Chemical Composition and Bioactivity of Essential Oil and Extracts of Salvia limbata C.A. Mey (Lamiaceae)

Background: This research tried to evaluate the chemical composition of the essential oil of Salvia limbata, and also bioactivity of essential oil and the plant extract. S. limbata is an aromatic plant with medicinal uses, such as analgesic, anti-inflammatory activities. Methods: The essential oils of the flowers of Salvia limbata were obtained using a Clevenger-type apparatus, and its chemical composition was identified by chromatography methods. The study was also conducted to evaluate antioxidant and antibacterial activities of the plant extracts. For this purpose, the leaves and flowers of S. limbata were extracted using methanol and then fractioned by n-hexane and Ethyl Acetate (EtOAc). Total Phenolic Content (TPC), Total Flavonoid Content (TFC), Radical Scavenging Activity (RSA) and the Disc-Diffusion (DD) assay were run to study the bioactivity of the plant extracts. Results: The oil was dominated by the oxygenated hydrocarbon of 5-methylene-2,3,4,4-tetramethylcyclopent- 2-enone (37.1%), oxygenated sesquiterpene of occidol (22.8%), and sesquiterpene hydrocarbon of β-bourbonene (12.1%). The EtOAc extract of the flowers showed the TPC with 66.2±0.62 mg GAE/g (mg of gallic acid equivalents per gram of extract) and RSA with IC50 of 17.96±0.54 μg/mL. Although the EtOAc extract of the leaves exhibited the highest TFC with 61.96±4.73 mg RuE/g (mg of rutin equivalents per gram of extract). The extracts inhibited the growth of Escherichia coli, Citrobacter frurdi, Pseudomonas aeruginosa, and Streptococcus pneumonia. The extracts were inactive against Staphylococcus coagulase, Enterobacter aerogenes, Acinetobacter baumannii, Serratia marcescens, and Klebsiella pneumoni. Conclusion: According to the results, the plant extracts were potent agant in antioxidant activity.

Background: Rhus pentaphylla Desf. (Anacardiaceae) widely grown in Algeria, is an underutilized plant and remains poorly studied. To investigate for the first time, the chemical composition profile of essential oil and hydrosol extract; besides Total Phenolic (TPC), Flavonoid (TFC) and Condensed Tannins Contents (CTC) of fruits, leaves and roots, the fatty acid composition was assessed. The radical scavenging activity of R. pentaphylla extracts was then evaluated. Methods: Essential oil and hydrosol extract were isolated respectively by hydrodistillation and liquidliquid- extraction from the aerial parts of R. pentaphylla. The antioxidant activity was evaluated using 2,2-Diphenyl-1-Picrylhydrazyl (DPPH) radical and ferric reducing antioxidant power (FRAP). Chemical compositions of various extracts were investigated using Gas Chromatography (GC) and GC-Mass Spectrometry (GC-MS). Results: The results of GC and GC-MS analysis revealed 83 components in essential oil and 75 components in hydrosol extract representing respectively 96.9% and 92.5% of the total extract composition. The main constituents were hexadecanoic acid (31.5%) followed by spathulenol (14.9%) in the oil while the higher amount present in the hydrosol extract was of spathulenol (14.2%). Otherwise, 13 and 18 fatty acids were identified in roots and fruits respectively. The highest levels of TPC and CTC were found in roots extracts while the highest quantity of TFC has been recorded in the leaves extract. Besides, hydrosol extract was able to scavenge DPPH and FRAP free radicals more efficiently than essential oil. Conclusion: R. pentaphylla contains bioactive substances that could be used as a new promising source of antioxidant compounds in different applications.

Free radical scavenging activity, total phenolic content and the chemical composition of the essential oil isolated by steam distillation from Artemisia dracunculus L. was investigated. The isolation yield was 0.24% (v/w) based on the fresh plant material (leaves). GC-MS investigation identified 21 components, accounting 99.93% of the total amount. The major components were sabinene (42.38%), isoelemicin (12.91%), methyl eugenol (9.09%), elemicin (7.95%) and beta-ocimene (6.46%). The free radical scavenging activity of the essential oil of Artemisia dracunculus L. was evaluated in vitro by the DPPH assay (IC50 = 0.730 � 0.213 mg/mL), BHA and alpha-tocopherol were used as a positive control. The total phenolic content of the tarragon essential oil was evaluated by the Folin-Ciocalteu method (GAE = 0.451 � 0.001 mg/g sample). In view of these data, we consider that tarragon essential oil could represent a new antioxidants source as a reliable option to reduce the usage of synthetic additives.

Background: The essential oil from the Acantholimon genus have been an integral part of the traditional food additive in Middle East. Most of the plants in Acantholimon genus have not been studied scientifically. The aim of this study is to investigate the chemical composition, antibacterial, insecticidal and anti-oxidant activities of three Acantholimon species including Acantholimon atropatanum, A. gilliatii and A. tragacanthium. Method: The essential oils of the aerial parts were extracted by hydrodistillation. Chemical constitutions were identified by gas chromatography- mass spectroscopy technique, also their toxicities were assessed against the two important grain products pests, Oryzeaphilus mercator and Tribolium castaneum. Antibacterial activity was assessed against the three foodborne bacteria that include Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus based on the disc diffusion assay. Free-radical-scavenging property was identified based on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. Results: 2-hexahydrofarnesyl acetone was the main compound in A. gilliatii and A. tragacanthium, whilst farnesyl acetone, heptacosane and germacrene D were the principal components of A. atropatanum essential oil. These oils exhibited 40-90% mortality of O. mercator and/or T. castaneum at a dose of 12 μl/l air after 48h of exposure, and exhibited significant free-radicalscavenging property (RC50 = 3.7 × 10-3 - 8.3 × 10-3 mg/ml). The oils of A. tragacanthium and A. gilliatii showed a weaker antibacterial activity compared to A. atropatanum. Conclusion: A. atropatanum, A. gilliatii and A. tragacanthium essential oils had significant insecticidal and anti-oxidant properties. They also showed week to moderate antibacterial activity against P. aeruginosa and S. aureus.

In the honey industry, heat treatments are usually applied to maintain honey’s quality and shelf life. Heat treatment is used to avoid crystallisation and allow the easy use of honey, but treatment with heat might affect the antioxidant and antibacterial activities, which are the immediate health effects of honey. This study will determine the effect of heat treatment on Malaysian and Australian stingless bee honey (SBH) produced by the common bee species in both countries. Eighteen honey samples were subjected to heat at 45 °C, 55 °C and 65 °C for one hour and subsequently analysed for their total phenolic content (TPC), total flavonoid content (TFC), DPPH radical scavenging activity, ferric reducing antioxidant power (FRAP) and minimum inhibitory concentration (MIC). The results show that all samples had high TPC, TFC and antioxidant activities before the treatment. The heat treatments did not affect (p < 0.05) the TPC, TFC and antioxidant activities in most samples, but did inhibit the antibacterial activities consistently in most of the samples, regardless of the bee species and country of origin. This study also confirms a strong correlation between TPC and TFC with FRAP activities for the non-heated and heated honey samples (p < 0.05). Other heat-sensitive bioactive compounds in SBH should be measured to control the antibacterial properties present.

This study was undertaken to determine the antioxidant and antimicrobial effect of essential oil and extracts of Echinophora sibthorpiana Guss. (fam. Apiaceae) collected in Macedonia. The chemical composition of E. sibthorpiana essential oil was characterized by the presence of methyl eugenol (60.40%), p-cymene (11.18%) and ?-phellandrene (10.23%). The free radical scavenging activity of extracts and essential oil was evaluated by DPPH and ABTS assays. The aqueous extract of aerial parts exhibited the strongest scavenging activity (IC50=1.67 mg/ml); results of the ABTS test showed that the most effective was the ethanol extract of aerial parts (1.11 mg vit. C/g). The essential oil showed stronger antioxidant activity compared to hydroxyanisole, ascorbic acid and quercetin that were used in the DPPH and ABTS tests, respectively. The total phenolic and flavonoid concentrations in the extracts ranged between 38.65-60.72 mg GA/g, and 3.15-19.00 mg Qu/g, respectively. The antimicrobial properties of the extracts and essential oil were investigated using a micro-well dilution technique against human pathogenic strains. The results were comparable with the effects of the positive controls, streptomycin and fluconazole. These findings indicate that E. sibthorpiana extracts and oil can be used in preventive treatments and as an alternative for synthetic preservatives.

Chemical composition of essential oil and the potential of Prangos gaubae for the management of public health problems such as Alzheimer's disease, obesity, Diabetes mellitus, and skin diseases were evaluated for the first time. In this direction, enzyme inhibitory effects, antioxidant activity, and total bioactive contents of the plant were determined. EO showed high acetylcholinesterase (2.97 mg GEs/g oil), α-amylase (1.35 mmol ACEs/g oil), α-glucosidase (38.84 mmol ACEs/g oil), and lipase (1.59 mmol OEs/g oil) inhibitory activities. Moreover, strong antioxidant effects were observed in antiradical (DPPH and ABTS), reducing power (CUPRAC and FRAP), total antioxidant, and metal chelating assays. Methanol extract exhibited promising DPPH radical scavenging activity (0.47 mmol TEs/g extract) and also high reducing power in CUPRAC (0.89 mmol TEs/g extract) and FRAP (0.52 mmol TEs/g extract) assays. All extracts showed low total flavonoid but high total phenolics content. Furthermore, they exhibited strong skin-care effect in tyrosinase inhibition assay. EO analysis showed the presence of germacrene D (26.7%), caryophyllene oxide (14.3%), ( E)-caryophyllene (13.8%), and spathulenol (11.3%) as the major volatile components. Results indicated that P. gaubae has promising potential for possible uses in food, cosmetic, and pharmaceutical industries due to its valuable phytoconstituents and biological activities.

Oregano (Poliomintha longiflora) essential oil (Ooil) is a product of high commercial value and many applications, including chemotherapy. Aiming to achieve the best use of this resource, the present study focuses on the characterization of separated fractions of Ooil by fractional vacuum distillation at low pressure. Four fractions (F1–F4) and undistilled oil (Unoil) were separated from Ooil and analyzed for their chemical composition and biological activities, such as antioxidant and antimicrobial activities. Gas chromatography–mass spectrometry shows differences in the composition among the fractions and Ooil. The amount of monoterpenes oxygenated (MO), sesquiterpenes hydrocarbon (SeH) and monoterpenes hydrocarbon (MH) varied between the fractions in ranges of 1.51–68.08, 3.31–25.12 and 1.91–97.75%, respectively. The major concentrations of MO and SeH were observed in F4 and Unoil. On the other hand, the highest concentrations of MH were found in F1 and F2, while the lowest were in F4 and Unoil. These results were correlated with the biological activity. Free-radical scavenging activity varied among fractions, with F4 and Unoil showing the highest activity. The antimicrobial test showed that F4 and Unoil had the highest activity in almost all cases. The correlation between the variables studied in the different fractions allows the definition of the particular properties for each one of them.

The essential oil obtained by hydrodistillation of the flowers of Rhanterium adpressum Coss. & Durieu was analyzed using GC and GC-MS. The essential oil was very rich in monoterpene compounds. The major components identified were the monoterpene hydrocarbons: camphene (21.8%), myrcene (19.3%) and α-pinene (17.4%). Other compounds, including limonene, β-pinene and terpinol-4-ol, were present in low content (4–6%). The composition of the fatty acids in the lipid extract obtained from the flowers was also investigated by GC and GC-MS. The main fatty acids identified were palmitic (47.4%), oleic (12.9%) and stearic acids (10.6%). The total phenolic contents and the antioxidant activities were also evaluated for both extracts. The total phenolic contents were determined using the Folin-Ciocalteu reagent and the antioxidant activities were measured using three different assays: DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenging activity, FRAP (ferric reducing antioxidant potential) and a molybdenum assay. As a result of these tests, the lipid extract exhibited the highest antioxidant activities in comparison with the essential oil extract.

The present study examined the effect of Lactobacillus plantarum ATCC14917 fermentation on the chemical composition and antioxidant activity of apple juice. Apple juice was fermented and examined of its antioxidant activity using chemical models and cellular antioxidant assay. Furthermore, the chemical composition of fermented apple juice was characterized by LC-MS/MS. Lactobacillus plantarum ATCC14917 fermentation showed an increase in DPPH and ABTS radical scavenging activity as well as cellular antioxidant activity of apple juice. However, fermentation decreased the total phenolic and flavonoid content. Subsequent LC-MS/MS analysis of the phenolic profile indicated that the content of 5-O-caffeoylquinic acid (5-CQA), quercetin, and phloretin with strong antioxidant activity was increased significantly after fermentation. The modified phenolic composition may contribute to the increased antioxidant activity of fermented apple juice. Our findings showed that Lactobacillus plantarum ATCC14917 fermentation may be an efficient way to enhance the bioavailability of phenolic compounds and to protect cells from oxidative stress.

Ethnobotanic surveys have revealed the use of Mangifera indica L. (Anacardiaceae) bark for the treatment of infectious diarrhea. The essential oil of M. indica bark is described for the first time for its chemical composition, radical scavenging activity (DPPH method) and antimicrobial properties. The total phenols content of its water and ethanol bark extracts as well as their radical scavenging and antimicrobial properties were also evaluated. Four commercial plant extracts were also studied for a comparison purpose. The antimicrobial activities were measured for all samples against three Gram (-): Escherichia coli, Salmonella enteritidis, Shigella and two Gram (+): Staphylococcus aureus and Bacillus cereus bacteria. The M. indica bark essential oil was characterized by the association of two major sesquiterpenes: ( E)-β-caryophyllene (60.3%) and α-humulene (36.7%). It presented the lowest ratio of concentration to inhibition zone diameter on all the strains. The aqueous and ethanol extracts of M. indica bark were characterized by high contents of total phenols compounds and high radical scavenging activity compared to the essential oil. Finally, the interesting combination of the antibacterial and antiradical activities of the aqueous M. indica bark extract justifies the traditional use of this plant part in decoction form for the treatment of diarrheal infections.
