scholarly journals Phenotypic and molecular characterization of fluoroquinolone resistant Pseudomonas aeruginosa isolates in Palestine

Abstract Fluoroquinolones are important antimicrobial agents for the treatment of Pseudomonas infections. A total of 11 isolates of P. aeruginosa were collected from different clinical samples from different medical centers in the North West Bank-Palestine during 2017. In this study, resistance to fluoroquinolones and secretions of β-lactamases were detected by phenotypic methods, while presence of β-lactamase gene sequences and other virulence factors were detected by PCR technique. PCR product for gyrA, parC and parE genes were sequenced for further analyses. The phylogenetic analyses, population diversity indices and haplotypes determination were conducted using computer programs MEGA version 6, DnaSP 5.1001 and median-joining algorithm in the program Network 5, respectively. Results of this study showed that the MIC for ciprofloxacin and norfloxacin had a range of 32-256 µg/ml. In addition, all isolates carried either exoT or exoT and exoY genes, different β-lactamase genes and 82% of these isolates harbored class 1 integrons. Analyses of the gyrA, parC and parE sequences were found to be polymorphic, had high haplotype diversity (0.945-0.982), low nucleotide diversity (0.01225-0.02001) and number of haplotypes were 9 for each gyrA and parE genes and 10 haplotypes for parC gene. The founder haplotypes being Hap-1 (18%), Hap-2 (27.3%) and Hap-6 (9.1%) for gyrA, parC and parE genes, respectively. Two of ParE haplotypes were detected as indel haplotypes. The Median-joining- (MJ) networks constructed from haplotypes of these genes showed a star-like expansion. The neutrality tests (Tajima’s D test and Fu’s Fs test) for these genes showed negative values. Palestinian fluoroquinolone resistant P. aeruginosa strains showed high MIC level for fluoroquinolones, β-lactamase producers, carried type III secretion exotoxin-encoding genes, most of them had integrase I gene and had high level of mutations in QRDR regions in gyrA, parC and parE genes. All these factors may play an important role in the invasiveness of these strains and make them difficult to treat. Isolation of these strains from different medical centers, indicate the need for a strict application of infection control measures in Medical centers in the North West Bank-Palestine that aim to reduce expense and damage caused by P. aeruginosa infections. Molecular analyses showed that Palestinian fluoroquinolone resistant P. aeruginosa haplotypes are not genetically differentiated; however, more mutations may exist in these strains.

A combination of historical, geographical, and epidemiological studies has given sufficient insight into the ecology of sleeping sickness to enable the main factors influencing the development and spread of an epidemic to be traced.The evidence shows that in West Africa sleeping sickness is not primarily a disease of the forest, where tsetse flies are most abundant, but belongs essentially to the dry country in the north of the savanna woodland zone, where the earliest occurrences and severest outbreaks have been located.The first mention of sleeping sickness comes from the upper Niger and dates back to the 14th century. By the beginning of the present century intense though localised epidemics were devastating parts of the Mossi, Grounsi and Lobi country of the upper Volta rivers. At this time the disease was unknown on the coast and of sporadic occurrence only in the forest. A severe trans-Volta epidemic covering 60,000 square miles, developed between 1924 and 1940, but was confined to the north of the inland savanna zone with nothing comparable in the forest.The epidemic spread in three principal ways : (1) Outwards from original foci of infection because of the dynamic nature of the disease. This produced a concentration of infection around headwaters, a feature characteristic of advanced epidemics. (2) Through the agency of travellers, originally from north to south but subsequently in both directions : a rapid method of spread producing linear distribution of infection along trade routes. The tempo was greatly increased on the pacification and development of West Africa after 1900. (3) A gradual southward shift in the main epidemic zone appears to be resulting from a long-term change in the African climate which is combining with man's activities to produce a southerly extension of xerophytic vegetation types and a regression of forest.The most important spread was that caused by the trading caravans, more especially the cola traders, who have been coming down to the cola-nut areas in the Ashanti forest from the big markets on the Niger and Upper Volta since the 11th century. The caravans were formerly very large, up to one or two thousand strong, and were frequently made up of Mossi and Grounsi from the territory that was so heavily infected by the beginning of the present century. It is certain that a continuous introduction of infection would have been taking place into the forest ever since trypanosomiasis was prevalent in the north, that is for 100 years at least. And infection has been known in the forest for about that period, yet always to a mild degree, never reaching epidemic form. It has been sought for, because conditions in the forest, with the vector Glossina palpalis in contact with every village and path, appeared to be ideal for the transmission of infection and this drew the particular attention of the early workers from 1908 onwards. But the most that could be found was a threatened epidemic in north-west Ashanti, very significantly centering on the big cola markets which formed the termini for the northern traders.This historical evidence and the reasoning from epidemiology lead to the conclusion that conditions in the forest are not conducive to the development of epidemic sleeping sickness and that the low state of endemicity found there is maintained by the constant introduction of infection from the true epidemic areas in northern savanna.From this conclusion arises a practical point of the greatest importance. If the sources from which infection is introduced into the forest could be eliminated the disease there should eventually die out and the tsetse, from the human point of view, would be harmless. Tsetse control in the forest may prove difficult and expensive, and if it is attempted by clearing this might end in the literal destruction of the forest. Such measures would be hard to justify, so many other factors of possibly greater importance than trypanosomiasis are involved, both the intrinsic value of a forest for its products and the wider value through its influence on climate, soil and water.In formulating a plan for the control of sleeping sickness, the habits of both vectors, human as well as insect, should be considered. The tsetse plays a major role in the development of the high infection rates characterising the epidemic outbreaks in northern savanna ; the human vector distributes infection from these sources along trade routes and into the forest. The elimination of the disease at its source, in true epidemic centres, which can be most effectively accomplished by eradication of the tsetse, will check the distribution of infections to the secondary areas of lighter infection which could then be cleared up by quite minor control measures or might even disappear spontaneously.This plan is now in operation in the Gold Coast. The validity of the arguments on which it was based is being shown by the results that are already apparent : the high rates of reduction in the epidemic areas and the pronounced lowering of infection in neighbouring, uncontrolled areas, more particularly in the forest region of north-west Ashanti where it is entered by a trade route coming from the previously heavily infected country.

Aims. This study was aimed at knowing the prevalence of vancomycin and high level aminoglycoside resistance in enterococcal strains among clinical samples.Study Design. It was an investigational study.Place and Duration of Study. It was conducted on 100Enterococcusisolates, in the Department of Microbiology, Pt. BDS PGIMS, Rohtak, over a period of six months from July to December 2014.Methodology. Clinical specimens including urine, pus, blood, semen, vaginal swab, and throat swab were processed andEnterococcusisolates were identified by standard protocols. Antibiotic sensitivity testing of enterococci was performed using Kirby-Bauer disc diffusion method.Results. High level gentamicin resistance (HLGR) was more common in urine samples (41.5%) followed by blood (36%) samples. High level streptomycin resistance (HLSR) was more common in pus samples (52.6%) followed by blood samples (36%). Resistance to vancomycin was maximum in blood isolates.Conclusion. Enterococci resistant to multiple antimicrobial agents have been recognized. Thus, it is crucial for laboratories to provide accurate antimicrobial resistance patterns for enterococci so that effective therapy and infection control measures can be initiated.

We used both molecular and craniometrical data to test the presence of Hylomyscus walterverheyeni in the Cameroon Volcanic Line (CVL). Our molecular (cytochrome b and 16S gene sequencing) and morphometrical data (discriminant analyses) clearly show the presence of H. walterverheyeni in the CVL, north-west of the Sanaga River. Hylomyscus walterverheyeni occupies both lowland and mountain forests (up to 2000m asl). In our phylogenetic analyses, the CVL specimens form a monophyletic group. This clade seems to reflect the role of the Sanaga River as a barrier to gene flow within the species.

The flatid planthopper, Metcalfa pruinosa (Hemiptera: Flatidae), which is an invasive species, is widespread in Korea. We sequenced a fragment of the COI from 536 individuals collected mainly in Korea and the European countries and combined these sequence data with the public data, totaling 830 individuals worldwide. The identification of one shared haplotype only between Korea and the USA, the presence of this haplotype only in the North-West region of Korea, and the highest haplotype diversity in this region suggested that the North-West region is another point of entry in addition to the South-East region, which is the presumed sole point of entry to Korea. Furthermore, it suggested that North-West entry involves the M. pruinosa originating from the USA. In an effort to find further variable regions in the mitochondrial genome, one region provided substantially increased variability compared to that of the fragment of COI. FST estimation, PCoA, and BAPS analysis, using the concatenated sequences of COI and the newly detected variable region to infer the expansion pattern in Korea, indicates that the main highway, running obliquely between the North-West and South-East regions, appears to be responsible for the current population genetic structure of M. pruinosa in Korea, facilitating gene flow through this highway traffic.

A novel Gram-negative, aerobic, motile and rod-shaped bacterium was isolated from Qurugöl Lake located in a mountainous region near Tabriz city in the north-west of Iran. Growth occurred at pH 6–10 (optimum, pH 7±0.5) and at 10–40 °C (optimum, 30 °C). Strain RCRI4T was able to grow in the absence and presence of NaCl to 3 % (w/v). The major fatty acids were C17 : 0, C16 : 1ω7c/C15 iso3-OH, C17 : 1ω8c and C16 : 0. The G+C content of genomic DNA was 45.3 mol%. Based on the 16S rRNA and gyrB gene sequences, phylogenetic analyses indicated that strain RCRI4T associated with the genus Alishewanella , and closely related type strains include Alishewanella agri BLO6T (97.8 %), Alishewanella aestuarii B11T (97.7 %), Rheinheimera perlucida BA131T (97.5 %), Alishewanella fetalis CCUG 30811T (97.3 %) and Alishewanella jeotgali MS1T (97.1 %). The level of DNA–DNA relatedness between strain RCRI4T and phylogenetically the closest related strains, A. agri BLO6T and R. perlucida BA131T, was 9 and 14 %, respectively. On the basis of phenotypic, chemotaxonomic and phylogenetic results, it is suggested that strain RCRI4T represents a novel species of the genus Alishewanella , for which the name Alishewanella tabrizica sp. nov. is proposed. The type strain is RCRI4T ( = LMG 26473T = JCM 17275T = KCTC 23723T).

Ancient DNA is becoming increasingly recognised as a tool in conservation biology to audit past biodiversity. The widespread loss of Australian biodiversity, especially endemic mammal populations, is of critical concern. An extreme example occurred on Depuch Island, situated off the north-west coast of Western Australia, where an unidentified species of rock-wallaby (Petrogale sp.) became extinct as a result of predation by red foxes. Two potential candidate species, Petrogale lateralis and P. rothschildi, both have ranges adjacent to Depuch Island, making identification based on geography difficult. A museum bone (one of the only surviving Depuch Island specimens) was subjected to standard ancient DNA analyses and procedures. Mitochondrial DNA cytochrome b and hypervariable control region were targeted for species identification. Ancient DNA was successfully recovered from the bone: 200 base pairs (bp) of control region and 975 bp of the cytochrome b gene. Bayesian phylogenetic analyses were employed to model the Depuch Island rock-wallaby DNA sequences together with sequences of other rock-wallaby taxa from GenBank. Evidence suggests that of the two Petrogale lateralis subspecies proposed to have inhabited Depuch Island, Petrogale lateralis lateralis was identified as the most likely. The identification of the Depuch Island rock-wallaby population may assist in the reintroduction of an insurance population of Petrogale lateralis lateralis, which is becoming increasingly threatened on mainland Australia.

Relapsing fever is caused by theBorreliaspecies of spirochetes. Louse-borne epidemics of the disease may happen but the endemic disease is generally transmitted to humans by the bite of an infected tick (Ornithodorus). Clinical and laboratory findings of tick-borne relapsing fever in children in the north-west of Iran, Qazvin, were evaluated. This study was conducted from September 1992 to September 2012. Records from 53 cases of tick-borne relapsing fever (TBRF) were reviewed. In positive cases, febrile illness, and spirochetes were recognized in peripheral blood preparations. Of the 53 children younger than 12 years, fifty two percent were male and about one third (34%) of the patients were in the age range of 7–12 years. The disease is recorded through the whole year but its peak occurs during summer (52.8%) and autumn (32.1%). Sixty eight percent of patients were living in urban areas but had frequent travel to rural area. Thirty two percent of the cases were living in rural areas where their dwellings were close to animal shelters. All (100%) of the 53 subjects were febrile. Travellers to the rural areas with high prevalence of the disease should be attentive of the risk of tick-borne relapsing fever and use suitable control measures. Consequently relapsing fever should be considered when patients who live in or have vacationed in north-west of Iran show a recurring febrile illness.

The hoplonemertean Vieitezia luzmurubeae, gen. et sp. nov. is described from specimens collected in a national park on the north-west Iberian Peninsula, the Parque Nacional Marítimo-Terrestre das Illas Atlánticas de Galicia. The species, previously mistaken as the Mediterranean species Tetrastemma vittigerum (Bürger, 1904), is frequently associated with two common species of sea squirt, Phallusia mamillata (Cuvier, 1815) and Ciona intestinalis (Linnaeus, 1767), inside which the nemertean completes its life cycle. Some of the specimens examined were protandrous hermaphrodites. Data on morphology and anatomy are provided with illustrations. Sequences of the nuclear ribosomal gene 18S rRNA and the mitochondrial gene cytochrome c oxidase subunit I were compared with those of other hoplonemertean species and all phylogenetic analyses suggested that Vieitezia is sister to the genus Gononemertes, which parasitises ascidians, within a clade also containing the genera Oerstedia and Nemertellina. In contrast, the morphologically similar genus Tetrastemma appears in a separate clade. This study stresses the need for combining molecular and morphological data when studying nemertean biodiversity.
