Background : The treatment of some diseases caused by free radicals and pathogenic bacteria usually by using antioxidants and antibiotics. Due to excessive use of antibiotics and other environmental cues, some bacteria are now resistant to certain antibiotics or even to multiple antibiotics. Some Vibrio cholerae bacterial strains are multiresistant to many antibiotics.Objective : The antioxidant and antibacterial activities of Brugueira gymnorrhiza stem extracts against pathogenic bacteria V. cholerae.Method : The B. gymnorrhiza stem was extracted by gradient maceration method. The DPPH method was used to determine the antioxidant activity and the disc diffusion method was used to determine the antibacterial activities. The column chromatography method was used to fractionate the selective extract with the best activity. The LC-MS/MS method was used to identify the compound obtained from the fraction with the best antioxidant and antibacterial activity.Result : Ethyl acetate extract of B. gymnorrhiza stem had the best antibacterial activity with MIC and MBC values of 62.50 mg/L. Ethyl acetate extract also showed the best value of antioxidant activity as indicated by an IC50 value of 255.03 mg/L. The results of fractions test showed that fraction 3 had the best antibacterial and the best antioxidant activities with both the MIC and MBC values of 7.90 mg/L and IC50 value of 348.91 mg/L, respectively.Conclusion : Ethyl acetate extract of B. gymnorrhiza stem has good potential as antioxidant and antibacterial. The compound which is thought as antioxidant and antibacterial from Ethyl acetate extract is 2-Ethyl-4-methyl-1H-imidazole.

Research on the antioxidant activity of the local onion roots of Palu (Allium cepa Var Aggregatum L.) with various polarity solvents has been carried out. This study aims to determine the potential of Palu local shallot root extract as an antioxidant and determine the IC50 value of the Palu local shallot root antioxidant activity. The method applied in this research is maceration using n-hexane, ethyl acetate, and ethanol. The extracts from each solvent were tested for their antioxidant activity by the DPPH method. The results of the study showed that the IC50 value in n-hexane extract, ethyl acetate extract, and ethanol extract were 579.98 ppm, 518.28 ppm 315.83 ppm and as a comparison used ascorbic acid obtained IC50 of 53.69 ppm. Based on the results obtained, the best antioxidant activity is in polar solvents (ethanol extract) compared to other solvents. Dumortiera hirsuta is one of the liverworts that can be used as a medicinal to prevent infection by pathogenic bacteria. The content of secondary metabolites of D. hirsuta has potential as antibacterial properties includes flavonoids, alkaloids and steroids. This research is to analyze the antibacterial activity of moss D. hirsuta against pathogenic bacteria that will be beneficial to humans. Liverworts of D. hirsuta were extracted using ethyl acetate solvent and tested against three types of pathogenic bacteria using the agar well-diffusion method. The results of this study indicated that the ethyl acetate extract of D. hirsuta at 100% concentration could inhibit the growth of Escherichia coli, Staphylococcus aureus, and Salmonella typhi bacteria. The range of antibacterial activity categories of the ethyl acetate extract of D. hirsuta to E. coli, S. aureus, and S. typhi between weak to moderate.

Abstract This study aimed to compare antioxidant, antibacterial and toxicity tests of various types of kesambi tree bark extract (Schleichera oleosa (Lour) Oken). The research stages included extraction of kesambi stem skin from Mancak Serang sub-district with multilevel maceration techniques using 3 solvents: Methanol, n-hexane, and ethyl acetate. The extract was then tested for antioxidant activity, antibacterial, and toxicity. The results of antioxidant testing using the DPPH method, antibacterial with agar diffusion method, and toxicity test with BSLT method, the results showed that ethyl acetate extract had better antioxidant activity (IC50 7,723 ppm.) compared to extract of methanol (IC50 7,801 ppm), and n-hexane extract (IC50 8,568 ppm). Antibacterial activity showed the ability to inhibit the growth of Streptococcus aureus compared to Escherichia coli at a concentration of 10000 ppm, and the results of the toxicity test showed that ethyl acetate extract had better activity (LC50 305,17 ppm) than n-hexane extract (LC50 374, 96 ppm) and methanol extract (LC50 431,26 ppm).Keywords: DPPH, BSLT, Agar Diffusion, Schleichera oleosa (Lour) Oken

This study aimed to investigate phytochemical screening and antioxidant activity of n–hexane, ethyl acetate and ethanol extract from lakoocha leaves. The powdered simplicia was macerated with n–hexane, ethyl acetate and ethanol 96% successively, filtered, then concentrated using rotary evaporator to obtain n–hexane extract, ethyl acetate extract and ethanol extract. Phytochemical screening and antioxidant activity was performed against these extracts. Antioxidant activity was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method using ultraviolet-visible spectrophotometer at wavelength of 516 nm after incubated for 60 minutes in dark place. Quercetin was used as positive control. The result of phytochemical screening showed n-hexane extract contains steroid, ethyl acetate extract contain steroid, tannin, glycoside, flavonoid and saponin, whereas ethanol extract contain tannin, glycoside, flavonoid and saponin. The IC50 value of n–hexane, ethyl acetate and ethanol extract was 1062.03±1.42 ppm, 323.18±0.02 ppm and 99.23±0.07 ppm respectively, whereas for quercetin was 2.32±0.01 ppm. This study showed that ethanol extract had antioxidant activity with strong category whereas n-hexane extract and ethyl acetate extract had inactive antioxidant activity with very weak categories.       Keyword: Antioxidant Activity, DPPH, Lakoocha leaf

Objective: The objective of this research is to evaluate the antioxidant and antibacterial activity of methanol, n-hexane, ethyl acetate, and flavonoid total of Dendrophthoe pentandra (L.) Miq extract.Methods: The leaf of D. pentandra (L.) Miq was gradually extracted using methanol, n-hexane, and ethyl acetate. From that extraction process, various isolates were obtained such as isolate of methanol, n-hexane, ethyl acetate, and flavonoid total. The obtained flavonoid total was continued into separation process using column system with eluent chloroform:methanol in ratio (9:1, 8:2, 7:3, and 6:4) and from this process was obtained six fractions (F1-5).Results: All isolates and fractions that obtained during the extraction and separation process were continued to the activity measurement, such as antioxidant activity using 1,1-diphenyl 2-picrylhydrazyl method and antibacterial activity using agar diffusion method to Salmonella typhi, Pseudomonas sp., Escherichia coli, and Staphylococcus aureus. The antioxidant activity (IC50) of the isolate of flavonoid total and fraction of F4 and F5 was 6.69, 5.98, and 5.95 μg/mL, respectively. On the antibacterial activity, measurement showed a linear correlation between the activity and concentration (10 and 100 μg/mL) of all isolates and fractions. The antibacterial activity was dominated by flavonoid total isolate.Conclusions: The results obtained in this research work showed that the extracts and fractions of D. pentandra (L.) Miq have a potency as antioxidant and antibacterial, especially flavonoid total that acts as antioxidant and fractions F4 and F5 that act as antibacterial.

Tujuan penelitian ini untuk mengetahui golongan senyawa metabolit sekunder dan aktivitas antioksidan dari setiap ekstrak kulit batang Chisocheton sp. C.DC Harms (Meliaceae). Hasil ekstraksi 200 g serbuk kulit batang Chisocheton sp. C.DC Harms (Meliaceae) dengan n-heksana, etil asetat dan metanol masing-masing dengan 2000 mL berturut-turut menghasilkan 7,193 g ekstrak pekat n-heksana, 8,798 g ekstrak pekat etil asetat dan 18,683 g ekstrak pekat metanol. Hasil uji fitokimia menunjukkan adanya golongan senyawa flavonoid, triterpenoid dan tanin. Hasil uji aktivitas antioksidan dengan metode DPPH menunjukkan bahwa ekstrak n-heksana memberikan nilai IC50 sebesar 337,28 µg/mL, ekstrak metanol sebesar 216,73 µg/mL, dan ekstrak etil asetat sebesar 199,89 µg/mL yang berarti etil asetat memiliki kemampuan yang paling besar dalam menangkap radikal bebas.ABSTRACTThe objective of this study was determine the class of secondary metabolite compounds and antioxidant activity of each stem bark extract of Chisocheton sp. C.DC Harms (Meliaceae). The results of the extraction of 200 g Chisocheton sp. C.DC Harms (Meliaceae) with n-hexane, ethyl acetate and methanol with 2000 mL each yielded 7,193 g of concentrated extract n-hexane, 8,798 g of concentrated extract of ethyl acetate and 18,683 g of concentrated methanol extract. Phytochemical test results showed the presence of flavonoid, triterpenoid and tannin compounds. The results of the antioxidant activity test using the DPPH method showed that the n-hexane extract gave an IC50 value of 337,28 µg/mL, methanol extract was 216,73 µg/mL, and ethyl acetate extract was 199,89 µg/mL which means that ethyl acetate has the ability to greatest in capturing free radicals.

Skin care is very important in cosmetics, especially facial treatments. Black mulberry is rich in phenols and is therefor usable in the treatment of acne. It also contains anthocyanin, a well-known antioxidant and a potential source of sun protection. This research aimed to develop a gel dossage from black mulberries (Morus nigra) extracts that would function as an antibacterial, antioxidant and sun blocking facial treatment. This research started with black mulberry fruit extracted through the maceration method by using ethanol (96%). Then, the antibacterial activity of the extract was determined by the disc-diffusion method, while the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) were determined by the microdilution method. Antioxidant activity of the extract was determined using the 1,1- diphenyl-2-picrilhidrazil (DPPH) method with vitamin C as a comparison. Furthermore, the extracts were formulated into gel formulas with variations of HPMC, Na-CMC, carbopol 934 and extract concentrations. The products were then physically evaluated for organoleptics, homogeneity, pH, viscosity, and dispersion as well as undergoing a hedonic test, an irritation test, an antioxidant activity test and determining the SPF value of the preparation. The results showed that the black mulberry fruit extract has antibacterial activity with MIC value of 0,025 g/mL against S. epidermidis and P. acnes, while MBC values were 0,025 g/mL and 0,05 g/mL, respectively. The black mulberry extract had antioxidant activity with IC50 value, i.e 146,73 μg/mL vitamin c i.e. 3,17 μg/mL. Formulation with best physical evaluation results showed ina formula containing a carbopol gel base of 934 0,015 g/mL with an extract cconcentration of 0,075 g/mL. This formula resulted in antioxidant activity with an IC50 value, i.e 1004,6 μg/mL, antibacterial activity with inhibition zone 6.83 ± 1.4 mm against and 6.76 ± 0.9 mm against S. epidermidis and P. acnes respectively and an SPF value of 1.9.

The phytochemical analysis and antibacterial and antioxidant activity of Ardisia silvestris extracts were carried out. The bioactive compounds of Ardisia silvestris leaves were extracted with petroleum ether, ethyl acetate, ethanol and water by immersion and microwave method. The DPPH method was used to determine the antioxidant activity of Ardisia silvestris extracts. The antibacterial properties of Ardisia silvestris tested against of Staphylococcu saureus, Samonella sp., and Escherichia coli were determined by using agar diffusion method. The agar diffusion method was used to determine the antibacterial effects of both plant extracts on the test organisms. The results showed that Ardisia silvestris leaves contained compounds such as essential oils, fats, alkaloids, flavonoids, coumarins, tannins, anthocyanoids, carotenoids, organic acids, reducing agents, proanthocyanidins, saponins and anthraquinones. Ardisia silvestris leaves had a total polyphenol content of 0.26% dry matter, tannin of 8.8%, and a total flavonoid of 1.44 mg/g. The ethyl acetate extract and water extract of the leaves had the antioxidant activity and were 4.2 and 4.4 times lower than ascorbic acid, respectively. The ethyl acetate extract of Ardisia silvestris had the highest oxidative activity. The zone of inhibition of the plant extract diameters at the concentration of 100 µl/ml ranged between 9.67mm and 20.67mm for ethyl acetate and ethanol extracts, respectively on E. coli. Similarly, the zones of inhibitionof ethyl acetate and ethanol extracts diameters were 14.67 and 15.33 mm, respectively on Samonella sp., however, it was not shown for Staphylococus aureus.

AbstrakKembang leson adalah racikan jamu untuk mandi yang terdiri dari berbagai obat-obatan herbal yang umum ditemukan di Jawa. Kembang leson mengandung minyak atsiri utama, yaitu camphene 1,29%, benzene methyl cymene 4,93%, camphor 4,75%, cyclohexane methanol 7,56%, dan curdione 4,83%. Golongan senyawa minyak atsiri dapat menghambat radikal bebas 2,2-diphenyl-1-picrylhydrazyl (DPPH) dan memiliki aktivitas antibakteri yang ditunjukkan dengan uji aktivitas antibakteri terhadap Staphylococcus aureus dan Escherichia coli. Tujuan penelitian ini adalah untuk mengetahui aktivitas antioksidan dan antibakteri dari minyak atsiri kembang leson. Minyak Atsiri kembang leson diekstraksi dengan metode destilasi. Minyak atsiri yang diperoleh diuji aktivitas antioksidan dengan metode DPPH, serta aktivitas antibakteri dengan metode difusi. Hasil penelitian menunjukkan bahwa minyak atsiri kembang leson memiliki aktivitas antioksidan dengan persentase penghambatan DPPH sebesar 56,16%, dan memiliki nilai IC50 (half maximal inhibitory concentration) sebesar 825,78 ppm. Aktivitas antibakteri minyak atsiri kembang leson juga efektif menghambat pertumbuhan E. coli, tetapi belum mampu menghambat pertumbuhan S. aureus. Minyak atsiri kembang leson  dapat digunakan sebagai sumber antioksidan alami. Abstract Kembang leson is an herbal concoction used for bathing. It comprises various herbal medicines found in Java. Kembang leson contains essential oils such as camphene 1.29%, benzene methyl cymene 4.93%, camphor 4.75%, cyclohexane methanol 7.56% and curdione 4.83%. It is known that essential oils can reduce DPPH free radicals and have antibacterial activity against S. aureus and E. coli.  Our research to determine the antioxidant and antibacterial activity of kembang leson essential oil. The essential oil was extracted by distillation. The essential oil obtained was tested for antioxidant activity using the DPPH method, and antibacterial activity using the diffusion method. The results show that kembang leson essential oils have antioxidant and antibacterial activity. The percentage of DPPH inhibition was 56.16% and the IC50 value was 825.78 ppm. Antibacterial activity of kembang leson essential oil inhibited the growth of E. coli but could not inhibit the growth of S. aureus. Thus, kembang leson essential oils have antioxidant and antibacterial activity against E. coli, but only have antioxidant activity against S. aureus. Further studies are needed to determent the main ingredients that play an important role in the mechanism of antioxidants and antibacterial of kembang leson essential oil.

Objective: The study was carried out the phytochemical and antibacterial activity of Knoxia sumatrensis (Retz.) Dc. using hexane and ethyl acetate extracts. Methods: The phytochemical screening was extracted from hexane and ethyl acetate solvent and its screening was analyzed by standard procedure. GC-MS technique was analyzed in ethyl acetate extract to identify the components present in the extract. The hexane and ethyl acetate extract of the plant was tested for antibacterial activity against human pathogenic bacteria using disk diffusion method. Results: The phytochemical screening was revealed the presence of phenols, saponins, flavonoids, cardiac glycosides, steroids, and tannins. The GC-MS results showed that the presence of seven bioactive compounds in ethyl acetate extract. The major compounds were identified such as N-Hexadecanoic Acid (9.336), 2-Piperidinone, N-[4-Bromo-N-Butyl] - (37. 883). The ethyl acetate extract showed good antibacterial activity. The maximum zone of inhibition was noticed in S. aureus (8.25 mm) using ethyl acetate extracts at 50 μg. Conclusion: Thus, this study the information regarding the phytochemical constituents present in the both extract. Hence, it can be used for further therapeutic applications in the near future study.
