the risk of cancer, and in the treatment of inflammation and neurodegenerative diseases. This study aims to explore the total phenolic and flavonoid content of two new basil hybrids growing in Romania, namely “Aromat de Buzau” (AB) and “Macedon” (MB). The antioxidant capacity of those two species was also analyzed by DPPH and cyclic voltammetry. Six different flavonoids, such as catechin (+), rutin, hyperoside, naringin, naringenin, and genistein, were separated, identified, and quantified by HPLC–DAD chromatography, for the first time, from romanian basil hybrids. The main flavonoid of the extracts was found to be naringin which is present in the highest amount (26.18 mg/kg) in “Aromat de Buzau” (O. basilicum) methanolic extract. These results suggest that dietary intake of these new hybrids can be a source of antioxidant compounds.

Bioassays provide an authentication of the traditional knowledge of medicinal plants. Antibacterial, antioxidant and phenolic content of nine medicinal plants (Achillea millefolium, Prunella vulgaris, P. laciniata, Lythrum salicaria, Epilobium angustifolium, E. hirsutum, Pedicularis comosa, Agrimonia eupatoria and Verbena officinalis) were investigated in connection with folkloric usages. Aerial parts of nine plant species were extracted with water and methanol (MeOH). Disc diffusion method was performed to evaluate the antibacterial activity of the extracts against ten pathogenic bacteria. Antioxidant activity was determined by using 2,2-diphenyl-1- picrylhydrazil (DPPH) radical photometric assay. Total phenolic and flavonoid content were investigated by using Folin-Ciocaltaeu and aluminum chloride (AlCl3) colorimetric method, respectively. Quantitative analysis of phenolic constituents of nine plants species were performed by high performance liquid chromatography-diode array detector (HPLC-DAD) via chosen ten phenolic standards (gallic acid monohydrate, caffeic acid, rutin hydrate, luteolin-7-O-β-D glucoside, kaempferol, rosmarinic acid, myricetin, quercetin, coumarin and apigenin). E. hirsutum showed the best antibacterial activity against gram-positive bacteria (S auerus, S. epidermidis and S. pyogenes). L. salicaria also demonstrated strong and broad spectrum antibacterial activity. P. laciniata, L. salicaria and E. angustifolium showed potent antioxidant activity. The highest phenolic and flavonoid content was observed with E. hirsutum and A. millefolium, respectively. L. salicaria also had very high amount of total phenol and flavonoid content. HPLC-DAD analysis displayed that P. laciniata and A. eupatoria were the best sources of rosmarinic acid. Furthermore, V. officinalis and P. vulgaris also had remarkable amount of rutin. This study revealed the scientific rationale behind the traditional knowledge of the tested plants. Consistent with traditional usages, the most prominent plants were L. salicaria, E. hirsutum and E. angustifolium in regard to strong antibacterial and antioxidant potentials. These plants may be proper natural sources with potential applications in pharmaceutical and food industry.

Objectives: The objectives of this study are to analyze phytoconstituents in Rhododendron arboreum leaves (qualitatively and quantitatively) and to estimate the immunomodulatory effects of extracts using different in vitro methods for intracellular and phagocytic killing potency of neutrophils, which are subsequent, involved in the process of phagocytosis by neutrophils.Methods: R. arboreum leaves extracts were separately extracted in 100% methanol, 50% methanol, and aqueous by a hot percolation technique using Soxhlet apparatus and concentrated with a vacuum rotary evaporator apparatus. The percentage yield of the extracts wares calculated. The extract was used to analyze phytoconstituents qualitatively, and total phenolic and flavonoid content were estimated. In another study, already purified methanolic extract of R. arboreum leaves, namely cytoplasmic male sterility (CMS-3) (kaempferol), RAM fr2, and transcranial magnetic stimulation (TMS-10) (ursolic acid) was used to evaluate immunomodulatory activities by nitroblue tetrazolium test, phagocytosis of killed Candida albicans.Results: Phytochemical screening of the extracts showed the presence of major classes of phytochemicals, i.e., phytosterols, flavonoids, phenols, alkaloids, carbohydrate, glycoside, sterols and steroids, terpenoids, and tannin while saponin was absent in all extract. The percent yield of leaves extract was 12.97%. Total phenolic content (TPC) expressed as gallic acid equivalents (GAE) ranged 1327.5±0.009 (in 100% methanolic extract), 1309.5±0.011 (in 50% methanolic), and 1468.5±0.006 mg GAE g−1 dry weight (in aqueous extract). Total flavonoid content expressed as rutin equivalents (RE) ranged 219±0.025 (in 100% methanolic extract), 184±0.02 (in 50% methanolic), and 262±0.58 mg (in aqueous extract) mg RE g−1 dry weight. R. arboreum purified extract CMS-3 (kaempferol), RAM fr2, and TMS-10 (ursolic acid) stimulated the neutrophils to phagocytic activity to the extent of 67.67±2.08%, 40.3±1.5%, and 79.67±0.57%, respectively. The above-said extracts stimulated the phagocytosis of killed C. albicans. The mean particle numbers for CMS- 3, RAM fr2, and TMS-10 were found to be 6.7, 6.3, and 7, respectively, at concentration 1 mg/ml.Conclusion: The phytochemical screening of all the extracts showed the presence of various phytochemicals that are biologically important. The total phenolic and flavonoid content of the plant are comparable to other medicinal plants. Even though the enormous progress on the phytochemistry and pharmacology of R. arboreum have been made, there still require more conclusive studies on the safety, efficacy, and in vivo toxicity of extracts and pure compounds to gain a better understanding.

Kepel or Stelechocarpus burahol is an exotic fruit originating from Java, Indonesia. This research was intended to evaluate the antioxidant activities of extracts and fractions of kepel fruit pulp (KFP) based on radical scavenging capability towards 2,2′-azinobis(3-ethylbenzo thiazoline-6-sulphonic acid) diammonium salt (ABTS) and 2,2′-diphenildiphenyl-1-picrylhydrazil radical (DPPH). In this study, the radical scavenging activities were also correlated with total phenolics and flavonoid contents. Among the evaluated samples, the ethyl acetate fraction exhibited the highest antiradical antioxidant activity towards ABTS radical with IC50 of 0.35 μg/ml, lower than that of vitamin C (as positive control). It also showed highest antiradical activity using DPPH based assay. Methanolic extract had total phenolic content of 58.28 ± 0.37% wt/wt gallic acid equivalent, higher than its fraction. Meanwhile the petroleum ether soluble fraction revealed flavonoid content (76.06 ± 11.9%) as rutin equivalent, among the extract and fractions evaluated. Based on coefficient (R2) values, phenolics and flavonoids contents contributed to 73.09% and 30.99%% towards antiradical scavenging activities, respectively. Dhaka Univ. J. Pharm. Microscopic fungi synthesize a wide range of phenolic compounds that play an important role in cell wall formation, as well as perform signaling and protective functions. One of the important functions of phenolic compounds, primarily polyphenols, is antioxidant activity. The most active polyphenols are flavonoids, their synthesis in microscopic fungi has not been studied enough. The aim of the study was to determine the total phenols and flavonoids content and antioxidant activity of Talaromyces funiculosus strains isolated from different econiches. Methods. 20 strains of the microscopic fungus T. funiculosus – 10 endophytes and 10 saprotrophs were used in the experiments. Cultivation of filamentous fungi was performed under submerged conditions with shaking at 210–230 rpm in Erlenmeyer flasks containing 200 ml of Czapek liquid medium at a temperature 26±2°C for 6 days. The biomass of the strains was filtered, dried at 70°C and extracted with 95% methanol (1:50 w/v). The total phenols content was determined using Folin-Ciocalteu reagent by the method of Arora, Chandra (2010) with some modifications and expressed as mg of gallic acid equivalents (GAE) per g of dry weight mycelium. Flavonoids in each extract were determined spectrophotometrically with AlCl3 and expressed as mg of quercetin equivalents (QE) per g of dry weight mycelium. Antioxidant activity of methanolic extracts was measured as radical scavenging activity by the percentage inhibition of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. Results. The total phenolic content of methanolic extracts of T. funiculosus strains ranged from 0.12±0.007 to 1.97±0.150 mg GAE/g. The highest level of phenolic compounds was found for saprotroph UCM F-16790 (1.97±0.150) and endophyte UCM F-16795 (1.71±0.137 mg GAE/g). The flavonoid content in methanolic extracts of the studied strains varied in the range of 0.02±0.001–0.39±0.041 mg QE/g. All studied strains of T. funiculosus showed antioxidant activity against DPPH radical, which was 12.19±2.34–60.37±7.24% inhibition. Statistical analysis revealed a strong positive correlation between the total phenols and flavonoids content (Pearson’s correlation coefficient r=0.92); between antioxidant activity, phenols (r=0.82) and flavonoids (r=0.81) content in methanol extracts of T. funiculosus strains. The total content of phenols and flavonoids and antioxidant activity did not depend on the trophic group of the studied strains. The moderate correlation between flavonoids content and antioxidant activity of methanolic extracts of endophytic T. funiculosus strains (r=0.46) may be related to their ability to synthesize a wider range of polyphenolic compounds with antioxidant activity. Conclusions. The studied T. funiculosus strains have the ability to synthesize phenolic compounds and flavonoids in different concentrations. Correlation analysis revealed strong correlations between total phenolic and flavonoid contents and antioxidant activity. The reason for phenomenon of moderate correlation (r=0.46) between flavonoid content and antioxidant activity in endophytic strains may be the synthesis of other compounds by individual strains. The determination of individual components synthesized by the most active strains is of scientific interest for further study. T. funiculosus UСM F-16795 and UСM F-16790 strains had a quite high level of phenols and flavonoids. Obtained data may be promising for the search for phenolic compounds with antioxidant properties to increase plant resistance to negative impacts of environmental stress factors.

Fig vinegar is a special product which has a long history and is handed down from generation to generation. In the current study, characteristics of homemade fig vinegars supplied from different cities of Turkey were compared in terms of their physicochemical, antioxidant and antimicrobial properties. Physicochemical properties of the samples were very diverse. Total phenolic and flavonoid content, and ABTS+ and DPPH radical scavenging activities of the samples were ranged from 313.5 to 594.25 mg GAE/L and 42.52-132.42 mg catechin/L, and 0.44-3.98 µg TE/mL and 1.25-7.51 µg TE/mL, respectively. The antioxidant activity of vinegars is highly correlated with their phenolic and flavonoid contents. Furthermore, vinegar samples exhibited inhibitive effect on all test microorganisms and the minimum inhibition concentration values were ranged between 0.39 and 25% (v/v). These results indicated the distinct properties of homemade fig vinegars and the potential of them as antimicrobial and antioxidant substances.
