Essential oil chemical composition, antimicrobial, anticancer, and antioxidant effects of Thymus convolutus Klokov in Turkey

Abstract In this study, the chemical composition, antimicrobial, antioxidant, and anticancer effects of Thymus convolutus Klokov oil and its main compound camphor were investigated. The oil was isolated from T. convolutus using hydrodistillation method, analyzed by gas chromatography/mass spectrometry (GC–MS), and 66 compounds were identified. The main component was determined as camphor at 16.6%. The antioxidant properties were identified with the DPPH (2,2′-diphenyl-1-picrylhydrazyl) radical-scavenging method and, 33.39 ± 0.25% DPPH was scavenging in 1000 μg/mL of essential oil. The strong antimicrobial activity was observed against Escherichia coli, Enterobacter aerogenes, Proteus vulgaris, and Pseudomonas aeruginosa with MIC values of 125 μg/mL. Aspergillus flavus was more sensitive (28%) against T. convolutus essential oil than other fungi. The cytotoxic effect of oil was analyzed by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) method. Camphor was effective on human hepatoma cells (Hep3B) at concentrations of 1 mg/mL, 500, 250, and 125 μg/mL, while essential oil of T. convolutus was found to be effective at concentrations of 250 and 125 μg/mL. A reduction in cell proliferation was observed in colon carcinoma cells (HT-29) treated with 500 μg/mL camphor for 48 h. No statistically significant effect was found in Umbilical Vein Endothelial Cells (HUVEC) treated with essential oil and camphor.

Background: The essential oil from the Acantholimon genus have been an integral part of the traditional food additive in Middle East. Most of the plants in Acantholimon genus have not been studied scientifically. The aim of this study is to investigate the chemical composition, antibacterial, insecticidal and anti-oxidant activities of three Acantholimon species including Acantholimon atropatanum, A. gilliatii and A. tragacanthium. Method: The essential oils of the aerial parts were extracted by hydrodistillation. Chemical constitutions were identified by gas chromatography- mass spectroscopy technique, also their toxicities were assessed against the two important grain products pests, Oryzeaphilus mercator and Tribolium castaneum. Antibacterial activity was assessed against the three foodborne bacteria that include Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus based on the disc diffusion assay. Free-radical-scavenging property was identified based on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. Results: 2-hexahydrofarnesyl acetone was the main compound in A. gilliatii and A. tragacanthium, whilst farnesyl acetone, heptacosane and germacrene D were the principal components of A. atropatanum essential oil. These oils exhibited 40-90% mortality of O. mercator and/or T. castaneum at a dose of 12 μl/l air after 48h of exposure, and exhibited significant free-radicalscavenging property (RC50 = 3.7 × 10-3 - 8.3 × 10-3 mg/ml). The oils of A. tragacanthium and A. gilliatii showed a weaker antibacterial activity compared to A. atropatanum. Conclusion: A. atropatanum, A. gilliatii and A. tragacanthium essential oils had significant insecticidal and anti-oxidant properties. They also showed week to moderate antibacterial activity against P. aeruginosa and S. aureus.

The investigation aimed to study the in vitro and in silico antioxidant properties of Melissa officinalis subsp. officinalis essential oil (MOEO). The chemical composition of MOEO was determined using GC–MS analysis. Among 36 compounds identified in MOEO, the main were beta-cubebene (27.66%), beta-caryophyllene (27.41%), alpha-cadinene (4.72%), caryophyllene oxide (4.09%), and alpha-cadinol (4.07%), respectively. In vitro antioxidant properties of MOEO have been studied in 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free-radical scavenging, and inhibition of β-carotene bleaching assays. The half-maximal inhibitory concentration (IC50) for the radical scavenging abilities of ABTS and DPPH were 1.225 ± 0.011 μg/mL and 14.015 ± 0.027 μg/mL, respectively, demonstrating good antioxidant activity. Moreover, MOEO exhibited a strong inhibitory effect (94.031 ± 0.082%) in the β-carotene bleaching assay by neutralizing hydroperoxides, responsible for the oxidation of highly unsaturated β-carotene. Furthermore, molecular docking showed that the MOEO components could exert an in vitro antioxidant activity through xanthine oxidoreductase inhibition. The most active structures are minor MOEO components (approximately 6%), among which the highest affinity for the target protein belongs to carvacrol.

Oregano (Poliomintha longiflora) essential oil (Ooil) is a product of high commercial value and many applications, including chemotherapy. Aiming to achieve the best use of this resource, the present study focuses on the characterization of separated fractions of Ooil by fractional vacuum distillation at low pressure. Four fractions (F1–F4) and undistilled oil (Unoil) were separated from Ooil and analyzed for their chemical composition and biological activities, such as antioxidant and antimicrobial activities. Gas chromatography–mass spectrometry shows differences in the composition among the fractions and Ooil. The amount of monoterpenes oxygenated (MO), sesquiterpenes hydrocarbon (SeH) and monoterpenes hydrocarbon (MH) varied between the fractions in ranges of 1.51–68.08, 3.31–25.12 and 1.91–97.75%, respectively. The major concentrations of MO and SeH were observed in F4 and Unoil. On the other hand, the highest concentrations of MH were found in F1 and F2, while the lowest were in F4 and Unoil. These results were correlated with the biological activity. Free-radical scavenging activity varied among fractions, with F4 and Unoil showing the highest activity. The antimicrobial test showed that F4 and Unoil had the highest activity in almost all cases. The correlation between the variables studied in the different fractions allows the definition of the particular properties for each one of them.

In previous work [1], the essential oil of the aerial parts of Marrubium vulgare L. obtained by hydrodistillation was analysed by gas chromatography coupled to mass spectrometry (GC-MS) in order to determine their chemical composition. Fifty (50) components in the oil of M. vulgare were identified. The results demonstrated that the major components of the essential oil were: 4,8,12,16-Tetramethyl heptadecan-4-olid (16.97 %), Germacrene D-4-ol (9.61 %), α- pinéne (9.37 %), Phytol (4.87 %), Dehydro-sabina ketone (4.12 %), Piperitone (3.27 %), δ-Cadinene (3.13 %), 1-Octen-3-ol (2.35 %) and Benzaldehyde (2.31 %). In this study, the antioxidant properties of essential oil were examined. The results showed that this oil can be considered an effective source of antioxidants of natural origin. This is the first report on chemical composition of M. vulgare essential oil cultivated in Algeria and the original study on the antioxidant activity of M. vulgare essential oil. The antioxidant activity was investigated with one method: 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method.

Background: This research tried to evaluate the chemical composition of the essential oil of Salvia limbata, and also bioactivity of essential oil and the plant extract. S. limbata is an aromatic plant with medicinal uses, such as analgesic, anti-inflammatory activities. Methods: The essential oils of the flowers of Salvia limbata were obtained using a Clevenger-type apparatus, and its chemical composition was identified by chromatography methods. The study was also conducted to evaluate antioxidant and antibacterial activities of the plant extracts. For this purpose, the leaves and flowers of S. limbata were extracted using methanol and then fractioned by n-hexane and Ethyl Acetate (EtOAc). Total Phenolic Content (TPC), Total Flavonoid Content (TFC), Radical Scavenging Activity (RSA) and the Disc-Diffusion (DD) assay were run to study the bioactivity of the plant extracts. Results: The oil was dominated by the oxygenated hydrocarbon of 5-methylene-2,3,4,4-tetramethylcyclopent- 2-enone (37.1%), oxygenated sesquiterpene of occidol (22.8%), and sesquiterpene hydrocarbon of β-bourbonene (12.1%). The EtOAc extract of the flowers showed the TPC with 66.2±0.62 mg GAE/g (mg of gallic acid equivalents per gram of extract) and RSA with IC50 of 17.96±0.54 μg/mL. Although the EtOAc extract of the leaves exhibited the highest TFC with 61.96±4.73 mg RuE/g (mg of rutin equivalents per gram of extract). The extracts inhibited the growth of Escherichia coli, Citrobacter frurdi, Pseudomonas aeruginosa, and Streptococcus pneumonia. The extracts were inactive against Staphylococcus coagulase, Enterobacter aerogenes, Acinetobacter baumannii, Serratia marcescens, and Klebsiella pneumoni. Conclusion: According to the results, the plant extracts were potent agant in antioxidant activity.

Increasing resistance of pathogens towards conventional antibiotics presents a major threat to public health because it reduces the effectiveness of antibiotic treatment. Mentha pulegium L., also known as pennyroyal, is an aromatic herb that belongs to the family Lamiaceae. Its essential oil has been traditionally used in medicine, aromatherapy, and cosmetics. The purpose of this work was to study the chemical composition of a pennyroyal essential oil and to evaluate their bioactivities, specifically, antioxidant, antimicrobial, anti-quorum sensing, and anti-inflammatory. A special focus was given to the antibacterial activity of the essential oil against Acinetobacter baumannii. The chemical composition of the essential oil was studied by GC-MS/GC-FID. The DPPH free radical scavenging assay and the β-carotene/linoleic acid system were used to evaluate the antioxidant properties. The antimicrobial and anti-quorum sensing activities were evaluated by disk diffusion assays and complemented with optical microscopy observations. The results showed that pulegone was the major compound (88.64%) of the pennyroyal essential oil. Regarding the antimicrobial activity, the action against Acinetobacter baumannii stands out, which, together with the capacity of the essential oil to inhibit the quorum sensing mechanisms, may suggest the use of the pennyroyal essential oil to further develop surface disinfectants for hospitals.

Cymbopogon species from the Poaceae family are widely distributed in the Himalayan region of India and commonly used as flavors, fragrances, cosmetics, and pharmaceuticals. It is known to contain compound citral, which give the lemon scent to many of the plants of the cymbopogon genera. The essential oil of  Cymbopogon flexuosus has high polyphenolic content which is responsible for antioxidant properties.  Beside citral is also used for the synthesis of vitamin B and Ionones. The bioactive potential of Lemongrass and constituent are rapidly increasing which is reflected from growing number of reports being published.  The present study was to know the chemical composition and in vitro antioxidant activity of essential oil of C. flexuosus from Uttarakhand. The essential oils of Cymbopogon collected in the region of Uttarakhand were obtained by hydrodistillation of the leaves and analyzed for chemical composition by GC/MS. The antioxidant activity of essential oils at different concentrations was determined against DPPH radical activity and vitamin C as the standard antioxidant compound.  The IC50    value and percentage of DPPH inhibition were recorded.  Twenty-five compounds were identified in essential oil extracted from leaves representing 93.15% of the oil composition. The yield of essential oil of Cymbopogon was 0.6 + 0.1 %and the major compound in the essential oil was citral (a racemic mixture of two isoforms geranial and nearl) followed by heptenone(1.98%) , linalool(1.65%), geraniol (1.47%), ?-caryophyllene (1.14% ) , limonene (0.92%),  nearl acetate (0.82%), citronellal(0.44 %) and citronellol (0.22%). Radical scavenging capacity (Inhibition, %) of the C. flexuosus essential oil was high (78.19+1.11) at the concentration level of 150 ?g/ml and  IC50 value of the essential oil was 43.67?g/ml.  The data of this study encourages to consider the essential oil of C. flexuosus  as a source of bioactive compounds which may add great industrial value to this crop.

The hydro-distilled essential oil of Citrus reticulata Blanco var. kinnow was analyzed by Gas chromatography-mass spectrometry (GC-MS). Five constituents out of fifteen constituents were identified from seeded C. reticulata oil representing 74.66% of the oil. The major constituent of the oil was ?- phellandrene (62.00%). ?-pinene(6.53%), ?-myrcene(2.81%), limonene(2.81%) and caryophyllene(0.51%) were present in considerable amount. From the low seeded C. reticulata oil, six components out of seventeen compounds were identified constituting 54.74% of the oil and the main component was ?-phellandrene (37.35%). ?-pinene(2.79%), ?-pinene(3.26%), ?-myrcene(4.16%), limonene(5.77%), caryophyllene(1.41%) were present in considerable amount. Introduction: Present study describes the antimicrobial activity and free radical scavenging capacity (RSC) of essential oil from Mentha longifolia (L.) Huds. Aim of this study to investigate the quality, antimicrobial andantioxidant activity of wild species Mentha longifolia essential oil from Bosnia and Herzegovina.Methods: The chemical profi le of essential oil was evaluated by the means of gas chromatography-mass spectrometry (GC-MS) and thin-layer chromatography (TLC). Antimicrobial activity was tested against 6bacterial strains. RSC was assessed by measuring the scavenging activity of essential oils on 2,2- diphenyl-1-picrylhydrazil (DPPH).Results: The main constituents of the essential oil of M. longifoliae folium were oxygenated monoterpenes,piperitone oxide (63.58%) and 1,8-cineole (12.03%). Essential oil exhibited very strong antibacterial activity.The most important antibacterial activity essential oil was expressed on Gram negative strains: Escherichia coli, Pseudomonas aerginosa and Salmonella enterica. subsp.enterica serotype ABONY. Antioxidant activity was evaluated as a RSC. Investigated essential oil was able to reduce DPPH radicals into the neutral DPPHH form (IC50=10.5 μg/ml) and this activity was dose –dependent.Conclusion: The study revealed signifi cant antimicrobial activity of the investigated essential oil. The examined oil exhibited high RSC, which was found to be in correlation to the content of mainly monoterpeneketones and aldehydes. These results indicate that essential oils could serve as safe antioxidant and antiseptic supplements in pharmaceuticals.
