ABSTRACT: Fipronil is a pesticide widely used for controlling fleas and ticks in domestic animals, and its short-term exposure can lead to serious effects on animals. However, the possible genotoxic effect of this compound has not been investigated in target animals. Based on the hypothesis that fipronil can induce genotoxicity, this study evaluated the effect of fipronil on DNA damage in peripheral blood cells. For that purpose, ten dogs of both sexes were used in the study. The product (6.7mg/kg) was applied on the dorsal neck region of each animal. Peripheral blood samples were collected immediately prior to application of the product, and at 3, 8 and 24 hours after the application. Samples were processed for comet assay. No statistically significant differences were found among the four time points. The current study suggests for the first time that a single exposure to this pesticide does not induce systemic genotoxic effect in dogs.

Pluripotent hemopoietic progenitors (CFU-GEMM) give rise to multilineage hemopoietic colonies in culture. We have examined the erythropoietin requirements of CFU-GEMM-derived erythroid progeny in patients with polycythemia vera (PV) and studied their proliferative activity by short-term exposure to 3HTdR. Mixed colonies with erythroid components were observed in all bone marrow and peripheral blood samples from patients with PV that were cultured without addition of exogenous erythropoietin. This response is consistent with previously reported growth patterns for CFU-E and BFU-E. The frequency of mixed colonies increased regularly when erythropoietin was added to the cultures. Short-term exposure of peripheral blood specimens to 3HTdR prior to plating yielded a reduction of the plating efficiency by 20%- 70% when compared to cells that were not exposed to 3HTdR. The observation of cycling CFU-GEMM in PV contrasts with the usually quiescent behavior of CFU-GEMM in peripheral blood of normal individuals under steady-state conditions. These results support the view that the increased proliferative rate observed for CFU-GEMM may be responsible for the increased formation of blood cells in PV.

Abstract Progenitors of blast cell colonies have been identified in acute leukemia. The peripheral blood of 18 of 25 patients with preleukemic states yielded low numbers of blast cell colonies, and the colony- forming cells were in an active proliferative state when assessed using short-term exposure to tritiated thymidine. The clinical significance of blast cell colonies is uncertain, but we suggest that further analysis of this cultural abnormality may lead to a better understanding of mechanisms and management in preleukemia.

Structural mitochondrial abnormalities as well as genetic aberrations in mitochondrial proteins have been known in Myelodysplastic syndrome (MDS) , yet there is currently little data regarding the metabolic properties and energy production of MDS cells. In the current study we used state-of-the-art methods to assess OXPHOS in peripheral blood cells obtained from MDS patients and healthy controls We then assessed the effect of food supplements- Coenzyme Q10 and carnitine on mitochondrial function and hematological response .We show here for the first time that in low risk MDS there is a significant impairment of mitochondrial respiration in peripheral blood cells and this can be improved with food supplements. We also show that such myelodysplastic syndrome, mitochondria, oxidative phosphorylation, coenzyme Q10, seahorse XF analyzer. supplements lead to improvement in cytopenia's and quality of life.

Chlorpyrifos (CPF) is an organophosphorus insecticide commonly used for domestic and agricultural purposes. The risk posed by environmental contamination from CPF is well acknowledged, and it has been detected worldwide in aquatic habitats and coastal areas. In addition, due to its slower degradation in seawater compared to freshwater, CPF is of particular concern for marine environments. Here, we investigated for the first time the morpho-functional alterations induced by CPF on the gills of Thalassoma pavo, a widespread species in the Mediterranean Sea. We tested the effects of two sublethal concentrations (4 and 8 µg/L) after 48 and 96 h. Our study demonstrates that the alterations induced by CPF are dose and time-dependent and highlight the harmful properties of this insecticide. After exposure to the low tested concentration, the more frequent alteration is an intense proliferation of the primary epithelium, whereas after exposure to the high concentration, the primary epithelium proliferation is less extensive, and the most evident effects are the thinning of secondary lamellae and the ectopia of chloride and goblet cells. CPF also modulated the expression of Na+/K+-ATPase. Dilation of lamellar apical tips, pillar cell degeneration, and appearance of aneurysms are often observed.

Hypoxia frequently associated with certain physiologic and pathologic conditions influences numerous cellular functions. Because the effects of short-term hypoxia are incompletely understood, we examined phagocytosis and cytokine production as well as the activation of the transcription factors HIF-1 and NFκB in peripheral blood cells of healthy volunteers exposed to an oxygen concentration equivalent to that found at a height of 5500 m. Furthermore, we analysed plasma HIF-1 and serum concentrations of various HIF-1-dependent genes. Results showed that short-term hypoxia increased phagocytosis in neutrophils without affecting monocyte phagocytosis. Hypoxia decreased basal TNFα concentration in monocytes and basal interferon γ concentration in CD4+T lymphocytes. In contrast, plasma HIF and serum VEGF concentrations were not affected by hypoxia, although serum EPO concentration was raised. In PBMC, hypoxia increased cytosolic HIF-1 concentration without affecting nuclear HIF-1 concentration and led to a rise in the nuclear NFκB in PBMC. Our results show that short-term hypoxia affects immune functions in healthy individuals. Furthermore, we speculate that the effects of hypoxia are not due to HIF-1, but are caused by the activation of NFκB .

Cellular responses to copper stress were investigated for the first time in a saxicolous lichen species, Lecanora polytropa (Hoffm.) Rabenh. Bright blue-green apothecia accumulated up to 1·3% Cu on a dry weight basis (205 μ mol Cu g−1), c. 50% in an exchangeable form. A bright turquoise-blue layer extended beneath the hymenium into the medulla, above and between a dentate photobiont layer. Oxalic (1·88 μ mol g−1), citric (0·83 μ mol g−1) and lower concentrations of malic (0·45 μ mol g−1) acids were determined by GC/MS analysis. Short-term exposure to high Cu2+ concentrations (40 and 400 μ mol g−1) under non-complexing conditions caused a dose-dependent decrease in chlorophyll a content; chlorophyll b and total carotenoid contents remained constant. The phaeophytinization quotient remained unchanged during Cu2+ exposure. Analysis of thiol peptides confirmed glutathione was reduced (GSH) in native L. polytropa (0·538 μ mol g−1), and phytochelatins (PC2 and PC3) oxidised. Short-term exposure to 40 μ mol g−1 Cu2+ oxidised c. 28% of the glutathione pool; oxidised phytochelatin concentrations remained unchanged. This is the first report of phytochelatin production and thiol peptide status in a crustose lichen. These represent two possible detoxification mechanisms in this Cu-tolerant species. Copper complexation by low molecular mass organic acids and non-protein thiols do not entirely account for its tolerance.

Abstract Peripheral blood cells from three patients with leukemia in relapse increased in numbers in short-term suspension cultures. This increase was dependent on the presence of either feeder populations containing a high percentage of blast cells or conditioned medium derived from such populations. Aneuploid cells present in direct marrow preparations were also prevalent in the cultures after 10 days. The apparent specificity of the increase for leukemic populations may provide a new approach to the study of leukemic blast cells.

Although peripheral blood mononuclear cells (PBMCs) are widely used as a valuable tool able to provide biomarkers of health and diseases, little is known about PBMC functional (biochemistry-based) metabolism, particularly following short-term nutritional challenges. In the present study, the metabolic capacity of minipig PBMCs to respond to nutritional challenges was explored at the biochemical and molecular levels. The changes observed in enzyme activities following a control test meal revealed that PBMC metabolism is highly reactive to the arrival of nutrients and hormones in the circulation. The consumption, for the first time, of a high fat–high sucrose (HFHS) meal delayed or sharply reduced most of the observed postprandial metabolic features. In a second experiment, minipigs were subjected to two-month HFHS feeding. The time-course follow-up of metabolic changes in PBMCs showed that most of the adaptations to the new diet took place during the first week. By comparing metabolic (biochemical and molecular) PMBC profiles to those of the liver, skeletal muscle, and adipose tissue, we concluded that although PBMCs conserved common features with all of them, their response to the HFHS diet was closely related to that of the adipose tissue. As a whole, our results show that PBMC metabolism, particularly during short-term (postprandial) challenges, could be used to evaluate the whole-body metabolic status of an individual. This could be particularly interesting for early diagnosis of metabolic disease installation, when fasting clinical analyses fail to diagnose the path towards the pathology.
