The aim of the study was selection of optimal conditions for obtaining carotenoid drugs of mycelium origin from the basidiomycete strains Laetiporus sulphureus Ls-08, Fomes fomentarius Ff-1201 and Fistulina hepatica Fh-18 and the study of antibacterial and total antioxidant activity of these compounds. The strains were surface grown on a glucose-peptone medium modified for each producer. The homogenized pigments of the mycelium strains were extracted with ethanol and the solvent was separated under vacuum at 60 ºC. The absorption spectra of the carotenoid drugs were recorded for alcoholic solutions at 350–500 nm. The antibacterial activity of the carotenoids was determined by the agar diffusion method, and the total antioxidant activity was determined by the DPPH-method. It was found that the optimum temperature for carotenoid extraction is 60 °C. The absorption spectra of carotenoid drugs showed three peaks in 420, 450 and 470 nm. These results respond to the β-carotene absorption spectra. The highest antioxidant activity was noted for carotenoid drugs from F. hepatica Fh-18 and L. sulphureus Ls-08 strains obtained at an extraction temperature of 40 and 60 °C respectively. The antibacterial activity of carotenoid drugs against the test cultures was not species dependent. Carotenoid drugs with a 20% concentrate obtained from the L. sulphureus Ls-08 strain had the highest antibacterial activity against the test cultures Staphylococcus aureus and Escherichia coli. Carotenoids from the mycelium of F. hepatica Fh-18 had the highest antibacterial activity against the test culture Candida albicans. Extraction temperature of 60 °C is optimal for mycelial yield of carotenoids from the studied strains. All preparations of carotenoids exhibited antibacterial activity against the test microorganism cultures. The carotenoid drugs obtained at 40 and 60 °C from the strains F. hepatica Fh-18 and L. sulphureus Ls-08 respectively showed the highest antioxidant activity.

<p>Penelitian ini merupakan lanjutan dari penelitian sebelumnya tentang produksi asam galat dari limbah kulit buah lokal di Lombok. Limbah kulit buah lokal yang digunakan yaitu kulit buah kepundung (<em>Baccaurea racemosa</em> Muell.Arg), kulit buah juwet (<em>Syzygium cumini</em>), dan kulit buah manggis (<em>Garcinia mangostana</em>). Pada penelitian ini dilakukan pengujian asam galat yang dihasilkan, untuk mengetahui aktivitas antioksidan dan antibakteri. Pengujian sifat antioksidan dilakukan dengan menggunakan metode DPPH, sedangkan aktivitas antibakteri diuji dengan menggunakan metode difusi agar pada bakteri <em>Escherichia coli</em> dan <em>Staphylococcus aureus</em>. Hasil penelitian menunjukkan bahwa asam galat dari ketiga kulit buah memiliki aktivitas antioksidan. Asam galat dari kulit buah kepundung, kulit buah juwet dan kulit buah manggis memiliki IC<sub>50</sub> masing-masing sebesar 5.95; 5,96; dan 5,92 ppm. Berbeda dengan kemampuan sebagai antioksidan, asam galat resistan terhadap bakteri <em>Escherichia coli</em> dan <em>Staphylococcus aureus </em>yang menunjukkan bahwa asam galat tidak memiliki aktivitas sebagai antibakteri.</p><p><strong>Antibacterial and antioxidant activity of gallic acid from peel local fruit which produced by tannase. </strong>This research is a continuation of the previous research about the gallic acid production from peel fruit local in Lombok. The local fruit waste<em> </em>used<em> </em>are<em> kepundung rind (Baccaurea racemosa</em> Muell.Arg)<em>, juwet rind </em>(<em>Syzygium cumini</em>) <em>and mangosteen rind </em>(<em>Garcinia mangostana</em>)<em>. </em>In this research, the produced Gallic acid was tested to determine its antioxidant and antibacterial activity. The antioxidant test conducted by DPPH method, meanwhile, the antibacterial activity was performed by agar diffusion method to bacteria <em>Escherichia coli</em> and <em>Staphylococcus aureus</em>. The results show that the produce gallic acid from those three rind provide antioxidant activity. Gallic acid from kepundung rind, juwet rind, and mangosteen rind have IC<sub>50</sub> values of 5,95; 5,96; and 5,92,  respectively. Gallic acid not showed inhibitory actions against the growth of Escherichia coli and Staphylococcus aureus. This result show that gallic acid from local fruit waste did not provide an antibacterial activity.</p>

Objective: This study estimated the Permangante Index (IKMnO4) and evaluated its correlation with parameters as total phenols (FT) and total antioxidant activity (TAA) of commercial teas of Camellia sinensis, fermented, unfermented and blended with other plants.Methods: The aqueous extracts were prepared from commercial samples of green and black teas, green tea blended with lemon (Camellia sinensis with Citrus limonium), green tea blended with mint (Camellia sinensis with Mentha piperita), green tea blended with peach (Camellia sinensis with Prunus persica) and green tea blended with orange (Camellia sinensis with Citrus sinensis). The Permangante index was determined by titrating the tea extract with potassium permanganate solution according to AOAC method (AOAC, 1980) with modifications. The concentration of phenols in the extracts was determined by spectrophotometry according to the standard procedure of Folin-Ciocalteau. The antioxidant activity of the extracts was evaluated using the radical 1,1-diphenyl-2-picrylhydrazyl (DPPH—) method. All the determinations were performed in triplicate, and the results presented as mean ± standard deviation (SD). The data were evaluated by univariate statistical analysis (ANOVA), F-test and mean test (Tukey´s test) for comparative results. Pearson’s correlation was used to measure the degree of linear correlation and the quantitative variables were from a normal population.Results: The results for Permanganate Index show a variation ranged from 0.40 to 0.80 mEq/l. The ANOVA analysis to IKMnO4 shown there is significant statistically difference at the 5% probability level (p < 0.05) between the types of tea as well as the amount of herb used to prepare the infusion. Therefore, it was observed that there significant contrast between tea extracts, the mixture of different plants in blended extracts as well as the tea manufacture process affect the polyphenols tannins content influencing the permanganate index parameter. Studies correlation shown strong correlations (r > 0.7) among the parameters. For the Camellia sinensis extracts the high correlations between IKMnO4 and TAA; FT and TAA demonstrates the suitability of the permanganate index in the evaluation of antioxidant activity.Conclusion: The results suggest that the Permanganate Index could be used for quality control of tea as an additional parameter.

Background : The treatment of some diseases caused by free radicals and pathogenic bacteria usually by using antioxidants and antibiotics. Due to excessive use of antibiotics and other environmental cues, some bacteria are now resistant to certain antibiotics or even to multiple antibiotics. Some Vibrio cholerae bacterial strains are multiresistant to many antibiotics.Objective : The antioxidant and antibacterial activities of Brugueira gymnorrhiza stem extracts against pathogenic bacteria V. cholerae.Method : The B. gymnorrhiza stem was extracted by gradient maceration method. The DPPH method was used to determine the antioxidant activity and the disc diffusion method was used to determine the antibacterial activities. The column chromatography method was used to fractionate the selective extract with the best activity. The LC-MS/MS method was used to identify the compound obtained from the fraction with the best antioxidant and antibacterial activity.Result : Ethyl acetate extract of B. gymnorrhiza stem had the best antibacterial activity with MIC and MBC values of 62.50 mg/L. Ethyl acetate extract also showed the best value of antioxidant activity as indicated by an IC50 value of 255.03 mg/L. The results of fractions test showed that fraction 3 had the best antibacterial and the best antioxidant activities with both the MIC and MBC values of 7.90 mg/L and IC50 value of 348.91 mg/L, respectively.Conclusion : Ethyl acetate extract of B. gymnorrhiza stem has good potential as antioxidant and antibacterial. The compound which is thought as antioxidant and antibacterial from Ethyl acetate extract is 2-Ethyl-4-methyl-1H-imidazole.

We have demonstrated that potatoes contain significant levels of antioxidants important to human health; however, since potatoes are not consumed raw, it is important to determine the effects of cooking/processing on these levels. Therefore, the changes in phenolic and carotenoid content and total antioxidant activity in potatoes were investigated using combinations of storage and cooking methods. Fresh and stored tubers (110 days at 4 °C) of 17 potato cultivars, both raw and cooked (microwaved, boiled, baked, fried), were analyzed for antioxidant activity using the DPPH method. In addition, carotenoid levels were determined for each treatment based on the absorbance of the methanol extraction (oxygenated phenolics and carotenoids) at 445 nm and the hexane extraction (non-oxygenated carotenoids) at 450 nm. Total antioxidant activity as well as carotenoid levels were significantly affected by both genotype and cooking method. Across extraction methods, the microwave and fry cooking treatments were generally highest in antioxidant activity, while boiling was the lowest. Oxygenated carotenoids were significantly affected by storage, while the non-oxygenated carotenoids were unaffected.

Background: Conventional food additives were associated with potential harm. Palm seeds are an interesting source of high-added value bioactive compounds, regarding their potential use in food industries. Aim: Date seeds extract (DSE) of Algerian Phoenix dactyliefra L. was studied for its phytochemical, antioxidant, and antibacterial activity and to identify its potential uses in foods. Material and methods: A total soluble solid of DSE (°Brix) was determined by refractometry. The total phenolic contents (TPCs) were determined according to the Folin-Cioclateu method and the total flavonoid and flavonols were also estimated with the colorimetric method. The DPPH method was used to determine the antioxidant activity of DSE. The IC50 values were also calculated and the antioxidant activity index (AAI) was determined. The agar well diffusion method was employed for the antibacterial activity of the aqueous DSE against various pathogens. Results: Moisture, ash, and fat contents (%) determined were 11.06, 1.33, and 7.06, respectively. The physicochemical analysis of the extract showed a pH value of 4.67, titratable acidity value of 0.64 citric acid/100 g, °Brix value = 3%, sugars values of 0.12, 0.47, and 0.35 g/L for reducing sugar, total sugar, and sucrose, respectively. The determination of phenolic compounds was carried out on three groups, namely total phenols = 229.67 mg GAEq/g, flavonoids = 201.12 mg QEq/g, and flavonols = 173.03 mg QEq/g, respectively. The DSE demonstrates to be effective as an antioxidant “in vitro” and showed a strong antibacterial activity with different inhibition levels, depending on the bacterial strains. Conclusion: The current investigations present knowledge about the possible role of palm industry by-products as a novel perspective for the development of new foods. We suggest that date seeds can be exploited in some food applications utilizing their high levels of bioactive molecules. The </span><span style="font-size: 10pt;">objectives</span><span style="font-size: 10pt;" lang="en-us" xml:lang="en-us"> of this research </span><span style="font-size: 10pt;">were</span><span style="font-size: 10pt;" lang="en-us" xml:lang="en-us"> to observe their total antioxidant activity values and the effect of intervention of high-antioxidant beverages (tomato juice and rice bran beverage) to total antioxidant activity on breast cyst women patient’s blood serum. Subject consumed each of the beverages for 14 days. Blood samples were taken before the intervention, at the end of tomato juice intervention period and at the end of rice bran bevera</span><span style="font-size: 10pt;">g</span><span style="font-size: 10pt;" lang="en-us" xml:lang="en-us">e intervention period. Total antioxidant activity on serum was determined by DPPH method. The ascorbic acid equivalent antioxidant capacity (AEAC) of rice bran greater than tomato juice is 28.74 mg/100g and 1.87 mg/100g respectively. The total antioxidant activity on serum was decreasing after tomato juice intervention (p&gt;0.05) but the decreasing of it was significantly shown after rice bran intervention (p&lt;0.05)</span><span style="font-size: 10pt;">.</span></p>

Skin care is very important in cosmetics, especially facial treatments. Black mulberry is rich in phenols and is therefor usable in the treatment of acne. It also contains anthocyanin, a well-known antioxidant and a potential source of sun protection. This research aimed to develop a gel dossage from black mulberries (Morus nigra) extracts that would function as an antibacterial, antioxidant and sun blocking facial treatment. This research started with black mulberry fruit extracted through the maceration method by using ethanol (96%). Then, the antibacterial activity of the extract was determined by the disc-diffusion method, while the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) were determined by the microdilution method. Antioxidant activity of the extract was determined using the 1,1- diphenyl-2-picrilhidrazil (DPPH) method with vitamin C as a comparison. Furthermore, the extracts were formulated into gel formulas with variations of HPMC, Na-CMC, carbopol 934 and extract concentrations. The products were then physically evaluated for organoleptics, homogeneity, pH, viscosity, and dispersion as well as undergoing a hedonic test, an irritation test, an antioxidant activity test and determining the SPF value of the preparation. The results showed that the black mulberry fruit extract has antibacterial activity with MIC value of 0,025 g/mL against S. epidermidis and P. acnes, while MBC values were 0,025 g/mL and 0,05 g/mL, respectively. The black mulberry extract had antioxidant activity with IC50 value, i.e 146,73 μg/mL vitamin c i.e. 3,17 μg/mL. Formulation with best physical evaluation results showed ina formula containing a carbopol gel base of 934 0,015 g/mL with an extract cconcentration of 0,075 g/mL. This formula resulted in antioxidant activity with an IC50 value, i.e 1004,6 μg/mL, antibacterial activity with inhibition zone 6.83 ± 1.4 mm against and 6.76 ± 0.9 mm against S. epidermidis and P. acnes respectively and an SPF value of 1.9.

Chalcone analogs have been reported to have a variety of exciting biological activities, such as anticancer, anti-inflammatory, antioxidant, photoprotective, antibacterial, and antidiabetic activities. Therefore, analogs of these compounds have been widely synthesized as intermediate compounds or target molecules in the discovery of bioactive compounds to be applied in the pharmaceutical field. The purpose of this study is to synthesize chalcone analog compounds (E)-1-(4-hydroxyphenyl)-3-(4-methoxyphenyl) prop-2-en-1-on with the microwave irradiation method and explore some of the biological activities of these compounds, including testing the antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) method, in vitro sunscreen activity by microplate method, and antibacterial activity by disk diffusion method. DPPH test results indicate that the compound has weak antioxidant activity, with an IC50 value of 300.43 µg/mL. However, the compound showed excellent potential as a UV B and UV A filter at a concentration of 150 µg/mL with a value of %Te of 0.73±0.10% (sunblock), %Tp of 0.07±0.00% (sunblock), SPF value of 21.10±1.46 (ultra-protection) and potentially better than benzophenone-3 as a standard sunscreen. Then, disk diffusion testing showed that the compound had weak antibacterial activity against Staphylococcus aureus and did not show antibacterial activity against Escherichia coli bacteria at test concentrations of 30, 60, and 120 µg/disk.

AbstrakKembang leson adalah racikan jamu untuk mandi yang terdiri dari berbagai obat-obatan herbal yang umum ditemukan di Jawa. Kembang leson mengandung minyak atsiri utama, yaitu camphene 1,29%, benzene methyl cymene 4,93%, camphor 4,75%, cyclohexane methanol 7,56%, dan curdione 4,83%. Golongan senyawa minyak atsiri dapat menghambat radikal bebas 2,2-diphenyl-1-picrylhydrazyl (DPPH) dan memiliki aktivitas antibakteri yang ditunjukkan dengan uji aktivitas antibakteri terhadap Staphylococcus aureus dan Escherichia coli. Tujuan penelitian ini adalah untuk mengetahui aktivitas antioksidan dan antibakteri dari minyak atsiri kembang leson. Minyak Atsiri kembang leson diekstraksi dengan metode destilasi. Minyak atsiri yang diperoleh diuji aktivitas antioksidan dengan metode DPPH, serta aktivitas antibakteri dengan metode difusi. Hasil penelitian menunjukkan bahwa minyak atsiri kembang leson memiliki aktivitas antioksidan dengan persentase penghambatan DPPH sebesar 56,16%, dan memiliki nilai IC50 (half maximal inhibitory concentration) sebesar 825,78 ppm. Aktivitas antibakteri minyak atsiri kembang leson juga efektif menghambat pertumbuhan E. coli, tetapi belum mampu menghambat pertumbuhan S. aureus. Minyak atsiri kembang leson  dapat digunakan sebagai sumber antioksidan alami. Abstract Kembang leson is an herbal concoction used for bathing. It comprises various herbal medicines found in Java. Kembang leson contains essential oils such as camphene 1.29%, benzene methyl cymene 4.93%, camphor 4.75%, cyclohexane methanol 7.56% and curdione 4.83%. It is known that essential oils can reduce DPPH free radicals and have antibacterial activity against S. aureus and E. coli.  Our research to determine the antioxidant and antibacterial activity of kembang leson essential oil. The essential oil was extracted by distillation. The essential oil obtained was tested for antioxidant activity using the DPPH method, and antibacterial activity using the diffusion method. The results show that kembang leson essential oils have antioxidant and antibacterial activity. The percentage of DPPH inhibition was 56.16% and the IC50 value was 825.78 ppm. Antibacterial activity of kembang leson essential oil inhibited the growth of E. coli but could not inhibit the growth of S. aureus. Thus, kembang leson essential oils have antioxidant and antibacterial activity against E. coli, but only have antioxidant activity against S. aureus. Further studies are needed to determent the main ingredients that play an important role in the mechanism of antioxidants and antibacterial of kembang leson essential oil.
